Verfahren zur Herstellung von Sulfonylharnstoffen
Es wurden bereits Verbindungen der Formel R"'40-NH-CO-NH-R", worin R"' einen gegebenenfalls durch ein oder zwei Alkyl-oder Alkoxyreste mit vorzugsweise bis zu 8 Kohlenstoffatomen oder durch Halogen substituierten Phenylrest oder einen aliphatischen bzw. cycloaliphatischen Kohlenwasserstoffrest mit 3 bis 8 Koh- lenstoffatomen oder einen Naphthalin- (2)-, einen 5, 6, 7, 8-Tetrahydronaphthalin- (2)- oder einen 4 Phenoxy-phenylrest und R"einen Alkyl-, Alkenyl-, Cycloalkyl-oder Cycloalkylalkylrest mit 2 bis 8 Koh lenstoffatomen bedeuten, als wertvolle Arzneimittel mit blutzuckersenkender Wirkung vorgeschlagen.
Es wurde nun gefunden, dass auch Verbindungen der Formel R-SO2-NH-CO-NH-R', worin R einen gegebenenfalls durch Alkyl-und/oder Alkoxyreste mit 1 bis 8 Kohlenstoffatomen und/oder Halogen substituierten Phenylrest, einen aliphatischen bzw. cycloaliphatischen KoMenwasserstoffrest mit 3 bis 8 Kohlenstoffatomen, einen Naphthalin- (2)-, 5, 6, 7, 8-Tetrahydfonaphthalin- (2) oder einen 4 Phenoxy-phenylrest und R'einen Rest der Formel -(CH2)n-C6H5, wobei n für eine ganze Zahl von 1 bis 4 steht, bedeuten, und deren Salze wertvolle Arzneimittel darstellen und sich insbesondere durch eine starke und langanhaltende Senkung des Blutzuckerwertes sowie durch eine sehr geringe Toxizität auszeichnen.
Im einzelnen können für R folgende Reste stehen : Phenyl und durch Alkyl oder Alkoxy monosubsti- tuierte Phenylreste, wie Methylphenyl, insbesondere p-Methyl-phenyl, Athylphenyl, Propylphenyl, Butylphenyl, Pentylphenyl und Hexylphenyl. Die Alkyloder Alkoxy-Substituenten im Phenylrest können sowohl geradkettig als auch verzweigt sein ; ausser in p-Stellung können sie auch an anderen Stellen, insbesondere in m-Stellung, des Phenylrestes gebunden sein. Weiterhin kommen für R in Betracht : Halogenphenylreste, wie Chlorphenyl und Bromphenyl. Ferner seien durch Alkyl und/oder Alkoxy disubstituierte Phen, ylreste, wie Dialkyl-, Dialkoxy-oder ALkyl alkoxy-phenylreste erwähnt.
Ausserdem können auch Phenylreste, die sowohl eine Alkyl-und/oder Alkoxy- gruppe als auch ein Halogen gleichzeitig enthalten, beispielsweise Methyl-chlor-phenyl, herangezogen werden. Auch die Halogensubstituenten können sich in beliebiger Stellung am Benzolkem befinden.
Schliesslich kann R für Alkylreste, beispielsweise fur einen Propyl-, Butyl-, Pentyl-, Hexylrest, für Cycloalkylreste, beispielsweise für den Cyclohexylrest, und für Cycloalkylalkylreste, beispielsweise für den Hexahydrobenzylrest, stehen. Soweit es sich dabei um ali phatische Reste handelt, können diese sowohl geradkettig als auch verzweigt sein.
Das erfindungsgemässe Verfahren zur Herstellung solcher Verbindungen ist dadurch gekennzeichnet, dass man Thioharnstoffe der Formel R-SO2-NH-CS-NH-R' mit den Schwefel durch Sauerstoff ersetzenden Mitteln behandelt.
