Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Alkyl-(7R,S)-7-deoxy-7-(co-substituiert-alkylthio)-a-thiolincosaminiden.

3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man als wasserfreie niedere Kohienwasserstoffcarbonsäure Eisessig verwendet.
4. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man als Ausgangsmaterial Methyl-(6R,7R)-N-acetyI-6,7-aziri-dino-6-deamino-7-deoxv-ce-thiolincosaminid und als Thiol 2-

Beispiele des Alkylrestes mit nicht mehr als 4 Kohlenstoffatomen sind der Methyl-, Ethyl-, Propyl-, Isopropyl-, Butyl-, sek.-Butyl-, Isobutyl- oder tert.-Butylrest.
Die Wellenlinie, die die Substituenten an das Kohlenstoffatom in Stellung 7 in den Formeln I, IVb, V und VII bindet, zeigt an, dass diese Verbindungen in beiden epimeren Formen (7R) und (7S) vorliegen.
Die Wellenlinie, welche die Methylgruppe und das Wasserstoffatom in den Formeln IIb und VI an das Kohlenstoffatom 7 bindet, zeigt an, dass diese Verbindungen in den beiden epimeren Formen (6R. 7R) und (6R, 7S) vorliegen.
Unter einem Carboxyacylrest werden in der Beschreibung und den Ansprüchen Acvlreste einer Kohienwasserstoffcarbonsäure oder einer durch einen inerten Rest substituierten Kohienwasserstoffcarbonsäure verstanden. Bevorzugt als Carboxacyl-reste sind die Acvlreste von Kohlenwasserstoffcarbonsäuren und

Beispiele für Acylreste von Kohlenwasserstoffcarbonsäuren sind die Acylreste (a) gesättigter und ungesättigter, geradkettiger oder verzweigter aliphatischer Carbonsäuren wie z. B. der Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, tert-Butylessigsäure, Valeriansäure, Isovaleriansäure, Capron-säure, Caprylsäure, Decansäure, Dodecansäure, Laurinsäure, Tridecansäure, Myristinsäure, Pentadecansäure, Palmitinsäure, Margarinsäure, Stearinsäure, Acrylsäure. Crotonsäure, Undecy-lensäure, Ölsäure, Hexynsäure, Heptynsäure, Octynsäure und dergleichen, (b) gesättigter oder ungesättigter alicyclischer Carbonsäuren wie z. B. derCyclobutancarbonsäure, Cyclopentan-carbonsäure, Cyclopentencarbonsäure, Methylcyclopentencar-bonsäure, Cyclohexancarbonsäure, Dimethylcyclohexencarbon-säure, Dipropylcyclohexancarbonsäure und dergleichen, (c) gesättigter oder ungesättigter alicyclischer aliphatischer Carbonsäuren wie z.B. der Cyclopentanessigsäure. Cyclopentanpro-pionsäure, Cyclohexanbuttersäure,Methylcyclohexanessigsäure und dergleichen, (d) aromatischer Carbonsäuren wie z.B. der Benzoesäure, Tolylsäure, Naphthoesäure, Äthylbenzoesäure, Isobutylbenzoesäure, Methylbutylbenzoesäure und dergleichen und (e) aromatisch-aliphatischer Carbonsäuren wie z. Unter durch einen inerten Rest substituierten Kohlenwasserstoffcarbonsäuren werden beispielsweise Säuren verstanden, in welchen ein oder mehrere direkt an ein Kohlenstoffatom gebundene Wasserstoff atome durch einen Rest ersetzt sind, welcher unter den nachstehend beschriebenen Reaktionsbedingungen zur Herstellung der erfindungsgemäss herstellbaren Verbindungen der Formel I inert ist. Beispiele für derartige Substituenten sind die Halogene, die Nitro-, Hydroxy-, Amino-, Cyan-, Thio-cyan- oder Alkoxygruppen. «ar» im Thiolnamen in Tabelle I bedeutet, dass sich der Substituent am aromatischen Ring befindet.
Als wasserfreie niedere Kohlenwasserstoffcarbonsäuren werden zum Beispiel trockene niedere Alkancarbonsäuren mit 2 bis 8 Kohlenstoffatomen, wie beispielsweise Essigsäure, Propionsäure und Octansäure, und aromatische Säuren mit 7 bis 8 Kohlenstoffatomen, wiez. B. Benzoesäure und Tolylsäure, verwendet. Die bevorzugte wasserfreie Kohienwasserstoffcarbonsäure ist der Eisessig.
Die als Ausgangsmaterial verwendeten Aziridinverbindun-gen der Formel IIb sind bekannt, vergleiche beispielsweise die US-PS 3671647 und 3702322. Sie können erhalten werden, wenn man eine Verbindung der Formel IV

