技术领域
本发明涉及化学领域,更具体地说涉及应用于食品工业和食品添加物的化学领域。本发明提供这样的新的化合物,它是由氨基酸和/或铵盐稳定的矿物盐,具有更高的水溶解度、更好的感官特性和更高的生物利用度。
背景技术
文献中已充分说明了可补充动物和人类对必需矿物质的需求的适当饮食的重要性。有大量报道显示出由必需矿物质的缺乏引起的问题,以及这些问题通过矿物质的摄入而好转的情况。矿物质在细胞代谢过程中必不可少,它们可作为酶的辅助因子,亦可作为电子转移链的一部分,等等。
矿物质由一组不向有机体提供能量但具有重要的调控功能以及在作为许多组织一部分时具有结构功能的营养素(约30种)组成。它们是骨骼和牙齿的组成部分,它们控制细胞内和细胞外液体的组成,并且它们是生命必需的分子——酶和激素——的一部分。其中具有两大类:常量元素和微量元素或痕量元素。之所以称为常量元素是因为它们必须通过饮食以较高的量供应或者因为它们在身体组织中具有较高的比例,其包括:钙、磷、镁、硫、钾、钠和氯。其它的例如钴、铜、铁、铬、氟、碘、锰、硒和锌也是必需的,但量较少,因此被称为微量元素或痕量元素。
同样,提供适量的氨基酸也很重要。氨基酸是蛋白质的主要成分。我们所消耗的食物为我们提供蛋白质。但是,这种蛋白质通常不以这样一种组合物的形式吸收;而是在由消化过程引起蛋白质裂解(“水解”或分解)后,经过肠壁以氨基酸和短链肽的形式吸收,即已知的“肠肝循环”。最初这些物质进入血流,并由此处被分配至需要它们以形成蛋白质的组织中,所述蛋白质在生命循环过程中被消耗。公知除了20种已知的蛋白质氨基酸以外,还有8种人类生命不可缺少(或必需)的氨基酸和2种“半必需”的氨基酸。这10种氨基酸必须通过有机体的日常食物摄取进入有机体,并且主要在有机体最需要它们时——机能障碍或疾病发生时——进入有机体。最成问题的必需氨基酸是色氨酸、赖氨酸和蛋氨酸。
谷物和块茎植物组成饮食主要部分的人群通常缺乏必需氨基酸。必需氨基酸的缺乏对儿童的影响比成人大。有必要指出,只要缺乏它们(必需氨基酸)中的一种,则不可能合成任何其中需要所述氨基酸的蛋白质。根据限制性氨基酸的不同,这会导致不同类型的营养不良。
值得注意的是,若为了获得合适的营养,则饮食中必须含有适量的必需金属,其中所述金属的大部分天然存在形式实际上是不能被利用的,因为含有这些金属的大部分化合物不能被高效地吸收、分配和使用。
用于对抗人类、动物或植物中缺失综合征的无机酸或有机酸的矿质营养素盐在其使用方面具有一些限制性因素,例如限制其可接受性的金属口味和/或限制其吸收和生物利用度的低水溶解度。必需元素的吸收往往取决于所用盐所具有的溶解度。因此,一般而言溶解度越高吸收越好。
其它相关的因素是一旦元素被吸收后的生物学行为。这意味着该元素必须遵循与已知的被当作参照的盐相同的代谢途径。
使用氨基酸和/或铵盐与有机酸或无机酸的盐共同形成的产品口味更好并且溶解度更高。
以前,开发了1∶1比例的必需金属和氨基酸的不同配合物,以提高所述金属在它们消耗后的生物利用度(参见专利No.3,941,818、3,925,433、3,950,372、4,021,569和4,067,994)。在所述专利中,所得的化合物形成螯合物。螯合物是一个非常简单的术语,它是指在配位化合物中包含金属中心的环结构。在一个具有一个以上“齿”的配体与同一金属中心配位时即产生这种化合物结构。