Nach dem Verfahren gemäss der Erfindung kann man zur Entschwefelung der Sulfonylthioharnstoffe Schwermetalloxyde in Gegenwart von Lösungsmitteln verwenden ; dabei kann es von Vorteil sein, anstelle der Schwermetalloxyde entsprechende Metallsalze, beispielsweise Bleil, Kupfer-oder Silbersalze, einzusetzen. Als weitere übliche Methode zur Entschwefe- lung kann man auch die Oxydation der Sulfonylthio- harnstoffe mit Natriumperoxyd oder salpetriger Säure durchführen.
Als Alkylreste, die, z. B. ein-oder zweimal, als Substituenten im Phenylrest, gegebenenfalls über Sauerstoff gebunden, auftreten können, kommen vorzugsweise Reste mit niederem Molekulargewicht in Betracht. Mit besonderem Vorteil verwendet man Reste mit 1 bis 6 Kohlenstoffatomen. Man kann auch Reste mit bis zu 8 Kohlenstoffatomen heranziehen, jedoch geht die Wirksamkeit der Verfahrenserzeugnisse darüber hinaus stark zurück. Anstelle der gege benenfalls durch Alkyl-oder Alkoxyreste substituier- ten Benzolsulfonyl-Verbindungen kann man auch solche aromatische Verbindungen heranziehen, die im Phenylrest ein-oder zweimal durch Halogen, vorzugsweise Chlor oder Brom, bzw. durch ein Halogen neben einem Alkyl-oder Alkoxyrest substituiert sind.
Als Ausführungsformen des Verfahrens zur Herstellung der genannten Sulfonylharnstoffe kommen auch bei halogensubstituierten Benzolsulfonyl-Verbindun- gen die gleichen in Frage, wie sie oben beschrieben sind.
Auch zur Herstellung von Sulfonylharnstoffen der angegebenen Formel, worin R einen 4-Phenoxy phenyl-, Naphthalin- (2)- oder 5, 6, 7, 8-Tetrahydro naphthalin-(2)-rest bedeutet, können die erwähnten Ausführungsformen herangezogen werden.
Bei den für das Verfahren gemϯ der Erfindung verwendeten Ausgangsstoffen handelt es sich vielfach um literaturbekannte Verbindungen. Als Ausgangsstoffe seien beispielsweise auf der der Sulfonylgruppe abgewandten Seite der Thioharnstoffgruppierung durch z. B. den ss-Phenyl-äthyl-, y-Phenyl-propyl- oder γ-Phenyl-butylrest substituierte nachstehende Verbindungen genannt :
Benzolsulfonylthioharnstoffe, 4 - Methyl - benzolsulfonylthioharnstoffe, 4-Athyl benzolsulfonylthioharnstoffe, 4-n-Propyl-benzolsulfo nylthioharnstoffe, 4-Isopropyl-benzolsulfonylthioharnstoffe, 4-n-Butyl-benzolsulfonylthioharnstoffe, 4-tert.-Butyl-benzolsulfonylthioharnstoffe, 4-n-Hexyl benzolsulfonylthioharnstoffe, 4-Methoxy-benzolsulfo nylthioharnstoffe, 4-Athoxy-benzolsulfonylthioham- stoffe, 4-n-Butoxy-benzolsulfonylthiohamstoffe. Anstelle solcher Verbindungen, die im Benzolkern in 4-Stellung substituiert sind, können auch die entsprechenden in 2-oder besser 3-Stellung substituierten Verbindungen eingesetzt werden.
Weiterhin kommen als Ausgangsstoffe in Betracht : entsprechende HaIogenbenzoIsulf onylthioharnstoffe, vorzugsweise die Chlor-und Bromverbindungen, wobei sich das Halogen in beliebiger Stellung im Benzolkern befinden kann.
Weiterhin können als Ausgangsstoffe dienen : entsprechende Dihalogenbenzolsulfonylthioharnstoffe, wobei sich die Halogenatome in beliebigor Stellung am Benzolkem befinden können. Ebenso können die entsprechenden Dimethylbenzolsulfonyl-Verbindun- gen oder auch die entsprechenden Dimethoxy-benzol- sulfonyl-Verbindungen oder Alkyl-halogen-benzol-, Alkoxy-halogen-benzol- und Alkoxy-alkyl-benzolsul fonylthiohamstoffe als Ausgangsstoffe verwendet werden. Ferner kommen in Betracht : entsprechende Alkyl-, Cycloalkyl-, Cycloalkylalkyl-, Naphthalin- (2)-, 5, 6, 7, 8-Tetrahydronaphthalin- (2)- und 4 Phenoxy-phenyl-sulfonylthioharnstoffe.
