虽然在等离子体解吸情况下,利用限定不良的束,但是,实际上在同时发展的二次离子质谱(SIMS)采用被电势差加速的离子的限定良好的分析束[Bennighoven,A.,Surface Scicence 28(2)541-(1971)]。由于离子束的小直径,SIMS提供优良的空间分辨率,但是,经历SIMS电离的分子的分子量范围却受到限制。也可以将该方法用于深度分析(in-depthanalysis),然而,在这种情况下,由于所形成的离子包含仅仅1个或2个原子,因此分子量限制更苛刻。在SIMS电离的情况下,首次发展了对液体样品的研究[liquid SIMS;LSIMS,Aberth,W.,Analytical Chemistry,54(12):2029-2034(1982)]。与原始方法相比,LSIMS具有放宽的范围限制,例如,可以通过其来电离小的蛋白质分子。LSIMS的缺点在于,必须使样品溶解在具有高表面张力和低蒸汽压的溶剂例如甘油中。该步骤通常涉及溶解度问题,并且,固体样品的溶解明显地排除获得有关样品分子的空间分布的信息的可能性。
LSIMS的另一已发展的方案是“快原子轰击”(FAB)方法[Williams,D.H.et al.,JACS,103(19):5700-5704(1981)]。该技术基于惰性气体离子的静电加速、随后中和,获得可用于电离的仍具有高能级的惰性气体原子的中性束。FAB电离也适于分析液相样品。
SIMS技术的另一发展方向已被引导至所谓的大簇碰撞(MCI)电离[Massive cluster impact;MCI,Mahoney,J.F.,Rapid Communications inMass Spectrometry,5(10):441-445(1991)],其利用多电荷(multiplycharged)甘油簇来替代常规采用的金离子。该技术可被用于分析固体表面,并且在实践上不限制所分析的分子的重量。与SIMS相比,该技术的另一个优点在于,形成可以通过质谱来更有效分析的多电荷离子。
上述方法的共同缺点在于,它们都严格地工作在高真空条件下。因此,样品被引入质谱仪的高真空状态,这需要严格限制样品的成分和尺寸。
从二十世纪八十年代早期开始,已经研发了其中激光被用作分析束的激光解吸电离方法[Cooks,R.G.et al.,JACS,103(5):1295-1297(1981)]。与SIMS类似地,简单的激光解吸电离给出差的电离效率,并且它们仅仅可被用于研究非常有限范围的分子。通过施加所谓的基体化合物(matrixcompound),激光解吸方法的应用领域被显著地扩展。大量过剩地存在的基体化合物通常被以溶液相混合至样品,混合物被共结晶到固体载体上,并且通过激光解吸即其中激光被用作分析束的手段,分析所获得的结晶样品。该新方法被命名为基体辅助的激光解吸电离(MALDI)[Karas,Hillenkamp,Analytical Chemistry,60(20):2299-2301(1988)]。该技术通常可被用于分析大分子化合物,例如聚合物、蛋白质、碳水化合物和核酸。MALDI的主要缺点在于,该技术需要将分析物分子嵌入到基体晶格中,因此对自然表面的分析是成问题的。
最近,对在大气条件下工作的解吸电离方法的需求已经有所增加。大气压解吸电离方法的优点包括:(1)样品不被引入质谱仪的真空状态中,这使得分析过程较快,(2)由于样品不进入真空,不需要去除挥发性组分,(3)可以这样地调查研究任意对象,(4)可以直接研究活的生物体。不能在大气压条件下使用利用原子或离子的高速束的解吸电离方法,这是因为,由于与气体分子的连续碰撞,粒子不能在高压下被加速至合适的速度,该现象导致粒子束的发散。
