Description
Die Erfindung betrifft ein neues Verfahren zur Herstellung von bekannten Prostaglandinzwischenprodukten der allgemeinen Formel I
worin n, R2 und R4 die oben angegebene Bedeutung haben, A ein Alkalikation und Rs eine Alkylgruppe bedeutet, umsetzt.
Als Alkylgruppen Ri, R2, R3, Rtund Rs kommen vozugs-weise gerad- und verzweigtkettige Alkylreste mit 1 -5 Kohlenstoffatomen in Betracht, wie beispielsweise der Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, tert.-Butyl-und Pentylrest. Bevorzugt ist die Methyl- und Äthylgruppe.
Die Hydroxygruppe Ri kann geschützt sein, beispielsweise durch Verätherung oder Veresterung.
Als Äther- und Acylreste kommen die dem Fachmann bekannten Reste in Betracht. Bevorzugt sind leicht abspaltbare Ätherreste, wie beispielsweise der Tetrahydropyranyl-, Tetrahydrofuranyl-, a-Äthoxyäthyl-, Trimethylsilyl-, Dimethyl-, tert.-butyl-silyl- und Tri-p-benzyl-silylrest. Als Acylreste seien beispielsweise Acetyl, Propionyl, Butyryl, p-Phenyl-benzoyl und Benzoyl genannt.
Als Alkalikationen A kommen insbesondere Li+ und Na+ in Betracht, vorzugsweise das Lithiumkation.
Als Halogenatome R2 und R3 kommen insbesondere Fluor und Chloratome in Betracht, wobei Fluoratome bevorzugt sind.
Bedeutet R.4 einen Arylrest, so kann dieser bevorzugt durch Halogen, Trifluormethyl, Hydroxyl, Aryl, Alkyl und/oder Alkoxyl mit je 1-4 C-Atomen l-3fach substituiert sein. Als bevorzugte Substituenten des Restes R4 seien Halogen, insbesondere Chlor und/oder Alkoxyl oder Alkyl mit 1-3 C-Atomen, insbesondere Methoxyl oder Methyl, und die Tri-fluormethylgruppe genannt. Besonders bevorzugt sind diejenigen Verbindungen, die im Benzolkern unsubstituiert oder einfach mit Chlor, Fluor oder Trifluormethyl substituiert sind.
Es ist bekannt, dass Oxa-Prostaglandine sehr gut pharmakologisch wirksame Verbindungen darstellen.
Beispielsweise ist 16-Aryloxy-Prostaglandin-F2a unter dem Namen Equimate® im Handel als hoch wirksames Luteoly-tikum für Tiere.
Die Synthese von Oxa-Prostaglandinen wird in den deutschen Offenlegungsschriften 22 23 365,23 22 673 und 26 06 051 beschrieben. Daraus ist ersichtlich, dass Phenoxypropyl-phosphonatlösungen bei Temperaturen von —78°C mit einer Butyllithiumlösung umgesetzt werden bzw. unter Eiskühlung mit einer NaH-Suspension. Anschliessend wird in dieses Gemisch der entsprechende Aldehyd eingetragen. Nach der Aufarbeitung erhält man das gewünschte Keton in Ausbeuten von 10-40%. Selbst bei sorgfältigster Reaktionsdurchführung ist es nicht möglich, reproduzierbare Ausbeuten zu erhalten, da bei in situ durchgeführter Reaktion unerwünschte und unkontrollierbare Nebenreaktionen ablaufen, wie beispielsweise die Abspaltung der Schutzgruppe in Ri bei geringem Alkaliüberschuss.
Es war daher die Aufgabe der vorliegenden Erfindung, ein Verfahren zur Herstellung von Oxa-prostaglandinzwischen-produkten zu entwickeln, das bei Raumtemperatur durchgeführt werden kann und in sehr guten reproduzierbaren Ausbeuten abläuft, wodurch auch eine technische Verwertbarkeit gewährleistet ist.
Es wurde nun gefunden, dass nach dem beanspruchten Verfahren die gewünschten Zwischenprodukte auch bei Raumtemperatur in reproduzierbaren Ausbeuten von 87% erhalten werden können. Nach dem erfindungsgemässen Verfahren wird das Anion des Phosphonats in der Regel nicht in situ erzeugt, sondern es wird bevorzugt als kristalline Substanz in Form seines Alkalisalzes isoliert und anschliessend mit den äquimolaren Mengen des Aldehyds umgesetzt.