Die Dehydrohalogenierung erfolgt, indem man beispielsweise eine Verbindung der Formel VII in einem inerten Lösungsmittel in Gegenwart eines Säureakzeptors erhitzt. Beispielsweise kann man ein Reaktionsgemisch aus Ausgangsverbindung, wasserfreiem Natriumcarbonat und Dimethylformamid kurz auf Rückflusstemperatur erhitzen, das Lösungsmittel entfernen und das Produkt aus einem geeigneten Lösungsmittel, wiez. Durch Acylieren von Verbindungen der Formel I mit einer L-2-PyrroIidincarbonsäure können Verbindungen der Formel V erhalten werden, in denen Ac den Acylrest der L-2-Pyrrolidin-carbonsäure bedeutet. Stellen beispielsweise Alk den Methylrest, n die Zahl 0, R, R| und R2 Wasserstoff dar und ist die L-2-Pyrrolidincarbonsäure die trans-l-Methyl-4-propyl-L-2-pyrroli-dincarbonsäure, wobei die Konfiguration (S) ist, so erhält man die Verbindung (7S)-7-Deoxy-7-(2-hydroxyethylthio)linco-mycin.

Mit sämtlichen Thiolen gemäss Tabelle I erzielt man die gewünschte Thiolyse durch einfaches Erhitzen des Alkyl-N-
acetyl-6,7-aziridino-6-deamino-7-deoxy-a-thiolincosaminidsmit dem betreffenden Thiol in beispielsweise Eisessig oder einer anderen wasserfreien niederen Kohienwasserstoffcarbonsäure. Die Reaktion verläuft im allgemeinen befriedigend innerhalb eines breiten Temperaturbereichs, das heisst zwischen etwa 25 und etwa 180°C. Vorzugsweise wird das Verfahren im Temperaturbereich von etwa 60 bis etwa 150° C und insbesondere von etwa 80 bis etwa 110° C durchgeführt. Die Mengen der Reaktionsteilnehmer IIb und III sind für den Reaktionsverlauf gewöhnlich nicht kritisch, beeinflussen jedoch die Ausbeute an S Produktverbindungen I. Man kann stöchiometrische Mengen verwenden, d. h. im wesentlichen äquimolare Mengen. Beste Ausbeuten werden normalerweise erzielt, wenn man das Thiol III im Überschuss anwendet, d. h. in einem molaren Überschuss und vorzugsweise in Mengen von mindestens 2 Moläquivalenten, 10 insbesondere von etwa 5 bis etwa 60 Moläquivalenten .Die Reaktion wird durch Säure katalysiert. Sie erfolgt daher unter sauren Bedingungen, wie man sie mit einer wasserfreien niederen Kohienwasserstoffcarbonsäure erzielt, z. B. mit Eisessig. Die wasserfreie niedere Kohienwasserstoffcarbonsäure wird zweck-15 mässig in einer molaren Menge von 1 bis 7 Mol, bezogen auf das Acylaziridin, eingesetzt.
Zweckmässig wird die Umsetzung in Gegenwart eines geeigneten organischen Lösungsmittels durchgeführt. Ein für diesen Zweck geeignetes Lösungsmittel muss mindestens das Thiolinco-20 saminid IIb teilweise lösen und darf den gewünschten Reaktionsverlauf nicht nachteilig beeinflussen. Beispiele für geeignete organische Lösungsmittel sind Dioxan, Tetrachlorkohlenstoff, Chloroform, Methylenchlorid, Benzol, Toluol, n-Hexan und ähnliche organische Lösungsmittel. Als organisches Lösungsmit-25 tel bevorzugt wird ein Überschuss der Thiolverbindung III, das heisst eine Menge über der für obige Reaktion benötigten Menge, unter der Voraussetzung, dass die Thiolverbindung III die obigen Kriterien hinsichtlich der Lösung des Reaktionsteilnehmers IIb bei der zur Durchführung des Verfahrens gewählten 30 Temperatur befolgt.
Im allgemeinen ist obige Umsetzung nach etwa 1 bis etwa 20 h beendet, je nach den Resten Aci, R, R,, R2 und R3 in den Formeln IIb und III. Die Beendigung der Umsetzung kann durch konventionelle analytische Verfahren festgestellt werden, bei-35 spielsweise durch Dampfphasenchromatographie, Dünnschichtenchromatographie und ähnliche Verfahren, die das Verschwinden der Ausgangsverbindung IIb und das Auftreten der gewünschten Produktverbindung IVb anzeigen.
Nach beendeter Thiolyse kann überschüssiges Lösungsmittel 40 und Reagens III aus dem Reaktionsgemisch entfernt werden, und ohne Abtrennung des Zwischenprodukts IVb wird das Reaktionsgemisch dann beispielsweise mit einem Acylhalogenid oder Säureanhydrid einer Kohienwasserstoffcarbonsäure mit 2 bis 12 Kohlenstoffatomen wie z.B. Acetanhydrid in bekannter 45 Weise acyliert, wobei man das betreffende Alkyl-N-acyl-2,3,4-tri-0-acyl-7-deoxy-7-(co-acyloxyalkylthio)-a-thiolincosaminid, falls R Wasserstoff ist, das Alkyl-N-acyl-2,3,4-tri-0-acyl-7-deoxy-7-(co-alkoxy-alkylthio)-a-thiolincosaminid, falls R Alkyl ist, oder Alkyl-N-acyl-2,3,4-tri-0-acyI-7-deoxy-7-((ü-aralkoxyal-50 kyIthio)-a-thiolincosaminid, falls R Alkyl bedeutet, erhält.
Nach beendeter Umsetzung kann das Reaktionsgemisch bekannten Verfahren unterworfen werden wie z. 5,0 g (1 Moläquivalent) Methyl-(6R,7R)-N-acetyl-6,7-aziri-dino-6-deamino-7-deoxy-a-thiolincosaminid werden in 50 g (35 Moläquivalente) 2-Hydroxyäthanthiol unter Rühren in einem Ölbad von 100°C gelöst. Dann werden 7,56 g (7,2 ml, 7 Moläquivalente) Eisessig zugegeben und das Gemisch wird unter Ausschluss von Feuchtigkeit 17 h auf 100° C erhitzt. Danach werden flüchtige Anteile aus der Reaktionslösung durch Destillation im Hochvakuum (ca. 0,2 mm) aus einem Ölbad von 100° C entfernt. Dabei erhält man einen sirupösen Rückstand, der im Dünn-schichtenchromatogramm (Silikagel, Methanol/Chloroform 1:5) eine Hauptzone mit Rf = 0,28 zeigt. Beim Chromatographieren an Silikagel in Methanol/Methylenchlorid (Volumenverhältnis 1:8, Säulenabmessung 5,8x 99 cm, 1200 g Kieselsäure) erhält man das gewünschte Material in den Fraktionen 152 bis 255 ( Auffangen von 50 ml-Fraktionen nach einem Vorlauf von 1450