在上述方法中,由于激光束不与空气分子相互作用,仅仅MALDI电离可以在大气压下实施而在仪器上没有变化。2002年由Laiko等人研发了大气压MALDI。然而,由于低的离子产率,其中通过在大气界面中的显著的离子损失,进一步降低了该低的离子产率,因此该技术没有被广泛推广,并且工作场所安全问题通常是与开放的实验设备中激光的使用相关联的。
最近研发的解吸电喷雾(electrospray)电离(DESI;Takats et al,Science,306(5695):471-471,2004)实际上是大气压版的上述MCI技术,但是却有着重大差异,即小滴是通过电喷雾方法产生的,且通过超声气体流替代静电场梯度而被加速到希望的速度。然而,DESI已实现了与大气压解吸电离方法相关联的所有期望,因此其打开了通往与化学成分和尺寸无关地对任意对象进行质谱分析的大门。在DESI处理的过程中,高速的电喷雾小滴与样品表面碰撞。碰撞小滴溶解存在于表面上的分子,并且发射同样带电的二次小滴。这些带电的二次小滴最终通过溶剂的完全蒸发而产生离子。
虽然与先前研发的解吸方法相比DESI技术具有多个优点,但其也显示出在几个应用领域的缺点。首先,DESI是严格的表面分析方法,因此其缺乏在多数样品的情况下进行深度分析的能力。其次,由于在带电粒子之中的库仑斥力,带电小滴的大气压束固有地发散,其阻碍高分辨率化学成像。此外,虽然从分析的观点DESI与生物组织完全兼容,但DESI的活体内应用被证明在多种动物标本中会引起栓塞。据推测,栓塞是与用于加速小滴的超声氮气流相关联的,该超声氮气流将气泡泵送到组织中并引起栓塞。
为了提供以上列举的问题的解决方案,需要一种可供选择的大气压解吸电离方法,其采用准直的且高能的分析束,其中该束不是由通过高速气体加速的小滴构成的。我们的研究的目的是研发一种大气压电离方法,其能够深度分析样品、高分辨率化学成像以及活体内分析。
发明内容
作为我们的研究工作的结果,研发了一种新的方法和装置,其在大气压下工作,其中高速、连续的液体射流被用作分析束,该液体射流优选带有电荷。已经注意到,在不具有电荷的液体束的情况下,仍可发生离子的形成,但是,如果液体射流本身具有带电粒子,则液体射流的碰撞可以产生数量高得多的带电粒子。正电势产生带正电荷的离子,而负电势产生带负电的离子。
本发明是基于这样的共识,即具有高能的液体射流(其可以是任何液体,例如水、水溶液、其他极化溶剂、或它们的任何混合物)能够使得粒子从样品移位,由此确保深度分析的可能性。该方法的另一个优点隐藏在这样的事实中,即所述高速液体射流较不发散,由此能够以微米级的分辨率化学成像,此外,该方法可应用于对生物系统的活体研究。
本发明的主题是一种用于将凝聚相样品的特定组分转换成气态离子及其分析的方法,其中通过从解吸单元释放的分析束,使得离子或可转换成离子的样品粒子从样品移位,并且分析所获得的气态离子,包括以下步骤:
应用液体射流作为分析束,使得当所述液体轰击表面时形成的小滴的液体蒸发或者允许其被蒸发,并且,如果需要,在所述分析之前电离所述液体小滴或通过所述液体的所述蒸发获得的样品粒子。
通过已知的方法执行所述分析,所述已知的方法根据所述样品离子的电荷、尺寸、质量或其它特性来区分所述样品的组分。优选地,应用质谱仪或离子迁移谱分析。

本发明的详细描述
下面详细地给出以上限定的重要要素。以对于本领域技术人员显而易见的通常方式应用未限定的其它要素(例如,发射、组分)。
1.凝聚相样品
可以将在此描述的方法和装置(下面称为“根据本发明的方法”)用于分析包含可以被电离以形成气相离子的组分的任意的固体或液体材料。样品可以具有同质的或者任意异质的结构(例如,人类或者动物组织、骨、木材等)。
可以将根据本发明的方法有利地用于分析从溶液沉积的且干燥的样品,优选其中为了医学诊断或者药理学目的而执行分析的生物样品。这样的生物样品的实例为流体,即血液、尿、溶液(liquor)等。明显地,还可以以这种方式研究任意样品的萃取物。
