Description
Verfahren zur Herstellung von Carotinoiden Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Carotinoiden der Formel EMI1.1 in der R und R'gegebenenfalls in 3-, 4-oder 5-Stellung eine Hydroxyl-oder Oxogruppe enthaltende 1, 2, 2-Trimethyl-cyclopentylcarbonyl-Reste oder R' zusätzlich auch den gegebenenfalls eine Hydroxyloder Oxogruppe enthaltenden 2, 6, 6-Trimethyl-cyclo- hexen-(l)-yl-Rest darstellen oder von an der Dreifachbindung partiell hydrierten Derivaten davon, welche zufolge ihrer intensiv roten Färbung als Farbstoffe z. B. für Nahrungsmittel und Futtermittel sehr geeignet sind. Die Verbindung der Formel I, in welcher die beiden Reste R und R'den 1, 2, 2-Trimethyl- 4-hydroxy-cyclopentylcarbonylrest darstellen, entspricht vermutlich einem als Capsorubin bezeichneten Paprika-Farbstoff, währenddem die Verbindung, bei welcher R den 1, 2, 2-Trimethyl-4-hydroxy-cyclo- pentylcarbonylrest und R'den 2, 6, 6-Trimethyl-4 hydroxy-cyclohexenr (l)-yl-Rest bedeuten, einem andern Paprika-Farbstoff, dem Capsanthin, zu entsprechen scheint. Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man einen Polyenaldehyd der Formel EMI1.2 oder an der Dreifachbindung partiell hydrierte Derivate davon, in welcher Formel Rl den gegebenenfalls eine Hydroxyl-, Oxo-oder ketalisierte Oxogruppe enthaltenden 2- [2', 6', 6'-Trimethyl-cyclohexen- (l')-yl]-äthenylrest oder den Formylrest darstellt, mit einem Keton der Formel EMI1.3 in der R2 Wasserstoff, eine Hydroxyl-, Oxo-ode ketalisierte Oxogruppe darstellt, kondensiert und allenfalls vorhandene ketalisierte Oxogruppen hydrolysiert. Eine im Kondensationsprodukt gegebenenfalls vorhandene Dreifachbindung kann durch partielle Hydrierung in eine Zweifachbindung übergeführt werden. Die als Ausgangsmaterialien verwendbaren Polyendialdehyde der Formel II können beispielsweise nach den Angaben in Helv. Chim. Acta, 39, 463 (1956) hergestellt werden. Die Herstellung von Verbindungen der Formel II, in welchen Rl den 2- [2', 6', 6'-Trimethyl-cyclohexen- (')-yl]-äthenylrest darstellt, wird in Helv. Chim. Acta, 42, 854 (1959) beschrieben. Die entsprechende Verbindung mit einem Hydroxylsubstituenten in 4-Stellung des Cyclo hexenrings ist eine unter der Bezeichnung ¯-Citraurin bekannteVerbindung [vgl. Karrer et al. carotinoidso, Elsevier Publishing Company (1950), 218]. Durch Oxydation der Hydroxylgruppe kann die entsprechende 4-Oxoverbindung gewonnen werden. Die als Ausgangsmaterialien benötigten Ketone der allgemeinen Formel III können beispielsweise aus Säuren der Formel EMI2.1 durch Behandlung mit Methyllithium gewonnen werden. In den Beispielen wird die Gewinnung von Verbindungen der Formel III näher beschrieben. Die Kondensation der Polyenaldehyde der Formel II mit den Ketonen der Formel III wird zweckmässig unter alkalischen Bedingungen vorgenommen. Vorzugsweise erfolgt die