ml) nach Entfernen des Lösungsmittels als nicht-kristallinen Feststoff in einer Menge von 2,98 g. Das erneute Chromatographieren zum Entfernen von Spurenvereinigungen erfolgt mit einem Hochleistungs-Flüssigkeitschromatogramm (241 g Silikagel, Säulenabmessungen 2,8 x 104 mm) in Methanol/Methylenchlorid (Volumenverhältnis 1:8) bei einer Fliessgeschwindigkeit von 3,5 ml/min und einem Druck von 0,84 kg/cm2. Es werden 50 ml-Fraktionen aufgefangen und das gewünschte Rohmaterial findet sich in den Fraktionen 21 bis 27. Es wird nach Entfernung des Lösungsmittels als farbloser amorpher Feststoff isoliert.

Der gemäss Beispiel 3, Teil A-3 erhaltene Feststoff wird acetyliert. indem man ihn in 10 ml Pyridin löst und 5 ml Acetanhvdrid zusetzt, worauf die Lösung unter Ausschluss von Feuchtigkeit 48 h bei Raumtemperatur stehengelassen wird. Flüchtige Anteile werden im Vakuum entfernt, der Rückstand wird in Methylenchlorid gelöst, die Lösung wird mit verdünnter wässriger Salzsäure, Wasser, gesättigter Natriumbicarbonatlö-sung und Wasser gewaschen und über wasserfreiem Natriumsulfat getrocknet. Nach Entfernen des Lösungsmittels im Vakuum erhält man einen farblosen Feststoff, der im Dünnschichtenchro-matogramm ( Silikagel, Aceton/Skellysolve B 1:1 ) eine einzige Zone vom Rf = 0,44 zeigt.

tat/Cyclohexan (Volumenverhältnis 1:1:1:3) einer Gegenstrom-verteilungunterworfen, wobei man ein Hauptprodukt bei einem K-Wert von 0,48 erhält. Durch Vereinigen des Materials aus den Röhrchen 130 bis 199 nach 500 Übergängen und Entfernung des Lösungsmittels im Vakuum werden 1,60 g ( 16 % Ausbeute) Methyl-(7S)-N-acetyl-2,3,4-tri-ö-acetyl-7-deoxy-7-(2-acetoxy-äthylthio)-a-thiolincosaminid als farbloser Feststoff erhalten. Daraus erhält man beim Umkristallisieren aus Äthylacetat/ Skellysolve B Nadeln vom F. 206—207°C. Der Schmelzpunkt sinkt nicht nach Vermischen mit Material aus2-Acetoxyäthyl-methylsulfid(U5-PS3915954 Beispiel 10,TeilA-10B)und Äthylensulfid (US-PS 3 767 649. Beispiel 1, Teil A-1 ), das mit dieser Verbindung im IR- und NMR-Spektrum identisch ist.

