背景技术

EDTA已经被用于各种食品、饮料和其它可食用产品中。US4299853描述了用于酒精饮料的防腐剂。所述饮料在贮存期间具有生物不稳定性所以要使用某种EDTA添加剂,该EDTA添加剂包括EDTA的水溶性碱金属或碱土金属盐,例如EDTA的钠、钾和钙盐。US3956513描述了用于食品或饮料调味料的固体产品,其中含有水不溶性钙、镁、镍、锰或锌异葎草酮络合物和EDTA的碱金属化合物或碱金属盐混合物(例如,EDTA的二钠、二钾、四钠或四钾盐),碱金属化合物与异葎草酮络合物的摩尔比至少是0.8至1。在US4820520中,公开了将NaEDTA用于与防腐剂相结合以增强食品和饮料中的抗真菌活性。US4937085描述了一种食品防腐剂组合物,用于防止马铃薯变色。该组合物包括柠檬酸、半胱氨酸、抗坏血酸和微量EDTA。US4020158、US4830716和US5516925提供了金属(包括铁)氨基酸螯合物作为膳食补充剂提供给人或其它动物服用,一种液体铁补充剂由Albion Laboratories,Inc.(Clearfield,Utah)以FerrochelTM为商标市售得到。US5653987描述了一种适于口服或鼻腔传输的液体药物制剂,其包括一种蛋白质药物制剂、水和至少两种吸收增强化合物,所述化合物包括EDTA二钠。
可以意识到,还有存在着对用于强化含铁饮料的方法的需要,即用以提供增强的铁的生物可利用性和稳定性。本发明实现了这种需要,和其它的需求和目的,以下对本发明实施方案的详细描述可以使它们变得显而易见。
发明概述
本发明提供了用EDTA铁作为铁源的铁强化饮料。本发明的用EDTA铁强化的饮料具有出色的铁生物可利用性。而且,本发明的EDTA铁强化饮料具有出色的储藏稳定性。即使在铁含量相对较高的情况下,它们的口味仍然很好,基本上没有不可接受的怪味道或异味。
在本发明中,十分惊奇地发现EDTA铁中的铁并没有与其它通常存在于添加了维生素/矿物质混合料(例如,钠、钙、钾、锌、碘、维生素C和维生素E等)的饮料中的其它阳离子进行大量的互相置换。结果,在该溶液中并没有象其它可溶铁形式那样,明显地产生自由铁用来反应,并产生异味或颜色。据认为EDTA铁充分地将铁螯合,使其不能发生反应,即使以相对稀释的水溶液形式用于本发明的非限制性实施方案的饮料中也是这样。本发明饮料的这个优点既可以适用于立即可饮(RTD)的含有效量EDTA铁的液体形式,也可以适用于含有效量EDTA铁的源自干粉混料的其复水还原形式。对于本发明的目的,技术术语“EDTA铁”指的是“乙二胺四乙酸铁钠”即“NaFeEDTA”。
在本发明的一个方面,提供的饮料组合物既可以是液体产品,也可以是可以被复水还原的干混产品,通常每液量盎司饮料的含铁量为至少约1%的U.S.DV。优选,每液量盎司的饮料,本发明饮料含铁量约1至约30%,且更优选约1至约5%的U.S.DV。典型地,6至8盎司含有这样的含铁量的本发明饮料的液体可以提供给个人U.S.DV所需铁量的很大部分。例如,在一个优选的实施方案中,在每6盎司的本发明饮料中,饮料组合物一般含约18%至约33%的U.S.DV,这是通过引入EDTA铁而实现的。为了实现这些配比,优选将EDTA铁添加进本发明的产品中,对于每液量盎司的成品饮料,添加量大于约0.2mg Fe或更多,优选约0.2至约3.0mg Fe,且更优选约0.4至约1.2mg Fe。