主要的其它应用领域是对自然对象(其中实际对象用作样品)的研究,其中可以任意选择该自然对象(例如,土壤、岩石、食物、活的组织)。其它重要的应用领域如下:
对活的组织的新陈代谢过程的研究;
确定在活的生物组织,例如,在脑中的药物分子(或肽、脂肪等)的空间分布;
在外科手术期间获得有关正被切割的组织的信息(例如,其是否包含癌细胞),以提高组织切除的效率,并使得被去除的健康组织的总质量最小;
调研木材对象,以检测有机(例如真菌)或无机(例如抗真菌剂)污染;
从皮肤确定消耗的材料(误用的药物、酒精、药品、咖啡、尼古丁等);
例如在现场研究中,分析土壤样品(用于环境污染物和细菌或真菌生物标志物(biomarker));
确定人类和动物食物的消耗的适合度以及不希望的化学试剂(例如抗生素)的水平;以及
优选地与ICP-MS方法相结合,确定任意对象的元素组成的空间分布。
2.液体射流/液体
在根据本发明的方法中,理论上每种合适的液体都可用作液体射流的液体,但是优选地,采用导电液体,其可以是水、水溶液、包含水的溶剂系统、任何极化溶剂(甲醇、乙醇、二甲基甲酰胺、二甲基亚砜、乙腈等)、它们的混合物,其优选包含在这些溶剂例如乙酸、蚁酸、溴化四甲基铵等中经历电离分解的添加剂。在所有的溶剂系统中,高度优选水。
对所使用的液体的其它要求是具有足够高的表面张力,以阻碍在小滴中射流的分裂(disruption)。
液体可以包含经历与样品化学反应的化合物。液体也可以为悬浮液,这在深度分析的情况下尤其有利。
3.解吸单元
解吸单元能够将具有合适的速度、直径和压力的连续液体发射到合适的方向和“从样品取样”。优选地,解吸单元是端部为喷嘴3的管道2B或11。
在根据本发明的方法的优选实施例中,通过直径在1至100μm、优选在1至60μm、更优选在1至5μm的范围的喷嘴,在范围为50至1500巴、优选为100至1000巴、更优选200至600巴的压力下,对液体加压。
4.粒子的解吸和电离
当液体射流与样品碰撞时,则形成小滴,这些小滴带走样品的粒子(样品粒子)。
当溶剂蒸发时,带电荷的小滴产生气态离子。
小滴可以在形成时带电荷,或者可以通过使用强电势差梯度,或者通过使它们与其它带电荷的液体小滴碰撞,或者通过光电电离,或者通过等离子体(例如,通过感应耦合等离子体,ICP),在稍后的步骤中使小滴电离。在前三种情况下电离的小滴产生分子离子,在后一情况下形成原子离子。因此,在前三种情况下,获得主要关于分子级的分析信息,而在后一情况下,可以确定元素成分。
由于液体射流的电势便于使所述小滴或者粒子在形成时带电,优选地通过在液体射流与地之间施加范围在1至8kV、优选地2-6kV、更优选地4至5kV的高电压,在导电的液体射流与地电势之间建立电势梯度。
5.蒸发
由于气态离子应被传送到分析器单元,必须预先使液体从小滴中完全蒸发。这可以自然方式发生(即,液体在所使用的温度下蒸发),或者可以通过加速蒸发的装置(例如,加热器或者高温气体流)的采用而促进蒸发。
6.分析器单元
分析器单元能够检测离子,因此分析器单元优选为质谱仪或者离子迁移谱。
7.样品收集器单元
通过使用样品收集器单元(优选为在其中产生压力差的管道),由该方法产生的气态离子被传送到分析器单元。这仅仅在上述步骤没有在分析器单元中直接实施时才使用(后者是优选实施例,参见下述内容)。
8.用于承载样品的表面
可用于承载样品的表面优选为可以耐受所施加的液体射流的电绝缘表面。样品承载架(sample carrier)优选由有机玻璃、玻璃、陶瓷或石英等制成。如果样品承载架的表面导电,那么需要通过合适的电源在该表面与分析器单元的入口之间产生电势差。
其它优选实施例
