Description
Die vorliegende Erfindung bezieht sich auf in 2-Stellung substituierte oder unsubstituierte 6-(2-Oxo-l,3-dioxolan-4-yl)-2-15 penem-3-carbonsäurederivate und auf in 2-Stellung substituierte oder unsubstituierte 6-(l-Hydroxy-2-alkyliden)-2-penem-3-car-bonsäurederivate der folgenden Formeln Ia und Ib sowie auf Verfahren zur Herstellung derselben und auf solche Verbindungen enthaltende pharmazeutische Präparate, wobei diese Ver-20 bindungen den folgenden Formeln entsprechen ch2oh

worin R, R1 und R2 die obigen Bedeutungen haben, in einem geeigneten Lösungsmittel unter Rückfluss erhitzt, um einen Penemring zu bilden.
5. Verfahren zur Herstellung von Verbindungen der Formel Ia gemäss einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass man eine Verbindung der folgenden Formel worin R das Wasserstoffatom, ein Leichtmetall oder eine Carbo-xyschutzgruppe, R1 eine Alkylgruppe und R2 das Wasserstoffatom, den Methylrest oder einen substituierten Methylrest bedeuten.
35 Bei der obigen Definition soll die Bezeichnung «Leichtmetall» ein Metall aus der zweiten bis vierten Periode der Gruppen I bis III des periodischen Systems bedeuten, welches ein physiologisch annehmbares Ion in menschlichen oder tierischen Körperflüssigkeiten liefert. Beispiele von solchen Leicht-40 metallen sind Lithium, Natrium, Kalium, Magnesium, Aluminium und Calcium.
Die Bezeichnung «Carboxyschutzgruppe» bezieht sich auf eine beliebige Schutzgruppe, welche üblicherweise in der Penicil-lin-Cephalosporin-Chemie zum Schützen der in der 3- oder 4-45 Stellung einer Penicillin- oder Cephalosporinverbindung vorhandenen Carbonsäuregruppe zur Anwendung gelangt und sich leicht ohne Beeinträchtigung des übrigen Teiles des Moleküls entfernen lässt. Beispiele solcher Schutzgruppen sind Aralkylre-ste, wiez. B. Benzyl, Methylbenzyl, Dimethylbenzyl, Methoxy-50 benzyl, Ethoxybenzyl, Nitrobenzyl, Aminobenzyl, Diphenylme-thyl, Phthalidyl, Phenacyl usw. ; substituierte Alkylreste, wie z. B. Trichlorethyl, tert.-Butyl, Allyl usw. sowie Arylreste, wie z. B. Pentachlorphenyl, Indanylusw. Diese Bezeichnung umfasst ebenfalls Esterreste, die gebildet werden bei der Umsetzung 55 zwischen der Carbonsäure an der 3-Stellung und einer N-Hy-droxyaminoverbindung, wiez. B. Acetonoxim, Acetophenon-oxim, Acetoaldoxim, N-Hydroxysuccinimid oder N-Hydroxy-phthalimid, Säureanhydridreste, wie sie bei der Umsetzung zwischen einer Carbonsäure und Kohlensäure oder einer beliebi-60 gen Carbonsäure gebildet werden, und starke reaktionsfähige Amidreste, die bei der Umsetzung zwischen einer Carbonsäure und einem Amin gebildet werden, z. B. ein substituiertes Amin oder ein substituiertes Hydrazin.
Die oben erwähnten Carboxyschutzgruppen können als Sub-65 stituenten einen Alkyl-, Cyanoalkyl-, Carboxyalkyl-, Carba-moylalkyl-, Hydroxyamoylalkyl, Alkoxyamoylalkyl-, Aminoal-kyl-, N-alkylierten Aminoalkyl-, Ureidoalkyl-, Halogenalkyl-, Carbamoylhalogenalkyl-, Arylthioalkyl-, heterocyclischen
Ethioalkyl-, Sulfamoylalkyl-, Alkenyl-, Carboxy-, Amino-, Nitro-, Hydroxyl- oder Oxorest oder ein Halogenatom enthalten. Diese Substituenten können überdies weitere Substituenten der soeben genannten Gruppen und Atome aufweisen. Ferner kann der Carboxyl-, Carbamoyl-, Amino-, Hydroxy- oder Oxorest, welcher in den Substituenten enthalten ist, durch eine übliche Schutzgruppe modifiziert sein. Zudem können zwei oder mehrere dieser Substituenten eine Ringbildung eingehen.
Die oben erwähnten Substituenten werden nachstehend als «übliche Substituenten» bezeichnet.
Besonders bevorzugte Carboxyschutzgruppen sind alkyl-esterbildende Gruppen, wie z. B. Alkyl, Alkanoyloxyalkyl, Alk-oxyalkyl oder Aminoalkoxyalkyl, aralkylesterbildende Gruppen, wie z. B. Benzyl, p-Nitrobenzyl, Phenacyl oder Phthalidyl sowie arylesterbildende Gruppen, wie z. B. Phenyl oder Halo-genphenyl.
Die Bezeichnung «Alkylgruppe» bzw. «Alkylrest», wie dies in Verbindung mit dem Symbol R1 verwendet wird, bezieht sich auf beliebige niedrige Alkylgruppen, wie z. B. Methyl oder Ethyl.
Schliesslich umfasst die Bezeichnung «substituiertes Methyl» in der Definition für das Symbol R2 Halogenmethyl-, Acyloxy-methyl-, heterocyclische Thiomethyl- und Dimethyl-tert.-butyl-silyloxygruppen.
Chlormethyl wird als Halogenmethylrest bevorzugt.
Bevorzugte Beispiele von Acyloxymethylgruppen sind Alk-anoyloxymethylgruppen, wie z. B. Acetyloxymethyl, Acyloxymethylgruppen, die sich von der Kohlensäure ableiten, Carba-moyloxymethylgruppen und solche Gruppen in einer geschützten Form. Als Acyloxymethylgruppen werden jedoch die Aral-kanoyloxymethyl-, Aroyloxymethyl-, Alkylsulfonyloxymethyl-und Arylsulfonyloxymethylgruppen bevorzugt.
Der heterocyclische Teil in der heterocyclischen Thiomethyl-gruppe ist eine 5- oder 6-gliedrige, mono- oder bicyclische, konjugierte heterocyclische Gruppe mit mindestens einem Hete-roatom, wobei das Heteroatom aus der Gruppe, bestehend aus Stickstoff-, Sauerstoff- und Schwefelatomen, gewählt wird. Die heterocyclische Gruppe kann gegebenenfalls einen oder mehrere der vorgenannten üblichen Substituenten enthalten.
5-gliedrige heterocyclische Gruppen mit drei oder vier Hete-roatomen werden besonders bevorzugt. Spezifische Beispiele solcher besonders bevorzugten heterocyclischen Gruppen sind Triazol, Oxadiazol, Thiadiazol, Tetrazol sowie Imidazol, Triazin und Dihydrotriazin.
Die Verbindungen der Formeln Ia und Ib besitzen starke antibiotische Wirkungen in bezug auf verschiedene Mikroorganismen und ebenfalls in bezug auf Mikroorganismen, welche gegenüber verschiedenen Antibiotika resistent sind.
Somit sind die erfindungsgemässen Verbindungen der Formeln Ia und Ib wertvolle Antibiotika gegen verschiedene grampositive und gramnegative Bakterien und sind daher wertvolle Arzneimittel für Mensch und Tier. Sie lassen sich für die Behandlung oder die Verhinderung von Infektionen anwenden, welche durch grampositive Bakterien, z.B. Staphylococcus aureus, Streptococcus pyogenes, Bacillus subtilis, Bacillus cereus, Diplococco pneumoniae, Corynebacterium diphteriae und gramnegative Bakterien, wie z. B. Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, Proteus vulgaris, Proteus rettgeri, Proteus morganii, Enterobacter cloacae, Shigella sonnei, Salmonella paratyphi, Salmonella typhi, Serratia marcescens, einschliesslich anaerober Bakterien, wie z.B. Bacteroides fragilis, Clostridium perfringens, verursacht werden. Sie lassen sich auch als Desinfektionsmittel für die Verhinderung des Zerfalles von verderblichen Materialien, als Zusatzstoffe zu Nahrungsmitteln oder zur Verhinderung eines bakteriellen Wachstums bei Hygieneartikeln verwenden.
Die Verbindungen der Formeln Ia und Ib lassen sich als solche oder als Verbindung mit anderen Wirkstoffen für die

