Die Erfindung betrifft ein Verfahren zur Herstellung von bicycüschen Laktonen der allgemeinen Formel

Als Alkylgruppen Ri, R2, R3 und R5 mit 1—5 Kohlenstoffatomen kommen gerad- und verzweigtkettige
Alkylreste in Betracht, wie beispielsweise der Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-,
tert.-Butyl- und Pentylrest. Bevorzugt sind die Methyl-
und Äthylgruppe.
Als Äther- und Esterreste Ri kommen die dem Fachmann bekannten Reste in Betracht. Bevorzugt sind
leicht abspaltbare Ätherreste, wie beispielsweise der Tetrahydropyranyl-, Tetrahydrofuranyl-, a-Äthoxyäthyl-,
Trimethylsilyl-, Dimethyl-, tert.-Butyl-silyl- und
Tri-Benzyl-silylrest. Als Esterreste seien beispielsweise der Acetyl-, Propionyl-, Butyryl-, p-Phenyl-benzoyl- und
Benzoylrest genannt.
Die Verfahrensprodukte stellen Zwischenprodukte für die Herstellung von Prostaglandinen dar. Es ist
bekannt, daß Oxa-Prostaglandine pharmakologisch sehr
wirksame Verbindungen darstellen.
Beispielsweise ist ein 16-Aryloxy-Prostaglandin-F2« als hoch wirksames Luteolytikum für Tiere im Handel.
Aus J. Amer. Chem. Soc. 91 (1969), 5675, Tetrahedron
Letters 49 (1971), 4753 und DE-OS 2261 496 ist die Umsetzung bicyclischer Aldehyde mit 2-Oxoalkyl-phosphonaten
bekannt. Während dieses Corey'sche Verfahren mit 2-Oxoalkyl-phosphonaten in Form ihrer in situ
erzeugten Alkaliderivate als Ausgangsverbindungen gute Ausbeuten an Prostaglandinzwischenprodukten
ergibt, sinken die Ausbeuten an Prostaglandinzwischenprodukt auf 15%, wenn man an Stelle der oben
beschriebenen Phosphonate 2-Oxo-oxaalkyl-phosphonate
(als Na- oder Li-Salze) umsetzt.
Die Synthese von Oxa-Prostaglandinen wird in den DE-OS 22 23 365, 23 22 673 und 26 06 051 beschrieben.
Aus der DE-OS 22 23 365 ist es bekannt, Phenoxypropylphosphonatlösungen bei Temperaturen von -78° C
mit einer Butyllithiumlösung umzusetzen. Anschließend wird in dieses Gemisch der entsprechende Aldehyd
eingetragen. Nach der Aufarbeitung erhält man das gewünschte Keton in Ausbeuten von 10—40%. Selbst
bei sorgfältigster Reaktionsdurchführung ist es nicht möglich, reproduzierbare Ausbeuten zu erhalten, da bei
in situ durchgeführter Reaktion unerwünschte und unkontrollierbare Nebenreaktionen ablaufen, wie beispielsweise
die Abspaltung der Schutzgruppe in Ri bei geringem Alkaliüberschuß.
Es war daher die Aufgabe der Erfindung, ein Verfahren zur Herstellung der als Oxa-prostaglandinzwischenprodukte
geeigneten betreffenden bicyclischen Lactone zu entwickeln, das bei Raumtemperatur
durchgeführt werden kann und in sehr guten reproduzierbaren Ausbeuten abläuft, so daß auch eine
technische Verwertbarkeit gewährleistet ist
Das erfindungsgemäße Verfahren ermöglicht nun die Herstellung der betreff enden bicyclischen Lactone auch
bei Raumtemperatur in erheblich höheren und reproduzierbaren Ausbeuten von bis zu 87% als bei den
bekannten Verfahren. Bei dem erfindungsgemäßen Verfahren wird das Anion des Phosphonats nicht in situ
erzeugt, sondern es wird als kristalline Substanz in Form seines Lithiumsalzes eingesetzt und mit den äquimolaren
Mengen des betreffenden Aldehyds umgesetzt.
Es war überraschend und nicht voraussehbar, daß hierdurch gegenüber den bekannten Arbeitsweisen eine
wesentliche Verbesserung in der Herstellung der betreffenden bicyclischen Lactone erzielt werden
könnte.
Die Umsetzung des betreffenden Aldehyds mit dem betreffenden Lithiumphosphonat erfolgt zweckmäßigerweise
wie üblich in Gegenwart einer Inertgasatmosphäre, wie Stickstoff und Argon, bei Temperaturen
zwischen —70° bis 80° C, vorzugsweise bei Raumtemperatur, in einem inerten Lösungsmittel, wie beispielsweise
Äther, Methylenchlorid, THF, Glyme, Dioxan, Chloroform, Hexan, Benzol.
Die Darstellung des Lithiumsalzes erfolgt nach den dem Fachmann bekannten Methoden, beispielsweise
durch Umsetzung äquimolarer Mengen etwa entsprechenden Phosphonats mit Butyllithium unter Inertgasatmosphäre.
Die Reaktion erfolgt zwischen -70^C bis 8O0C, vorzugsweise bei Raumtemperatur in inerten
Lösungsmitteln, wie beispielsweise Pentan, Hexan, Benzol, Toluol, Äther, Dioxan.