本发明的盐不是上文中所描述体系的螯合化合物,但它们由配位键(dative bond)稳定并具有很高的溶解度(高于50b/w或每升水中400gr的溶液),这使得可以简单地将它们加入食物中,而不会改变水相的体积。
发明内容
因此,本发明的一个目的是获得与一种氨基酸和/或铵盐结合的有机酸或无机酸的矿物盐产品,它具有以下通式或结构:

本发明的另一个目的是提供用氨基酸和/或铵盐稳定的矿质营养素的盐,其中酸选自纯的或结合的(pure or associated)富马酸、苹果酸、盐酸(chloridic acid)或硫酸。
另外,本发明的另一个目的是提供用氨基酸和/或铵盐稳定的矿质营养素的盐,其中氨基酸选自氨基乙酸(甘氨酸)和/或赖氨酸,铵盐为有机的或无机的铵盐。
此外,本发明的另一个目的是这样的含有用氨基酸和/或铵盐稳定的矿物盐的食品补充剂,它可通过有效地增加矿物质的吸收、分配和使用从而改善人和动物的营养和健康。
另外,本发明的另一个目的是这样的用氨基酸和/或铵盐稳定的矿质营养素的盐,其中具有适当的介质,例如水、苏打水、包括天然果汁或合成果汁的液体或粉末形式的果汁、酸乳酪、乳制品甜品、黄油、奶酪和人造黄油。
此外,本发明的另一个目的是这样的用氨基酸和/或铵盐稳定的矿质营养素的盐,其中的介质或选定的形式包括用于药物产品、饮食补充剂或兽医用途的药丸、泡腾剂粉、粉末、片剂、糖果、硬胶囊或软胶囊。
此外,本发明的另一个目的是可作为淀粉类产品一部分的用氨基酸和/或铵盐稳定的矿质营养素的盐,这些淀粉类产品例如面包、面团、饼干、油酥点心、脱水的或浓缩的食品,如炖制品、汤、点心、果冻、颗粒状或或挤出成型的谷类薄片、谷物棒、整粒豆(whole grain soy)或面粉制成的粉末状或薄片形式的副产品,以及燕麦。
此外,本发明的另一个目的是这样的用氨基酸和/或铵盐稳定的矿质营养素盐:当所用介质含水时,所得溶液可用作叶面肥料或其它肥料。
另外,本发明的另一个目的是一种用氨基酸和/或铵盐稳定的矿质盐的合成方法,其特征在于包括以下步骤:
a)将之前形成盐溶于氨基酸和/或铵盐的水溶液
b)适当地加热并调节pH值。
c)获得在水溶液中形成的产物。通过将水蒸发,可以获得干燥粉末形式的产物。可以使用产物的溶液或干燥粉末。

c)过滤,以分离所沉淀的硫酸钙和仍在水溶液中的产物。
d)将水蒸发,直至获得干燥粉末形式的产物。
另外,本发明的另一个目的是一种用于合成盐的方法,其特征在于包括以下步骤:
a)溶解在氨基酸和/或铵盐的水溶液中,以形成通式的关系。
b)加热溶液并调节pH值,
c)所得产物保留在水溶液中,并可通过蒸发水而获得干燥粉末。
其中所述盐是硫酸铜、硫酸锌、硫酸钾、硫酸钠、硫酸锰、硫酸铁和硫酸亚铁,氯化钠、氯化钾或氯化钙。

具体实施方式
已开发出与氨基酸和/或铵盐结合的纯的或阳离子组合物的形式的有机阴离子的产物和无机阴离子的产物,其中有机阴离子例如富马酸根、苹果酸根、葡糖酸根、乳酸根,无机阴离子例如硫酸根或氯离子,所述阳离子例如钙、镁、铁、锌、铜、锰、钠、钾、钴和镍,所述氨基酸例如甘氨酸(氨基乙酸)和/或L-赖氨酸。
当将所形成的盐与作为标准的未结合的盐一起评价,并且具有相同的需要提供的金属离子浓度时,其显示出显著更高的水溶解度,并具有可接受的口味。