Von den Alkyl-, Cycloalkyl- und Cycloalkylalkyl-sulfonylthio- harns. toffen ! seien diejenigen genannt, die beispielsweise folgende Reste R enthalten : Propyl-, Butyl- (l)-, Butyl-(2)-, 2-Methyl-propyl-(1)-, Pentyl-(2)-, Pentyl (3)-, 3-Methyl-butyl-(1)-, 2-Methyl-butyl-(2)-, Hexyl (1)-, Cyclohexyl und Cyclohexylmethyl.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Verbindungen zeichnen sich durch grosse Stabilität aus. Gegenüber den in der Chemotherapie zur Bedeutung gelangten Aminobenzolsulfonamiden ist insbesondere ihre Beständigkeit gegenüber oxydierenden Einflüssen bemerkenswert.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Verbindungen sind wertvolle Arzneimittel und zeichnen sich insbesondere durch eine beachtliche blutzuckersenkende Wirkung aus. Sie unterscheiden sich von den bekannten Aminobenzolsulfonami- den vor allem auch dadurch, dass sie infolge Fehlens einer p-ständigen Aminogruppe keine den Sulfanilamiden vergleichbare chemotherapeutische Wirkung besitzen. So wird beispielsweise die Darmflora nicht beeinflusst und weiterhin ist eine bei dem Dauergebrauch zu befürchtende Gewöhnung pathogener Keime nicht beobachtet worden. Die neuen Verbindungen lassen sich auf einfachere Weise herstellen als die bekannten Aminobenzolsulfonylharnstoffe.
Pharmakologische Versuche am Kaninchen haben ergeben, dass beispielsweise die Verfütterung von 400 mg N-Cyclohexansulfonyl-N'- (j-phenyl-äthyl)- harnstoff in Form des Natriumsalzes pro kg und per os eine Senkung des Blutzuckerspiegels um durch schnittlich 45 /o bewirkt. Nach Verfütterung von beispielsweise N- (3, 4-Dime-thyl-benzolsulfonyl)-N'-(4- phenyl-äthyl)-harnstoff beobachtet man eine Senkung des Blutzuckerspiegels um durchschnittlich 400/o. Die am Kaninchen gemachten Beobachtungen konnten durch Untersuchungen an anderen Versuchstieren be stätigt werden.
Verfüttert man beispielsweise den nach dem Verfahren gemäss der Erfindung erhältlichen N- (4-Methyl-benzolsulfonyl) !- N- (jss-phenyl-äthyl)- harnstoff in einer Dosis von 100 mg pro kg und per os am nüchternen Hund, so beobachtet man folgende Senkung des Blutzuckerwertes in Prozent : 33 O/o nach zwei Stunden 370/o nach drei Stunden 37 /o nach sechs Stunden 15-200/9 nach 24 Stunden O'/o. nach 48 bzw. 72 Stunden.
Weiterhin führt beispielsweise der N- [Naphthalin- (2)-sulfonyl]-N'- (p-phenyl-äthyl3-harnstoff beim Kaninchen zu einer Blutzuckersenkung von 45 /o, der N- (4-Isopropyl-benzolsulfonyl)-N'-(¯-phenyl-Ïthyl)hamstoff zu einer Blutzuckersenkung von ebenfalls 45 /o, der N- (4-Methyl-benzolsulfonyl)-N'- (3'-phenyl- propyl)-harnstoff zu einer Blutzuckersenkung von 40 < '/o und der N- (3-Chlor-4-methyl-benzolsulfonyl) N'-(¯-phenyl-Ïthyl)-harnstoff zu einer Blutzuckersenkung von 35%.