verschiedensten Arten der oralen oder parenteralen Dosierungsformen verwenden. Die pharmazeutischen Präparate können in Form einer Mischung von 0,01 bis 99% mindestens einer Verbindung der Formel Ia oder Ib mit einem pharmazeutischen Trägermittel angewendet werden, wobei das Trägermittel ein festes oder flüssiges Material sein kann, in welchem die Verbindungen gelöst, dispergiert oder suspendiert sind. Sie können in einer Einheitsdosierungsform vorliegen. Die festen Präparate können in Form von Tabletten, Pulvern, trockenen Sirupen, Pastillen, Granulaten, Kapseln, Pillen, Suppositorien oder dgl. vorliegen. Die flüssigen Präparate können in Form von Injektionslösungen, Salben, Dispersionen, Inhaliermitteln, Suspensionen, Lösungen, Emulsionen, Sirupen oder Elixieren vorliegen. Sie können geschmackskorrigierende Mittel enthalten und gefärbt sein, wobei die Tabletten, Granulate und Kapseln mit einem Überzug versehen sein können.
Man kann sämtliche Verdünnungsmittel, wiez. B. Stärke, Saccharose, Lactose, Calciumcarbonat oder Kaolin; Füllmittel, wiez. B. Lactose, Zucker, Salz, Glycin, Stärke, Calciumcarbonat, Calciumphosphat, Kaolin, Bentonit, Talk oder Sorbitol; Bindemittel, wie z. B. Stärke, Akaziengummi, Gelatine, Glucose, Natriumalginat, Tragacanthgummi, Carboxymethylcellu-lose, Sirup, Sorbitol oder Polyvinylpyrrolidon; Auflockerungsmittel wiez. B. Stärke, Agar, Carbonate oder Natriumlaurylsul-fat; Gleitmittel, wie z. B. Stearinsäure, Talk, Paraffin, Borsäure, Siliciumdioxid, Natriumbenzoat, Polyethylenglycol, Kakaoöl oderMagnesiumstearat; Emulgiermittel, wiez. B. Lecithin, Sor-bitanmonooleat, Glycerindioctanoat oder Akaziengummi; Suspendiermittel, wie z. B. Sorbitol, Methylcellulose, Glucose, Zuckersirup, Gelatine, Hydroxyethylcellulose, Carboxymethyl-cellulose, Aluminiumstearatgel oder hydrierte Fette; Lösungsmittel, wiez. B. Wasser, Puffermittel, Erdnussöl, Sesamöl oder Methyloleat; Präservativa, wie z. B. Methyl- oder Ethyl-p-hydro-xybenzoat oder Sorbinsäure; geniessbare Färbemittel, aromatische Substanzen, löslich machende Mittel, Puffermittel, Stabilisiermittel, Analgetika, Dispergiermittel, Netzmittel, Antioxidantien und dgl. verwenden, vorausgesetzt, dass solche Mittel die Wirkung der erfindungsgemässen Verbindungen nicht nachteilig beeinflussen.
Die erfindungsgemässen Verbindungen der Formeln Ia und Ib, worin R ein Leichtmetall darstellt, sind in Wasser löslich und werden üblicherweise in Form von Lösungen für eine orale Verabreichung oder für eine intravenöse, intramuskuläre oder subcutane Injektionslösung in an sich bekannter Weise verwendet. Die Verbindungen lassen sich in wässrigen oder öligen Lösungsmitteln für Injektionszwecke auflösen, wobei man Lösungen in Ampullen bilden kann. Im allgemeinen lässt sich eine verlängerte Lagerungsdauer dadurch bewirken, dass man eine Ampulle herstellt, welche Kristalle, Pulver, Mikrokristalle oder Lyophilisate von Verbindungen der Formel Ia oder Ib enthält, wobei man die Arznei vor der Anwendung in einem der erwähnten Lösungsmittel löst oder suspendiert, worauf man solche Lösungen oder Suspensionen für Injektionszwecke verwenden kann. Eine solche Ampulle oder eine solche Injektionslösung lässt sich einem Patienten bei einer täglichen Dosierungsmenge von beispielsweise 0,2 bis 5 g verabreichen, und zwar je nach der Art der infizierten Bakterien, je nach dem Zustand des Patienten und je nach Intervallen der Verabreichung.
Die Verbindungen der Formeln Ia und Ib in Form von pharmazeutisch zulässigen Estern, wiez. B. Indanyl-, Acetoxy-methyl-, Pivaloyloxymethyl-, Ethoxycarbonyloxy ethyl-, Phenacyl-, Phthalidyl-, Phenyl-, Tolyl-, Xylyl-, Methoxyphenylestern, lassen sich durch den Verdauungstrakt in gewissem Masse absorbieren und können in Form von Pulvern, Tabletten, Granulaten, Kapseln, trockenem Sirup, Emulsionen, Lösungen, Suspensionen und ähnlichen oralen Präparaten an Mensch und Tier verabreicht werden. Sie können in Form von reinen Verbindungen oder als mindestens eine Verbindung der Formel Ia oder Ib

