Abstract
本发明涉及一种cDNA分子,其编码源自一种经批准的疫苗株的麻疹病毒(MV)的全长反基因组(+)RNA链的核苷酸序列。本发明还涉及使用所述cDNA制备免疫原性组合物。
Description
麻疹病毒是单负链病毒目(mononegavirales)的一个成员,即具有非节段负链RNA基因组的病毒。麻疹病毒(MV)的非节段(non-segmented)基因组具有一种反信使极性,其产生一种当纯化时既不在体内或体外翻译也非感染性的基因组RNA。
对非节段负链RNA病毒的转录和复制及其装配为病毒颗粒在病毒学领域已经特别加以研究和报道(3rdedition,vol.1,1996,Lippincott-Raven publishers-Fields BN et al)。麻疹病毒的转录和复制不涉及感染细胞的核,而是在所述感染细胞的细胞质中发生。麻疹病毒的基因组包含编码来自6个基因(称为N、P、M、F、H和L)的6个主要结构蛋白及来自P基因的额外的两个非结构蛋白的基因。所述基因顺序如下:3’,N,P(包括C和V),M,F,H及在5’末端的大聚合酶蛋白L。该基因组在基因间区域M/F进一步包含非编码区;这个非编码区含有大约1000个非翻译RNA的核苷酸。所述基因分别编码前导肽(I基因)、病毒的核壳蛋白即核蛋白(N)、磷蛋白(P)及大蛋白(L),它们围绕基因组RNA装配以提供所述核壳。其它基因编码病毒包膜蛋白,包括血凝素(H)、融合蛋白(F)和基质蛋白(M)。
麻疹病毒已经被分离而且从1954年分离的Edmonston MV中通过在原代人肾细胞或羊膜细胞上连续传代已经获得活的减毒疫苗(Enders,J.F.,and T.C.Peebles.1954.Propagation in tissue cultures odcytopathogenic agents from patients with measles.Proc.Soc.Exp.Biol.Med.86:277-286.)。然后使所用的毒株适应鸡胚成纤维细胞(CEF)以产生Edmonston A和B种子(seed)(Griffin,D.,and W.Bellini.1996.Measles virus,p.1267-1312.In B.Fields,D.Knipe,et al.(ed.),Virology,vol.2.Lippincott-Raven Publishers,Philadelphia)。Edmonston B于1963年作为第一个MV疫苗被批准。Edmonston A和B在CEF上进一步传代产生更减毒的Schwarz和Moraten病毒(Griffin,D.,and W.Bellini.1996.Measles virus,p.1267-1312.In B.Fields,D.Knipe,et al.(ed.),Virology,vol.2.Lippincott-Raven Publishers,Philadelphia),最近已经表明它们的序列是相同的(Parks,C.L.,R.A.Lerch,P.Walpita,H.P.Wang,M.S.Sidhu,and S.A.Udem.2001.Analysis of the noncodingregions of measles virus strains in the Edmonston vaccine lineage.J Virol.75:921-933;Parks,C.L.,R.A.Lerch,P.Walpita,H.P.Wang,M.S.Sidhu,and S.A.Udem.2001.Comparison of predicted amino acidsequences of measles virus strains in the Edmonston vaccine lineage.JVirol.75:910-920)。由于Edmonston B疫苗是反应原性的,因此其于1975年被废弃并以Schwarz/Moraten疫苗代替,后一疫苗是目前在世界最广泛应用的麻疹疫苗(Hilleman,M.2002.Current overview of thepathogenesis and prophylaxis of measles with focus on practicalimplications.Vaccine.20:651-665)。也使用一些其它疫苗株:日本使用AIK-C、Schwarz F88、CAM70、TD97,俄罗斯使用Leningrad-16及Edmonston Zagreb。CAM70和TD97中国疫苗株不是衍生自Edmonston。Schwarz/Moraten和AIK-C疫苗在CEF上生产。Zagreg疫苗在人二倍体细胞(WI-38)上生产。
在值得注目的开创性研究中,Martin Billeter及其同事克隆了相应于Edmonston MV的反基因组的一个感染性cDNA,并确定了一种独创和有效的反遗传学程序(reverse genetics procedure)以拯救(rescue)相应的病毒(Radecke,F.,P.Spielhofer,H.Schneider,K.Kaelin,M.Huber,K.,G.Christiansen,and M.Billeter.,1995.Rescue of measles viruses from cloned DNA.EMBO Journal.14:5773-5784和WO 97/06270,在此并入参考)。
然而,序列对比(见下文)表明在这个载体中克隆的基因组与Edmonston B序列不同,而是与Edmonston-wt更接近,Edmonston-wt是Edmonston分离株在Vero细胞上的一个早期传代(Parks,C.L.,R.A.Lerch,P.Walpita,H.P.Wang,M.S.Sidhu,and S.A.Udem.2001.Analysis of the noncoding regions of measles virus strains in theEdmonston vaccine lineage.J Virol.75:921-933;Parks,C.L.,R.A.Lerch,P.Walpita,H.P.Wang,M.S.Sidhu,and S.A.Udem.2001.Comparison of predicted amino acid sequences of measles virus strains inthe Edmonston vaccine lineage.J Virol.75:910-920),并具有与任何Edmonston亚群均不相关的10个氨基酸取代。另外,尽管这个载体在表达CD46并缺失IFN I型受体的小鼠中是免疫原性的(19),但本发明人在如下的实验研究中示出当其以104TCID50标准剂量接种时在非人灵长类动物中不是免疫原性的。因此,尽管其的确有助于理解MV复制的一些方面,但是从25年前被废弃的疫苗株中产生的而且其序列如此不同的这个载体看起来不适合作为接种载体,尤其在人体内。
