Description
Verfahren zur Herstellung neuer Phosphorsäureester Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Phosphorsäureestern der Formel I: EMI1.1 In dieser Formel bedeuten: X Sauerstoff oder Schwefel, Rt und R2 unabhängig voneinander je einen niederen Alkyl-Rest, R3 Wasserstoff, einen Alkoxycarbonyl- oder Halogenmethyl-Rest, R4 einen niederen Alkoxy-, Alkylthio- oder Dialkylamino-Rest, R5 einen niederen Alkoxy-Rest, oder R4 einen niederen Alkylthio-Rest und R5 einen Dialkylamino-Rest. In der Formel I sind unter niederen Alkylresten Rt und R2 Reste mit 1-5 Kohlenstoffatomen, wie Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl, sec.-Butyl, Isobutyl und tert.-Butyl, sowie die Pentyl-Reste zu verstehen. Die Reste Rt und R2 können untereinander gleich oder verschieden sein. Solche niederen Alkylreste stellen auch den Alkylteil von Alkoxy- und Alkylthio-Resten R4, R5 dar, sie treten ferner als Substituenten einer Dialkylaminogruppe R4, R5 auf und bilden den Alkylteil in Alkoxycarbonyl-Resten R3. Unter einem Halogenmethyl-Rest R3 ist insbesondere der Trifluormethyl Rest zu verstehen. Die Phosphorsäureester der Formel I werden erfindungsgemäss hergestellt, indem man eine Verbindung der Formel II: EMI1.2 in der Me ein Alkalimetallatom bedeutet, mit einem Phosphorsäurehalogenid der Formel II: EMI1.3 in der Hal Chlor oder Brom bedeutet, umsetzt. In den Formeln II und III haben die Symbole Rl-R5 und X die unter Formel I angegebenen Bedeutungen. Die Umsetzungen werden in einem gegenüber den Reaktionsteilnehmern inerten Lösungs- oder Verdünnungs mittel durchgeführt. Als solche haben sich aromatische Kohlenwasserstoffe und Halogenkohlenwasserstoffe, wie Benzol, Toluol, Xylole, Chlorbenzol, bewährt, ferner kommen aliphatische Kohlenwasserstoffe, Äther etc. in Betracht Die Reaktionstemperaturen liegen im allgemeinen zwischen 0 und 1000 C, vorzugsweise zwischen 20 und 800 C. Die Ausgangsstoffe der Formel II sind bekannte Verbindungen oder lassen sich nach bekannten Verfahren durch Kondensation entsprechender Ester in Gegenwart von Kondensationsmitteln herstellen. Als Esterkomponenten kommen einerseits Alkoxy-essigsäureester, Alkylthio-essigsäureester, Alkylthio-essigsäureamide, andererseits Ameisensäureester, Halogenmethyl-essigsäureester, Oxalsäureester in Betracht; als Kondensationsmittel werden die den Estern entsprechenden Alkalialkanolate und Alkalimetallhydride verwendet. Die neuen Phosphorsäureester der Formel I besitzen ausgezeichnete insektizide und gute akarizide Eigenschaften. Aufgrund ihrer guten Frassgift- und Kontaktgift-Wirkung sind sie zur Bekämpfung fressender und saugender Insekten im Pflanzen- und Vorratsschutz geeignet. Die günstige Warmblütertoxizität erlaubt ferner ihren Einsatz auf dem Veterinär- und Hygiene-Sektor. Bei den neuen Wirkstoffen der Formel I beträgt die DL50 für Ratte p.o. bis 500 mg/kg Körpergewicht; demgegenüber liegt die DL30 von bekannten und im Handel befindlichen Phosphorsäureestern, wie 1. von 0,0-Dimethyl-0-(1-methy-2-chlor-2- diäthylcarbamyl-vinyl)-phosphorsäureester [vgl. belgisches Patent Nr. 552 284, Handelsname Phosphamidon ] und 2. von 0, 0-Dimethyl-0-(1-methyl-2-methoxycarbonyl vinyl)-phosphorsäureester [vgl. amerikanisches Patent Nr. 2 685 552, Handelsname Phosdrin ] für Ratten p. o. unter 50 mg/kg Körpergewicht. Ausserdem tritt bei den erfindungsge- mässen Wirkstoffen die Wirkung schneller ein und hält länger an als bei den erstgenannten, bekannten Phosphorsäureestern Ferner wurde festgestellt, dass diese Wirkstoffe der Formel I insbesondere eine ausgezeichnete, langandauernde Wirkung gegen Insekten der Familie Muscidae, Stomoxidae und Culicidae, z. B. gegen die polyvalent-resistenten und normal sensiblen Stubenfliegen (Musca domestica), Wadenstecher (Stomoxys calcitrans) und Stechmücken (z.B. Aedes aegypti, Culex fatigans, Anopheles