1.在该实施例中,高速、带电荷的连续液体射流被用作分析束。当液体射流碰撞表面时,液体在表面上聚集,并且在真空的帮助下,所述液体通过位于碰撞区域附近的管道系统被连续地排出。该设置消除了由偶然的液体聚集引起的表面交叉污染,例如,通过所聚集的液体相,分析物分子向先前它们不存在于其中的位置处转移。
2.在该实施例中,所分析的样品和/或液体射流喷嘴被安装在允许三维(3D)移动的承载台上。该仪器设置能够提供与样品的化学成分的位置相关的信息。
3.在该实施例中,在表面上的碰撞点和碰撞角度受到控制。通过3D线性移动承载台,实现在表面上碰撞点的受控制的移动,以控制样品和发射液体的喷嘴相对于彼此的位置。通过使用旋转的承载台,以希望的角度旋转样品或者喷嘴,来控制液体射流的碰撞角度。
4.在该实施例中,通过电连接高压电源的出口与用作分析束的导电液体射流,建立离子形成所必需的电势梯度,并且解吸单元被用作反电极。
5.在该实施例中,液体射流被保持在或者接近地电势(<150V),并且样品收集器单元被保持在具有相对于所产生的离子的极性相反的极性的高电势(>1000V)。
6.在该实施例中,相对于样品收集器单元的入口,液体射流被保持在高电势,并且所述液体射流被高速气体射流所包围。该实施例减轻了射流的减速,并且还防止在大气压下射流直径的增大。
7.在该实施例中,相对于样品收集器单元的入口,液体射流被保持在高电势,并且承载样品的台的温度被控制在-50至+300℃的范围内。该实施例的优点在于,在室温下为液体或者软的样品可以被固化,用于在降低的温度下分析。该实施例的另一个优点在于,在较高的温度下,可以提高特定组分的电离效率。
8.在该实施例中,将多个连续的液体射流用于电离,相对于样品收集器单元的入口,它们被保持在高电势。该实施例的优点在于,这样可以调研较大的表面,并且也可实现较高的离子电流。
9.在该实施例中,液体射流包含经历与样品的特定组分瞬时化学反应的化合物。在特定的情况下,分析所述化学反应的生成物,而在其它情况下,化学反应被用于抑制组分的不希望的电离。
10.在该实施例中,施加降低的压力(p<1巴)。在这种情况下,由于可以在质谱仪的真空室中进行电离,因此所形成的离子不需要穿过大气界面被传递至质谱仪,导致大量的离子损耗。
11.在另一优选实施例中,液体射流被用于在样品中切割(即,液体射流被用作切割器件)。该实施例提供关于正被切割的对象的化学成分的实时信息。在液体射流的分裂时形成的小滴通过移动穿过电势梯度而带电荷,并且使用质谱来分析所形成的离子。通过使用高电压电源,在被切割的样品与质谱仪之间建立电势差。
12.在另一优选实施例中,使用已知的外科手术工具(电外科手术工具、手术刀、激光等)来在样品中切割,并且使用上述方法分析样品的这样暴露的部分。这样,通过传导由所述液体形成的小滴和穿过电势梯度的所述样品并且通过质谱分析所得到的气态离子,已知的外科手术方法与根据本发明的方法的结合连续地提供关于正被切割的样品的成分的信息。通过使用高压电源来在样品和质谱仪之间建立电势梯度。在又一实施例中,优选在电离步骤之后,通过使用气体流(吹入或抽吸),所述小滴和样品的粒子也可以被传送至分析器单元。

图1示例了根据本发明的装置的工作原理。在根据本发明的方法的实施例期间,通过将导电液体2泵送经过喷嘴3,产生高速液体射流1。通过管道2B,将液体2传送到喷嘴3。通过使用高压电源4,在地与液体射流1之间产生几千伏量级的电势差。被导向表面5上的液体射流1产生液体小滴7,如果表面5是绝缘体,或者导电但与地电绝缘,这些液体小滴7就带上电荷。带电荷的小滴7包含被沉积到表面5上的样品6的粒子(例如分子),这些粒子可溶于液体2中。溶剂2从带电荷的小滴7的蒸发导致形成可溶于液体2中的样品6的组分的气态离子8,这些气态离子8可通过样品收集器单元9(优选地大气界面)而被传送到分析器单元10(优选为其中可通过质谱方法分析离子的质谱仪)中。通过该方法,可以采集关于表面5的信息,或者,如果表面5是惰性的,则可以采集关于被沉积到表面5上的样品6的信息。