Kondensation in Gegenwart einer alkoholischen L¯sung eines Alkalimetallhydroxyds, z. B. äthanolischer Kalilauge. Die all fällige partielle Hydrierung der Dreifachbindung wird zweckmässig unter Verwendung eines mit Blei vergifteten Palladiumkatalysators [vgl. Helv. Chim. Acta, 35 (1952), 446] durchgeführt. Beisplel 1 Eine L¯sung von 3, 5 g 1-Carbaxy-1, 2, 2-trimethyl-cyclopentan in 40 ml Äther wird tropfenweise unter Rühren zu einer aus 10 g Methyljodid bereiteten L¯sung von Methyllithium in 50 ml Äther gegeben. Die Reaktionsmischung wird während 18 Stunden unter Rückfluss gekocht und anschliessend abgekühlt. Nach sorgfältigem Zufügen von Eiswasser werden 2, 0 g 1-Acetyl-1, 2, 2-trimethyl-cyclo- pentan isoliert ; Siedepunkt 88-92 C/23 mm, amas 1692 cm-1. Eine Mischung von 0, 5 g dieses Ketons und 50 mg Crocetindialdehyd in 3 ml 5% igem äthano lischem Kaliumhydroxyd wird 4 Stunden stehengelassen und während dieser Zeit gelegentlich geschüttelt und erwärmt. Anschliessend wird die Mischung während 30 Minuten unter Rückfluss gekocht, dann abgekühlt und mit Benzol verdünnt. Die Lö- sung wird nun mit Wasser gewaschen, über Natriumsulfat getrocknet und eingeengt. Durch Chromatographie des Rückstandes in einem Lösungsmittel- gemisch von Benzol und Petroläther (Siedebereich 60-80 ) im Verhältnis 1 : 1 an Aluminiumoxyd [vgl. Brockmann, Berichte (1941), Bd. 74, S. 73 ; Stufe IV], Isolierung der roten Hauptzone und Umkristallisation aus Benzol/Petroläther (Siedebereich 60-80 ) erhÏlt man 22 mg 1, 20-Di-[1', 2', 2'-trimethyl-cyclopentyl]- 4, 8, 13, 17-tetramethyl-eicosanonaen (2, 4, 6, 8, 10, 12, 14, 16, 18)-dion- (1, 20) als violette Kristalle ; Schmelzpunkt 205-206 ; Amax 517 und 483 mu., a = 95 000 und 105 000 (in Benzol) ; vmax 1664 cul7-1. Beispiel 2 4, 0 g Kaliumborhydrid werden in kleinen Anteilen zu einer warmen L¯sung von 10, 0 g 1-Carboxy 1, 2, 2-trimethyl-cyclopentanon- (3) und 3, 3 g Natriumhydroxyd in 130 ml Wasser gegeben. Die erhaltene Mischung wird auf dem Dampfbad während 16 Stunden erhitzt und anschliessend abgekühlt. Man fügt 13, 3 ml konz. Schwefelsäure in 70 ml Wasser hinzu und erwärmt die Reaktionsmischung während einer Stunde auf dem Dampfbad, kühlt ab und extrahiert gründlich mit ¯ther. Die ätherische L¯sung wird mit gesättigter Natriumbicarbonatlösung extrahiert. Der Extrakt wird angesäuert, das ausgefallene Produkt durch Behandlung mit Ather isoliert und aus einer Mischung aus Athylacetat und Petroläther (Siedebereich 60-80 ) umkristallisiert. Man erhÏlt 5, 7 g einer Mischung der epimeren Formen von 1-Carboxy 1, 2, 2-trimethyl-3-hydroxy-cyclopentan ; Schmelzpunkt 220-222 ; fmax 1695 cl-1. Der Methylester dieser Verbindung siedet bei 132-135 /17 mm ; v 3450 und 1718 cm-1. 