und mindestens einen pharmazeutischen Trägerstoff enthaltende Mittel angewendet werden. Die Herstellung lässt sich nach an sich für solche Zwecke ähnlichen Methoden durchführen, wobei man dem Patienten eine tägliche Dosis von beispielsweise 1 bis 2 g je nach dem Zustande des Patienten und der in Frage kommenden Krankheit, verabreichen kann.
Für die Behandlung oder die Verhinderung von menschlichen und Veterinären bakteriellen Infektionen kann man eine Verbindung der Formel Ia bzw. Ib bei einer täglichen Dosis von beispielsweise 0,2 bis 5 g für Injektionszwecke oder beispielsweise 1 bis 2 g für orale Verabreichung oder aber 10 |ig bis 1 g für die topische Anwendung bei Intervallen von beispielsweise 3 bis 12 Stunden verabreichen.
Die therapeutischen Indikationen sind Krankheiten, welche durch Bakterien verursacht werden, die auf Verbindungen der Formeln Ia und Ib ansprechen, wie z.B. Pneumonie, Bronchitis, Pneumonitis, Empyem, Nasopharyngitis, Tonsillitis, Rhinitis, Dermatitis, Pustulosis, Ulcusbildung, Abszesse, Wund- und Weichteilinfektionen, Ohreninfektionen, Osteomyelitis, Septi-cämie, Gastroenteritis, Enteritis, Infektionen der Luft- oder Harnwege und Pyelonephritis.
Die Verbindungen der Formeln Ia und Ib werden dem Patienten vorzugsweise in Form von pharmazeutischen Präparaten, wie z. B. Pulvern, trockenen Sirupen, Tabletten, Pastillen, Granulaten, Kapseln, Pillen, Suppositorien, Injektionslösungen, Salben, Dispersionen, Inhalierungsmitteln, Suspensionen, Lösungsmitteln, Emulsionen, Sirupen und Elixieren verabreicht. Sie können in einer Einheitsdosierungsform, z. B. in Form von Tabletten, Pastillen, Kapseln, Injektionslösungen, Ampullen, Granulaten oder Pulvern in einem separaten Behälter oder Verpackung vorliegen.
Sämtliche oben erwähnten pharmazeutischen Präparate lassen sich in bekannter Weise herstellen.
Selbstverständlich wird der Fachmann auch erkennen können, dass die Verbindungen der Formeln Ia und Ib auch als keimtötende Mittel oder als Antiseptika zur Anwendung gelangen können. Überdies sind sie als Ausgangsmaterialien für die Herstellung von gewissen andern Verbindungen der Formeln Ia und Ib und als Antibiotika für das Testen der Empfindlichkeit von Mikroorganismen wertvoll.
Bevorzugte Verbindungen der Formeln Ia und Ib, wie sie erfindungsgemäss angestrebt werden, sind solche, in welchen der Rest RNatrium, Kalium, Diphenylmethyl, p-Methoxybenzyl, Phthalidyl, 2-Oxo-dioxolenylmethyl, Acetoxymethyl- l-(Eth-oxycarbonyloxy)-ethyl- oder Pivaloyloxymethyl bedeutet.
Die Verbindungen der Formeln Ia und Ib lassen sich nach verschiedenen Verfahren herstellen, welche nachstehend ausführlich beschrieben werden:
Die Methoden für die Herstellung eines Salzes einer Verbindung der Formel Ia, wie sie oben erwähnt worden ist, sind auch anwendbar für die Herstellung von Salzen von Verbindungen der Formel Ib.