图2图示了根据本发明的装置的优选实施例。在50至1500巴的压力下,通过不锈钢管道11,泵送导电液体2,例如0.1mM的HCl水溶液。通过将高压电源4连接到不锈钢管道11,在液体2与地之间产生1至8kV的高电势。通过被不锈钢连接器12、不锈钢密封件13和没有螺丝的螺旋夹具14夹持的不锈钢或者蓝宝石喷嘴15,液体2以100至1000m/s的线速度从管道11射出。通过螺旋夹具12,不锈钢管道11被安装在旋转的且三维线性移动台上。移动系统提供对喷嘴3、样品6和样品收集器单元9的相对位置、以及碰撞角15和收集角16的适当控制。喷嘴3与表面5之间的最佳距离在1至20mm的范围内,并且最佳的碰撞角15在60至90度的范围内。通过排泄管17排出在表面上有时聚集的过量液体,该排泄管17的远端被连接到泵。
图3图示了在实例2中描述的根据本发明的装置的优选实施例。与图2不同,喷嘴3是通过焊接来密封1/16”外径的不锈钢管道11的端部而产生的,并且通过激光钻孔的手段在端壁中钻出直径为1μm的0.2mm长的孔。为了在深度上磨蚀表面18,与样品6的表面平行地移动液体射流1,而可溶于所采用的液体的被侵蚀的表面材料的特定组分被转换成气态离子8,通过单元9,这些气态离子8被传送至质谱仪。聚乙烯排泄管17被用于在合适的泵的辅助下排出在表面上聚集的过量液体。
图4图示了在实例3中详细描述的根据本发明的装置的优选实例。与图2和3不同地,图4的装置包括由熔融二氧化硅(silica)制成的喷嘴3,该喷嘴3是通过使用毛细管牵引(puller)设备制成的。与在图2中给出的实施例类似地,通过连接器13将熔融二氧化硅喷嘴3连接到不锈钢管道11。与图2和3中图示的实施例不同地,不通过质谱仪的大气界面,而是通过使用连接到分析器单元(质谱仪)的长1m、外径为1/8”、内径为2mm的铜管道20,直接收集离子。通过加热器21,加热铜管道20,该加热器21通过使用从温度计22反馈的温度而受到控制。
该装置不包括表面5,因为其主要功能是对对象直接取样,并且也可以被用作外科手术切割设备。通过夹具19来确保该装置的部件的紧固。
与图2和3的实施例类似地,也应用聚乙烯排泄管17来排出在表面上偶然聚集的过量液体。通过夹具19来提供装置的部件的相对位置。图4中所示的装置的目的不是分析样品6的表面,而是通过方法23来切割样品6,并且采集关于被切割样品的成分的化学信息。
图5示出通过在图2中图示的装置获得的质谱。在聚(异丁烯酸甲酯)表面上沉积且干燥10μl的包含100ng母鸡蛋白溶菌酶的水溶液。使用Thermo Finnigan LCQ Duo质谱仪分析从表面解吸的溶菌酶的气态离子。在该谱中,可以识别出具有10、9和8个电荷(10倍、9倍、8倍的质子化形式)的溶菌酶离子。
图6示出通过在图2中图示的装置获得的质谱。在玻璃表面上干燥10μl的包含10ng缓激肽(bradykinin)的水溶液。使用Thermo Finnigan LCQDuo质谱仪分析从表面解吸的肽的气态离子。在该谱中,可以识别出具有2和1个电荷(1倍和2倍的质子化形式)的缓激肽离子。
图7示出通过在图3中图示的装置获得的质谱。在玻璃表面上沉积20μm厚的鼠脑冷冻切片。通过解吸电离获得样品的脂类成分的负离子,并且使用Thermo Finnigan LCQ Duo质谱仪分析这些负离子。在该谱中,可以识别出脂肪酸和磷脂的离子。
图8示出通过在图4中图示的装置获得的质谱。使用Thermo FinniganLCQ Duo质谱仪分析从外科手术暴露的心脏的表面通过水射流方法电离的组分的负离子。在该谱中,可以识别出在心脏的新陈代谢过程中起重要作用的离子。