在本发明中,通过EDTA铁将铁引入饮料或饮料粉中。优选将EDTA铁用作饮料或饮料混合粉料的唯一铁源。该EDTA铁并不是必须与其它铁源结合,来提供足够量的生物可利用铁。事实上,存在其它补充性铁源,例如硫酸亚铁、富马酸亚铁、柠檬酸亚铁等等,能够降低或损害由单独使用EDTA铁强化饮料而带来的有益的稳定性和可口性方面的特性打折,因此优选应避免使用。
本发明还针对一种预防或治疗缺铁性贫血的方法,是通过病人饮用有效量的本发明的强化饮料而实现的。

发明详述
本发明总地涉及用EDTA铁强化铁的饮料。这样的铁强化饮料包括立即可饮(RTD)饮料和干混粉料饮料,所述的干混粉料饮料可以采用合适的液体进行复水还原。用于本发明的铁的形式(即EDTA铁或EDTA铁钠)是水溶性的,且将由此明显地在高水分的和水性食品/饮料产品中具有高反应性,并容易产生有异味和不佳色泽。换句话说,如果饮料中有非常高的水分含量和活性,通常情况下人们将预期在EDTA铁中的铁离子与通常在饮料中常见的其它阳离子(例如,钠或钙)互相置换,从而使自由铁离子反应并形成异味或不佳色泽(与可溶铁离子形式的通常情况一致)。令人惊奇的是,本发明的采用EDTA铁的强化饮料还没有发生这样的事情。
如以下实施例所显示的,不论在复水还原的饮料粉,还是立即可引的饮料中,EDTA铁并没有产生令人不快的金属异味或不佳色泽。而且,与用做比较的用其它铁形式强化的饮料相比,根据本发明的用高于前述饮料中铁含量的EDTA铁强化的饮料其口味是可以接受的。也就是说,通过对根据本发明的用EDTA铁强化的饮料的取样,分析员没有发现与铁相关的通常伴随铁强化剂的使用而产生的感官缺陷,包括金属异味、氧化味和/或不新鲜的味道。
在本发明的实践中,将优选溶质,包括EDTA铁,以饱和量或低于饱和量添加到饮料的液体介质中,这样在贮存容器中就不会形成或堆积明显的沉淀或固体残渣。在一种情况下,对于每液量盎司的饮料,本发明的强化铁饮料一般含有的EDTA铁的量至少是约1%,优选约1至约30%,更优选约1至约5%的U.S.DV。为了实现这样的铁含量,一般添加到本发明饮料溶液中的EDTA铁的比例是每液量盎司本发明的立即可饮饮料或复水还原饮料加入约0.2mg或更多的铁,优选约0.2至约3.0mg铁,更优选约0.4至约1.2mg铁。EDTA铁或EDTA铁钠一般含有约12%的铁。由此,为了达到上述的铁添加量而加入的铁的含量是易于计算和实施的。
对于本发明的立即可饮饮料或复水还原饮料,主要的共有成分是EDTA铁(有助于满足U.S.DV要求的有效量)和能溶解可饮用液体EDTA铁的。本发明的饮料和饮料混合粉料中可以包括任意可食用成分,例如风味剂、甜味剂、维生素、矿物质等,只要这些成分不会产生异味、不佳色泽或其它令人不快的感官特性就行。对于本发明的立即可饮饮料或复水还原饮料,优选主要的共有成分是EDTA铁(有助于满足U.S.DV要求的有效量)、风味料和能溶解可饮用液体EDTA铁和风味料的。优选唯一添加的铁的形式是EDTA铁。
能溶解EDTA铁、风味料和任意其它成分的可饮用液体可以是水基的、醇基的或对于这些成分而言其它任意合适的并且是可饮用的液体溶剂。一般情况下,水基液体介质在本发明的实施中是优选的。液体的比例一般应足以使这些成分溶解,并且还足以实现所需风味剂所要达到的浓度/稀释度。一般,本发明所涉及的用于水果饮料中水的配比,无论是立即可饮饮料还是复水还原形式,应该是约50至约99%。