2. Eliminierung der Carboxyschutzgruppe Eine Verbindung der Formel Ia, worin R eine Carboxyschutzgruppe darstellt, kann nach irgendeiner beliebigen der üblichen, Schutzgruppen entfernenden Reaktionen der nachstehend 10 beschriebenen Art in eine Verbindung der Formel Ia übergeführt werden, worin R das Wasserstoffatom darstellt.
In der folgenden Beschreibung wird die Carboxyschutzgruppe hin und wieder mit dem Namen wiedergegeben, welcher der Gruppe entspricht, die durch die Umsetzung zwischen der 15 Carbonsäure und der für das Schützen der Carbonsäure zur Anwendung gelangenden Verbindung gebildet wird, dies aber lediglich zwecks Vermeidung einer Komplexizität der Beschreibung. So wird die Schutzgruppe «R» im Rest der Formel

1. Herstellung von Salzen
Die Umsetzung einer Verbindung der Formel Ia, worin R das Wasserstoffatom darstellt, mit einer Base oder einem Salz einer schwächeren Carbonsäure liefert eine Verbindung der Formel Ia, worin R ein Leichtmetall bedeutet. Die Umsetzung kann in an sich für den Fachmann bekannter Weise durchgeführt werden. Bevorzugte Methoden bestehen darin, dass man eine freie Säure der Formel Ia mit einem Leichtmetallbicarbonat neutralisiert. Eine weitere Methode ist die Austauschreaktion einer freien Säure der Formel Ia mit einem Salz einer niederen Carbonsäure in einem polaren, organischen Lösungsmittel, wie z. B. Alkohol, Keton oder Ester, worauf man ein Lösungsmittel zugibt, in welchem das gewünschte Salz der Formel Ia spärlich löslich ist.
Die obigen Umsetzungen benötigen eine bis zehn Minuten, sofern sie bei einer Temperatur von weniger als 50 °C durchgeführt werden. Nötigenfalls kann man das Reaktionsgemisch während einer längeren Zeitdauer auf sich wirken lassen, vorausgesetzt dass keine Nebenreaktionen auftreten.
aus Zweckmässigkeitsgründen auch als «Ester» bezeichnet.
a) Die stark reaktionsfähigen Schutzgruppen aufweisenden 25 Verbindungen der Formel Ia können dadurch beseitigt werden,
dass man sie mit einer Säure, einer Base, einem Puffer oder einem Ionenaustauschharz in einer wässrigen Lösung behandelt. Weniger reaktionsfähige Schutzgruppen, wiez. B. Trichlore-thyl-, p-Nitrobenzyl- oder Phenylacylester können dadurch beseitigt werden, dass man solche Verbindungen mit einem Metall und einer Säure oder mit einem Dithionat behandelt oder aber katalytisch reduziert.
b) Aralkylester können durch Hydrierung unter Verwendung von beispielsweise Palladium oder Nickel als Katalysator beseitigt werden.
c) Aralkylester, Cyclopropylmethylester undSulfonylethyl-ester lassen sich durch Solvolyse unter Anwendung einer Mineralsäure, einer Lewissäure, einer Sulfonsäure oder einer starken Carbonsäure und nötigenfalls in Gegenwart eines Kationenspül-mittels entfernen.
d) Phenylacylester, AlkenylesterundHydroxyaralkylester können durch Einwirkung einer Base oder eines Nucleophils entfernt werden. Fotochemisch aktive Phenylacylester können durch Lichtbestrahlung eliminiert werden.
e) 2-Alkinylester können durch Umsetzung mit einem Alkali-metallalkanoat und Palladium-Triphenylphosphin in ein Alkalimetallsalz übergeführt werden.
f) Es lassen sich im übrigen auch die anderen für die Beseitigung von Carboxyschutzgruppen bekannten üblichen Verfahren im Zusammenhang mit der vorliegenden Erfindung anwenden.
Die Verbindungen der Formel Ib, worin R eine Carboxyschutzgruppe darstellt, können in der gleichen Weise, wie dies oben in Verbindung der Formel Ib, worin R das Wasserstoffatom darstellt, beschrieben worden ist, übergeführt werden.