由于这些原因,本发明人决定针对于儿童的麻疹载体需要从经批准的疫苗株(approved vaccine strain)中产生,并因此已经从广泛使用的麻疹病毒的Schwarz/Moraten株的病毒颗粒中开始克隆了一种感染性cDNA。这种cDNA使得可以产生Schwarz/Moraten疫苗原液(vaccine stock)而无需依赖于种子原液的可得性。基于一种为安全性原因在CEF上生长的批准有效的疫苗株,其还可以用作重组接种载体并在世界范围应用。这种载体在需要其存在的某些环境中对成年人也可以应用。
发明描述
本发明涉及一种cDNA分子,其编码源自一种经批准的疫苗株的麻疹病毒(MV)的全长反基因组(+)RNA链的核苷酸序列。
上文中术语“编码”是指cDNA使全长反基因组(+)RNA转录的能力,所述cDNA尤其作为转录的模板。因此,当该cDNA是双链分子时,其中一条链与麻疹病毒的反基因组(+)链RNA具有相同的核苷酸序列,但该cDNA中U核苷酸被T取代。
图5示出了本发明的DNA分子的序列,其包含上述的一个cDNA序列,所示出的该cDNA的链与MV株的反基因组(+)RNA链的序列相同,除了U由T取代之外。
上述cDNA分子当置于合适的条件下时,可以产生能产生麻疹病毒感染性颗粒的感染性反基因组(+)RNA。
获得的cDNA特别具有所述病毒RNA的天然反基因组(+)链的原始5’和3’末端。另外,获得的cDNA符合六倍规则,这一规则是表达感染性病毒颗粒所需的。
六倍规则(rule of six)是指cDNA中存在的核苷酸总数等于6的倍数,这个规则使得可以充分复制麻疹病毒的基因组RNA。这个规则在以上引用的Fields Virology第1197页加以描述。
衍生自一个MV经批准的疫苗株的本发明的cDNA分子可以得自Schwarz或Moraten株。
这些毒株已经在一些出版物中加以描述并用于制备目前使用的疫苗。本发明人特别提议应用可得自Aventis Pasteur(France)的Schwartz株。
根据本发明的另一特定的实施方案,所述cDNA分子置于一个异源表达控制序列的控制下。
当在不能使具有其天然控制序列的cDNA全部转录的细胞类型中寻求这个cDNA的表达时,插入这样一个控制序列对cDNA表达是有利的。
根据本发明的一个特定的实施方案,所述异源表达控制序列包含T7启动子和T7终止子序列。这些序列分别位于MV的全长反基因组(+)RNA链的编码序列的5’和3’末端,并从这个编码序列的临近序列开始。
在本发明的一个特定实施方案中,上述cDNA分子被修饰,即包含额外的核苷酸序列或基序或者在所述cDNA内包含缺失或取代。
在优选的实施方案中,本发明的cDNA分子在其5’末端进一步包含与编码MV的批准疫苗株的全长反基因组(+)RNA链的核苷酸序列的第一个核苷酸相邻的一个后接一个锤头状核酶序列的GGG基序,并且其在3’末端包含与编码全长反基因组(+)RNA链的所述核苷酸序列的最后一个核苷酸相邻的一个核酶序列。丁型肝炎病毒核酶(δ)适于进行本发明。
在与上述编码序列的第一个核苷酸相邻的5’末端放置的GGG基序改良了所述cDNA编码序列的转录效力。正确装配麻疹病毒颗粒需要编码反基因组(+)RNA的cDNA符合六倍规则,当在所述cDNA的编码序列的5’末端加入GGG基序时,自GGG基序的3’开始,还加入一个核酶,以便能在MV的全长反基因组(+)RNA链的第一个编码核苷酸切割转录物。
因此,在加入GGG基序以改良转录效力的情况中,加入两个核酶以保证MV的全长反基因组(+)RNA链的编码序列的切割。
根据本发明,术语“cDNA”涵盖了通过RNA分子的逆转录而获得的DNA分子,RNA分子包括但非限于mRNA分子。
从本发明揭示的材料开始或者使用与所揭示的本发明的cDNA相关的特征可以使用制备DNA的任何其它技术,包括合成或PCR技术。
因此,用于描述本发明的分子的核苷酸序列的术语“cDNA”仅涉及这样的事实,即最初所述分子通过逆转录麻疹病毒的病毒颗粒基因组的全长基因组(-)RNA链而获得。这不应被认为是对于其制备方法的限制。因此纯化的核酸(包括DNA)涵盖在本发明的cDNA的范围内,条件是所述核酸尤其DNA达到上述定义的要求。
本发明还涉及根据上述定义之一的一种cDNA分子,其包含在能复制的一种质粒内。
可以制备许多质粒以达到本发明的要求,本发明尤其涉及图2所示的质粒pTM-MVSchw。
质粒pTM-MVSchw于2002年6月12日保藏于CNCM(法国巴黎),保藏号I-2889。这个质粒在以下实施例和图中加以描述。其是衍生自Bluescript的一种质粒载体,包含置于T7RNA聚合酶的启动子的控制下的编码麻疹病毒株Schwarz的全长序列,其大小为18967个核苷酸。
本发明特别还涉及一种cDNA分子,其优选使用先前报道的包含293-3-46辅助细胞的拯救系统(Radecke et al.and WO 97/06270),在使病毒颗粒装配的条件下能从所述cDNA生产MV经批准的疫苗株的感染性病毒颗粒,293-3-46辅助细胞表达MV的RNA基因组序列转录、复制所必需的蛋白质。
293-3-46细胞在制备病毒颗粒的实施例中用作例子。然而,它们可以由适于组成辅助细胞的任何其它合适的细胞系代替。
生产这种感染性颗粒的方法在本申请的实施例中给出。

本发明当然涉及上述每一个特定序列。
优选实现本发明的一个特定的cDNA分子是包含质粒pTMMVschw所含插入子(insert)的分子,所述质粒保藏在CNCM,保藏号I-2889,其中所述插入子编码麻疹病毒的全长反基因组(+)RNA链的一个核苷酸序列。一个特定的插入子是包含在由如下限制位点限定的序列内的插入子:NotI(在图5上位于1位)和NotI(在图5上位于16203位)。
在本发明的一特定的实施方案中,所述cDNA分子是从麻疹病毒的病毒颗粒中纯化的病毒RNA的逆转录产物。
