Verfahren zur Herstellung von Sulfonylharnstoffen
Es wurden bereits Verbindungen der Formel R"¯SO,-NH-CO-NH-R", worin R"' einen gegebenenfalls durch ein oder zwei Alkyl- oder Alkoxyreste mit vorzugsweise bis zu 8 Kohlenstoffatomen oder durch Halogen substituierten Phenylrest oder einen aliphatischen bzw. cycloaliphatischen Kohlenwasserstoffrest mit 3-8 Kohlenstoffatomen oder einen Naphthalin-(2)-, einen 5 ,6,7,8-Tetrahydronaphthalin-(2)- oder einen 4-Phenoxy-phenylrest und R" einen Alkyl-, Alkenyl-, Cycloalkyl- oder Cycloalkylalkylrest mit 2-8 Kohlenstoffatomen bedeuten, als wertvolle Arzneimittel mit blutzuckersenkender Wirkung vorgeschlagen.
Es wurde nun gefunden, dass auch Verbindungen der Formel R-SO,NH-CO-NH-R', worin R einen gegebenenfalls durch Alkyl- und/oder Alkoxyreste mit 1-8 Kohlenstoffatomen undloder Halogen substituierten Phenylrest, einen aliphatischen bzw. cycloaliphatischen Kohlenwasserstoffrest mit 3-8 Kohlenstoffatomen, einen Naphthalin-(2)-, 5 ,6,7,8-Tetrahydronaphthalin-(2)- oder einen 4 Phenoxy-phenylrest und R' einen Rest der Formel -(CH2) l,-C6H5, wobei n für eine ganze Zahl von 1 bis 4 steht, bedeuten, und deren Salze wertvolle Arzneimittel darstellen und sich insbesondere durch eine starke und langanhaltende Senkung des Blutzuckerwertes sowie durch eine sehr geringe Toxizität auszeichnen.
Im einzelnen können für R folgende Reste stehen: Phenyl und durch Alkyl oder Alkoxy monosubstituierte Phenylreste, wie Methylphenyl, insbesondere p-Methyl-phenyl, Äthylphenyl, Propylphenyl, Butylphenyl, Pentylphenyl und Hexylphenyl.
Die Alkyl- oder Alkoxy-Substituenten im Phenylrest können sowohl geradkettig als auch verzweigt sein; ausser in p-Stellung können sie auch an anderen Stellen, insbesondere in Stellung, des Phenylrestes gebunden sein. Weiterhin kommen für R in Betracht: Halogenphenylreste, wie Chlorphenyl und Bromphenyl. Ferner seien durch Alkyl undloder Alkoxy disubstituierte Phenylreste, wie Dialkyl-, Dialkoxy- oder Alkyl-alkoxyphenylreste, erwähnt. Ausserdem können auch Phenylreste, die sowohl eine Alkyl- und/oder Alkoxygruppe als auch ein Halogen gleichzeitig enthalten, beispielsweise Methyl-chlorphenyl, herangezogen werden. Auch die Halogensubstituenten können sich in beliebiger Stellung am Benzolkern befinden.
Schliesslich kann R für Alkylreste, beispielsweise für einen Propyl-, Butyl-, Pentyloder Hexylrest, für Cycloalkylreste, beispielsweise für den Cyclohexylrest, und für Cycloalkylalkylreste, beispielsweise für den Hexahydrobenzylrest, stehen. Soweit es sich dabei um aliphatische Reste handelt, können diese sowohl geradkettig als auch verzweigt sein.
Das erfindungsgemässe Verfahren zur Herstellung solcher Verbindungen ist dadurch gekennzeichnet, dass man Verbindungen der Formel RSO2-Hat mit Isoharnstoffäthern der Formel

umsetzt, wobei R2 für einen aliphatischen Kohlenwasserstoffrest steht und Hal ein Halogen bedeutet, und die Kondensationsprodukte hydrolysiert.