5, 7 g 1-Carboxy-1, 2, 2-trimethyl-3-hydroxy-cyclopentan werden in 50 ml Ather gelöst und die erhaltene L¯sung tropfenweise unter Rühren einer aus 18 g Methyljodid bereiteten ätherischen L¯sung von Methyllithium zugefügt. Die Mischung wird während 18 Stunden unter Rückfluss erwärmt und anschlie ssend abgekühlt. Nun wird vorsichtig Eiswasser hinzugefügt und das gebildete neutrale Produkt iso- liert. Man erhÏlt auf diese Weise 1 g 1-Acetyl-3 hydroxy-1, 2, 2-trimethyl-cyclopentan ; ?max 3425 und 1686 cl-1. Eine Mischung des nach dem vorhergehenden Absatz gewonnenen Ketons mit 100 mg Crocetindialdehyd und 3 ml 5 % igem methanolischem Ka liumhydroxyd wird während 19 Stunden bei 20 gehalten und anschliessend während 21/2 Stunden unter Rückfluss erwärmt. Das Reaktionsgemisch wird nun abgekühlt, mit Benzol verdünnt, mit Wasser gewaschen, über Natriumsulfat getrocknet und eingeengt. Durch Isolierung gemäss Beispiel 1 und Umkristallisation aus Benzol/Petroläther (Siedebereich 60-80 ) erhÏlt man 66 mg 1, 20-Di- [3-hydroxy-1, 2, 2-trimethyl-cyclo pentyl]-4, 8, 13, 17-tetramethyl-eicosanonaen (2, 4, 6, 8, 10, 12, 14, 16, 18)-dion- (1, 20) als violette Kristalle ; Schmelzpunkt 187-188 C ; rmax 3401 und 1661 mit ; A.,,, (in benzolischer Lösung) 520 und 485 m, u., E = 104 000 und 118 000. Beispiel 3 Eine L¯sung von 190 g 4-Carbäthoxy-4, 5, 5-tri methyl-cyclohexadion- (1, 3) und 4, 5 g p-Toluolsulfo- säure in 1, 5 1 Athanol und 900 ml Benzol wird während 4 Tagen unter einem < Dean-Stark) > -Ab- scheider unter Rückflussbedingungen erwärmt. Das im Abscheider angesammelte Kondensat wird wiederholt entfernt und das. Lösungsmittel in der Reak tionsmischung entsprechend nachgefüllt. Anschlie ssend wird die Reaktionsmischung abgekühlt, unter vermindertem Druck eingeengt, mit Petroleumäther (Siedebereich 40-60 ) verdünnt, mit gesättigter Natriumcarbonatlösung gewaschen, über Magnesiumsulfat getrocknet und eingeengt. Die Destillation des Rückstandes ergibt bei einem Siedepunkt von 118¯/ 0, 35 mm als farbloses Í1 176 g 6-Carbäthoxy-3- Ïthoxy-5, 5, 6-trimethyl'-cyclohexen- (2)-on- (1) ; nD = 1, 4878 ; man 1721, 1653 und 1608 mit ; max (in äthanolischer Lösung) 256 mou, a = 5700. Eine L¯sung von 10 g dieses Enoläthers in 50 ml Ather fügt man zu 2, 0 g Lithiumaluminiumhydrid in 150 ml ¯ther. Die Mischung wird unter Rückflussbedingungen während 20 Stunden erwärmt, dann abgekühlt und mit verdünnter Schwefelsäure im Dberschuss behandelt. Aus der Reaktionsmischung können 7, 0 g Rohprodukt isoliert werden. Durch Chromatographie an Aluminiumoxyd erhÏlt man 2, 4 g 4-Hydroxymethyl-4, 5, 5-trimethyl-cyclohexen (2)-on-(1), welches aus Petroläther (Siedebereich 40-60 ) umkristallisiert wird ; Schmelzpunkt 151 ; ?max (in Ïthanolischer L¯sung) 230 mut, c = 7800 ; max (in Chloroform) 3680, 1672 und 1600 cl-1. Das 2, 4-Dinitro-phenylhydrazon dieser Verbindung weist einen Schmelzpunkt von 174 auf ; Amax (in äthanolischer L¯sung) 373 m, u, e = 26 000. 