MethyI-(7S)-7-deoxy-7-(2-hydroxyäthylthio)-a-thiolincos-aminid kann aus Methyl-(7S)-N-acetyl-2,3,4-tri-0-acetyl-7-(2-acetoxyäthylthio)-a-thiolincosaminid nach dem Verfahren der US-PS 3767649, Beispiel 1, Teil B-l hergestellt werden.

- 632 5192PATENTANSPRÜCHE worin R. R|, R2, R3 und n die vorstehend angegebene Bedeutung besitzen,1. Verfahren zur Herstellung einer Verbindung der Formel in Gegenwart einer wasserfreien niederen Kohlenwasserstoffcar-Q |-| bonsäure erhitzt unter Bildung einer Verbindung der Formel I VbNH;R1R2 R3I I f eS-C-C-(C)n-0-RIIIH H H(I)S-Al k worin Alk einenAlkylrest mit nicht mehr als 4 Kohlenstoffatomen,Ri Wasserstoff, Methyl oder Ethyl,R2 Wasserstoff, Methyl oder Ethyl,R3 Wasserstoff, Methyl oder Ethyl,n die Zahl 0 oder 1,R Wasserstoff, einen Alkylrest mit 1 bis 6 Kohlenstoffatomen oder einen Arylrest mit 7 bis 12 Kohlenstoffatomen darstellen, und die Wellenlinie, welche die Substituenten an das Kohlenstoffatom in Stellung 7 bindet, anzeigt, dass die Verbindungen in beiden epimeren Formen (7R) und (7S) vorliegen,dadurch gekennzeichnet, dass man eine Verbindung der Formel IIbHAcìN\CH,(IIb)RiR2 R3I I IHS-C-C-(C)n-0-R111H H H(ili)303540S-Al k worin Alk die vorstehend angegebene Bedeutung besitzt, Aci einen Carboxacylrest darstellt, und die Wellenlinie, welche die Methylgruppe und das Wasserstoffatom an das Kohlenstoffatom 7 bindet, anzeigt, dass die Verbindungen in den beiden epimeren Formen (6R, 7R) und (6R, 7S) vorliegen,mit einem Thiol der Formel IIIC H11015Aci NHR1R 2 R «I I fS-C-C-(C)n-0-RII I H H H2025(Ivb)S-Al k worin R, Ri, R2, R3, Aci, Alk und n die vorstehend angegebene Bedeutung besitzen,die alkoholischen Hydroxylgruppen dieser Verbindung acyliert unter Bildung des betreffenden Alkyl-N-acyl-2,3,4-tri-0-acyl-7-deoxy-7-(a)-acyloxyalkylthio)-a-thiolincosaminids, falls R Wasserstoffist, Alkyl-N-acyl-2,3,4-tri-0-acyl-7-deoxy-7(co-alkoxyal-kylthio)-a-thiolincosaminids, falls R ein Alkylrest mit 1 bis 6 Kohlenstoffatomen ist, oder Alkyl-N-acyl-2,3,4-tri-0-acyl-7-deoxy-7-(©-aralkoxyalkylthio)-a-thiolincosaminids, falls Rein Aralkylrest mit 7 bis 12 Kohlenstoffatomen ist, und diese Verbindung deacyliert unter Bildung des gewünschten Produktes.

Verfahren nach Anspruch 1 zur Herstellung von Verbindungen der Formel IVb (S)4550CH;556065H,N-RiR2 R31 1 1■C-C-(C)n-0-RI I IH H HY\■Alk(IVb (S))worin Alk, R, R], R2, R.-sund n die in Anspruch 1 angegebene Bedeutung besitzen, dadurch gekennzeichnet, dass man eine Verbindung der Formel3632 519AcxNHydroxyethanthiol verwendet und als Endprodukt Methyl-(7S)-(2-hydroxyethylthio)-7-deoxy-a-thiolincosaminid erhält.
- 5. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass man als Ausgangsmaterial Methyl-(6R,7R)-N-acetyl-6,7-aziri-5 dino-6-deamino-7-deoxy-a-thiolincosaminid und als Thiol 2-Hydroxyethanthiol verwendet und als Endprodukt Methyl-(7S)-(2-hydroxyethylthio)-7-deoxy-a-thiolincosaminid erhält.S-Al k worin AC| und Alk die in Anspruch 1 angegebene Bedeutung besitzen, mit einem Thiol der Formel III10
