Verfahren zur Herstellung von neuen Benzolsulfonylharnstoffen

Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Benzolsulfonylharnstoffen der allgemeinen Formel :

worin X Sauerstoff oder Schwefel und Rl einen gesättigten oder ungesättigten alicyclischen Kohlenwasserstoffrest mit 3 - 8 Kohlenstoffatomen, wobei diese Reste, falls sie 4 - 8 Kohlenstoffatome enthalten, auch durch Sauerstoff und bzw. oder Schwefel unterbrochen sein können, oder einen Benzyl- oder a-Phenyl- äthylrest bedeuten, und deren Salzen, welche blutzuckersenkende Eigenschaften besitzen und auf Grund ihrer starken Wirksamkeit als oral verabreichbare Antidiabetika geeignet sind.
Man erhält die Verfahrenserzeugnisse, indem man die entsprechenden substituierten Benzolsulfonyl- - thioharnstoffe herstellt und aus diesen den Schwefel in üblicher Weise, beispielsweise mit Hilfe von Oxyden oder Salzen von Schwermetallen oder auch durch Anwendung von Oxydationsmitteln, wie Wasserstoffperoxyd, Natriumperoxyd. und salpetrige Säure, eliminiert.
Als Ausgangsstoffe kommen für das Verfahren gemäss der Erfindung p-Trifluormethoxy- und p-Trifluormethyl-mercaptobenzolsulfonyl-thioharnstoffe in Betracht. In ihnen kann der Substituent Rt beispiels - weise folgende Bedeutung haben : Cyclopentenyl, Cyclopentyl, Cyclohexyl, Cyclohexenyl, Cycloheptyl und 4-Methyl-cyclohexyl, 2,5-Endomethylen-cyclohexyl, Bornyl, ferner Cycloalkyl-alkyl, wie Cyclohexyl-methyl und Cyclohexyl-äthyl, ausserdem Tetrahydro-a-furfuryl, 2-Phenyläthyl- (1) und Benzyl.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinen hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise können die Umsetzungen unter Verwendung von Lösungsmitteln bei Zimmertemperatur oder bei erhöhter Temperatur durchgeführt werden.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Benzolsulfonylharnstoffe stellen wertvolle Arzneimittel dar, die sich insbesondere durch gute blutzuckersenkende Wirksamkeit bei geringer Toxizität auszeichnen. Beispielsweise bewirkt der N- (4-Trifluormethylmercapto-benzolsulfonyl) -N' -cyclohe- xyl-harnsto'ff am Kaninchen in einer Dosierung von 400 mg/kg des Natriumsalzes per os eine Senkung des

(4-Trifluormethoxy-ben-Die Verfahrenserzeugnisse zeigen infolge des Fehlens einer p-ständigen Aminogruppe im Benzolkern keinen den therapeutisch bei Infektionskrankheiten verwendeten Sulfonamiden vergleichbaren Effekt, so dass auch bei jahrelanger Medikation keine Resistenzerzeugung zu befürchten ist.
Aus dem gleichen Grunde treten auch Nebenerscheinungen, die auf eine Schädigung der Darmflora zurückzuführen sind, nicht auf.
Die Verfahrenserzeugnisse sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirkung zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise herangezogen werden :
Alkalische Mittel wie Alkali-oder Erdalkalihydroxyde,-carbonate oder-bicarbonate, ferner physiologisch verträgliche organische Basen.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Hilfs- und Trägerstoffe, wie Talkum, Stärke, Milchzucker, Traganth, Magnesiumstearat usw. enthalten.