3. Einführung einer heterocyclischen Thiogruppe Die Verbindungen der Formel Ia, worin R2 eine heterocyclische Thiomethylgruppe darstellt, können durch Umsetzung einer Verbindung der Formel Ia, worin R2 einen Methylrest 60 darstellt, der als Substituenten eine abspaltbare Gruppe aufweist, mit einem entsprechenden heterocyclischen Thiol oder einem reaktionsfähigen Derivat davon erhalten werden. Bevorzugte abspaltbare Gruppen sind Halogenatome, Sulfonyloxy-und Alkanoyloxygruppen. Bevorzugte reaktionsfähige Derivate 65 des heterocyclischen Thiols sind Alkalimetallsalze und Ammoniumsalze von Thiol. Die Reaktion kann bei einer Temperatur zwischen 0 und 60 °C in einem wasserfreien Lösungsmittel oder in einem wässrigen Lösungsmittel durchgeführt werden. Die

Umsetzung lässt sich gegebenenfalls durch Zugabe eines Dehy-dratisierungsmittels oder von Phosphorylchlorid beschleunigen.
Die obige Beschreibung ist auch anwendbar für die Herstellung von Verbindungen der Formel Ib.
4. Cyclisierung Wie in Beispiel 2 erläutert wird, lassen sich die Verbindungen der Formel Ia oder Ib gemäss Erfindung durch Erhitzen unter Rückfluss einer geeigneten Carbonylalkylidenphosphoranver-bindung in einem geeigneten Lösungsmittel herstellen, wobei die Verbindung eine Ringschlussbildung eingeht und dies unter Bildung eines Penemrings. Die bevorzugten Lösungsmittel für diese Umsetzung sind Benzol, Toluol und Xylole.
5. Umwandlung einer Verbindung der Formel Ia in eine Verbindung der Formel IB Die Dioxolanylverbindungen der Formel Ia können in Hydroxy alkylidenverbindungen der Formel Ib dadurch übergeführt werden, dass man die ersteren mit einer katalytischen Menge von l,5-Diazabicyclo-[5,4,0]-5-undecen (DBU) oder Natriumbicarbonat in einem Lösungsmittel, wie z. B. Chloroform, Benzol, Acetonitril oder Wasser, behandelt.
6. Herstellung von pharmazeutisch aktiven Estern Die Verbindungen der Formel Ia, worin R eine Carboxy-schutzgruppe darstellt, lassen sich nach der Eliminierung der Schutzgruppe gemäss dem oben erwähnten Verfahren in eine Verbindung der Formel Ia überführen, welche eine pharmazeutisch aktive Estergruppe trägt. So kann man beispielsweise eine Verbindung der Formel Ia, worin R den Pivaloyloxymethylrest darstellt, dadurch herstellen, dass man eine Verbindung der Formel Ia, worin R das Wasserstoffatom darstellt, mit Pivaloyl-oxymethyliodid in Dimethylformamid (DMF) bei einer Temperatur zwischen 0 °C und Zimmertemperatur in Gegenwart einer Base, wie z. B. Triethylamin, umsetzt.
Die Verbindungen der Formel Ib, welche eine pharmazeutisch wirksame Estergruppe aufweisen, lassen sich in eben dergleichen Weise, wie dies soeben beschrieben worden ist, herstellen.
7. Schutz der Carbonsäuregruppen bzw. anderer reaktionsfähiger funktioneller Gruppen Bei der Durchführung der vorgenannten, verschiedenen Reaktionen oder bei der Überführung einer Verbindung der Formel Ia bzw. Ib in eine andere Verbindung der Formel Ia bzw. Ib mag es hin und wieder erforderlich sein, andere reaktionsfähige, funktionelle Gruppen zu schützen als die für die beabsichtigte Umsetzung in Frage kommende, reagierende Gruppe. Für diesen Zweck dienen verschiedene übliche Arten zum Schützen von Gruppen. Solche Methoden finden sich beispielsweise in der Literatur, wie z. B. J.F.W. McOmie Ed., «Protective Groups in OrganicChemistry», S. 183, Pleum Press, N.Y., 1973; S. Patai Ed., «The Chemistry of Functional Groups», S. 505, Interscience Pubi., John Wiley & Sons Ltd., London, 1969; und Flynn Ed., «Cephalosporins and Penicillins», AcademicPress, N.Y., 1972. Typische Beispiele zum Schützen von reaktionsfähigen, funktionellen Gruppen sind die Acylierung und Veretherung im Falle von Hydroxylgruppen, die Acylierung, Enaminierung und Sily-lierung im Falle von Aminogruppen und die Veresterung, Ami-dierung und die Bildung eines Säureanhydrids im Falle von Cabronsäuren.
Es sei daraufhingewiesen, dass die Bezeichnung «Schützen von Carbonsäuren», wie dies hier verwendet wird, die Veresterung einer Carbonsäure in der 3-Stellung zum Zwecke der