Zu einer Lösung von 548 mg (lS,5R,6R,7R)-6-Formyl-7-benzoyloxy-2-oxabicyclo[3.3.0]-octan-3-on
(Journal American Chemical Society Bd. 96, S. 5865 [1974]) in 20 ml absolutem Tetrahydrofuran fügt man 581 mg des
Lithiumsalzes von Dimethyl^-oxo-S-phenoxypropylphosphonat
und rührt 1,5 Stunden bei Raumtemperatur unter Argon. Anschließend setzt man 03 ml Eisessig zu,
engt im Vakuum ein, versetzt mit 100 ml Methylenchlorid,
schüttelt nacheinander mit 10 ml 5%iger Natriumbicarbonatlösung und zweimal mit je 10 ml Wasser aus.
Man trocknet die organische Lösung mit Magnesiumsulfat, dampft im Vakuum ein und löst den Rückstand in
Methylenchlorid, versetzt mit etwa der lOfachen Menge
Isopropyläther und dampft überschüssiges Methylenchlorid auf dem Dampfbad ab, bis die Substanz
kristallisiert Man erhält 710 mg der Titelverbindung als
is farblose Kristalle vom R 132°C (Ausbeute 87,5% der
Theorie).
Das oben verwendete Lithiumsalz von Dimethyl-2-oxo-3-phenoxypropylphosphonat
ist wie folgt hergestellt worden:

in 430 ml abs. Äther tropft man bei 5° C unter einer Argonatmosphäre eine Mischung

anschließend rührte man 10 Minuten.
Nach dem Absaugen erhielt man 14,1 g des Lithiumsalzes als farbloses kristallines Pulver.
Schmelzpunkt: 184° C.

Zu einer Lösung von 7,06 g (lS,5R,6S)-6-Formyl-2-oxa-bicyclo-[3.3.0]-octan-3-on
(E. J. Corey und Mitarbeiter, Journal of Organic Chemistry, Bd. 39, S. 256 [1974])
in 450 ml abs. Tetrahydrofuran fügt man 13,3 g des
4(i Lithiumsalzes von Dimethyl^-oxo-S-phenoxyoropylphosphonat
und rührt 2 Stunden bei Raumtemperatur unter Argon. Anschließend neutralisiert man mit
Eisessig, engt im Vakuum ein, versetzt den Rückstand mit 300 ml Methylenchlorid, schüttelt nacheinander mit
v> 30 ml 5%iger Natriumbicarbonatlösung, zweimal mit je 30 ml Wasser aus, trocknet die organische Lösung mit
Magnesiumsulfat und dampft im Vakuum ein. Nach Umkristallisation des Rückstands aus Isopropanol
erhielt man 11,0 g (84,5% d. Th.) der Titelverbindung als
><> farblose Kristalle vom F. 9i0C.
Claims (1)
- Patentanspruch:Verfahren zur Herstellung von bicyclischen Laktonen der allgemeiner FormelR.3 ein Wasserstoffatom oder einen Alkylrest mit 1 bis 5 C-Atomen, R4 einen ggf. durch ein Chlor- oder Fluoratom oder die Trifluormethylgruppe substituierten Arylrest bedeuten und π die Zahl 0,1,2 oder 3 darstellt, das dadurch gekennzeichnet ist, daß man einen Aldehyd der allgemeinen Formel
