Butyl. Ebenso kommen die entsprechenden Alkoxygruppen, wie
Methoxy, Äthoxy, n-Propoxy, Isopropoxy, n-Butoxy,
Isobutoxy oder tert. Butoxy, in Frage.
So beträgt z.
Process for the preparation of benzenesulfonylureas
It is already known that certain benzenesulfonyl urea derivatives have blood sugar-lowering properties and are therefore suitable as antidia betics which can be administered orally (see, for example, drug research, 8 [1958], pages 448-454). Especially the N- (4-methylbenzenesulfonyl) -N'-n-butyl-urea has gained great importance in diabetes therapy due to its good blood-sugar-lowering properties and its good tolerance.
It has now been found that benzenesulfonylureas of the formula
EMI1.1
wherein R is hydrogen, a halogen atom, an alkyl or. Alkoxy group with 1-4 carbon atoms or the trifluoromethyl group, R 'is an alicyclic radical of the formula
EMI1.2
n denotes the numbers 0 or 1 and -ZZ'- denotes the groupings -CH ° CHr or -CH = CH-, and the salts thereof have blood-sugar-lowering properties and are suitable as orally administrable antidiabetics due to their strong effectiveness.
The present invention relates to a process for the preparation of such benzenesulfonylureas, which is characterized in that benzenesulfonylthioureas of the formula
EMI1.3
treated with the sulfur atom by an oxygen atom replacing agents and, if necessary, treated the compounds obtained with alkaline agents such as alkali or alkaline earth metal hydroxides, carbonates, bicarbonates or organic bases. The correspondingly substituted benzenesulfonylthioureas are here prepared by known processes. From these, the sulfur is then in the usual way, for example with the help of oxides or salts of heavy metals, or by using oxidizing agents such as hydrogen peroxide, sodium peroxide and nitrous
Acid, eliminated.
Such benzenesulfonylthiourea are used as starting materials for the process according to the invention
Derivatives are considered, which preferably contain an alkylated or alkoxylated benzene radical. As alkyl groups are mentioned: methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl, sec. Butyl and tert. Butyl. The corresponding alkoxy groups, such as
Methoxy, ethoxy, n-propoxy, isopropoxy, n-butoxy,
Isobutoxy or tert. Butoxy, in question. Particularly suitable are: methyl, ethyl, methoxy and 2ithoxy groups in the p- and m-positions.
If, on the other hand, halogen-substituted benzenesulfonyl compounds are used as starting materials, those with a fluorine, chlorine or bromine atom or with the trifluoromethyl group are preferably suitable as substituents.
Benzenesulfonylthioureas which can be used are, for example, those which contain the radicals R1, l-amino -2,5-endomethylene-cyclohexyl, 1-amino-2,5-endomethylene-X3-cyclohexenyl, 1-aminomethyl-2, 5-endo methylenecyclohexyl or l -Aminomethyl- 2,5-endome- ethylene-X3-cyclohexyl wear.
The embodiments of the process according to the invention can be largely varied with regard to the reaction conditions and adapted to the respective conditions. For example, the reactions can be carried out using solvents, at room temperature or at elevated temperature. In order to obtain the process products in the purest possible form, the process product is taken up in very dilute ammonia, undissolved constituents are filtered off and the desired benzenesulfonyl urea is obtained by acidification.
The new benzenesulfonylureas obtainable by the process according to the invention are valuable medicaments which are distinguished in particular by their blood sugar-lowering effectiveness.
For example, N- (4-methylbenzenesulfonyl) -N'-2 ', 5'-endomethylene-8' -cyclohexen- (1 ') -yl] methyl urea is effective in rabbits at a dosage of 400 mg / kg orally a reduction in blood sugar by 30 to 40%. Corresponding results are obtained when testing the N- (4-methoxy-benzenesulfonyl) - N '- [2', 5'-endomethylene-d3'-cyclohexen- (l ') - yl] -methyl urea and N- (4-Ethoxy-benzenesulfonyl) -N '-. [2', 5'-endomethylene -'-cyclohexen- (1 ') - yl] -methyl-urea. The numerical values given were obtained by comparing control animals kept similarly. The blood sugar values were determined by hourly analyzes according to Hagedorn / Jensen.
The good blood sugar-lowering effect of the new process products can also be demonstrated if the test is carried out at a considerably lower dose (threshold dose). For example, the N- (4-methylbenzenesulfonyl) -N '- [2', 5'-endomethylene-Z13-cyclohexen- (1 ') - yl] methylurea in a test on rabbits Dose of 50 mg / kg reduced blood sugar by 28%. This value is retained for 6 hours.
Furthermore, the N- (4-bromo-benzenesulfonyl) -N '[2', 5'-endomethylene-cyclohexyl- (1)] - urea, when administered intravenously to rabbits, already lowers blood sugar at a threshold dose of 5 mg / kg, while the corresponding threshold dose for intravenous administration to rabbits for the known N- (4-methylbenzenesulfonyl) -N'-n-butylurea is 20 mg / kg. The new compound thus proves to be four times more effective than the known compound with regard to the intravenous threshold dose in rabbits. The N- (4-chloro-benzenesulfonyl) -N '- [2', 5 '- endomethylene cyclohexyl- (1) 3-urea is already effective at a threshold dose of 2.5 mg / kg when administered intravenously to rabbits.
In this particular test, the new compound is therefore about eight times as effective as the well-known N- (4-methyl-benzenesulfonyl) -N'-n-butyl urea.
For a long-term therapeutic, such as the oral anti-diabetic agents, it is of great importance that the compounds used are not very toxic; these requirements are met by the new process products. So z. B. the LD50 per os on the white mouse for N- (4-methylbenzenesulfonyl) -N'-t2 ', 5'-endomethylene-A3, -cyclohexen- (1') -yl] -methyl-urea more than 7 g / kg.
As a result of the lack of a p-amino group in the benzene nucleus, the products of the process show no effect comparable to the sulfonamides (used therapeutically in infectious diseases), so that no resistance is to be feared even after years of medication. For the same reason, there are no side effects that can be traced back to a disturbance in the intestinal flora.
The products of the process should preferably be used for the production of orally administrable preparations with a blood sugar lowering effect for the treatment of diabetes mellitus and can be applied as such or in the form of their salts or of substances which lead to salt formation. For salt formation, for example, the following can be used: alkaline agents such as alkali or alkaline earth metal hydroxides, carbonates or bi-carbonates, as well as physiologically compatible organic bases. Tablets which, in addition to the products of the process, contain the usual auxiliaries and carriers, such as talc, starch, lactose, tragacanth, magnesium stearate, etc., are preferred as medical preparations.
example
33.6 g of N- (4-methyl-benzenesulfonyl) -N '- [2', 5'-endo methylene-dg '-cyclohexenyl- (1') - methyl] thiourea (prepared by reacting p-toluenesulfonamide with (2,5-Endomethylene-J3'-cyclohexenyl- (l ') - methyl mustard oil with a boiling point of 120-1230 C in the presence of ground potassium carbonate and acetone) are dissolved in acetone and with stirring at room temperature with a solution of 7 6 g of sodium nitrite in a little water are added, 72 ml of 5N acetic acid are then added dropwise to the mixture, which has been cooled to 50 ° C. After stirring for three hours at 5-100 ° C., the precipitated sulfur is suctioned off and the solution is concentrated under reduced pressure.
The precipitate of N- (4 methylbenzeneszònyl) -N "-12", 5 "-endomet 8 'cyclohexen- (l') - yl] -methylurea is sucked off, rubbed with water, sucked off again and unicrystallized from methanol: Melting point 175-1770 C.