Herstellung eines pharmakologisch aktiven Esters umfasst. Die Veresterung von Verbindungen der Formel Ia bzw. Ib, welche eine freie Cabonsäure enthalten, kann dadurch bewirkt werden, dass man die Säure mit einer Base unter Bildung eines Carboxy-5 lates neutralisiert und das letztere mit einem Säurehalogenid behandelt, das einen geeigneten Esterrest enthält.
8. Reaktionsbedingungen 10 Die meisten der in den obigen Absätzen 1 bis 7 aufgezählten Reaktionen werden üblicherweise bei einer Temperatur zwischen -50 °C und 100 °C und insbesondere zwischen -20 °C und 50 °C während 10 Minuten bis 5 Stunden in einem geeigneten Lösungsmittel und nötigenfalls unter wasserfreien Bedingungen 15 durchgeführt.
Beispiele von für diese Verfahren verwendbaren Lösungsmittel sind Kohlenwasserstoffe, wiez. B. Pentan, Hexan, Octan, Benzol, Toluol oderXylol; halogenierte Kohlenwasserstoffe, wie z. B. Dichlormethan, Chloroform, Tetrachlorkohlenstoff, 20 Dichlorethan, Trichlorethan oder Chlorbenzol; Ether, wie z. B. Diethylether, Methylisobutylether, Dioxan oder Tetrahydro-furan; Ketone, wie z. B. Aceton, Methylethylketon oder Cyclo-hexanon; Ester wie z.B. Ethylacetat, Isobutylacetat oder 25 Methylbenzoat,Nitrokohlenwasserstoffe, wiez. B. Nitromethan oder Nitrobenzol; Nitrile, wie z.B. Acetonitril oder Benzonitril; Amide, wie z. B. Formamid, Acetamid, Dimethylformamid, Dimethylacetamid oder Hexamethylphosphorotriamid; Sulf-oxide, wiez. B. Dimethylsulfoxid; Carbonsäuren, wie z. B. 30 Ameisensäure, Essigsäure oder Propionsäure; organische Basen, wiez. B. Diethylamin, Triethylamin, Pyridin, Picolin, Collidin oder Chinolin; Alkohole, wie z. B. Methanol, Ethanol, Propanol, Hexanol, Octanol oder Benzylalkohol ; Wasser oder andere industriell zugängliche Lösungsmittel und Mischungen 35 davon.
Die auf diese Weise schliesslich erhaltenen Produkte der Formel Ia bzw. Ib der vorliegenden Erfindung lassen sich aus dem Reaktionsgemisch nach irgendeiner der üblichen bekannten Methoden, z. B. durch Absorption, Eluierung, Destillierung, 40 Ausfällung, Konzentration, Chromatographie und dgl. isolieren, nachdem man Verunreinigungen, wiez. B. noch verbleibende Ausgangsmaterialien, Nebenprodukte und Lösungsmittel in an sich bekannter Weise, z. B. durch Extraktion, Eindampfen, Waschen, Filtrieren, Trocknen, usw., entfernt hat.
45 Praktische und besonders bevorzugte Ausführungsformen der vorliegenden Erfindung werden in den folgenden Präparaten und Beispielen erläutert, worin die physicochemischen Daten der Produkte in den Tabellen I bis VI aufgezählt werden. Die Infrarotdaten (IR) und die magnetischen Kernresonanzdaten 50 (NMR) werden durch v (cm1) und ò(ppm)-Werte bzw. die Kupplungskonstante J durch HZ wiedergegeben. Die Teile und Prozentsätze sind jeweils Gewichtsteile bzw. Gewichtsprozente, sofern nichts anderes ausgesagt wird. 1,1g des rohen Ausgangsmaterials der Formel II-l werden in 100 ml trockenem Toluol gelöst und diese Lösung dann während 2 h auf 85 °C erhitzt. Nach Beendigung der Umsetzung wird das Lösungsmittel unter vermindertem Druck entfernt und der Rückstand durch Chromatographie über Kieselgel unter Verwendung einer Mischung von Benzol und Aceton im Mischungsverhältnis von 4:1 als Eluiermittel chromatographiert. Die in der obigen Überschrift erwähnte Verbindung (2-1) wird in Form eines Schaumes in einer Ausbeute von 670 mg erhalten.

