Verfahren zur Herstellung neuer Benzolsulfonylharnstoffe
Es ist bekannt, dass gewisse Benzolsulfonylharnstoff-Derivate blutzuckersenkende Eigenschaften aufweisen und somit als per os verabreichbare Antidiabetika geeignet sind (vgl. beispielsweise Arzneimittel-For schung, Band 8 (1958), Seiten 448-454). Insbesondere der N-(4-Methylbenzolsulfonyl)-N'-n-butyl-harnstoff hat aufgrund seiner guten blutzuckersenkenden Eigenschaften und seiner guten Verträglichkeit in der Diabetes-Therapie grosse Bedeutung erlangt.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer Benzolsulfonylharnstoffe der Formel

worin X Sauerstoff oder Schwefel und R1 einen gesättigten oder ungesättigten, gegebenenfalls durch Sauerstoff und/oder Schwefel unterbrochenen aliphatischen oder alicyclischen Kohlenwasserstoffrest mit 2-8 Kohlenstoffatomen bzw. bei ringförmigen Verbindungen mit 3-8 Kohlenstoffatomen oder einen Benzyl- oder p-Phenyläthylrest bedeuten, und deren Salzen, welche blutzuckersenkende Eigenschaften besitzen und auf Grund ihrer starken Wirksamkeit als oral verabreichbare Antidiabetika geeignet sind.
Das Verfahren ist dadurch gekennzeichnet, dass man in entsprechend substituierten Benzolsuifonylthio- harnstoffen der Formel

das Schwefelatom der Thioharnstoffgruppe mit den Schwefel durch Sauerstoff ersetzenden Mitteln behandelt. Man kann hierzu Oxyde oder Salze von Schwermetallen oder Oxydationsmittel verwenden.
Die erhaltenen Verbindungen können durch Behandlung mit Basen in die entsprechenden Salze übergeführt werden.
Als Oxydationsmittel eignen sich beispielsweise Wasserstoffperoxyd, Natriumperoxyd und salpetrige Säure.
Als Ausgangsstoffe kommen für das Verfahren gemäss der Erfindung p-Trifluormethoxy-und p-Trifluormethyl-mercaptobenzolsulfonyl-Thioharnstoffe in Betracht.
Zur Herstellung der Trifluormethoxy- bzw. Trifluormethylmercapto-benzolsulfonsäure-Derivate geht man zweckmässig von den bekannten Trifluormethoxybzw. Trifluormethyl-mercaptoanilinen aus, die man zunächst nach der Methode von Meerwein in die entsprechenden Sulfonsäurechloride überführt.
Hinsichtlich des Substituenten R1 kommen für das Verfahren Verbindungen in Betracht, die sich von folgenden Aminen ableiten: Äthyl-, n-Propyl-, Isopropylamin, Butylamin-(l), Butylamin-(2), 2-Methyl-propylamin-(l), 2-Methylpropylamin-(2), Pentylamin-(l), Pentylamin-(2), Pentylamin-(3), 3-Methylbutylamin-(l), Hexylamine, wie Hexylamin-(l) und 2-Methyl-pentylamin-(1), Heptylamine, wie Heptylamin-(l), Heptylamin-(4), Octylamine, wie Octylamin-(l). Weiterhin seien beispielsweise als Alkenylamine Allylamin und Crotylamin, als Cycloalkylamine, Cyclopentenylamin, Cycloheptylamin und 4-Methyl cyclohexylamin, 2,5 -Endomethylencyclohexylamin, Bornylamin und als Cycloalkylalkylamine Cyclohexylmethylamin und Cyclohexyläthylamin genannt.
Als aliphatische oder cycloaliphatische Verbindungen, die durch Sauerstoff oder Schwefel unterbrochen sind, kommen beispielsweise in Frage:
3-Methoxy-propylamin, 3 -Äthoxy-propylamin, 4-Methoxy-butylamin, Tetrahydro-a-furfurylamin, 3 -Methylmercaptopropylamin und 3 -Äthylmercapto propylamin. Weiterhin kommen in Frage: 2-Phenyläthylamin-(l) und Benzylamin.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinen hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise können die Umsetzungen unter Verwendung von Lösungsmitteln, bei Zimmertemperatur oder bei erhöhter Temperatur durchgeführt werden.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Benzolsulfonylharnstoffe stellen wertvolle Arzneimittel dar, die sich insbesondere durch gute blutzuckersenkende Wirksamkeit bei geringer Toxizität auszeichnen. Beispielsweise bewirkt der N-(4-Trifluor methylmercaptobenzolsulfonyl) -N' - cyclohexylharnstoff am Kaninchen in einer Dosierung von 400 mg/kg des Natriumsalzes per os eine Senkung des Blutzuckers um maximal 40 0, o. Die entsprechenden Werte betragen für den N-(4-Trifluormethoxy-benzolsulfonyl)-N'-cyclo- heptylharnstoff 300in, für den N-(4-Trifluormethoxybenzolsulfonyl)-N'-cyclooctylharnstoff 300/0 und für den N-(4-Trifluormethoxy-benzolsulfonyl)-N'-isobutyl- harnstoff 40 Oio.
Die Verfahrenserzeugnisse zeigen infolge des Fehlens einer p-ständigen Aminogruppe im Benzolkern keinen den therapeutisch bei Infektionskrankheiten verwendeten Sulfonamiden vergleichbaren Effekt, so dass auch bei jahrelanger Medikation keine Resistenzerzeugung zu befürchten ist. Aus dem gleichen Grunde treten auch Nebenerscheinungen, die auf eine Schädigung der Darmflora zurückzuführen sind, nicht auf.
Die Verfahrenserzeugnisse sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirkung zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. unter Zusatz von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise herangezogen werden:
Alkalische Mittel, wie Alkali- oder Erdalkalihydroxyde, -carbonate- oder -bicarbonate, ferner physiologisch verträgliche organische Basen.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die die Verfahrenserzeugnisse in einer Dosierung von 0,1 bis 0,5 Gramm und zusätzlich die üblichen Hilfs- und Trägerstoffe, wie Talkum, Stärke, Milchzucker, Traganth, Magnesiumstearat usw. enthalten.

N-(4-Trifluormethoxy-benzolsulfonyl)-N'-cyclo heptyl-harnstoff
30 g N-(4-Trifluormethoxy-benzolsulfonyl)-N'-cycloheptyl-thioharnstoff vom Schmelzpunkt 153-1550 C (hergestellt durch Umsetzung von 4-Trifluormethoxybenzolsulfonamid mit Cycloheptylsenföl in Gegenwart von gemahlenem Kaliumcarbonat und Aceton als Lösungsmittel) werden in 450 ml Aceton gelöst. Die Lösung wird bei einer Temperatur von ca. 60 C unter Rühren in kleinen Anteilen innerhalb von 45 Minuten mit einer Lösung von 5,2 g Natriumnitrit in 25 mol Wasser und anschliessend innerhalb von 60 Minuten mit 45 ml 5n-Essigsäure versetzt.
Man rührt 31/2 Stunden bei 0 bis +5 C nach, dampft den grössten Teil des Acetons unter vermindertem Druck ab, saugt den bei der Reaktion entstandenen elementaren Schwefel ab und verdünnt mit Wasser. Der ausgefallene kristalline Niederschlag wird abgesaugt und aus Methanol umkristallisiert. Die Ausbeute beträgt 22 g; Schmelzpunkt 141-143 C.