从病毒纯化的RNA中制备cDNA有利地限制了细胞成分尤其是细胞DNA或RNA的存在,所述细胞成分可以存在于细胞中用于病毒的培养。其尤其限制了病毒基因组RNA的存在,所述RNA是不完整的或突变的并存在于细胞中,而且限制了大量存在于细胞中的病毒mRNA的存在。
本发明进一步涉及具有上述特征的一种cDNA分子,当将其在体内给予时能诱导针对麻疹病毒的至少一种抗原的免疫应答。
本发明还涉及一种重组单负链病毒目病毒,该重组病毒包含根据上述任一限定的麻疹病毒的cDNA分子及一种RNA病毒的DNA序列,所述重组病毒能激发针对麻疹病毒或者所述RNA病毒或者麻疹病毒和RNA病毒的体内体液和/或细胞应答。
本发明还涉及上述的一种重组cDNA分子,该重组分子进一步包含在使其表达为异源氨基酸序列的条件下克隆的一个异源DNA序列,所述克隆以能使获得的重组cDNA符合六倍规则的方式进行。
异源编码序列尤其是DNA序列是在能表达抗原或抗原表位的,具有免疫原性性质及尤其在接受其的宿主体内具有激发或帮助免疫原性应答的能力的序列中优选的。这种异源DNA序列可以例如衍生自病原体抗原。
本发明有利地使得可以在称为附加转录单位(additionaltranscription unit,ATU)尤其Billeter等于WO 97/06270中揭示的ATU的序列中插入这种异源DNA序列。
这种ATU在图4中特别表示。
当用于进行本发明时,该ATU优选位于编码MV的反基因组的全长(+)RNA链的cDNA分子的N末端序列中,而且尤其位于这个病毒的P和M基因之间或者位于H和L基因之间。已经观测到MV的病毒RNA的转录是从5’至3’末端梯度进行。这解释了当插入cDNA的编码序列的5’末端时,所述ATU将可以更有效地表达其含有的异源DNA序列。
本发明还涉及一种包含上述包括重组cDNA的cDNA分子载体。一特殊的载体是克隆和/或表达这个cDNA的载体。
根据本发明的一个优选的实施方案,该载体是一个质粒尤其是于2002年6月12日保藏于CNCM、保藏号为No.I-2889的pTM-MVSchw。
本发明还包括其它载体,如于2002年6月12日保藏于CNCM,保藏号为I-2890的pTM-MVSchw2-gfp,或者于2003年5月26日保藏于CNCM,保藏号为I-3034的pTM-MVSchw2-GFPbis。
这些载体衍生自pTM-MVSchw,并因此是衍生自Bluescript的质粒载体,其包含置于T7RNA聚合酶的启动子的控制下的编码麻疹病毒株Schwarz的全长序列,并进一步包含编码GFP蛋白的gfp基因,所述基因插入ATU中第2位(即在MV的N和P基因之间)。
pTM-MvSchw的大小是18967个核苷酸。pTM-MVSchw2-gfp的大小是19800个核苷酸。
pTM-MVSchw2-gfp和pTM-MVSchw2-GFPbis之间的差异相应于ATU序列中的一个突变,其中如图4B所示在ATU的末端的C核苷酸被取代,以提供pTM-MVSchw2-GFPbis。
本发明还涉及一种制备感染性麻疹病毒颗粒的方法,包括:
1)在一种辅助细胞系中在使得病毒颗粒装配的条件下从所述cDNA表达根据上述限定之一的本发明的cDNA或者含有这种cDNA的载体,所述辅助细胞系也表达MV的反基因组(+)RNA序列的转录、复制和包壳所必需的蛋白质;
2)回收所表达的病毒颗粒。
根据这种方法的一特定的实施方案,其包括:
1)用上述cDNA或载体转染辅助细胞,其中所述辅助细胞能表达辅助功能,以表达一种RNA聚合酶及表达MV病毒的N、P和L蛋白;
2)将步骤1)的所述转染的辅助细胞与适于作为所述cDNA来源的MV疫苗株传代的传代细胞共培养;
3)回收所产生的感染性MV病毒颗粒。
根据一优选的实施方案,辅助细胞衍生自人胚胎肾细胞系293,该细胞系保藏在ATCC,保藏号为No.CRL-1573。
根据这种方法的另一方面,适于传代的细胞是CEF细胞。
CEF细胞可以从得自法国EARL Morizeau,8rue Moulin,28190Dangers的受精鸡卵或者任何其它生产者的受精鸡卵中制备。
根据本发明揭示的方法有利地用于生产适用作疫苗组合物的感染性麻疹病毒。
本发明因此涉及一种免疫原性组合物,其有效成分(activeprinciple)包含通过上述方法获得的感染性麻疹病毒颗粒。
本发明还涉及一种疫苗组合物。有利的是,这种疫苗组合物具有一种有效成分,该成分包含从上述载体的cDNA中拯救的麻疹病毒颗粒,其在基于辅助细胞的挽救系统中表达。
这种疫苗组合物适于抗麻疹病毒起保护作用。根据其中所述cDNA与编码免疫原性氨基酸序列的一种异源DNA序列重组的实施方案,所述疫苗组合物可进一步适于抗免疫原性DNA序列衍生自其中的病原体而起保护作用。
本发明还涉及一种细胞,其与本发明的一种cDNA分子或者上述一种载体重组。一种优选的细胞是原核细胞如大肠杆菌(E.coli)或沙门氏菌(Salmonella)。
另外优选的细胞是真核细胞,尤其是在酵母中选择的细胞,如酿酒酵母(Saccharomyces cerevisiae)。
本发明限定范围内的细胞可以根据一特定的实施方案通过这样的事实鉴定,即其包含表达辅助功能的核苷酸序列,所述辅助功能是表达一种RNA聚合酶及表达MV病毒的N、P和L蛋白所必需的。这种细胞因此可用于病毒颗粒的拯救。

EdB-tag和麻疹疫苗株之间的序列对比

病毒颗粒从Aventis Pasteur(法国里昂)惠赠的一批麻疹Schwarz疫苗中纯化。这种大量的疫苗制品(50ml,3104TCID50/ml)是通过刮擦感染的CEF细胞,冻溶细胞和培养基并过滤细胞碎片而获得的。将颗粒通过30%蔗糖垫层离心而浓缩。使用一种基于硅胶的膜(QlAmp,Qiagen)从裂解的颗粒中纯化病毒RNA。将病毒RNA逆转录为cDNA中,使用随机六聚体(pdN6,1μM)和代表MV基因组前32个核苷酸的一种特异性寡核苷酸(MVSchwRT15′-ACCAAACAAAGTTGGGTAAGGATAGTTCAATC-3′,10μM)的混合物作为引物。为保证逆转录的保真度和全长产物的产量,使用SuperScript II DNA聚合酶(GibcoBRL)。使用PfuTurbo DNA聚合酶(Stratagene)和一组与独特的限制位点邻近的特异性引物通过PCR产生一组6个覆盖全长病毒cDNA的重叠片段(图2中编号为1-6)。