当本发明的强化饮料是水果味饮料时,可以使用天然和合成的,水溶的和水可分散的风味剂,包括,例如市售的水果饮料风味剂用于本发明的一般水果风味物质,包括例如桔子油、酸橙油和柠檬油等。其它风味料也可以在用于水果饮料的公开配方中找到。另外,可以向饮料中添加天然浓缩果汁以提供或增强所需的水果味。这些浓缩果汁一般是液体、果浆或果汁的形式。浓缩果汁一般含有至少约45%的果汁。
在一个实施方案中,饮料中的风味剂是桔子味的。能在市场上得到的浓缩桔子香精含有萜烯。这些萜烯非常易于氧化,并由此易于产生异味。没有充分稳定的铁会氧化这些桔子味成分。EDTA铁成功地使这些铁稳定,使其不能与饮料中的桔子香精反应。
也可以将食用色素,例如U.S.Certified Food Colors(美国允许使用的食用色素)添加到本发明饮料中。在本发明饮料中还可以添加防腐剂;例子如包括苯甲酸钠、抗坏血酸、柠檬酸、乳酸、苹果酸、酒石酸和丙二醇等。另外,在本发明的饮料干混粉料的情况中,使用者通常可以使用自来水来进行复水还原。由于含有溶解的矿物盐,因此自来水通常略微有些偏碱性,所以也可以含有酸化剂,例如柠檬酸、磷酸或苹果酸等,以中和自来水中的碱度,或用于其它目的。优选的,防腐剂是可食用酸,例如抗坏血酸、柠檬酸、乳酸、苹果酸、酒石酸和其混合物;这样的可食用酸自然可以中和所使用的复水还原液体中存在的任何碱度。对于本发明的立即可饮饮料,在其包装之前,还可以任选地使该饮料碳酸化。本发明的饮料干混粉料可以任选地在碳酸化液体中复水还原或它们可以任选地在复水还原之后碳酸化。
本发明的饮料也可以含有甜味剂。甜味剂可以选自例如蔗糖、葡萄糖、果糖、水解玉米淀粉、麦芽糖糊精、玉米糖浆干粉、乳糖,高果糖玉米糖浆、果寡糖、人工甜味剂和其混合物的一种或多种。合适的人工甜味剂包括,例如阿斯巴甜、氯化蔗糖衍生物、糖精、环磺酸盐和双氧噻嗪钾等。在本发明的干混粉料的情况中,在复水为饮料之前,甜味剂一般以细颗粒的形式存在于混合料中。甜味剂的量可以是不同的,但一般如果含有,则含量占立即可饮饮料或复水还原饮料的0至约50%,更优选约5至约25%。
在本发明的实施方案中涉及了非脂饮料产品,所需的通常由脂肪提供的口感可以改为由非脂稳定剂提供所述的非脂稳定剂,例如,但不仅限于纤维素,例如羧甲基纤维素、羧甲基纤维素钠、羟甲基纤维素、羟丙基甲基纤维素、纤维素凝胶和黄原胶、角叉菜胶、瓜尔豆胶和阿拉伯树胶等,及其混合物。在本发明的饮料中,这样的稳定剂的量一般是0至约30%,更优选约5至约15%。
脂肪也可以选择性地作为饮料的成分添加,例如以乳脂、大豆油、氢化豆油、分馏的椰子油、高油酸红花油、玉米油、低芥酸菜籽油和可可脂等,以及其混合物形式添加。蛋白质也可以选择性地作为饮料成分添加,例如以乳蛋白质、植物蛋白(例如大豆、小麦)等,及其混合物的形式添加。例如大豆饮料可以制备成本发明实施方案的立即可饮饮料或由干混粉料复水还原的饮料。
本发明的饮料干混粉料可以由含EDTA铁的干风味剂基料形成。这样的风味剂基料能够通过本技术的公知的常规方式烘干,例如喷雾干燥、蒸发干燥、真空干燥和冷冻干燥等,或能够通过干混合各种成分而制备。优选的,该烘干方法提供平均粒径是约10至约1000微米的速溶颗粒。糖和/或稳定剂也可以用作该干风味剂基料的其它成分的载体。
饮料混合粉料包括水果味饮料粉、乳类饮料和植物基饮料(例如,豆奶)等。立即可饮饮料包括水果味饮料粉、乳类饮料和植物基饮料(例如,豆奶)等。本发明的强化饮料也可以被冷冻,形成冰棍、加果汁冰水等,或用于贮存和食用的其它常规形式(例如,浓缩饮料、甜食等)。