所形成产品的这些特性使其可极好地用于提供饮食补充剂、药物产品和食品及饮料中矿物质常量营养素和微量营养素含量,方便地将其增加或富化。
所形成的化合物的通用化学结构对应于以下图示:
{[Ac]n-·Men+←n[氨基酸和/或铵盐]}·x H2O
其中:
n取1至3的值,并且
x取0至10的值。
金属Men+可以是钙、镁、锌、铁、铜、锰、钠、钾、钴或镍。
酸可以是纯的或结合的富马酸、乳酸、葡糖酸、苹果酸、盐酸或硫酸。
氨基酸可以是氨基乙酸(甘氨酸)和/或赖氨酸。
铵盐可以是有机的或无机的铵盐。
对所形成产物的结构进行的研究,显示出这样一种产物的结构,其中氨基酸和/或铵盐的氨基氮通过配位共价键结合至金属上(参见图1和2)。
与氨基酸的氨基氮配位的共价键的功能鉴定通过与茚三酮的反应表征(参见图1和2)。
图1示出与甘氨酸结合的富马酸锌(Zn的生物学富马酸盐)样品和甘氨酸标准样品。
图2示出与茚三酮反应后的相同的样品。甘氨酸标准样品的紫色显示出甘氨酸是游离的。黄色对应于用茚三酮再生(restore)的产物的反应,所述反应显示出甘氨酸的羧基是游离的,而氨基与金属(本例中为Zn)结合。如果甘氨酸是游离的,颜色会为紫色。但是,如果甘氨酸的两种官能团(氨基和羧基)都结合至金属上(如在氨基螯合化合物中出现的情况),就不会存在任何与茚三酮的反应,则颜色为无色。
产物的特征在于不会形成螯合物,并且所得的盐比盐本身具有更好的溶解度和口味。
因此,本发明的目的是获得与氨基酸和/或铵盐结合的有机或无机酸的矿物盐产品,其具有以下通式:
{[Ac]n-·Men+←n[氨基酸和/或铵盐]}·x H2O
它具有更好的口味和更高的水溶解度,从而具有更高的生物利用度。
产物的制备方法可包括以下步骤:
a)将之前形成的盐溶于氨基酸和/或铵盐水溶液中,以一种适当的方式加热并调节pH值。获得水溶液中形成的产物。通过将水蒸发可获得干燥粉末形式的产物,可使用产物的溶液或干燥粉末。
b)所形成的钙盐可分别通过加入铁或亚铁、铜或锰的硫酸盐被铁或亚铁、铜或锰的离子取代。所形成的硫酸钙沉淀出来,而具有相同阴离子的铁或亚铁、铜或锰的盐保留在溶液中,并与氨基酸和/或铵盐结合。过滤,以分离所沉淀的硫酸钙和仍留在水溶液中并可通过蒸发水被变成干燥粉末的所得产物。
c)还可将例如硫酸铜、硫酸锌、硫酸钾、硫酸钠、硫酸锰、硫酸铁或硫酸亚铁的盐以通式中所示的关系溶解于氨基酸的水溶液中,加热该溶液并调节pH。所得的产物保留在水溶液中,并可通过蒸发水而获得干燥粉末。
d)还可将例如氯化铜、氯化锌、氯化钾、氯化钠、氯化锰、氯化铁或氯化亚铁的盐以通式中所示的关系溶解于氨基酸的水溶液中,加热该溶液并调节pH。将所得的产物保留在水溶液中,并可通过蒸发水获得干燥粉末。
溶解度的比较数据
1.富马酸亚铁(参见4094 Index Merck XII Ed.,第686页)SP169.90,Fe++的含量为32.87%,AW Fe 55.847。
25℃下水中的溶解度:每升水1.4克。这容许每升水溶解0.460grFe++(亚铁)。用2摩尔甘氨酸(氨基乙酸)稳定的富马酸亚铁:
含有
富马酸亚铁169.90gr(53.09%)。
甘氨酸(2×75.07)150.14gr(46.91%)。
产物320.04gr(17.45%的Fe)。
25℃下,用2摩尔甘氨酸稳定的富马酸亚铁在水中的溶解度:54.6克每升。