在病毒反基因组的5’末端的片段1通过PCR使用特异性引物工程化以含有一个具有为达全效所需的GGG基序的T7RNA聚合酶启动子,以及在T7启动子和第一个病毒核苷酸之间插入的一个锤头状核酶序列。为产生这个片段,将两个重叠寡核苷酸一起退火:前导序列1(5′-TATGCGGCCGCTAATA CGACT CACTATAGGGCCAACTTTGTTTGGTCTGA-3′)含有一个NotI位点,T7启动子(下划线处)及锤头状核酶序列的前19个核苷酸,前导序列2(5′-GGTGACCCGGGACTCCGGGTTTCGTCCTCACGGACTCATCAGACCAAACA-3′)含有一具有SmaI/XmaI位点的锤头序列。在PCR扩增后,将所得片段通过PCR延伸与第二个也通过PCR从Schwarz cDNA中产生的片段连接,使用与锤头状序列重叠(下划线)并覆盖MV Schwarz基因组1-15的寡核苷酸MVSchw1(5′-GAGTCCCGGGTCACCAA ACAAAGTTGGGTAAG-3′),和MVSchw160(5′-GGTTTGTCCTTGTTTCTTTT-3′,MV Schwarz基因组141-160)。片段2(长度为2173个核苷酸,见图2)使用寡核苷酸MVSchwRT1(5′-ACCAAACAAAGTTGGGTAAGGATAGTTCAATC-3′,MV Schwarz基因组1-32)和MVSchw2173(5′-ATTCCCTTAACCGCTTCACC-3′,MV Schwarz基因组2155-2174)扩增。片段3(长度为3142个核苷酸)使用寡核苷酸MVSchw1901(5′-CTATGGCAGCATGGTCAGAAATA-3′,MV Schwarz基因组1901-1924)和MVSch5043(5′-ATTGTCGATGGTTGGGTGCT-3′,MV Schwarz基因组5024-5043)扩增。片段4(长度为4349个核苷酸)使用寡核苷酸MVSchw4869(5′-AAACTTAGGGCCAAGGAACATAC-3′,MV Schwarz基因组4869-4891)和MVSchw9218(5′-GGACCCTACGTTTTTCTTAATTCTG-3′,MV Schwarz基因组9194-9218)扩增。片段5(长度为6200个核苷酸)使用寡核苷酸MVSchw9119(5′-AGATAGGGCTGCTAGTGAACCAAT-3′,MV Schwarz基因组9119-9142)和MVSchw15319(5′-ATCAGCACCTGCTCTATAGGTGTAA-3′,MVSchwarz基因组15295-15319)扩增。为获得片段6,将通过PCR产生的两个重叠片段一起退火。使用如下寡核苷酸:MVSchw15155(5′-GCAGCAGATAATTGAATCATCTGTGAGGACTTCAC,MVSchwarz基因组15155-15190)和MVSchw15570(5′-CCCGGAGTAAAGAAGAATGTGCCCCCAGAATTTGC-3′,MVSchwarz基因组15535-15570)。这个片段与预先通过PCR扩增p(MV+)质粒(得自M.Billeter)而获得的含有与T7终止子连接的丁型肝炎病毒(HDV)核酶的一个片段退火,使用寡核苷酸MVSchw15547(5′-GGCACATTCTTCTTTACTCCGGGAACAAAAAGTTG-3′,MVSchwarz基因组15547-15581)和MVSchwEnd(5′-ATAGGGCCCGCGGCCGCATCCGGATATAGTTCCTCCTTTCA-3′,含有与T7终止子的最后的核苷酸连接的一个ApaI限制位点)。将由此产生的6个片段克隆入pCR2.1-TOPO载体(Invitrogen,Groningen,Netherlands)中并测序。为装配MV Schwarz全长DNA,构建一种修饰的BlueScript KS(+)质粒:将产生NotI,KasI/NarI,SpeI,ApaI多接头的两个内部互补的寡核苷酸进行退火并插入用NotI/ApaI消化的pTM质粒中(pTM通过缺失T7启动子衍生自pBluescript KS(+)(Stratagene)(Tangy,F.,A.McAllister,and M.Brahic.1989.Molecular cloning of the complete genome of Theiler′s virus,strainGDVII,and production of infectious transcripts.J.Virol.63:1101-11066))。将6个MV Schwarz cDNA片段使用独特的限制位点逐步装配在一起。片段1和片段2使用MV序列中的BlpI位点(图2)和pCR2.1-TOPO载体主链中的.BglII位点装配在一起。使用MV序列中的SbfI位点和pCR2.1-TOPO载体主链中的BglII位点将所得质粒与片段3装配在一起,产生含有MV Schwarz片段1-3的质粒pCR2.1-TOPO-MVSchw-1-2-3。在用NotI/Narl消化这个质粒之后,将含有T7启动子、锤头状核酶和MV Schwarz反基因组的前4922个核苷酸的片段插入NotI/Narl消化的pTM载体中,产生pTM-MVL。同时,将片段5和6使用MV序列中的BsmBI位点(图2)和pCR2.1-TOPO载体主链中的BssHII位点装配在一起,产生含有MV Schwarz片段5-6的质粒pCR2.1-TOPO-MVSchw-5-6。在用SpeI/ApaI消化这个质粒之后,将含有MV Schwarz反基因组的最后6720个核苷酸、HDV核酶和T7终止子序列的片段插入SpeI/ApaI消化的pTM载体中,产生pTM-MVT。为进行最后的装配,制备4个片段并连接在一起:1)pTM-MVL的一个SapI/SapI片段(长度为4367个核苷酸),其含有一部分pTM主链,T7启动子,锤头状核酶及MV反基因组的前1813个核苷酸;2)pTM-MVL的一个SapI/NarI片段(长度为3110个核苷酸),其含有MV Schwarz反基因组的第1813-4923位核苷酸;3)pCR2.1-TOPO-MVSchw-3的一个NarI/SpeI片段(长度为4253个核苷酸),其含有MV Schwarz反基因组的第4923-12157位核苷酸;及4)pTM-MVT的一个SpeI/SapI片段(长度为7235个核苷酸),其含有MV Schwarz反基因组的第12157-15894位核苷酸、HDV核酶、T7终止子和一部分pTM载体主链。