根据一个例举的、非限制性的本发明实施方案,优选可溶的、饮料干混粉料含有基于干基的约0.01至约0.2%的EDTA铁,约40至约70%的柠檬酸、约5至约25%的风味剂、约5至约30%的的稳定剂、约0至约15%的色素和约5至约15%的维生素/矿物质混合物。这里所使用的术语“干”或“粉”一般指的是水分含量小于约3%的可以相对自由流动的物质。
本发明饮料组合物的成分能够使用常规技术混合。例如,干成分能够与液体载体通过搅拌混合至所需的程度,以溶解这些成分,并确保这些成分基本上均匀地分散。
如上所述,本发明的铁强化饮料包括饮料干混粉料和立即可饮(RTD)饮料。无论制备成立即可饮饮料还是复水还原饮料,用于本发明的强化饮料的EDTA铁是具有高生物利用率的。确实,其可与硫酸亚铁相媲美。硫酸亚铁一般被认为是度量铁生物利用率的通用标准。确实,在膳食抑制剂例如肌醇六磷酸酯或肌醇六磷酸存在的情况下,EDTA铁结合的铁比硫酸亚铁中的铁利用率更高。而且,EDTA铁并不导致令人不快的感官特性。这种特性通常存在于是结合硫酸亚铁的每液量盎司饮料铁的U.S.DV中。
以下的实施例是为了对本发明进行举例说明,并不意味着是对本发明的限制。除非特别说明,本文所采用的所有百分数均是重量百分数。
实施例1.表1所详列的饮料粉基料配方采用不同形式的铁制备。各种产品在保存期内的稳定性由经过训练的感官属性评味员进行评定。

该包胶的硫酸亚铁由Balchem Corp.(Slate Hill,NY)生产。FerrochelTM是具有营养功能的矿物质氨基酸螯合物,其含有用于强化食品的铁,由Albion Laboratories,Inc.(Clearfield,Utah)生产。
强化的饮料粉样品被密封在铝袋中(每份样品10袋,袋装有22g饮料混合物),并接着在控制的环境(即约85%的相对湿度和约90)下,储存16周。在这些储存条件下,储存一周相当于在正常储存条件下的约1.25至约1.5个月。
一开始(即,0周)对11种贮存样品的每种中的一袋进行感官评定,并且在此后每两周需要进行一次感官评定。为了达到评定的目的,将样品袋中装有的22g物料经混合溶于室温下烧杯中的180ml自来水中,以提供各自的饮料样品。由训练有素的评味员对感官特性进行评定。他们可以定性地鉴别出轻微、中度的和强烈的金属味之间的差异,并识别出一般与源自桔子香精的任何氧化萜烯相关的异味。该饮料样品在室温下,盛在任意编码的烧杯中提供给评味员(即盲检)。平均用约10位评味员(且没有少于5位的情况)来进行每一轮的感官测试。采用两套独立的分数对测试样品进行评定:一个是针对金属味的,而一个是综合分(即结合外观、风味和异味评定的分数)。采用的感官等级是由0至8;对于金属味的评定,0指的是没有金属异味,而8则指有明显的金属异味;对于综合分,0指的是非常差,而8是优秀。
感官评定的结果以附图中柱状图的形式显示。用EDTA铁钠(EDTA铁)制备的本发明饮料样品(样品3)是稳定的,且在16周的测试期间与负对照样品(即,无铁)相比较具有最低的异味,没有不佳的色泽。而且,本发明饮料样品被认为与“无铁的”对照样品的感官特性差不多一样,不需要任何附加的防腐剂或风味掩蔽剂。