Die Reaktion der Sulfonsäurehalogeniden der Formel R-SO2-Hal mit den Isoharnstoffäthern kann man beispielsweise im Falle der Verwendung von festen Sulfonsäurehalogeniden in wässrigem Medium in Gegenwart von Kaliumcarbonat durchführen.
Nach einer vorteilhaften Ausführungsform des Verfahrens setzt man die Isoharnstoffäther in Form von Salzen in wässriger acetonischer Lösung und unter Kühlung mit den Sulfonsäurehalogeniden in Gegenwart von Alkalilaugen um. Die aus der Lösung ausfallenden Sulfonylisoharnstoffäther werden zum Beispiel abgesaugt und gegebenenfalls aus verdünnten Alkoholen umkristallisiert. Sie werden beispielsweise mit konzentrierten Säuren, vorzugsweise Salzsäure, auf etwa 60-100" C erwärmt, wobei Gasentwicklung beobachtet werden kann.
Als Alkylreste, die zum Beispiel ein- oder zweimal als Substituenten im Phenylrest, gegebenenfalls über Sauerstoff gebunden, auftreten können, kommen vorzugsweise Reste mit niederem Molekulargewicht in Betracht. Mit besonderem Vorteil verwendet man Reste mit 1 bis 6 Kohlenstoffatomen.
Man kann auch Reste mit bis zu 8 Kohlenstoffatomen heranziehen, jedoch geht die Wirksamkeit der Verfahrenserzeugnisse darüber hinaus stark zurück.
Anstelle der gegebenenfalls, z. B. durch Alkyl- oder Alkoxyreste, substituierten Benzolsulfonylhalogenide kann man auch entsprechende aromatische Verbindungen heranziehen, die im Phenylrest ein- oder zweimal durch Halogen, vorzugsweise Chlor oder Brom, bzw. durch ein Halogen neben einem Alkyloder Alkoxyrest substituiert sind. Als Ausführungsformen des Verfahrens zur Herstellung der genannten Sulfonylharnstoffe kommen auch bei halogensubstituierten Benzolsulfonylhalogeniden die gleichen in Frage, wie sie oben beschrieben sind.
Auch zur Herstellung von Sulfonylharnstoffen der angegebenen Formel, worin R einen 4-Phenoxyphenyl-, Naphthalin-(2)- oder 5,6,7,8-Tetrahydro- naphthalin-(2)-rest bedeutet, können die erwähnten Ausführungsformen herangezogen werden.
Bei den für das Verfahren gemäss der Erfindung verwendeten Ausgangsstoffen handelt es sich vielfach um literaturbekannte Verbindungen. Als Ausgangsstoffe seien beispielsweise genannt:
Benzolsulfonsäurehalogenide,
4-Methyl-benzolsulfonsäurehalogenide, 4-Athyl-benzolsulfonsäurehalogenide,
4-n-Propyl-benzolsulfonsäurehalogenide,
4-Isopropyl-benzolsulfonsäurehalogenide, 4-n-Butyl-benzolsulfonsäurehalogenide,
4-tert.-Butyl-benzolsulfonsäurehalogenide,
4-n-Hexyl-benzolsulfonsäurehalogenide,
4-Methoxy-benzolsulfonsäurehalogenide, 4-Äthoxy-benzolsulfonsäurehalogenide,
4-n-Butoxy-benzolsulfonsäurehalogenide.
Anstelle solcher Verbindungen, die im Benzolkern in 4-Stellung substituiert sind, können auch die ent sprechenden, in 2- oder besser 3-Stellung substi tuierten Verbindungen eingesetzt werden.
Weiterhin kommen als Ausgangsstoffe in Be tracht: Halogenbenzolsuifonsäurehalogenide, vorzugsweise die Chlor- und Bromverbindungen, wobei sich das Halogenatom in beliebiger Stellung im Benzolkern befinden kann. Weiterhin können als Ausgangsstoffe dienen: Dihalogenbenzolsulfonsäurehalogenide, wobei sich die Halogenatome in beliebiger Stellung am Benzolkern befinden können, oder entsprechend disubstituierte Benzolsulfonsäurehalogenide, beispielsweise Dimethyl -benzolsulfonsäurehalogenide, Di methoxy-benzolsulfonsäurehalogenide oder die entsprechenden Alkyl-halogenbenzol-, Alkoxy-halogenbenzol- und Alkoxy-alkyl-benzolsulfonsäurehalogenide.