在连接和克隆后,获得几个完整构建体。对所得的称为pTM-MVSchw的质粒全部测序(Acc.Num.CNCM I-2889)。在这个cDNA与先前报道的Schwarz基因组序列(Parks,C.L.,R.A.Lerch,P.Walpita,H.P.Wang,M.S.Sidhu,and S.A.Udem.2001.Analysis of the noncoding regions of measlesvirus strains in the Edmonston vaccine lineage.J Virol.75:921-933;Parks,C.L.,R.A.Lerch,P.Walpita,H.P.Wang,M.S.Sidhu,and S.A.Udem.2001.Comparison of predicted amino acid sequences of measlesvirus strains in the Edmonston vaccine lineage.J Virol.75:910-920)之间未发现突变。
从pTM-MVSchw质粒中回收感染性Schwarz病毒
为从pTM-MVSchw cDNA中回收Schwarz病毒,我们使用Radecke等描述的(Radecke,F.,P.Spielhofer,H.Schneider,K.Kaelin,M.Huber,K.Dotsch,G.Christiansen,and M.Billeter.1995.Rescue ofmeasles viruses from cloned DNA.EMBO Journal.14:5773-5784)并经Parks等修改的(Parks,C.L.,R.A.Lerch,P.Walpita,M.S.Sidhu,and S.A.Udem.1999.Enhanced measles virus cDNA rescue and geneexpression after heat shock.J Virol.73:3560-3566)基于辅助细胞的拯救系统。使用磷酸钙方法对稳定表达T7RNA聚合酶及麻疹病毒N和P蛋白的入辅助细胞(293-3-46细胞,由Radecke等揭示(17))使用pTM-MVSchw质粒(5μg)和表达MV聚合酶L基因的质粒(pEMC-La,20ng,由Radecke等揭示(17))进行转染。在37℃温育过夜后,用新鲜培养基更换转染培养基并进行热激(43℃2小时)(12)。在37℃温育2天后,考虑到安全性,将转染的细胞移至一层CEF细胞上并在32℃温育以避免最初在CEF细胞上选择的并且目前生长在这些细胞上的Schwarz疫苗的任何适应。如下制备上述鸡胚成纤维细胞(CEF)。将受精的鸡卵(EARL Morizeau,8rue Moulin,28190Dangers,法国)在38℃温育9天。无菌收集鸡胚。除去头、肢翼和内脏并将鸡胚切片及在37℃用胰蛋白酶消化5-10分钟(Trypsine/EDTA 2.5g/L)。在过滤(70μm)及在DMEM高葡萄糖/10%FCS中洗涤几次后,接种细胞(5-7×106个细胞/平皿)并在37℃温育过夜。共培养后在第3-7天之间易于回收感染性病毒。在CEF中偶尔出现合胞体但不是系统性的。在37℃共培养转染的293-3-46辅助细胞和灵长类Vero细胞(非洲绿猴肾)之后也通过相同技术拯救Schwarz病毒。在这种情况中,在共培养2天后在所有的转染物中均系统地出现合胞体。为测试病毒对Vero细胞的适应,将在Vero细胞上拯救的克隆的Schwarz病毒制品在Vero细胞上传代两次。纯化病毒颗粒并如上述使用用于克隆的引物逆转录病毒RNA(见上述)。对病毒基因组全部测序。发现在拯救的/传代的病毒与用于转染的cDNA之间在15894个核苷酸中有两个核苷酸改变。这些突变分别在10个不同克隆中的7和8个克隆的相同区域中发现,表明在病毒群体之中突变的高百分率。另外,这两个突变均导致融合蛋白(F)中氨基酸改变:在266位G变为R,在365位Y变为S。
相反,在32℃在CEF细胞上回收和传代的病毒的基因组序列与原始的Schwarz病毒的序列相同。这个观测结果表明改变Schwarz病毒的宿主细胞导致快速适应,这可影响疫苗的性质。
拯救的病毒的生长能力
分析从cDNA中拯救的Schwarz病毒在CEF和Vero细胞上生长的能力,并与从中衍生的工业化批量Schwarz疫苗(得自AventisPasteur)及用于从其cDNA中拯救的EdB-tag病毒相对比。将于6孔板中的单层Vero细胞以不同的感染复数用病毒感染。在感染后(pi)不同时间点,将细胞刮入培养基中。在冻溶后,通过测定Vero细胞中的TCID50而确定感染效价。生长曲线:将6孔板中的单层Vero细胞以不同的感染复数(MOI)用病毒感染。在感染后(pi)不同时间点,将细胞刮入培养基中。在冻溶后,通过测定Vero细胞中的TCID50而确定感染效价。
TCID50效价:将Vero细胞接种于96孔板中(7500个细胞/孔)并用于DMEM/5%FCS中连续1∶10稀释的病毒样品感染。在37℃温育4-5天(Ed-B病毒)和7天(Schwarz病毒)后,将细胞用结晶紫染色,并确定产生50%测试单位感染的病毒稀释液。以组织培养感染剂量(TCID50)描述的50%终点是通过Karber方法计算的(Karber,G.1931.Beitrag zur kollektiven Behandlung pharmakologischerReihenversuche.Arch Exp Path Pharmak.162:480-483)。在Vero细胞上测试时,从其各自的cDNA中拯救的Schwarz和EdB-tag病毒的生长动力学是相似的(图3)。在Vero细胞上Schwarz病毒达到高产量(使用感染复数0.01在感染2天后达到107-108TCID50/ml)。在CEF细胞上测试时,Schwarz病毒在32℃与在37℃一样能良好生长,而EdB-tag不能(图3)。这个观测结果证实用于克隆EdB-tag的病毒不适应CEF细胞。Schwarz病毒在CEF上的产量低于在Vero细胞上的产量(使用感染复数0.05感染4天后达到106TCID50/ml)。当使用用于克隆的原始的Schwarz病毒感染CEF细胞时观测到相似的生长曲线和相似的效价(图3)。这些观测结果表明从其cDNA中拯救的Schwarz病毒与用于克隆的原始的一批疫苗具有相同的生长特性。
