Verfahren zur Herstellung von neuen Benzolsulfonylharnstoffen
Es ist bekannt, dass gewisse Benzolsulfonylharnstoff-Derivate blutzuckersenkende Eigenschaften aufweisen und somit als per os verabreichbare Antidiabetika geeignet sind (vgl. beispielsweise Arzneimittel-For schwung, 8 (1958), Seiten 448454). Insbesondere der N-(4-Methyl-benzol-sulfonyl)-N'-n-butyl-harnstoff hat auf Grund seiner guten blutzuckersenkenden Eigenschaften und seiner, guten Verträglichkeit in der Diabetestherapie grosse Bedeutung erlangt.

n die Zahlen 0 oder 1 und -Z-Z'- die Gruppierungen -CH2-CH2 - oder CH=CH-bedeuten, und deren Salze blutzuckersenkende Eigenschaften besitzen und auf Grund ihrer starken Wirksamkeit als oral verabreichbare Antidiabetika geeignet sind.
Die genannten Verbindungen erhält man erfindungsgemäss, indem man Benzolsulfonyl-isoharnstoff äther, Benzolsulfonyl-guanidine, bzw. Benzolsulfonylparabansäuren durch Hydrolyse in die gewünschten Benzolsulfonylharnstoffe überführt. Die der Hydrolyse zu unterwerfenden Verbindungen können hergestellt werden, indem man basifizierte Harnstoff-Derivate, z. B. Isoharnstoffäther, Guanidine, oder Salze von Mono-R'-substituierten Parabansäuren mit tert. Aminen. mit substituierten Benzolsulfonsäurehalogeniden zur Umsetzung bringt.
Als Ausgangsstoffe kommen für das Verfahren gemäss der Erfindung einerseits solche Benzolsulfonyl Derivate in Betracht, die vorzugsweise einen alkylierten oder alkoxylierten Benzolrest enthalten. Als Alkylgruppen seien erwähnt:
Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, sek.-Butyl und tert. Butyl. Ebenso kommen die entsprechenden Alkoxygruppen, wie Methoxy, Sith- oxy, n-Propoxy, Isopropoxy, n-Butoxy, Isobutoxy oder tert. Butoxy, in Frage. Insbesondere sind geeignet: Methyl-, 3ithyl-, Methoxy- und Äthoxy-Gruppen in pund m-Stellung. Werden andererseits halogensubstituierte Benzolsulfonylverbindungen als Ausgangsstoffe herangezogen, so sind als Substituenten vorzugsweise solche mit einem Fluor-, Chlor- oder Brom-Atom oder mit der Trifluormethylgruppe geeignet.
Als Reaktionspartner der Benzolsulfonylderivate kommen 2,5 Endomethylen-cyclohexyl-, 2,5-Endome thylen-G3'-cyclohexenyl-, 2, 5-Endomethylen-cyclohexylmethyl und 2,5-Endomethylen-a3'-cyclohexenylme- thylderivate von Isoharnstoffäthern, Guanidinen und Parabansäuren in Betracht.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise können die Umsetzungen unter Verwendung von Lösungsmitteln, bei Zimmertemperatur oder bei erhöhter Temperatur durchgeführt werden. Um die Verfahrensprodukte in möglichst reiner Form zu erhalten, nimmt man zweckmässig eine möglichst vollständige Abtrennung von den als Ausgangsstoffen eingesetzten oder im Verlauf der Reaktion entstehenden Benzolsulfonamiden vor, die vorteilhaft dadurch erreicht werden kann, dass man das Verfahrensprodukt in stark verdünntem Ammo niak aufnimmt, von ungelösten Bestandteilen abfiltriert und durch Ansäuern den gewünschten Benzolsulfonylharnstoff gewinnt.
Die nach dem Verfahren gemäss der Erfindung erhältlichen neuen B enzolsulfonylharnstoffe stellen wertvolle Arzneimittel dar, die sich insbsondere durch ihre blutzuckersenkende Wirksamkeit auszeichnen.
Beispielsweise bewirkt der N-(4-Methyl-benzolsulfo nyl-)-N'-[2',5'-endomethylen-#3'-cyclohexen-(1')-yl]-me- thyl-harnstoff am Kaninchen in einer Dosierung von 400 mg/kg per os eine Senkung des Blutzuckers um 30 > 40%. Entsprechende Resultate werden bei der Prüfung des N-(4-Methoxy-benzolsulfonyl)-N'-[2',5'-endo methylen-#3'-cyclohexen-(1')-yl]-methyl-harnstoffs und N-(4-;.thoxy-benzolsulfoayl) -N'-[2',5'- endomethylen- #3'-cyclohexen-(1')-yl]-methyl-harnstoff erhalten. Die angegebebenen Zahlenwerte wurden durch Vergleich mit gleich artig gehaltenen Kontrolltieren erhalten. Die Blutzuckerwerte wurden durch stündliche Analysen nach Hagedorn/Jensen bestimmt.