Ferner kommen in Betracht entsprechende Alkyl-, Cycloalkyl-, Cycloalkylalkyl-, die Naphthalin (2), 5,6,7,8-Tetrahydronaphthalin-(2)- 4-Phen- oxy-phwnylsulfonsäurehalogenide. Von den Alkyl-, Cycloalkyl- und Cycloalkylalkyl-sulfonylhalogeniden seien diejenigen genannt, die beispielsweise folgende Reste R enthalten: Propyl, Butyl-(l)-, Butyl-(2)-, 2-Methyl-propyl-(1)-, Pentyl-(2), Pentyl-(3)-, 3-Me thyl-butyl-( 1)-, 2-Methyl-butyl-(2)-, Hexyl-( 1)-, Cy clohexyl und Cyclohexylmethyl.
Für die Umsetzung mit den vorstehend genannten Verbindungen können erfindungsgemäss N-(l,- Phenyl--äthyl)-isoharnstoffäther, N-(;,-Phenyl-propyl)isoharnstoffäther und N-(y-Phenyl-butyl)-isoharn- stoffäther herangezogen werden.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Verbindungen zeichnen sich durch grosse Stabilität aus. Gegenüber den in der Chemotherapie zur Bedeutung gelangten Aminobenzolsulfonamiden ist besonders ihre Beständigkeit gegenüber oxydierenden Einflüssen bemerkenswert.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Verbindungen sind wertvolle Arzneimittel und zeichnen sich insbesondere durch eine be achtliche blutzuckersenkende Wirkung aus. Sie unterscheiden sich von den bekannten Aminobenzolsulfonamiden vor allem auch dadurch, dass sie infolge Fehlens einer p-ständigen Aminogruppe keine den Sulfanilamiden vergleichbare chemotherapeutische Wirkung besitzen. So wird beispielsweise die Darmflora nicht beeinflusst, und weiterhin ist eine bei dem Dauergebrauch zu befürchtende Gewöhnung pathogener Keime nicht beobachtet worden. Die neuen Verbindungen lassen sich auf einfachere Weise herstellen als die bekannten Aminobenzolsulfonylharnstoffe.
Pharmakologische Versuche am Kaninchen haben ergeben, dass beispielsweise die Verfütterung von 400 mg N-Cyclohexansulfonyl-N'-(-phenyl-äthyl)- harnstoff in Form des Natriumsalzes pro kg und per os eine Senkung des Blutzuckerspiegels um durchschnittlich 45 o/O bewirkt. Nach Verfütterung von beispielsweise N-(3,4-Dimethyl-benzolsulfonyl) N'-(, B-phenyl-äthyl)-harnstoff beobachtet man eine Senkung des Blutzuckerspiegels um durchschnittlich 40 ao. Die am Kaninchen gemachten Beobachtungen konnten durch Untersuchungen an anderen Versuchstieren bestätigt werden.
Verfüttert man beispielsweise den nach dem Verfahren gemäss der Erfindung erhältlichen N-(4-Methyl-benzolsulfonyl) - N'-(, 6-phenyl-äthyl)-harnstoff in einer Dosis von 100 mg/kg und per os am nüchternen Hund, so beobachtet man folgende Senkungen des Blutzuckerwertes in Prozent:
33 O/o nach 2 Stunden, 37 ovo nach 3 Stunden,
37 6/o nach 6 Stunden,
15-20 O/o nach 24 Stunden, 0 8/o nach 48 bzw. 72 Stunden
Weiterhin führt beispielsweise der N-[Naphtha lin-(2)-sulfonyl]-N' -phenyl-äthyl)-harnstoff beim Kaninchen zu einer Blutzuckersenkung von 45 8/0, der N-(4-Isopropyl - benzolsulfonyl) - (, 6-phenyl- äthyl)-harnstoff zu einer Blutzuckersenkung von ebenfalls 45 O/o, der N-(4-Methyl-benzolsulfonyl)-N'- (3'-phenyl-propyl)-harnstoff zu einer Blutzuckersenkung von 40 O/o und der N-(3-Chlor-4-methyl-ben zolsulfonyl)
-N'-(fl-phenyl - äthyl) - harnstoff zu einer Blutzuckersenkung von 35 O/o.