在Schwarz cDNA中导入一个附加转录单位
在报道EdB-tag病毒的克隆的先前研究中(17),作者揭示了一种独创的方法以使病毒cDNA适应作为适于外源转基因表达的载体。他们将一个附加转录单位(ATU)插入在病毒基因组的不同位置中。这个ATU是MVN-P基因间区域的一个拷贝,含有插入在多克隆位点盒中一个转基因的MV依赖性表达所必需的顺式作用序列。作者及我们自己进行了大量测试,发现根据在基因组中的插入位置,插入在这个ATU中的外源转基因的表达是非常有效的。沿着基因组转录梯度表达不同的MV基因,引起N基因的高表达及L基因的低表达(Lamb,R.,and D.Kolakofsky.1996.Paramyxoviridae:the viruses ans theirreplication,p.1177-1199.In B.Fileds,D.Knipe,et al.(ed.),FieldsVirology.Lippincott-Raven Publishers,Philadelphia)。
ATU的插入利用这个梯度,使得转基因根据插入位置而高或低表达。另外,在文中外源转基因使用与真MV基因相同的控制和途径被表达。
为将Schwarz cDNA转化作为载体,我们构建了一个相似的ATU,其插入在所述cDNA的两个不同位置中(图4)。对该cDNA测序并发现无突变。
从cDNA中回收的Schwarz MV在猕猴中的免疫原性
第一次实验:与Schwarz疫苗相对比
在猕猴(cynomolgus macaques)中对比从pTM-MVSchw质粒中拯救的并在CEF细胞上传代两次的病毒的免疫原性与Schwarz疫苗的免疫原性。传代条件如下:
在拯救后,从与293-3-46辅助细胞共培养的CEF细胞中挑取分离的合胞体并将一个单个合胞体在600μl的OptiMEM 1(Gibco)中稀释并涡旋。将这个接种体用于感染于35mm的孔中或T-25培养瓶中的新鲜CEF细胞(80-90%铺满)。在37℃吸附2小时后,将该接种体用DMEM/5%FCS代替并将细胞在32℃温育1-2天。当出现小的合胞体时,将感染的细胞在T-75培养瓶中扩大培养:将细胞用PBS洗涤并用PBS/1mM EDTA/0.25%胰蛋白酶分离1分钟,然后与新鲜CEF细胞一起移至T-75培养瓶中(T-75培养瓶1/4铺满)。在32℃在DMEM/5%FCS中温育4-7天后,收获病毒(1代):除去培养基并将感染的细胞刮入3ml的OptiMEM 1中。在进行一次冻溶循环后,通过离心弃去细胞碎片(1500rpm,5分钟,室温)。将这个原种保持在-80℃并用于以相同方式感染新鲜的CEF以制备2代原种。
使用得自Aventis Pasteur的未传代的批量制品及在CEF细胞上传代两次的相同制品测试不同配方的疫苗。如下制备病毒:将CEF细胞(得自温育9天的鸡胚)在0.05MOI感染并在32℃温育7天。通过刮擦感染的细胞、冻溶并低速澄清细胞碎片而纯化病毒。用于制备MV疫苗的稳定剂得自Aventis Pasteur。将相同剂量的有或无稳定剂的不同批次疫苗制品与冻干的终产物(Rouvax,Aventis Pasteur)进行对比。所有的疫苗制品均使用TCID50方法在Vero细胞上滴定。为猴进行皮下注射并在不同时间点取血样。为对比体液应答和细胞应答,通过ELISA(Trinity Biotech,USA)探寻血清中抗MV抗体的存在及通过ELISPOT在PBMC中探寻抗MV T细胞的存在。
第二次实验:与EdB-tag株对比
将针对猿D型逆转录病毒、猿T细胞嗜淋巴细胞病毒、猿免疫缺陷病毒和麻疹病毒为血清反应阴性的群体饲养的恒河猴(Macacamulatta)或猕猴(Macaca fascicularis)根据American Association forAccreditation of Laboratory Animal Care指导进行圈养。为猴经皮下接种于OptiMEM(GibcoBRL)中稀释的不同剂量的(103-105TCID50)EdB-tag或Schwarz MV或者在厂商提供的溶液中稀释的104TCID50的冻干Rouvax MV疫苗(Aventis Pasteur,Marcy l′Etoile,法国)。在接种后的不同时间点收集血样。
在免疫接种后1个月通过ELISA(Trinity Biotech,USA)探寻血清中抗MV抗体的存在。每次对1/20稀释的血清样品的确定均一式三份。来自阴性反应猴的5个样品的混合物用作阴性对照。为确定每个样品的免疫状态比率(ISR),将阴性对照的吸光度从阳性样品的吸光度中减去,并将结果除以ELISA试剂盒中提供的厂商推荐的校准吸光度。只有ISR值高于0.9才认为在这个测试中是阳性的。
细胞免疫应答通过γ-IFN ELISpot分析确定。将冷冻的PBMC解冻并在RPMI,10%FCS和4U/ml rh-IL2(Boehringer Mannheim)中温育过夜。将Multiscreen-HA 96孔板在4℃用PBS中的4μg/ml捕获抗γ-IFN(GZ-4,MAbTech)包被过夜,洗涤,然后用100μlRPMI,10%FCS在37℃温育1小时。将培养基用悬浮于100μl的RPMI-10%FCS中的5×105PBMC和100μl的刺激剂代替。所述刺激剂由107pfu重组的修饰的疫苗Ankara(32)组成,MVA-HMV或MVA-wt作为对照。将细胞在37℃刺激24小时。植物凝集素(phytohemaglutinin)A(2.5μg/ml,Sigma)用作阳性对照,RPMI用作阴性对照。将平板用PBS洗涤两次,用PBS、0.05%Tween 20(Sigma)洗涤4次及用PBS再洗涤两次。加入一种生物素酰化的抗γ-IFN抗体(7-B6-1,MabTech,100μl,1μg/ml于PBS中)并将平板再37℃温育2-4小时。加入链霉抗生物素-碱性磷酸酶(AP)缀合物(Roche,100μl,于PBS中1/2000稀释)并用1M Tris pH 9.5,1.5M NaCl,0.05M MgCl2中的BCIP/NBT(Promega)产生斑点。在室温干燥过夜后,使用自动成像分析系统(ELISpot Reader,Bio-Sys)计数斑点。减去MVA-wt刺激后获得的低背景并将结果以MVA-HMV特异性γ-IFN生产细胞/百万个PBMC表示。