Die Umsetzung des Aldehyds mit dem Alkali-Phosphonat erfolgt zweckmässigerweise in Gegenwart einer Inertgasatmosphäre, wie z.B. Stickstoff und Argon, bei Temperaturen zwischen -70°C bis 80°C, vorzugsweise bei Raumtemperatur, in einem inerten Lösungsmittel, wie beispielsweise Äther, Methylenchlorid, THF, Glyme, Dioxan, Chloroform, Hexan, Benzol u.a.
Die Darstellung des Alkalisalzes kann nach den dem Fachmann bekannten Methoden, wie beispielsweise durch Umsetzung äquimolarer Mengen von Phosphonat und Butyllithium

unter Inertgasatmosphäre, erfolgen. Die Reaktion erfolgt insbesondere zwischen -70°C bis 80°C, vorzugsweise bei Raumtemperatur in inerten Lösungsmitteln, wie beispielsweise Pentan, Hexan, Benzol, Toluol, Äther, Dioxan u.a.
Beispiel
(lS,5R,6R,7R)-6-[(E)-3-Oxo-4-phenoxy-l-butenyl]-7-ben-zoyloxy-2-oxabicyclo[3.3.0]-octan-3-on
Zu einer Lösung von 548 mg (lS,5R,6R,7R)-6-Formyl-7-benzoyloxy-2-oxabicyclo[3.3.0]-octan-3-on (J. Amer. Chem. Soc. 96,5865 [1974]) in 20 ml absolutem Tetrahydrofuran fügt man 581 mg des Lithiumsalzes von Dimethyl-2-oxo-3-pheno-xypropylphosphonat und rührt 1,5 Stunden bei Raumtemperatur unter Argon. Anschliessend setzt man 0,3 ml Eisessig zu, engt im Vakuum ein, versetzt mit 100 ml Methylenchlorid, schüttelt mit 10 ml 5%iger Natriumbicarbonatlösung und zweimal mit je 10 ml Wasser. Man trocknet mit Magnesiumsulfat, dampft im Vakuum ein und kristallisiert den Eindampfrückstand aus Methylenchlorid-Isopropyläther um. Dabei erhält man 710 mg der Titelverbindung als farblose Kristalle (Ausbeute 87,5% der Theorie).
Schmelzpunkt: 132°C
Das Lithiumsalz von Dimethyl-2-oxo-3-phenoxypropyl-phosphonat wurde wie folgt hergestellt:
Zu einer Lösung von 16,3 g Dimethyl-2-oxo-3-phenoxy-propyl-phosphonat in 430 ml abs. Äther tropft man bei 5°C unter einer Argonatmosphäre eine Mischung von 52,6 ml 1.2 molare Butyllithiumlösung in Hexan und 150 ml Hexan, wobei ein weisser Niederschlag ausfiel, anschliessend rührte man 10 Minuten.
Nach dem Absaugen erhielt man 14,1 g des Lithiumsalzes als farbloses kristallines Pulver.
Schmelzpunkt: 184°C
Beispiel 2
( 1 S,5R,6R)-6-[(E)-3-Oxo-4-phenoxy-1 -butenyl]-2-oxabi-cyclo-[3.3.0]-octan-3-on
Zu einer Lösung von 7,06 g (lS,5R,6R)-6-Formyl-2-oxa-bicyclo-[3.3.0]-octan-3-on (E.J. Corey et al., J. Org. Chem. 39, 256 [1974]) in 450 ml abs. Tetrahydrofuran fügt man 13,3 g des Lithiumsalzes von
Dimethyl-2-oxo-3-phenoxypropylphosphonat (Herstellung siehe Beispiel 1) und rührt 2 Stunden bei Raumtemperatur unter Argon. Anschliessend neutralisiert man mit Eisessig, engt im Vakuum ein, versetzt mit 300 ml Methylenchlorid, schüttelt mit 30 ml 5%iger Natriumbicarbonatlösung,
zweimal mit je 30 ml Wasser, trocknet mit Magnesiumsulfat und dampft im Vakuum ein. Nach Umkristallisation des Rückstands aus Isopropanol erhielt man 11,0 g (84,5% d. Th.) der Titelverbindung als farblose Kristalle.
Schmelzpunkt: 91°C