而且,本发明的EDTA铁强化饮料样品相对于所有被测试的铁形式提供了明显出色的结果。在铁强化样品中,只有本发明的EDTA铁强化饮料样品(样品3)在整个十六周的测试期间提供了令人满意的结果。相反,用硫酸亚铁、葡萄糖酸亚铁、富马酸亚铁和酒石酸亚铁强化的对照样品(分别是样品2、6、9、10),在一经配制马上就有了明显的金属异味,并且随着十六周试验的继续,味道越来越重,并伴随产生了氧化味。用包胶的硫酸亚铁、FerrochelTM、含铁糖类和柠檬酸亚铁强化的对照样品(分别是样品4、5、7、8)在试验的开始并没有明显的金属味;但在十六周试验结束之前,所有样品均产生中度至强烈的金属异味,并伴随产生了氧化味,这是不能令人接受的。负对照样品(不含铁的样品1),正如人们预料的,在十六周的试验期间,并没有产生金属异味或氧化异味。令人惊讶的是,与用不溶/低反应形式,例如富马酸亚铁和甘油磷酸亚铁配制的饮料样品(分别是样品7和9)相比,本发明的EDTA铁强化样品也显示出更好的稳定性。在正常贮存条件(即,未采取加速条件)下进行的贮存稳定性测试已经证明了加速贮存的评定结果。
实施例2.本实施例对用EDTA铁强化的本发明饮料样品的生物可利用性与用硫酸亚铁强化的对照样品进行了比较。使用实施例1中所述的样品2和3的同样方式和同样的配方制备饮料样品。使用如US6017713(在此引入作为参考)所主要描述的铁蛋白技术测定制备的每一个饮料样品的铁生物可利用性。简单地说,就是将0.9ml饮料样品随着标准餐引入一模拟的消化、胃蛋白酶和肠系统(Glahn等人,J.Nutrition,128,1555(1998))。得到的消化溶液被放在位于单层人体肠癌细胞(即Caco-2细胞)之上的透析膜顶部的井中。源自样品中可用的铁被Caco-2细胞吸收,并以铁蛋白的形式(贮存蛋白)贮存。接着用如US6017713所述方法测定铁蛋白含量。将源自本发明EDTA铁强化饮料样品的铁生物可利用性与硫酸亚铁强化的样品进行比较。
实施例3.本实施例对本发明的EDTA铁饮料与负(即,无铁)和正(即,硫酸亚铁)对照立即可饮的产品进行稳定性比较。采用以下表3所述的基本配方,在试验工厂中制备25磅批次的立即可饮(RTD)饮料,其中:批次1-负对照;批次2-本发明产品;和批次3-正对照。每200ml的量批次2和3的饮料提供约3.6mg Fe。将配制的立即可饮饮料在205巴氏杀菌30秒,热填充入袋,并密封。

除了将各样品的200ml等分试样在生产线上灌入铝袋中以外,对饮料进行了如实施例1所述的加速保存期测试,并在与实施例1相同的贮存条件下,立即进行和在贮存期内每周一次进行测评。如实施例1所述进行感官评定。采用与实施例1相同的0至8等级进行测评。感官评定的结果列于下表4中。

在采用加速贮存条件下,三周约等于3个月的正常贮存期。在这三周时间内,EDTA铁钠强化的样品(本发明样品,批次2)是稳定的,与无铁对照样品(批次1)相当。用硫酸亚铁强化的样品(批次3)显示出明显的异味,并得到了不能令人满意的分数。由于在加工期间的热处理,且立即可饮形式有稳定性,无需借助于对配方添加物料以增强稳定性或掩蔽异味,本发明EDTA铁钠样品的稳定性特别明显。
借助于对具体的工艺和产品实施方案的说明,本发明已经得到了特别具体的描述,应该理解在本发明公开的基础上可进行各种改变、改进以及适应性变动,并且它们是在下述权利要求所定义的本发明的实质和范围之内。

1.一种铁强化饮料,含有EDTA铁和有效量的至少溶解基本全部EDTA铁的饮用液体,其中在该饮料中不存在显著量的自由铁离子,并且与没有铁源的饮料相比,该饮料没有异常的感官特性。
2.根据权利要求1的铁强化饮料,还含有风味剂。
3.根据权利要求2的铁强化饮料,其中每液量盎司饮料,含有的EDTA铁的含量应能提供约1至约30%的U.S.DV的铁。