248 mg der Ausgangsverbindung II-l werden in einer Mischung von 10 ml Methylenchlorid und 1 ml Wasser gelöst. Zu dieser Lösung gibt man dann 200 mg des Natriumsalzes von 5-Mercapto-l-methyl-1,2,3,4-tetrazol und 20 mg Tetra-n-butylam-moniumbromid hinzu, worauf man das Gemisch während 2 h bei Zimmertemperatur rührt. Die organische Phase wird hernach abgetrennt, mit gesättigter, wässrigerNatriumbicarbonatlösung gewaschen und über Magnesiumsulfat getrocknet. Nach dem Verdampfen des Lösungsmittels wird der Rückstand über Kieselgel unter Verwendung einer Mischung von Benzol und Ethylace-tat im Mischungsverhältnis von 1:3 als Eluiermittel gereinigt, wobei man das Zwischenprodukt erhält, welches eine Tetrazolyl-thiomethylgruppe aufweist.
Ausbeute: 260 mg. IR (v™?3 ): 1805,1755.
Das so erhaltene Zwischenprodukt wird in 20 ml Toluol gelöst und die Lösung alsdann während 2 h auf 90 °C erhitzt. Nach Beendigung der Umsetzung wird das Lösungsmittel im Vakuum verdampft und der Rückstand durch Säulenchromatographie über Kieselgel unter Verwendung einer Mischung von Benzol und Ethylacetat im Mischungsverhältnis von 2:1 als Eluiermittel gereinigt, wobei man die gewünschte Verbindung (3-1) in Form eines Schaumes erhält. Die Ausbeute beträgt 130 mg.

40 mg der gemäss Angaben in Beispiel 1 erhaltenen Ausgangsverbindung (2-1) werden in einer Mischung von 2 ml Methylenchlorid und 0,5 ml Wasser gelöst. Diese Lösung wird hierauf mit 19 mg des Natriumsalzes von 5-Mercapto-l-methyl-1,2,3,4-tetrazol und 3 mg Tetra-n-butylammoniumbromid versetzt und das Gemisch während 2 h bei Zimmertemperatur gerührt. Die organische Phase wird abgetrennt, mit gesättigter wässriger Natriumbicarbonatlösung gewaschen und über Magnesiumsulfat getrocknet. Das organische Lösungsmittel wird im Vakuum verdampft und der Rückstand durch Chromatographie über Kieselgel unter Verwendung einer Mischung von Benzol und Ethylacetat im Mischungsverhältnis von 2:1 als Eluiermittel gereinigt. Die gewünschte Verbindung (3-1,3-4) wird auf diese Weise in Form eines Schaumes in einer Ausbeute von 38 mg erhalten.
In der gleichen Weise wie in den vorangehenden Beispielen lassen sich verschiedene Verbindungen der Formel Ia herstellen. Diese Verbindungen sind in den Tabellen II und III aufgezählt.

Die Reaktion endet spontan unter Bildung von Hydroxyalkylidenverbindungen (4-1 und 4-2) mit einem trans/cis-Verhältnis von 1:3. Nach Zugabe von 5 ml Ethylacetat wird das Reaktionsgemisch mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Durch Eindampfen des Lösungsmittels erhält man die gewünschten V erbindungen .Ausbeute: 32 mg.