小鼠免疫及体液免疫应答的鉴定。将CD46转基因杂合的FVB小鼠(33)与缺少I型干扰素受体的129sv IFN-α/βR-/-小鼠(30)杂交。将F1子代通过PCR筛选并将CD46+/-动物再次与129sv IFN-α/βR-/-小鼠杂交。选择IFN-α/βR-/-CD46+/-动物并用于免疫实验。这些小鼠对MV感染易感(27,29)。将6周龄的雌性CD46+/-或CD46+/-IFN-α/βR-/-(IFNAR)小鼠腹膜内接种104TCID50的不同疫苗制品(4只小鼠/组)。在接种后一个月收集的血清中通过ELISA(Trinity Biotech,USA)探寻抗MV抗体的存在。在这种情况中,一种抗小鼠IgG Mab(Amersham)用作二级抗体。每次确定均一式三份。用阴性小鼠血清混合物确定的吸光度要从在阳性小鼠中测定的吸光度中减去。因为在这种情况中使用ISR对比样品是不可能的,因此测试小鼠血清的连续稀释液以确定终点限度(endpoint limit)阳性稀释液。
结果:
在用EdB-tag和Schwarz MV疫苗接种后的猕猴和小鼠体液免疫应答的对比。EdB-tag MV是衍生自Edmonston B株的一种分子克隆的MV(16)。我们将其与Schwarz商购的MV疫苗免疫的猕猴中的免疫原性加以对比。EdB-tag病毒是在感染复数(MOI)为0.05感染的Vero细胞中制备的。当合胞体占据培养物的80-90%时,刮下细胞,将细胞和培养基冻溶并通过低速离心去除细胞碎片。得自Aventis Pasteur(Marcy l′Etoile,法国)的Schwarz MV是以相同方式从在32℃生长的感染的鸡胚成纤维细胞(CEF)中制备的,在此温度这个毒株适应CEF。这两种疫苗制品的效价均通过在Vero细胞中进行终点稀释分析(endpoint dilution assay)而确定并以TCID50表示。将不同剂量的(103-105TCID50)EdB-tag和Schwarz MV经皮下注射入猕猴中(每个剂量注射2只猴)。作为对照,将动物也注射104TCID50的冻干的商购Schwarz疫苗(Rouvax,Aventis Pasteur)。在接种后1个月收集的猕猴血清中通过ELISA确定抗MV抗体水平。用103和104TCID50的SchwarzMV接种的猕猴具有与标准剂量Rouvax疫苗诱导的相似的抗体水平(图6)。用104TCID50的EdB-tag病毒接种的猕猴保持阴性(未示出)。注射高10倍的剂量(105TCID50)仅诱导微弱应答,这个应答比用103TCID50的Schwarz MV观测到的应答低(图6)。用商购的疫苗接种诱导最佳应答,因为冻干的辅助作用所致。
在如材料和方法章节中所述获得的遗传修饰的小鼠中也测试了不同的疫苗制品。使用两种类型的小鼠:表达CD46(33),人MV疫苗株受体(34)的小鼠,及表达CD46和缺少I型IFN受体的小鼠(29)。为6周龄的小鼠经腹膜内接种104TCID50的不同疫苗制品(4只小鼠/组)。图7示出在在接种一个月后收集的两种类型的小鼠血清中抗MV抗体的检测。在CD46小鼠中,EdB-tag病毒的免疫原性低于Schwarz疫苗。用前者获得的平均效价为1/80,而后者为1/1280。EdB-tag病毒在缺少I型IFN受体的CD46小鼠中免疫原性也较低,但在CD46免疫感受态小鼠中差别不显著,可能表明在这两种毒株之间对IFNα/β的敏感性不同。
从cDNA中回收的Schwarz MV的免疫原性。将从pTM-MVSchw质粒中拯救的并在CEF或Vero细胞上传代两次的病毒对猕猴(cynomolgus macaques)的免疫原性与工业化的Schwarz疫苗的免疫原性相对比。在这个实验中使用猕猴(Cynomolgus macaques),因为难以获得MV阴性的中国恒河猴。这些猕猴与恒河猴同样对MV敏感,如一些研究所示(28,26)。将猴(每个制品2只动物)皮下注射104TCID50的来自Aventis的Schwarz MV疫苗或者拯救自pTM-MVSchw质粒并在CEF或Vero细胞上生长的Schwarz MV。在不同时间点收集的血清中确定抗MV抗体的存在(图8)。所有接种的猕猴均成为阳性的。在免疫1或2个月后在测试的不同疫苗制品之间观测到无统计学显著差别。这个结果表明自pTM-MVSchw质粒中拯救的病毒在非人灵长类动物中与亲代Schwarz疫苗具有相同的免疫原性。在CEF或Vero细胞上生长的拯救的病毒之间检测到无不同,表明通过在Vero细胞上传代在F蛋白中产生的两个突变不影响病毒的免疫原性。
在接种后第一个月期间观测白细胞(WBC)总数、淋巴细胞和单核细胞数的变化(图9)。正如先前在MV接种后所观测的结果(1),在第一周期间有轻度的白细胞减少。在第二周期间,观测到循环淋巴细胞和单核细胞数明显增加。这与通过γ-IFNELISpot分析检测到的MV特异性T淋巴细胞数达到峰值相一致(图9D)。在自质粒中拯救的Schwarz MV与由Aventis制备的Schwarz疫苗诱导的特异性细胞免疫应答之间检测到无统计学显著差异。

在本项研究中,我们描述了克隆及拯救麻疹病毒的Schwarz/Moraten减毒株、两种广泛应用的麻疹疫苗Attenuavax(Merckand Co.Inc.,West Point,USA)和Rouvax(Aventis Pasteur,Marcy l′Etoile,France)的组成,及组合的麻疹、流行性腮腺炎和风疹疫苗(MMR)的组成(35)。在儿科临床实验使用中,从一个cDNA中产生活的减毒MV必须是与亲代疫苗同样安全而且有效的。假定安全性和效力最终依赖于减毒株的基因组序列,因此我们从工业化疫苗中制备的病毒颗粒中使用克隆保真度最佳的方法克隆了MV Schwarz cDNA。结果,我们获得的克隆的序列与亲代Schwarz MV基因组的序列相同。为在拯救期间获得最大产量,将病毒反基因组cDNA置于具有全效所必需的GGG基序的T7RNA聚合酶启动子的控制下。将一锤头状核酶插入在这个GGG基序与第一个病毒核苷酸之间,以使得准确切割病毒RNA。在拯救期间为避免使Schwarz疫苗适应未被鉴定(non-certified)的细胞,将用工程化的cDNA转染的辅助细胞与CEF共培养,CEF是最初用于选择这个疫苗并现用于制备该疫苗的细胞。将拯救的病毒在CEF上传代两次并对其基因组进行完全测序。当将此序列与原始的病毒序列相对比时未发现突变。另外,拯救的病毒与原始的Schwarz病毒在CEF上的生长动力学和产量是相同的。