Die obigen Werte wurden durch Vergleich mit Blutzuckerwerten gleichartig gehaltener, nicht beL- handelter Kontrolltiere ermittelt. Die Blutzuckerwerte können durch stündliche Analysen nach Hagedorn/ Jensen ermittelt werden.
Die nach dem Verfahren der vorliegenden Erfindung erhältlichen Produkte sind sehr wenig toxisch.
So beträgt beispielsweise die LD50 an der weissen Maus f r den nach dem Verfahren gemäss der Erfindung erhältlichen N- (4-Methyl-benzolsulfonyl)-N'- (4-phenyl-äthyl)-harnstoff 5, 5 bis 6 g/kg.
Die Verfahrensprodukte sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirkung zur Behandlung d'es Diabetes mellitus dienen.
Die genannten Verfahrensprodukte können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, Verwendung finden. Zur Salzbildung können beispiels- weise herangezogen werden : Ammoniak, alkalische Mittel, wie Alkali-oder Erd'alkalihydroxyde, Alkalicarbonate oder-bicarbonate, ferner physiologisch verträgliche organische Basen.
In dem britischen Patent Nr. 604259 wurden Sulfonylharnstoffe der Formel

worin X eine Aminogruppe oder einen in eine Aminogruppe überführbaren Rest und Aryl einen substi- tuierten oder unsubstituierten Benzylrest bedeuten, beschrieben. Die Prüfung dieser Verbindungen, ins besondere des N- (4-Amino-benzolsulfonyl)-N'-benzyl- hamstoffes, hat ergeben, dass ihnen keine analoge blutzuckersenkende Wirkung zukommt, wie sie die Verbindungen der vorliegenden Erfindung in eindeutiger Weise besitzen. Es wurde ferner festgestellt, dal3 der N-Benzyl-sulfonyl-N'-butyl-harnstoff keine blutzuckersenkende Wirkung besitzt.
Daher war nicht zu erwarten, da¯ den vorliegenden Verfahrenserzeugnissen eine so gute antidia- betische Wirkung zukommen w rde. Vor allem war nicht damit zu rechnen, dass die vorliegenden Verbindungen trotz des Vorhandenseins von Arylalkylresten eine so geringe Toxizität besitzen wurden.
Beispiel
N-(4-Methyl-benzolsulfonyl)-N'-(¯-phenyl-Ïthyl) harnstoff
16, 7 g N-(4-Methyl-benzolsulfonyl)-N'-(¯-phenyl äthyl)-thio-harnstoff [hergestellt durch Umsetzung von p-Toluolsulfonamid-natrium mit jB-Phenyl-äthyl- senföl in acetonischer Lösung und in fast quantitativer Ausbeute ; Schmelzpunkt 144 bis 146 C (nach Umkristallisieren aus Methanol)] werden in 400 cm3 Aceton gelöst. Diese Lösung versetzt man unter Rühren mit 3, 8 g Natriumnitrat, gelöst in 25 cm3 Wasser. Die erhaltene Lösung wird auf etwa 5 C abgekühlt und ungeachtet einer geringfügigen Kristallabscheidung unter Rühren im Verlauf von einer Stunde tropfenweise mit 30 cm3 Sn-Essigsäure versetzt.
Man riihrt anschliessend noch drei Stunden bei Zimmertemperatur nach, verdünnt mit 21 Wasser und saugt den ausgefallenen Niederschlag ab. Dieser wird mit verdünntem Ammoniak (etwa 1 : 60) behandelt, der bei der Reaktion entstandene elementare Schwefel abfiltriert und das noch trübe Filtrat 20 Minuten mit Kohle auf dem Dampfbad erhitzt.
Die nach Filtration resultierende klare Lösung wird mit Salzsäure versetzt, wobei der gewünschte rohe Sulfonylharnstoff in fast quantitativer Ausbeute ausfällt. Nach dem Umkristallisieren aus Methanol erhält man 12 g N-(4-Methyl-benzolsulfonyl)-N'-(¯ phenyl-äthyl)-harnstoff in Form weisser Kristalle und vom Schmelzpunkt I45 bis 147 C.