104 mg Aluminiumchlorid werden in einem Gemisch von 2 ml Anisol und 0,2 ml Methylenchlorid gelöst. Diese Lösung wird dann mit 100 mg des als Ausgangsmaterial verwendeten Esters (3-1,3-4) bei-40°C versetzt, und das Gemisch wird während 1 h bei dieser Temperatur gerührt. Dann wird das Reaktionsgemisch bei-40 °C mit 10 ml eines 0,01 molaren Phosphatpuffers, enthaltend 295 mg Natriumbicarbonat, versetzt. Nach der Zugabe von 20 ml Methylenchlorid wird das Gemisch während 30 min bei 0 °C gerührt, filtriert und abgetrennt, wobei man eine wässrige Phase erhält. Diese wässrige Phase wird mittels HP-20 gereinigt und lyophilisiert, wobei man die gewünschte Verbindung in Form eines weissen Pulvers erhält. Ausbeute: 35 mg.
Verschiedene Verbindungen der Formel Ia, die man in der gleichen Weise wie gemäss Beispiel 5 erhält, finden sich in der Tabelle V.

120 mg der Ausgangsverbindung (2-2) werden in 50 ml rührt das Gemisch während einer Stunde. Nach Beendigung der
Ethylacetat gelöst und bei Zimmertemperatur in Gegenwart von Umsetzung wird das Gemisch mit 20 ml Ethylacetat versetzt und
100 mg eines 10%igen Palladium-auf-Kohle-Katalysators im Wasserstoffstrom bei Zimmertemperatur einer katalytischen Reduktion unterworfen. In einstündigen Intervallen werden jeweils weitere 100 mg Palladium-auf-Kohle-Katalysator hinzugegeben und die Umsetzung während 6 h weitergeffihrt. Nach Beendigung der Umsetzung wird das Lösungsmittel im Vakuum verdampft, wobei man einen Rückstand erhält, der als als Zwischenprodukt dienende Carbonsäure anzusehen ist. Die so erhaltene Carbonsäure wird in 5 ml DMF gelöst. Zu dieser Lösung gibt man alsdann 40 ul Triethylamin und 108 |xl Pivaloyl-oxymethyliodid (ICH20C02t-ICH20C0-2at-Bu) hinzu und

filtriert. Das Filtrat wird mit Wasser gewaschen, über Magne-15 siumsulfat getrocknet und im Vakuum eingedampft, um das Lösungsmittel zu entfernen. Der Rückstand wird durch Chromatographie über Kieselgel unter Verwendung einer Mischung von Benzol und Ethylacetat im Mischungsverhältnis von 2:1 als Eluiermittel gereinigt, wobei man die in der obigen Überschrift 20 erhaltene Verbindung erhält. Die Ausbeute beträgt 70 mg.

Claims (6)
- 661 730PATENTANSPRÜCHE 1. In 2-Stellung substituierte oder unsubstituierte 6-(-2-Oxo-1,3-dioxolan-4-yl)-2-penem-3-carbonsäurederivate der Formel Ia und in 2-Stellung substituierte oder unsubstituierte 6-(l-Hydroxy-2-alkyliden)-2-penem-3-carbonsäurederivate der Formel Ib h2°hccx)r(Ia)(Ib)coor worin R das Wasserstoffatom, ein Leichtmetall oder eine Carbo-xyschutzgruppe, R1 eine Alkylgruppe, R2 das Wasserstoffatom, die Methylgruppe oder eine substituierte Methylgruppe bedeuten.
- 2. Verbindungen nach Anspruch 1, worin R2 eine substituierte Methylgruppe darstellt.
- 3. Verbindungen nach einem der vorangehenden Ansprüche, worin R das Wasserstoffatom, Natrium, Kalium, Diphenyl-methyl, p-Methoxybenzyl, Acetoxymethyl, l-(Ethoxycarbonyl-oxy)-ethyl oder Pivaloyloxymethyl bedeuten.

Verfahren zur Herstellung einer der Verbindungen der Formel Ia gemäss einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass man eine Verbindung der folgenden Formel0=Ocr1coor worin X eine abspaltbare Gruppe darstellt, die aus der Gruppe entnommen ist, die aus Halogenen, Sulfonyloxy- und Alkanoyl-oxygruppen besteht, und R und R1 die obigen Bedeutungen haben, mit einer Verbindung der folgenden FormelHS-Het worin Het eine heterocyclische Gruppe darstellt, oder einem reaktionsfähigen Derivat davon umsetzt.
- 6. Verfahren zur Herstellung von Verbindungen der Formel Ib gemäss einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass man eine Verbindung der Formel Ia mit einer katalytischen Menge l,5-Diazabicylo[5,4,0]undec-5-en oderNatriumbicarbo-nat in einem geeigneten Lösungsmittel behandelt.
- 7. Antibakterielles Präparat, dadurch gekennzeichnet, dass es eine wirksame Menge einer Verbindung gemäss einem der Ansprüche 1 bis 3 und überdies mindestens ein übliches Trägermittel enthält.10
