Verfahren zur Herstellung von Sulfonylharnstoffen
Es wurde gefunden, dass sich Verbindungen der Formel R-SO2-NH-CO-NH-Rt, worin R einen aliphatischen oder cycloaliphatischen Kohlenwasserstoffrest mit 3-8 Kohlenstoffatomen und Rt einen gesättigten oder ungesättigten, offenkettigen oder ringförmigen, Sauerstoff oder Schwefel enthaltenden aliphatischen Kohlenwasserstoffrest mit vorzugsweise bis zu 8 Kohlenstoffatomen bedeuten, und die Salze dieser Verbindungen bei geringer Toxizität durch eine starke blutzuckersenkende Wirksamkeit auszeichnen.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zur Herstellung solcher Verbindungen, das dadurch gekennzeichnet ist, dass man Verbindungen der Formel RSO2.-Hal mit Isoharnstoffäthern der Formel

umsetzt, wobei R2 für einen aliphatischen Kohlenwasserstoffrest steht und Hal Halogen bedeutet, und die Kondensationsprodukte hydrolysiert.
Die Reaktion der Sulfonsäurehalogenide mit den Isoharnstoffäthern kann man beispielsweise im Falle der Verwendung von festen Sulfonsäurehalogeniden in wässerigem Medium in Gegenwart von Kaliumcarbonat durchführen. Nach einer vorteilhaften Ausführungsform des Verfahrens setzt man die Isoharnstoffäther in Form von Salzen in wässeriger acetonischer Lösung und unter Kühlung mit den Sulfonsäurehalogeniden in Gegenwart von Alkalilaugen zu den entsprechenden Sulfonylharnstoffäthern um.
Diese werden beispielsweise mit konzentrierten Säuren, vorzugsweise Salzsäure, auf etwa 60-100 C erwärmt, wobei Gasentwicklung beobachtet werden kann.
Als Ausgangsstoffe kommen für das Verfahren gemäss der Erfindung folgende Sulfonylverbindungen in Betracht: Alkyl-, Cycloalkyl- oder Cycloalkylalkylsulfonsäurehalogenide, die als Alkylreste den Propyl-, Butyl-(l)-, Butyl-(2)-, 2-Methyl-propyl-(l)-, Pentyl (1)-, Pentyl-(2)-, Pentyl-(3)-, 3-Methyl-butyl-(l)-, 2-Methyl-butyl-(l)-, Hexyl-(l)-, Hexyl-(2)-, 4-Methyl-pentyl- (2)-, 2-Athyl-butyl-(l)-, Heptyl-(l)-, Hetpyl-(2)-, Heptyl-(4) -, 2 2,4-Dimethyl-pentyl-(1)-, 2,4-Dimethyl-pentyl-(3)-, Octyl-(l)- oder Octyl-(2)rest, als Cycloalkylreste den Cyclohexyl- oder Cyclo heptylrest und als Cycloalkylakylreste den Cyclohexylmethyl- und Cyclohexyläthylrest tragen können.
Für die Umsetzung mit den vorstehend genannten Sulfonsäurehalogeniden können Isoharnstoffäther mit folgenden Resten R1 verwendet werden: Methoxypropyl, Sithoxy-propyl, Propoxy-propyl, Propoxy äthyl, Athoxy-äthyl, Äthoxy-äthylenoxy-äthyl, Meth oxy - äthylenoxy - äthyl, a- Tetrahydrofurfurylmethyl, 6-Propyl-dioxan-(1,3), B, 4-Diäthoxy-äthyl, Methylmercapto-propyl und 22ithyl-mercapto-propyl.
Die Verfahrenserzeugnisse stellen wertvolle Heilmittel dar, die sich insbesondere durch eine starke blutzuckersenkende Wirksamkeit und sehr geringe Toxizität auszeichnen. Die letztgenannte Eigenschaft lässt sie zur Verwendung als perorale Antidiabetik als besonders geeignet erscheinen, da derartige zur Behandlung der Zuckerharrrruhr geeignete Verbindungen unter Umständen über lange Jahre gegeben werden müssen. Die Verbindungen können als solche oder in Form ihrer Alkalisalze bzw. in Gegenwart von Stoffen, welche zu einer Salzbildung führen, Verwendung finden. Zur Salzbildung können beispielsweise herangezogen werden: Alkali- oder Erdalkalihydroxyde, Alkalicarbonate oder -bicarbonate, ferner physiologisch verträgliche organische Basen.