富马酸亚铁(53.9%):29.0克/升。容许每升水中溶解9.53克Fe++(亚铁),溶解度每升水增加:20.7倍(由以下得到:9.53gr的Fe/0.460gr的Fe)。
所述数据清楚地显示出一种例如富马酸亚铁的盐的水中溶解度增加,该盐用于提供强化食品中的食品补充剂的矿物质微量营养素离子。
2.富马酸钙3H2O(参见manufacturer Bartek Inc.,加拿大)SP208.18钙的含量为19.25%(aw Ca 40.08)。
25℃下水中的溶解度:每升水中1.22克。这容许每升水中溶解0.234克Ca++。用2摩尔甘氨酸(氨基乙酸)稳定的富马酸钙3H2O

溶解度增加:以钙计23.9倍(由以下得到:5.591/0.234gr)
实施例:
本文中提供的以下具体实施例是用于说明本发明的本质。这些实施例仅用于说明的目的,而不应被理解为本发明所要求的保护范围的限制因素。
实施例1:
使用氨基乙酸(甘氨酸)的5.0%溶液制备钙盐,如富马酸钙。
1.将以下物质装入2000CC的烧杯:
净化水:1800gr。称氢氧化钙(至少95%):24.84gr(检查纯度和湿度),使其分散均匀。
2.称氨基乙酸(至少98.5%):47.92克,并加入该烧杯中。水浴器中搅拌加热。
3.称富马酸(至少99.0%):37.05克,并加入该烧杯中。继续在水浴器中搅拌加热,以使溶液保持60至85℃不超过30分钟。
4.取一份样品,根据需要使用氢氧化钙或富马酸调节溶液的pH值至6.5(误差5%,6.2至6.8)。
5.pH值调好后检测钙,结果必须是钙不少于0.56gr%。
6.将所得的溶液通过滤纸过滤,并优选用一种产品促进过滤,由此获得结晶溶液。
7.所得的溶液可使用热空气干燥,直至其变干。粉末可含有不超过15%的结晶水。
8.获得约100至112克的产物,该重量取决于结晶水的含量。
9.在工业规模上,溶液在适当的反应器中制备,并通过喷雾装置干燥。
实施例2:
使用氨基乙酸(甘氨酸)的10%溶液制备亚铁盐,如富马酸亚铁:
1.将以下物质装入2000CC的烧杯:
净化水:900mL或gr。称氢氧化钙(至少95%):24.84gr(检查纯度和湿度),搅拌使其分散均匀。
2.称氨基乙酸(至少98.5%):47.92克,并加入该烧杯中。水浴器中搅拌加热。
3.称富马酸(至少99.0%):37.05克,并加入该烧杯中。继续在水浴器中搅拌加热,以使溶液保持60至85℃不超过30分钟。
4.取一份样品并调节溶液的pH值至4.5(误差5%)。
5.pH值调好后,立刻加入硫酸亚铁7H2O(至少19.8%Fe++):88.74gr,停止加热,继续搅拌不少于30分钟。
6.将上述溶液静置沉降,上层清液通过过滤处理进行过滤,获得暗绿色结晶溶液。
该溶液含有约1.75%的铁和10%(9.5至10.5%)的溶解产物。
7.所得溶液可使用热空气干燥,直至其变干。在105-108℃下干燥的损失不应超过10%。
8.获得约100至115克的产物,该重量取决于结晶水的含量。
9.在工业规模上,溶液在适当的反应器中制备,并通过喷雾装置干燥。
Claims (16)
1.用氨基酸和/或铵盐稳定的矿质营养素的盐,其中所述盐用有机酸的阴离子或无机阴离子和金属阳离子与氨基酸和/或铵盐结合获得,其特征在于金属阳离子通过一个离子键结合至阴离子上,并通过一个共价的配位键与氨基酸和/或铵盐的氨基结合,该盐具有一个如下的通式:
{[Ac]n-·Men+←n[氨基酸和/或铵盐]}·x H2O
其中:
-n取1至3的值,并且
-x取0至10的值。
2.权利要求1的用氨基酸和/或铵盐稳定的矿质营养素的盐,其特征在于所述金属“Men+”选自以下的阳离子:钙、镁、锌、铁、铜、锰、钠、钾、镍或钴。
3.权利要求1的用氨基酸和/或铵盐稳定的矿质营养素的盐,其特征在于所述酸选自纯的或结合的富马酸、苹果酸、盐酸或硫酸。
4.权利要求1的用氨基酸和/或铵盐稳定的矿质营养素的盐,其特征在于所述的氨基酸选自氨基乙酸和/或赖氨酸,并且所述铵盐可为有机的或无机的铵盐。
5.根据权利要求1的被稳定的矿质营养素的盐,其特征在于它们是被包覆的或制成胶囊的。
6.一种用于治疗人类、动物和植物的营养缺乏症,且是常量和微量矿质营养素来源的产品,其特征在于其含有前述权利要求中任一项的盐。
7.一种通过有效地增加矿物质的吸收、分配和使用从而改善人类和动物的营养和健康的食品补充剂,其特征在于含有前述权利要求的由氨基酸和/或铵盐稳定的矿物盐以及至少一种可接受的介质。
8.权利要求6的通过有效地增加矿物质的吸收、分配和使用从而改善人类和动物的营养和健康的食品补充剂,其特征在于所述介质选自水、苏打水、包括天然果汁或合成果汁的液体或粉末形式的果汁、酸乳酪、乳制品甜品、黄油、奶酪和人造黄油。
9.权利要求6的通过有效地增加矿物质的吸收、分配和使用从而改善人类和动物的营养和健康的食品补充剂,其特征在于所述介质选自可用于药物产品、饮食补充剂或兽医用途的药丸、片剂、泡腾剂粉、粉末、片剂、糖果、硬胶囊或软胶囊。
10.权利要求6的通过有效地提高矿物质的吸收、分配和使用从而改善人类和动物的营养和健康的食品补充剂,其特征在于它可以是淀粉类产品的一部分,所述淀粉类产品例如面包、面团、饼干、油酥点心、脱水的或浓缩的食品,如炖制品、汤、点心、果冻、颗粒状或挤出成型的谷类薄片、谷物棒、整粒豆或面粉制成的粉末状或薄片形式的副产品,以及燕麦。
11.一种可用作叶面肥料或其它肥料的产品,其特征在于它含有权利要求1的盐和含水介质。
12.一种用于合成权利要求1的用氨基酸和/或铵盐稳定的矿物盐的方法,其特征在于包括以下步骤:
a)在氨基酸和/或铵盐的水溶液中溶解之前制得的盐,使它们形成权利要求1中通式的关系,
b)在介于X和Y℃之间的一个温度下加热,并调节pH至一个A和B之间的值,以及
c)获得于水溶液中的所制得的产物。
13.权利要求12的方法,其特征在于将所述产物的水溶液中的水蒸发,直至获得干燥粉末形式的产物。
14.用于合成权利要求1的用氨基酸稳定的铜、铁和锰盐的方法,其特征在于包括以下步骤:
a)分别通过加入硫酸铁或硫酸亚铁、硫酸铜或硫酸锰,将所形成的钙盐用铁或亚铁、铜或锰的离子取代;
b)所形成的硫酸钙沉淀出来,从而使具有相同阴离子的铁或亚铁、铜或锰盐保留在溶液中,并结合至氨基酸和/或铵盐上;
c)过滤以将沉淀的硫酸钙分离出来,从而使所形成的产物保留在水溶液中;
d)将水蒸发,直至获得干燥粉末形式的产物。
15.权利要求1的方法,其特征在于所述的盐选自硫酸铜、硫酸锌、硫酸钾、硫酸钠、硫酸锰和硫酸铁或硫酸亚铁。
16.权利要求1的方法,其特征在于所述的盐选自氯化钠、氯化钾和氯化钙。