Die gute blutzuckersenkende Wirkung der neuen Verfahrensprodukte lässt sich auch nachweisen, wenn die Prüfung bei einer erheblich niedrigeren Dosierung (Schwelldosis) vorgenommen wird. So bewirkt beispielsweise der N-(4-Methyl-benzolsulfonyl)-N'-2',5'-endome thylen-#3'-cyclohexen-(1')-yl)-methylharnstoff bei der Prüfung am Kaninchen in einer Dosis von 50 mg/kg eine Senkung des Blutzuckers um 28%. Dieser Wert bleibt über 6 Stunden erhalten.
Weiterhin wirkt der N-(4-Brom-benzolsulfonyl)-N'- [2', 5'-endomethylen-cyclohexyl-(1)]-harnstoff bei intravenöser Verabreichung an Kaninchen bereits bei einer Schwellendosis von 5 mp/kg blutzuckersenkend, währen die entsprechende Schwellendosis bei intravenöser Verabreichung an Kaninchen für den bekannten N-(4 Methyl-benzolsulfonyl)-N'-n-butyl-harnstoff 20 mg/kg beträgt. Die neue Verbindung erweist sich somit hinsichtlich der intravenösen Schwellendosis am Kaninchen als viermal wirksamer als die bekannte Verbindung. Der N-(4-Chlor-benzolsulfonyl)-N'- [2', 5'-endomethylen-cy clohexyl-(l)]-harnstoff ist bei intravenöser Applikation an Kaninchen bereits in einer Schwellendosis von 2,5 mg/kg wirksam.
Somit ist die neue Verbindung bei diesem speziellen Test etwa achtmal so wirksam wie der be kannte N-(4-Methyl-benzolsulfonyl)-N'-n-butyl-harnstoff.
Für ein Dauertherapeuticum, wie es die oralen Antidiabetika darstellen, ist von grosser Bedeutung, dass die zur Anwendung kommenden Verbindungen wenig toxisch sind; diese Forderungen werden von den neuen Verfahrenserzeugnissen erfüllt. So beträgt z. B. die LD50 per os an der weissen Maus für den N-(4-Methyl-benzol sulfonyl)-N'-[2',5'-endomethylen-#3'-cyclohexen-(1')-yl]- methyl-harnstoff mehr als 7 g/kg.
Die Verfahrenserzeugnisse sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirkung zur Behandlung des Diabetes mellitus dienen und können als solche in Form ihrer Salze bzw. von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise herangezogen werden:
Alkalische Mittel wie Alkali- oder Erdalkalihydro xyde, -carbonate, -bicarbonate. ferner physiologisch verträgliche organische Basen.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Hilfs- und Trägerstoffe, wie Talkum, Stärke, Milchzucker, Traganth, Magnesiumstearat usw. enthalten.
Beispiel I
N-(4-Methoxy-benzolsulfonyl)-N'-[2',5'-endomethylen #3'-cyclohexen-(1')-yl]-methyl-barnstoff a) 11 g (2,5-Endomethylen-#3-cyclohexen-(1)-yl)- methyl-parabansäure (hergestellt aus [2,5-Endomethylen #3-cyclohexen-(1)-yl]-methyl-harnstoff und Oxalylchlorid), 5 g Triäthylamin und 10,3 g 4-Methoxy-benzolsulfochlorid werden in 200 ml Benzol gelöst und 4 Stunden am Rückflusskühler zum Sieden erhitzt. Man filtriert von ausgefallenem Triäthylaminitydrochlorid ab, engt das Filtrat ein und kristallisiert den Rückstand aus Isopropanol um.
Die erhaltene 1-(4-Methoxy-benzolsulfo nyl)-3-[2, 5-endomethylen-A3- cyclohexen-(1)-yl]-methyl- parabansäure schmilzt bei 149-150O C. b) 9 g 1-(4-Methoxy-benzolsulfonyl)-3-[2, 5-endome- thylen-A3-cyclohexen-(l)-yl]-methyl-parabansäure werden mit 150 ml 1n Natronlauge auf dem Dampfbad erwärmt. Nach kurzer Zeit ist Lösung eingetreten. Man verdünnt das Gemisch mit 100 ml Wasser, filtriert unter verwendung von Kohle und säuert das Filtrat an. Der abgeschiedene N-(4-Methoxy-benzolsulfonyl)-N'-[2',5' endomethylen-#3'-cyclohexen-(1')-yl]-methyl-harnstoff wird aus Äthanol umkristallisiert und schmilzt bei 145 147O C.