The compounds obtainable by the process according to the invention are distinguished by great stability. Compared to the aminobenzenesulfonamides which have become important in chemotherapy, their resistance to oxidizing influences is particularly remarkable.
The compounds obtainable by the method according to the invention are valuable medicaments and are distinguished in particular by a considerable blood sugar-lowering effect. They differ from the known aminobenzenesulfonamides above all in that, due to the lack of an amino group in the p position, they do not have any chemotherapeutic effect comparable to that of sulfanilamides. For example, the intestinal flora is not influenced, and furthermore, habituation to pathogenic germs, which is to be feared during long-term use, has not been observed. The new compounds can be prepared in a simpler manner than the known aminobenzenesulfonylureas.
Pharmacological tests on rabbits have shown that, for example, feeding 400 mg of N-cyclohexanesulfonyl-N'- (j-phenyl-ethyl) -urea in the form of the sodium salt per kg and per os reduces the blood sugar level by an average of 45% . After feeding with, for example, N- (3, 4-dimethyl-benzenesulfonyl) -N '- (4-phenyl-ethyl) -urea, a reduction in blood sugar level by an average of 400% is observed. The observations made on rabbits could be confirmed by studies on other test animals.

The above values were determined by comparison with blood sugar values of similarly kept, untreated control animals. The blood sugar values can be determined by hourly analyzes according to Hagedorn / Jensen.
The products obtainable by the process of the present invention are very low in toxicity.
For example, the LD50 on the white mouse for the N- (4-methylbenzenesulfonyl) -N′- (4-phenylethyl) urea obtainable by the method according to the invention is 5.5 to 6 g / kg.
The products of the process should preferably be used for the production of orally administrable preparations with a blood sugar-lowering effect for the treatment of diabetes mellitus.
The process products mentioned can be used as such or in the form of their salts or in the presence of substances which lead to salt formation. For salt formation, for example, the following can be used: ammonia, alkaline agents, such as alkali or alkaline earth metal hydroxides, alkali carbonates or bicarbonates, and also physiologically compatible organic bases.
In British Patent No. 604259, sulfonylureas have the formula

where X is an amino group or a radical which can be converted into an amino group and aryl is a substituted or unsubstituted benzyl radical. Testing of these compounds, in particular of N- (4-amino-benzenesulfonyl) -N'-benzylurea, has shown that they have no blood sugar-lowering effect analogous to that which the compounds of the present invention clearly have. It was also found that N-benzyl-sulfonyl-N'-butyl-urea has no blood-sugar-lowering effect.
It was therefore not to be expected that the present products of the process would have such a good anti-diabetic effect. In particular, it was not to be expected that the present compounds would have such a low toxicity despite the presence of arylalkyl radicals.

N- (4-methyl-benzenesulfonyl) -N '- (¯-phenyl-)thyl) urea
16.7 g of N- (4-methyl-benzenesulfonyl) -N '- (¯-phenyl ethyl) -thio-urea [produced by reacting p-toluenesulfonamide sodium with jB-phenyl-ethyl-mustard oil in acetone solution and in almost quantitative yield; Melting point 144 to 146 C (after recrystallization from methanol)] are dissolved in 400 cm3 of acetone. This solution is mixed with 3.8 g of sodium nitrate, dissolved in 25 cm3 of water, while stirring. The resulting solution is cooled to about 5 ° C. and, irrespective of a slight precipitation of crystals, with stirring, 30 cm 3 of Sn acetic acid are added dropwise over the course of one hour.
The mixture is then stirred for a further three hours at room temperature, diluted with water and the precipitate which has separated out is filtered off with suction. This is treated with dilute ammonia (approx. 1:60), the elemental sulfur produced during the reaction is filtered off and the still cloudy filtrate is heated with charcoal on the steam bath for 20 minutes.
The clear solution resulting after filtration is treated with hydrochloric acid, the desired crude sulfonylurea precipitating in almost quantitative yield. After recrystallization from methanol, 12 g of N- (4-methyl-benzenesulfonyl) -N '- (¯ phenyl-ethyl) -urea are obtained in the form of white crystals with a melting point of 145 to 147 C.