具体实施方式
通过以下的工作实例并且参考附图,详细描述根据本发明的方法,但不将权利要求的范围限制于这些实例和附图。
实例1
用来分析干燥的溶剂溶液小滴的用于质谱的水射流解吸离子源
1.1用于质谱的水射流解吸离子源包括以下部分:
-HPLC泵(Jasco),
-1/16”OD,1mm ID的不锈钢管道(11),
-连接器(Swagelok,Upchurch)(13),
-密封件(Swagelok,Upchurch)(14),
-5μm ID的蓝宝石喷嘴(3),
-2个用于3D线性移动的移动台(Newport),
-用于在一个维度上旋转的旋转台(Newport),
-高压电源(Bertan)(4),
-HDPE管道,1/16”OD,1mm ID(17),
-膜泵,
-质谱仪(Thermo Finnigan LCQ Duo)。
1.2用于质谱的水射流解吸离子源的结构
在图2中示出装置的示意图。不锈钢管道11通过HDPE管道而连接到HPLC泵,并且根据图2连接喷嘴3。利用合适的螺旋夹具12将具有喷嘴的不锈钢管道的端部安装在旋转台上,并且以使不锈钢管道与移动台电绝缘的方式,将所述旋转台安装在3D线性移动台系统上。所述移动台系统被安装在源平台上,通过利用合适的螺栓,该源平台被安装在质谱仪的大气界面部件9上。高压电源4的电出口被连接到不锈钢管道11。
通过利用螺钉,将由聚乙烯制成的样品承载板安装到另一3D线性移动台系统上。并且,以根据表1中给出的值来设定图2中限定的几何参数的方式,将3D线性移动台系统安装在所述源平台上。
以HDPE管道的端部距离喷嘴1mm的方式,将用于从表面排出过量液体的HDPE管道17安装到不锈钢管道11。HDPE管道17的远端(即,距离样品较远的一端)被连接到用于所述目的的膜泵。
1.3.使用水射流解吸离子源来研究干燥后的溶剂小滴
在1mm厚的聚(异丁烯酸甲酯)表面5上滴落且干燥溶解相的样品。通过以10μl/min的流速由HPLC泵经由喷嘴泵送0.1%的乙酸水溶液,产生被导向至表面上的液体射流,其中从喷嘴3射出的液体射流与样品表面成70度的碰撞角。以图中所示的收集角为20度的方式,定位样品和喷嘴。其它实验细节总结在表1中。

通过使用3D线性移动台系统,连续地研究在表面上沉积和干燥的样品6。图5示出了在表面上沉积的作为溶液滴的100ng缓激肽的质谱,而图6示出了在表面上作为溶液滴沉积的10ng细胞色素C的质谱。这些谱表明与通过电喷雾电离所获得的化合物的谱的高度相似性,并且基于相同总则进行对谱的解释。在射流解吸与电喷雾谱的特征之间的相似性是与实际上由多电荷小滴形成离子的事实相关联的。与电喷雾相比,射流解吸电离的主要优点之一是完全消除了在高吞吐量分析(每分钟多于10个样品)的情况下的交叉污染。
实例2

2.1用来定义样品中特定化合物的浓度的空间分布的用于质谱的水射流解吸离子源包括以下部分:
-HPLC泵(Jasco),
-1/16”外径、1mm内径的不锈钢管道,通过在0.2mm的长度上焊接,该不锈钢管道在一端上被密封,并且通过激光钻孔的手段,钻孔穿过被密封部分,以形成1μm直径的圆形截面小孔(11)
-连接器(Swagelok,Upchurch)(13),
-密封件(Swagelok,Upchurch)(14),
-2个用于3D线性移动的计算机控制的移动台(Newport),
-用于在一个维度上旋转的旋转台(Newport),
-高压电源(Bertan)(4),

2.2用来定义样品中特定化合物的浓度的空间分布的水射流解吸离子源的结构