Schwarz病毒也是在共培养转染的辅助细胞与非常适合MV的Vero细胞之后拯救的。然而在这种情况中,在Vero细胞上两次传代后在病毒融合蛋白(F)中出现两个突变。这种迅速适应与在Vero细胞上更融合的表型相关。相反,拯救的Schwarz MV在CEF上不融合(在感染的CEF中仅观测到很少的合胞体)。在F蛋白中出现两个突变(在266位G变为P,在365位Y变为S)。这些突变存在于在Vero细胞上生长的EdB-tag病毒中(见图6),它们也存在于与Edmonston株高度相关但不感染CEF的Hallé株中(31)。这两个突变因此看起来与Vero细胞中增强的融合相关。Schwarz病毒在Vero细胞上仅传代两次之后F蛋白就快速适应表明为保持其遗传完整性,该疫苗必须在CEF上生长。
在猕猴中从pTM-Schw质粒中拯救的病毒与亲代Schwarz疫苗具有相同免疫原性。强调的是在这些实验中猕猴是用低剂量的用于人体免疫的病毒接种的。因此,可以指导使用标准疫苗剂量(104TCID50)用这个病毒进行人体临床实验。相反,先前克隆的EdB-tag MV当以与克隆的Schwarz MV相同剂量使用时,其在猕猴中不是免疫原性的而且在CD46转基因小鼠中免疫原性较低。
什么是Schwarz MV株具有较高免疫原性的原因呢?用活的减毒病毒疫苗诱导良好的免疫原性需要在组织中以足以充分引发免疫系统的高水平复制。在Schwarz和EdB-tag MV基因组之间的一些突变位于P/V/C和L基因中,提示复制效力的不同。Schwarz MV在体内淋巴细胞中复制比EdB-tag MV更有效是可能的,即使它们在Vero细胞中以相同速度复制。在体内有效复制需要一些逃避IFN-α/β应答的机制。EdB-tag病毒适应的Vero细胞对IFN-α/β刺激不应答。因此EdB-tag MV是在没有IFN-α/β应答的情况中选择的并可以是对这种宿主防御机制特别敏感的。事实上,已经示出野生型MV在Vero细胞上的传代将病毒的表型从非IFN诱导物改变为IFN诱导物(36)。同样,Ed-tag MV在CD46受体转基因及还剔除IFN-α/β受体的小鼠中是免疫原性的这一事实,提示这个病毒是特别IFN敏感性的。令人感兴趣地,IFN-α/β应答帮助引发抗疫苗的特异性免疫应答。因此一个好的活疫苗必须同时诱导IFN-α/β应答及在一定程度上逃避这一应答。为此,在具有强IFN-α/β应答的原代细胞如CEF上选择减毒的病毒疫苗可能是一个上策。
提供IFN抗性的MV产物还未鉴别。然而,密切相关的Sendai病毒的非结构C蛋白已经示出抵消IFN诱导的抗病毒状态(37)。我们在EdB-tag P/V/C基因中发现的与任何Edmonston亚群均不相关的5个突变可能与其在猕猴中低免疫原性相关。另一方面,通过在Vero细胞上传代Schwarz病毒在F蛋白中产生的两个突变不影响其免疫潜力,表明病毒包膜蛋白的融合性质在免疫原性中不起明显作用。
将pTM-MVSchw质粒通过在基因组的不同位置导入两个ATU而工程化以表达外源基因。拯救的Schwarz重组MV表达绿色荧光蛋白,因此示出这种新的麻疹疫苗具有载体功能。总之,这种分子克隆将可以产生不依赖于原种的MV疫苗。就其ATU而言,可以将其用作载体以基于有效的并广泛应用的经批准的疫苗株而生产重组疫苗。

11、权利要求1-6或9任一项的cDNA分子,其通过缺失或取代Schwarz株或Moraten株的(+)RNA链的几个核苷酸而修饰,条件是所得cDNA符合六倍规则。
12、权利要求1-6或9任一项的cDNA分子,当在体内给予时,其能诱导针对麻疹病毒的至少一种抗原的免疫应答。
13、一种重组的cDNA分子,其由以下序列组成:
-权利要求1-12任一项的cDNA分子,以及
-能表达一种异源氨基酸序列并编码病原体免疫原性序列的一种异源DNA序列,
其中该重组cDNA进一步符合六倍规则。
14、权利要求13的重组的cDNA,其进一步包含一个用于克隆所述异源DNA序列的附加转录单位。
15、权利要求14的重组的cDNA序列,其中所述附加转录单位克隆在麻疹病毒的N基因的上游,或者克隆在麻疹病毒的P和M基因之间,或者克隆在H和L基因之间。
16、一种载体,其包含权利要求1-15任一项的cDNA分子。
17、权利要求16的载体,其是质粒pTM-MVSchw,该质粒于2002年6月12日保藏于CNCM,保藏号I-2889。

25、一种制备感染性麻疹病毒颗粒的方法,包括:
1)在一种辅助细胞系中在使得病毒颗粒可以装配的条件下从所述cDNA开始表达权利要求1-15任一项的cDNA或者权利要求16-24任一项的载体,所述辅助细胞系还表达麻疹病毒的反基因组(+)RNA序列的转录、复制和包壳所必需的蛋白质;
2)回收表达的病毒颗粒。
26、权利要求25的方法,包括
1)用权利要求1-15任一项的cDNA或者权利要求16-24任一项的载体转染辅助细胞,其中所述辅助细胞能表达辅助功能以表达一种RNA聚合酶及表达MV病毒的N、P和L蛋白;
2)将步骤1)的所述转染的辅助细胞与适于传代作为所述cDNA来源的MV疫苗株的传代细胞共培养;
3)回收产生的感染性MV病毒颗粒。
27、权利要求25或26的方法,其中转染步骤后用新鲜的培养基更换转染培养基并在温育之前应用热激。
28、权利要求25或26的方法,其中所述辅助细胞衍生自人胚胎肾细胞系293即ATCC CRL-1573。
29、权利要求25或26任一项的方法,其中所述适于传代的细胞是CEF细胞。
30、权利要求25或26任一项的方法,其用于生产适用作疫苗组合物中有效成分的感染性麻疹病毒。
31、一种免疫原性组合物,其有效成分包含通过权利要求25-30任一项的方法获得的感染性麻疹病毒颗粒。
32、一种疫苗组合物,其有效成分包含在基于辅助细胞的拯救系统中从权利要求1-15任一项的cDNA分子或权利要求16-24任一项的载体中拯救的麻疹病毒颗粒。
33、权利要求31的免疫原性组合物或者权利要求32的疫苗组合物,其适用于抗麻疹病毒的保护。
34、权利要求32的疫苗组合物,其适用于抗麻疹病毒以及抗衍生所述免疫原性序列的病原体的保护。
35、一种重组单负链病毒目病毒,其包含权利要求1-15任一项的cDNA分子及一种RNA病毒的DNA序列,其能激发体内抗麻疹病毒或所述RNA病毒或者抗麻疹病毒和RNA病毒这两者的体液和/或细胞应答。
36.制备权利要求1-15任一项的cDNA分子的方法,包括从Schwarz株或Moraten株的病毒粒子纯化病毒RNA的步骤。