21 g des nach dem vorhergehenden Absatz gewonnenen rohen Cyclohexenons werden in 50 ml Athanol gelöst und in Anwesenheit von 3, 0 g l Öliger PalljadiumkoMe bis zum Stillstand der Wasserstoffaufnahme mit Wasserstoff geschüttelt. Nach dem Entfernen des Katalysators und des Lösungsmittels erhÏlt man 4-Hydroxymethyl-4, 5, 5-tri methyl-cyclohexanon-(l), welches ohne Reinigung weiter verwendet wird ; vmax 3497 und 1704 cl-1. Eine abgekühlte L¯sung von 12 g dieser Verbindung in 150 ml Aceton fügt man zu 25 g Chrom trioxyd in 70 ml 2n Schwefelsäure. Die Mischung wird während 2 Stunden bei 20 ger hrt und an schliessend in 1 Liter Wasser gegossen. Das saure Produkt wird isoliert und aus BenzoVPetroläther (Siedebereich 40-60 ) umkristallisiert. Man erhÏlt 2, 0 g 4-Carboxy-3, 3, 4-trimethyl-cyclohexanon- (l) ; Schmelzpunkt 188 ; vmax (in Chloroform) 3000-3500 und 1706 cm-1. Das 2, 4-Dinitro-phenylhydrazon die ser Verbindung schmilzt nach dem Umkristallisieren aus Athanol bei 193 ; AmaX (in äthanolischer Lösung) 355 mu, c = 17 500. Eine L¯sung von 4, 4 g der nach dem vorher gehenden Absatz gewonnenen Ketosäure in 200 ml einer In Kalium-tert.-butylatlösung wird in Gegen wart von Sauerstoff bis zum Stillstand der Sauerstoff aufnahme geschüttelt. Die L¯sung wird in Wasser gegossen und mit 10 % iger Salzsäure angesäuert. Durch Extraktion mit Chloroform erhÏlt man 4, 6 g eines festen Rückstandes, der gemäss spektrophoto- metrischer Bestimmung eine Reinheit von 89 % aufweist. Das gereinigte 4-Carboxy-4, 5, 5-trimethyl-cy clohexandion- (1, 2) schmilzt bei 136-137 ; ?max3100 bis 3500, 1695 und 1672 mit ; (in lithanoli- scher L¯sung) 268 m, c, e == 6500 ;/tmax (in alkali- scher L¯sung) 312 m/, t, E 5200. Eine L¯sung von 4, 2 g des nach den Angaben im vorhergehenden Absatz gewonnenen 89 % igen Dions in 100 ml 5 % igem Kaliumhydroxyd wird unter Rückflussbedingungen gekocht, bis nach ungefähr 20 Stunden kein Absorptionsmaximum bei 312 m, u mehr festgestellt werden kann. Die L¯sung wird abgekühlt, durch Zufügen von Salzsäure bis zu pH 4 angesäuert und filtriert. Das. Filtrat wird mit dreimal 20 ml Ather extrahiert und anschliessend mit 9, 0 g Phosphorsäure und 9, 0 g Natriumwismutat versetzt. Die erhaltene Suspension wird über Nacht stehengelassen und anschliessend während 2 Stunden kräftig gerührt. Nun extrahiert man viermal mit je 50 ml Ather, engt die vereinigten Extrakte ein und kristallisiert den Rückstand aus. Benzol/Petroläther (Siedebereich 60-80 ). Das gebildete 3-Carboxy3, 4, 4-trimethyl-cyclopentanon- (l) schmilzt in einem geschlossenen Schmelzpunktröhrchen bei 216 [eine Mischung mit 3-Carboxy-2, 2, 3-trimethyl-cyclopenta- non-(1) vom Schmelzpunkt von 228 schmilzt bereits bei 207-209 ] ; ?max (in Chloroform) 3100-3500, 1736 und 1701 cm-1. Das 2, 4-Dinitro-phenylhy- drazon dieser Säure schmilzt nach dem Umkristalli sieren aus Methanol bei 223 . Diese Säure