在图3中示出装置的示意图。以喷嘴3为端部的不锈钢管道11通过HDPE管道而连接到HPLC泵。将不锈钢管道的端部安装在旋转台上,并且以使不锈钢管道与移动台电绝缘的方式,将所述旋转台安装在3D线性移动台系统上。所述移动台系统被安装在源平台上,通过利用合适的螺栓,该源平台被安装在质谱仪的大气界面部件上。高压电源4的电出口被连接到所述不锈钢管道11。
使用合适的螺钉,将由聚乙烯制成的样品承载板安装到另一3D线性移动台系统上。并且,以设定图2中限定的几何参数的方式,将3D线性移动台系统安装在所述源平台上。
以HDPE管道的端部距离喷嘴1mm的方式,安装用于从表面排出过量液体的HDPE管道17。HDPE管道17的远端(即,距离样品较远的一端)被连接到用于所述目的的膜泵。
2.3.使用水射流解吸离子源来定义样品中特定化合物的浓度的空间分布
将样品6,例如生物组织的解剖体,安装到样品承载板上,并且,如果需要,对其进行固定。对装置设定表2中列出的工作参数,并且以10μm/min的速度使样品相对于喷嘴和质谱仪沿图3中示出的19移动。在对表面扫描期间连续地获得质谱。由于样品的10至50μm厚的上层18被完全消融,因此可以重复扫描,直到样品被完全消耗掉。计算机控制的移动台使得所收集的数据能够相对于原位置显像。其能够确定特定组分的空间浓度分布。图7示出了通过在图中给出的装置而获得的鼠脑部分的质谱。

实例3
基于水射流解吸的外科手术设备
3.1基于水射流解吸的外科手术设备包括以下部分(该设备示于图4中):
-HPLC泵(Jasco),
-1/16”外径、1mm内径的不锈钢管道(11),
-连接器(Swagelok,Upchurch)(13),
-密封件(Swagelok,Upchurch)(14),
-具有1至5μm内径的牵引式二氧化硅毛细管喷嘴(3),

-HDPE管道,1/16”外径、1mm内径(17),
-膜泵,
-质谱仪(Thermo Finnigan LCQ Duo)。
3.2基于射流解吸的外科手术设备的结构
具有0.32mm外径和10μm内径的熔融二氧化硅毛细管在一端(3)被拉到1μm外径,而其另一端被连接到具有1/16”外径的HDPE管道,该HDPE管道被连接到HPLC泵。具有1m的长度、1/8”外径和2mm内径的铜管道(20)被连接到质谱仪的入口,其中铜管道(20)具有加热器(21)和温度计(22)。通过电子温度控制器来连接加热器和温度计。
喷嘴、连接到质谱仪的铜管道和用于抽吸过剩的水的HDPE管道被嵌入到由PEEK聚合物材料制成的夹具(19)中。
3.3.基于射流解吸使用外科手术设备
在HPLC泵上开关,该设备能够用于切割任意软对象,例如,生物组织。如图4所示,液体射流在样品(6)的表面上形成腔(23),其深度随着时间而增加,并且设备的横向移动切割样品对象。为了通过该设备产生关于组织的相关化学信息,发现表3中列出的参数是最优的。图8示出了在外科手术干预的过程中该设备所记录的关于鼠心脏的质谱。

Claims (33)
1.一种将凝聚相样品的特定组分转换成气态离子及其分析的方法,其中通过从解吸单元释放的分析束,使得离子或可转换成离子的样品粒子从样品移位,并且通过分析器单元分析所获得的气态离子,包括以下步骤:
应用液体射流作为分析束,使得当所述液体轰击表面时形成的小滴的液体蒸发。
2.根据权利要求1的方法,其中水、水溶液、或任何其它极化溶剂、或者它们的任何混合物被用作所述液体射流的液体组分。
3.根据权利要求1或2的方法,其中在所述分析器单元与等电势的所述液体射流和样品之间施加电势差。
4.根据权利要求1或2的方法,其中质谱仪或者离子迁移谱仪被用作所述分析器单元。
5.根据权利要求1或2的方法,其中所获得的所述气态离子通过样品收集器单元而被传送到所述分析器单元。
6.根据权利要求1或2的方法,其中所述小滴或者移位的样品粒子在所述分析器单元与样品之间被电离。