Im Tierversuch lässt sich die Wirkung auf den Blutzuckerspiegel beispielsweise von Kaninchen nachweisen. Verabreicht man normal gefütterten Kaninchen die genannten Verbindungen in einer einmaligen Dosis von durchschnittlich 400 mglkg in beispielsweise bicarbonat-alkalischer Lösung, so sieht man eine rasch einsetzende Senkung des Blutzuckerspiegels, die innerhalb von 3-4 Stunden ein Maximum von etwa 30 /o des Ausgangswertes erreicht.
Beispielsweise erniedrigt die angegebene Dosis von N 3-MethylL butan- (1)-sulfonylliNt-(3tmethoxy- propyl)-harnstoff den Blutzucker nach 2 Stunden um etwa 30 /o ; die Wirkung hält etwa 6 Stunden an. Im Falle der Verwendung von N-[3-Methyl-butan-(l) sulfonyll-N'-(3'-äthoxy-propyl)-harnstoff erhält man nach 2 Stunden eine Blutzuckersenkung um 20 /o, die nach 6 Stunden noch 160/0 beträgt. Im Falle der Verwendung von N-Cyclohexansulfonyl-N'-(3'-methoxypropyl)-harnstoff beträgt die Senkung des Blutzuckers nach einer Stunde etwa 300/0; ; diese Senkung hält über 6 Stunden hinaus an.
Im Falle der Verwendung von N- Cyclohexylmethansulfonyl -N'-(3'-methoxy- propyl)-harnstoff beträgt die Blutzuckersenkung nach
2 Stunden 40 /o und nach 6 Stunden noch 350/ob
Die Blutzuckerwerte können durch stündliche Analysen nach Hagedorn-Jensen ermittelt werden.
Die Blutzuckersenkung wird durch Vergleich mit den Blutzuckerwerten gleichartig gehaltener, nicht be handelter Kontrolltiere ermittelt.
Es ist bereits bekannt, dass der N-(4-Amino benzolsulfonyl) - N'-n-butyl-harnstoff blutzuckersen kende Eigenschaften aufweist. Weiterhin ist bekannt, dass diese Verbindung auf Grund ihres Sulfanilyl charakters auch chemotherapeutisch wirksam ist. Da für die Verwendung als orales Antidiabetikum die
Dauergabe über- einen langen Zeitraum hinaus erfor derlich ist, ist es jedoch wünschenswert, dass die applizierte Verbindung keinen Sulfanilylcharakter hat und somit von andersartigen Wirkungen möglichst frei ist, um eventuelle Schädigungen, beispielsweise der Darmflora, sowie Allergien und die Resistenzbildung pathogener Keime gegenüber Sulfonanilylamiden auszuschliessen.

N-(Cyclohexansulfonyl)-N'-(3'-methoxy-propyl) harnstoff
40 g Methoxypropylharnstoff werden mit 29 ml Dimethylsulfat auf dem Dampfbad erwärmt, bis sich das Reaktionsgemisch verflüssigt und die Temperatur durch die Reaktionswärme auf etwa 120"C angestiegen ist. Man lässt erkalten, löst in 150 ml Wasser, gibt 55 g Cyclohexansulfochlorid hinzu und tropft unter Rühren eine Lösung von 27 g Natriumhydroxyd in 180 ml Wasser zu, wobei die Temperatur unter 400 C gehalten wird. Der sich bei der Reaktion abscheidende ölige Cyclohexansulfonylisoharnstoffäther wird abgetrennt und mit der doppelten Menge konzentrierter Salzsäure im Bad etwa ¸ Stunde auf 80 bis 90"C erhitzt.
Es entwickelt sich ein Gas, und nach dem Erkalten scheidet sich eine kristalline Sub stanz aus. Man saugt ab, nimmt in 1 1 /oigem Ammo- niak auf, filtriert und fällt mit Säure den N-Cyclo hexansulfonyl - N'- (3'-methoxy-propyl)-harnstoff mit Salzsäure aus. Die Substanz schmilzt nach dem Umkristallisieren aus Athanol/Wasser bei 127-129 C.