stephensi); gegen Insekten der Familien Curculionidae, Bruchididae, Dermestidae, Tenebrionidae und Crysomelidae, z. B. Kornkäfer (Sitophilus granaria), Speisebohnenkäfer (Bruchidius obtectus), Speckkäfer (Dermestes vulpinus), Mehlkäfer (Tenebrio molitor), Kartoffelkäfer (Leptinotarsa decemlineata) und deren Larvenstadien; gegen Insekten der Familie Pyralididae, z. B. Mehlmottenraupen (Ephestia kühniella); der Familie Blattidae, z. B. Küchenschaben (Phyllodromia germanica, Periplaneta americana, Blatta orientalis); der Familie Aphididae, z.B. Blattläuse (Aphis fabae); der Familie Pseudococcidae, z.B. Schmierläuse (Planococcus citri) und der Familie Locustidae, z. B. Wanderheuschrecken (Locusta migratoria) aufweisen Die erfindungsgemässen Wirkstoffe der Formel I besitzen ausserdem eine gute Wirkung gegen die larvalen und adulten Stadien von Spinnentieren, z.B. der Familien Acarinae, Ixodidae, Arachnidae und Argasidae. In Mischungen mit Synergisten und ähnlich wirkenden Hilfsstoffen, wie Bernsteinsäuredibutylester, Piperonylbutoxyd, Olivenöl, Erdnussöl usw. wird das Wirkungsspektrum der erfindungsgemässen Wirkstoffe verbreitert und insbesondere die insektizide Wirkung verbessert. Ebenso lässt sich die insektizide Wirkung noch durch Zusatz von anderen Insektiziden, wie z. B. Phosphorsäure-, Phosphonsäure-, Thio- und Dithiophosphorsäureestern und -amiden, Halogen-Kohlenwasserstoffen und Analogen von DDT-Wirksubstanz sowie Pyrethrinen und ihren Synergisten, akariziden Wirksubstanzen, wie Halogenbenzilsäureestem, wie 4',4'-Dichlorbenzilsäureestern, 4',4'-Dibrombenzilsäureestern, insektiziden und akariziden Carbamaten, z. B. Oximcarbamaten, Arylcarbamaten und Carbamaten von enolisierbaren Heterocyclen, wesentlich verbreitern und an gegebene Umstände anpassen. Die folgenden Beispiele veranschaulichen die Herstellung der neuen Phosphorsäureester der Formel I. Sofern nichts anderes vermerkt ist, sind die Temperaturen in Celsius-Grad angegeben. Die nachfolgende Tabelle enthält weitere Beispiele von Phosphorsäureestern der Formel I, die nach den in den Beispielen beschriebenen Methoden hergestellt wurden. Beispiel 1 Ein Gemisch aus 26,4 g Äthoxyessigsäureäthylester und 14,8 g Ameisensäureäthylester wird langsam zu einer Suspension von 13,6 g Natriumäthylat in 300 ml Benzol (wasserfrei) getropft. Das Gemisch wird 7 Stunden bei Raumtemperatur gerührt. Anschliessend werden 29 g Chlorphosphorsäuredimethylester, gelöst in 25 ml Benzol (wasserfrei), zugetropft. Das Reaktionsgemisch wird 24 Stunden bei Raumtemperatur gerührt und dann mit 200 ml Wasser ausgeschüttelt. Die organische Phase wird abgetrennt, getrocknet und vom Lösungsmittel befreit. Das Rohprodukt wird im Vakuum destilliert. Der 0,0-Dimethyl-0-(2-äthoxy2-äthoxycarbonyl-vinyl)-phosphorsäureester hat den Siedepunkt 119-124 /0,45 mm. Analyse: Ber. C 40,3 H 6,4 P 11,5 Gef. C 40,3 H 6,3 P 11,3 Beispiel 2 a) Zu 8 g Natrium, emulgiert in 150 ml Toluol (wasserfrei), tropft man 32,2 g Äthanol (wasserfrei) und rührt 1 Stunde bei 95". Dann wird die Suspension auf 0-5 gekühlt und ein Gemisch aus 52,6 g 2-Äthylthio- N,N-dimethyl-acetamid und 26,5 g Ameisensäureäthylester in 50 ml Ather (wasserfrei) langsam zugetropft. Nach 16 Stunden Rühren bei 0-5" wird das ausgeschiedene Natriumsalz abfiltriert, mit 300 ml Äther (wasserfrei) gewaschen und im Vakuum getrocknet b) Zu einer Suspension von 9,9 g dieses Natriumsalzes in 100 ml Benzol (wasserfrei) tropft man eine Lösung von 8 g Chlorthiophosphorsäuredimethylester in 25 ml Benzol (wasserfrei). Das Reaktionsgemisch wird 24 Stunden bei Raumtemperatur gerührt. Nach Behandlung mit Aktivkohle wird das Filtrat am Vakuum eingedampft. Das so erhaltene 0,0-Dimethyl-0-(2-äthylthio-2-N,N- dimethylcarbamyl-vinyl)-thiophosphat wird durch Chromatographie an einer Kieselgelsäule gereinigt und hat den Brechungsindex nD20 = 1,5243. Analyse: Ber. C 36,1 H 6,1 N 4,7 P 9,7 S 21,4 Gef. C 36,0 H 6,0 N 4,6 P 10,2 S 21,0 Beispiel 3 a) Nach der in der deutschen Patentschrift 708 513 beschriebenen Arbeitsweise werden 6,9 g Natrium in 400 ml Isopropanol gelöst und in einen mit Stickstoff gespülten Autoklaven übergeführt. Bei 70" und 10 bar wird solange Kohlenstoffmonoxid aufgepresst, bis keine Druckabnahme mehr registriert werden kann. Dann werden 39,6 g Methoxyessigsäureisopropylester in den Autoklaven eingeführt, und bei 70" wird der Kohlenstoffmonoxiddruck bei 40 bar 7 Stunden gehalten. Nach dem Abkühlen wird anschliessend abfiltriert und das Filtrat eingedampft, wobei sich das gebildete Enol-Natriumsalz abscheidet. b) Das so erhaltene Natriumsalz wird in 500 ml Benzol (wasserfrei) suspendiert und auf 0-6 in Eisbad abgekühlt. Anschliessend werden 43,35 g Chlorphosphorsäuredimethylester, gelöst in 50ml Benzol zugetropft. Das Reaktionsgemisch wird 24 Stunden bei Raumtemperatur gerührt und dann zweimal mit 150 ml Wasser ausgeschüttelt. Die organische Phase wird abgetrennt, getrocknet und vom Lösungsmittel befreit. Das Rohprodukt wird in Vakuum destilliert. Der so erhaltene 0,0-Dimethyl-0-(2-methoxy-2isopropoxycarbonyl-vinyl)-phosphorsäureester siedet bei 97 /0,01 mm. Beispiel 4 In eine Suspension von 6 g Natriumhydrid (55-60 0/oig in Mineralöl) in 200ml Benzol (wasserfrei) wird bei 0-60 ein Gemisch aus 17,7 g Methoxyessigsäureäthylester und 21,9 g Oxalsäurediäthylester eingetropft. Anschliessend wird 2 Stunden bei 0-6 , 16 Stunden bei Raumtemperatur und 1 Stunde bei 4-50 gerührt. Dann werden bei 0-6" 21,6 g Chlorphosphorsäuredimethylester, gelöst in 25 ml Benzol (wasserfrei), zugetropft. Das Reaktionsgemisch wird 24 Stunden bei Raumtemperatur gerührt und dann mit 100 ml Wasser ausgeschüttelt. Die organische Phase wird abgetrennt, getrocknet und eingedampft. Das Rohprodukt wird im Vakuum destilliert. Der erhaltene 0,0-Dimethyl-0-(1 ,2-bisäthoxy-carbonyl)-2- methoxy-viny & -phosphorsäureester siedet bei 136 /0,005 mm. Beispiel 5 Die gemäss dem vorstehenden Beispiel 3a) erhaltene Lösung des Enol-Natriumsalzes kann direkt, ohne vorheriges Eindampfen mit Chlorphosphorsäuredimethylester umgesetzt werden. Unter Eisbadkühlung tropft man 43,35 g Chlorphosphorsäure dimethylester zu der erwähnten Salzlösung. Das Reaktionsgemisch wird 24 Stunden bei Raumtemperatur gerührt und anschliessend in Vakuum eingedampft. Der Rückstand wird mit 500 ml Äther extrahiert und die so erhaltene Lösung mit 150 ml Wasser ausgeschüttelt Die organische Phase wird abgetrennt, getrocknet und von Lösungsmittel befreit. Nach Destillation in Vakuum erhält man den 0,0-Dimethyl-0-(2-methoxy-2isopropoxycarbonyl-vinyl)-phosphorsäureester. Die folgende Tabelle enthält weitere Beispiele von Phosphorsäureestern der Formel I, die nach den in den vorstehenden Beispielen beschriebenen Methoden hergestellt wurden. - PATENTANSPRUCH Verfahren zur Herstellung von Phosphorsäureestern der Formel I EMI5.1 worin X Sauerstoff oder Schwefel, Rt und R2 unabhängig voneinander je einen niederen Alkyl-Rest R3 Wasserstoff, einen Niederalkoxycarbonyl- oder Halogenmethyl-Rest, R4 einen Niederalkoxy-, Niederalkylthio- oder Di-niederalkylamino-Rest, und R3 einen niederen Alkoxy-Rest bedeuten und, wenn R4 einen niederen Alkylthiorest darstellt, R3 auch einen Di-niederalkylamino-Rest bedeuten kann, wobei bei R1-R5 unter niederem Alkyl ein Rest mit 1-5 C-Atomen verstanden wird, dadurch gekennzeichnet, dass man eine Verbindung der Formel II EMI5.2 in der Me ein Alkalimetallatom bedeutet, R,, R4 und R3 die oben angegebene Bedeutung haben,mit einem Phosphorsäurehalogenid der Formel III EMI5.3 in der Hal Chlor oder Brom bedeutet, Rt, R2 und X die unter Formel I angegebene Bedeutungen haben, in Gegenwart eines gegenüber den Reaktionsteilnehmern inerten Lösungs- oder Verdünnungsmittel umsetzt.UNTERANSPRÜCHE 1.