kann nach den Angaben in Beispiel 2 durch Reduktion mit Kaliumborhydrid in 3-Carb- oxy-3, 4, 4-trimethyl-cyclohexanol-(1), durch Behandlung mit Lithiummethyl in 3-Acetoxy-3, 4, 4-trimethylcyclohexanol und schliesslich durch Umsetzung mit Crocetindialdehyd in Capsorubin übergeführt werden. Beispiel 4 Eine Mischung von 200 mg ss-Apo-8'-carotinal, 1, 0 g 1-Acetyl-1, 2, 2-trimethyl-cyclopentan und 15 mI 5 % iger äthanolischer Kaliumhydroxydlösung wird erwärmt und während 2 Tagen bei 20 gehalten. Schliesslich wird die Reaktionsmischung 30 Minuten unter Rückfluss erwärmt. Die Reaktionsmischung wird abgekühlt, mit 250 ml Petroläther (Siedebereich 70-80 ) verdunnt, mit wässrigem Methanol (1 : 9) und mit Wasser gewaschen. Die L¯sung wird ge trocknet und eingeengt. Durch Chromatographie des Rückstandes in benzolischer L¯sung an Aluminium oxyd und anschliessender Isolierung der roten Haupt zone und Umkristallisation des daraus gewonnenen Rückstandes aus Benzol/Methanol erhÏlt man 25 mg 1-[2', 6', 6'-Trimethyl-cyclohexen-(l')-yl]- 19- [I', 2', 2'-trimethyl-cyclopenty1] 3, 7, 12, 16-tetramethyl-nonadecanonaen- (1, 3, 5, 7, 9, 11, 13, 15, 17)-on- (19) in Form von Nadeln, Schmelzpunkt 164-165 ; itmas (in Benzollösung) 483 mu, s = 10 100 ; max (in Chloroform) 1667 cm-1, ? = 200.
Claims (1)
- PATENTANSPRUCH I Verfahren zur Herstellung von Carotinoiden der Formel EMI4.1 in der R und R'gogebenenfalls in 3-, 4-oder 5-Stel- lung eine Hydroxyl-oder Oxogruppe enthaltende 1, 2, 2-Trimethyl-cyclopentylcarbonyl-Reste oder R' zusätzlich auch den gegebenenfalls eine Hydroxyloder Oxogruppe enthaltenden 2, 6, 6-TrimethyI-cyclo- hexen- (1)-yl Rest darstellen, oder an der Dreifachbindung partiell hydrierten Derivaten davon, dadurch gekennzeichnet, dass man einen Polyenaldehyd der Formel EMI4.2 oder an der Dreifachbindung partiell hydrierte Derivate davon, in welcher Formel Ri den gegebenenfalls eine Hydroxyl-,Oxo-oder ketalisierte Oxogruppe enthaltenden 2-[2', 6', 6'-Trimethyl-cyclohexen- (l')-yQ-äthenyl'rest oder den Formylrest darstellt, mit einem Keton der Formel EMI4.3 in der R2 Wasserstoff, eine Hydroxyl-, Oxo-oder ketalisierte Oxogruppe darstellt, kondensiert und allenfalls vorhandene ketalisierte Oxogruppen hy- drolysiert.UNTBRANSPRÜCHE 1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine im Kondensationspro- dukt vorhandene Dreifachbindung partiell hydriert.2. Verfahren nach Patentanspruch I und Unteranspruch 1, dadurch gekennzeichnet, dass man die Kondensation unter alkalischen Bedingungen durchführt.3. Verfahren nach Patentanspruch I und den Un teranspruchen 1 und 2, dadurch gekennzeichnet, dass man die Kondensation in Gegenwart einer alkoholischen Lösung eines Alkalimetallhydroxyds durchführt.PATENTANSPRUCH II Verwendung von gemäss Patentanspruch I erhaltenen Carotinoiden als Farbstoffe in Lebensrnitteln und Futtermitteln.