Die obigen Werte wurden durch Vergleich mit Blutzuckerwerten gleichartig gehaltener, nicht behandelter Kontrolltiere ermittelt. Die Blutzuckerwerte können durch stündliche Analysen nach Hagedorn Jensen ermittelt werden.
Die nach dem Verfahren der vorliegenden Erfindung erhältlichen Produkte sind sehr wenig toxisch.
So beträgt beispielsweise die LD10 an der weissen Maus für den nach dem Verfahren gemäss der Erfindung erhältlichen N-(4-Methyl-benzolsulfonyl)-N' (F;-phenyl-äthyl)-harnstoff 5,5-6 g/kg.
Die Verfahrensprodukte sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirkung zur Behandlung des Diabetes mellitus dienen.
Die genannten Verfahrensprodukte können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, Verwendung finden. Zur Salzbildung können beispielsweise herangezogen werden: Ammoniak, alkalische Mittel, wie Alkali- oder Erdalkalihydroxyde, Alkalicarbonate oder -bicarbonate, ferner physiologisch verträgliche organische Basen.

The reaction of the sulfonic acid halides of the formula R-SO2-Hal with the isourea ethers can be carried out, for example, in the case of using solid sulfonic acid halides in an aqueous medium in the presence of potassium carbonate.

The compounds obtainable by the process according to the invention are distinguished by great stability. Compared to the aminobenzenesulfonamides that have become important in chemotherapy, their resistance to oxidizing influences is particularly remarkable.
The compounds obtainable by the process according to the invention are valuable medicaments and are distinguished in particular by a considerable blood sugar lowering effect. They differ from the known aminobenzenesulfonamides above all in that they have no chemotherapeutic effect comparable to the sulfanilamides due to the lack of an amino group in the p position. For example, the intestinal flora is not influenced, and furthermore, habituation to pathogenic germs, which is to be feared during long-term use, has not been observed. The new compounds can be prepared in a simpler manner than the known aminobenzenesulfonylureas.
Pharmacological tests on rabbits have shown that, for example, feeding 400 mg of N-cyclohexanesulphonyl-N '- (- phenyl-ethyl) - urea in the form of the sodium salt per kg and per os reduces the blood sugar level by an average of 45%. After feeding with, for example, N- (3,4-dimethyl-benzenesulfonyl) N '- (, B-phenyl-ethyl) -urea, a decrease in blood sugar level by an average of 40 ao is observed. The observations made on rabbits could be confirmed by studies on other test animals.
If, for example, the N- (4-methyl-benzenesulfonyl) -N '- (6-phenyl-ethyl) -urea obtainable by the process according to the invention is fed in a dose of 100 mg / kg and per os on an empty dog, so the following reductions in blood sugar levels can be observed in percent:

The above values were determined by comparison with blood sugar values of similarly kept, untreated control animals. The blood sugar values can be determined by hourly analyzes according to Hagedorn Jensen.
The products obtainable by the process of the present invention are very low in toxicity.
For example, the LD10 on the white mouse for N- (4-methylbenzenesulfonyl) -N '(F; -phenyl-ethyl) -urea obtained by the method according to the invention is 5.5-6 g / kg.
The products of the process should preferably be used for the production of orally administrable preparations with a blood sugar lowering effect for the treatment of diabetes mellitus.