Beispiel 2
N-(4-Methyl-benzolsulfonyl)-N'-[2',5'-endomethylen #3'-cyclohexen-(1')-yl]-methyl-harnstoff
16,6 g [2',5'-Endomethylen-#3'-cyclohexen-(1')-yl]- methyl-harnstoff werden mit 12,6 g Dimethylsulfat durch Erhitzen auf dem Dampfbad zur Reaktion gebracht.
Man lässt erkalten, löst das Reaktionsprodukt in 50 ml Wasser, setzt die Lösung von 19 g p-Toluolsulfochlorid in 50 ml Aceton zu und tropft unter Rühren eine Lösung von 9 g Natriumhydroxyd in 50 ml Wasser zu, wobei man die Reaktionslösung schwach alkalisch hält und die Temperatur nicht über 40O C steigen lässt.
Man rührt noch 1 Stunde nach, trennt den abgeschiedenen Sulfonylisoharnstoffäther ab und erhitzt ihn mit 50 ml konzentrierter Salzsäure auf dem Dampfbad, bis die Gasentwicklung beendet ist. Der Ansatz wird gekühlt, mit Wasser versetzt und das abgeschiedene Produkt abgesaugt. Man kristallisiert aus Methanol um und erhält den N-(4-Methyl-benzolsulfonyl) N'-[2', 5'-endomethylen- #3'-cyclohexen-(1')-yl]-methyl-harnstoff vom Schmelzpunkt 175-177 C.
Process for the preparation of new benzenesulfonylureas

The compounds mentioned are obtained according to the invention by converting benzenesulfonyl-isourea ether, benzenesulfonyl-guanidines or benzenesulfonylparabanic acids into the desired benzenesulfonylureas by hydrolysis. The reactants of the benzenesulfonyl derivatives are 2,5-endomethylene-cyclohexyl, 2,5-endomethylene-G3'-cyclohexenyl-, 2,5-endomethylene-cyclohexylmethyl and 2,5-endomethylene-a3'-cyclohexenylmethyl derivatives of isourea ethers, guanidines and parabanic acids.
The embodiments of the process according to the invention can be largely varied with regard to the reaction conditions and adapted to the respective conditions. For example, the reactions can be carried out using solvents, at room temperature or at elevated temperature. The new benzene sulfonylureas obtainable by the process according to the invention are valuable medicaments which are distinguished in particular by their blood sugar-lowering effectiveness.

The numerical values given were obtained by comparison with control animals kept in the same way. The blood sugar values were determined by hourly analyzes according to Hagedorn / Jensen.
The good blood sugar-lowering effect of the new process products can also be demonstrated if the test is carried out at a considerably lower dose (threshold dose). For example, the N- (4-methylbenzenesulfonyl) -N'-2 ', 5'-endome thylen- # 3'-cyclohexen- (1') - yl) methylurea when tested in rabbits in a dose of 50 mg / kg lower blood sugar by 28%. This value is retained for 6 hours.
Furthermore, when administered intravenously to rabbits, N- (4-bromo-benzenesulfonyl) -N'- [2 ', 5'-endomethylene-cyclohexyl- (1)] urea lowers blood sugar levels at a threshold dose of 5 mp / kg the corresponding threshold dose for intravenous administration to rabbits for the known N- (4-methylbenzenesulfonyl) -N'-n-butylurea is 20 mg / kg. The new compound thus proves to be four times more effective than the known compound with regard to the intravenous threshold dose in rabbits. The N- (4-chloro-benzenesulfonyl) -N'- [2 ', 5'-endomethylene-cy clohexyl- (l)] - urea is already effective in a threshold dose of 2.5 mg / kg when administered intravenously to rabbits .
The new compound is thus about eight times as effective in this particular test as the well-known N- (4-methylbenzenesulfonyl) -N'-n-butylurea.
For a long-term therapeutic, such as the oral antidiabetic agents, it is of great importance that the compounds used are not very toxic; these requirements are met by the new process products. So z. B. the LD50 per os on the white mouse for the N- (4-methyl-benzene sulfonyl) -N '- [2', 5'-endomethylene- # 3'-cyclohexen- (1 ') - yl] - methyl -urea more than 7 g / kg.

Example I.
N- (4-methoxy-benzenesulfonyl) -N '- [2', 5'-endomethylene # 3'-cyclohexen- (1 ') - yl] -methyl-urea) 11 g (2,5-endomethylene # 3-cyclohexen- (1) -yl) methyl-parabanoic acid (prepared from [2,5-endomethylene # 3-cyclohexen- (1) -yl] -methyl urea and oxalyl chloride), 5 g of triethylamine and 10.3 g 4-methoxy-benzenesulfochloride are dissolved in 200 ml of benzene and heated to boiling for 4 hours on a reflux condenser. 16.6 g of [2 ', 5'-endomethylene # 3'-cyclohexen- (1') - yl] methyl urea are reacted with 12.6 g of dimethyl sulfate by heating on the steam bath.