7.根据权利要求1或2的方法,其中所述样品被沉积在表面上。
8.根据权利要求1或2的方法,其中通过冷却或加热在外部控制所述样品的温度。
9.根据权利要求1或2的方法,其中在所述方法期间通过抽吸排出从所述液体射流产生的且未传送到所述分析器单元的过量液体。
10.根据权利要求1或2的方法,其中在所述分析之前电离所述液体小滴或通过所述液体的所述蒸发获得的样品粒子。
11.根据权利要求1或2的方法,其中采用多个液体射流。
12.根据权利要求1或2的方法,其中为了确定所述样品的成分的空间分布,使得一个或多个所述解吸单元相对于所述样品移动。
13.根据权利要求1或2的方法,其中为了确定所述样品的成分的空间分布,使得所述样品相对于一个或多个所述解吸单元移动。
14.根据权利要求1或2的方法,为了确定所述样品的成分的深度分布,所述液体射流被用于在所述样品中切割。
15.根据权利要求1或2的方法,其中所述样品是生物组织。
16.根据权利要求15的方法,其中通过任何已知的外科手术方法暴露所述样品。
17.根据权利要求10的方法,其中在电离之后,通过气体流注方法将所述小滴以及在所述液体射流和所述样品的相互作用时形成的样品粒子传送到所述分析器单元。
18.根据权利要求1或2的方法,其中化合物被混合到所述液体射流中,所述化合物与所述样品的特定组分反应。
19.根据权利要求1或2的方法,其中所述方法在与大气压不同的压力条件下执行。
20.一种用于将凝聚相样品的组分转换成气态离子的装置,包括:
用于承载样品(6)的表面(5);
至少一个解吸单元(A),用于使得离子或可转换成离子的样品粒子(8)从样品(6)移位;
样品收集器单元(9);
分析器单元(10),
其特征在于,所述解吸单元(A)具有提供液体射流(1)的喷嘴(3)、用于传送液体(2)的管道(2B),所述管道连接到所述喷嘴(3),并且所述喷嘴(3)被导向至所述用于承载所述样品(6)的表面(5)。
21.根据权利要求20的装置,其中所述液体射流是水、水溶液、或任何其它极化溶剂、或者它们的任何混合物。
22.根据权利要求20或21的装置,包括用于在所述液体射流(1)和所述表面(5)之间产生电势差的器件(4)。
23.根据权利要求20或21的装置,其中所述分析器单元(10)是质谱仪或者离子迁移谱仪。
24.根据权利要求20或21的装置,其中所述样品收集器单元(9)的出口(9A)被设置为紧靠着表面(5)。
25.根据权利要求20或21的装置,其中用于蒸发所述液体(2)的单元被设置在所述表面(5)与所述分析器单元(10)之间。
26.根据权利要求20或21的装置,其中用于电离所述样品粒子(8)的单元被设置在所述表面(5)与所述分析器单元(10)之间。
27.根据权利要求20或21的装置,其中至少一个所述解吸单元(A)具有相对于所述样品(6)的位置控制器,或者用于承载所述样品(6)的所述表面(5)具有相对于所述解吸单元(A)的位置控制器。
28.一种用于将凝聚相样品的组分转换成气态离子的装置,其可通过施加液体射流而用于在样品中挖出腔或者用于切割样品,包括:
至少一个解吸单元(A),用于使得离子或可转换成离子的样品粒子(8)从样品(6)移位;
管道(20),其是样品收集器(9)的一部分,且连接到分析器单元,
其特征在于,所述解吸单元(A)具有提供液体射流(1)的喷嘴(3)、用于传送液体(2)的管道(2B),所述管道连接到所述喷嘴(3),并且所述解吸单元(A)和连接到分析器单元的所述管道(20)通过夹具(19)被彼此紧固。
29.根据权利要求28的装置,其中所述液体射流是水、水溶液、或任何其它极化溶剂、或者它们的任何混合物。
30.根据权利要求28或29的装置,包括用于在所述液体射流(1)和连接到分析器单元的所述管道(20)之间产生电势差的器件(4)。
