Die Hydrolyse der genannten Sulfenyl-, Sulfinyl- oder Sulfonyl-parabansäuren,-isoharnstoffäther oder - isothioharnstoffäther erfolgt zweckmässig in alkalischem Medium. Isoharnstoffäther können auch in einem sauren Medium mit gutem Erfolg hydrolysiert werden. Der Ersatz des Schwefelatoms in der Harnstoffgruppierung von entsprechend substituierten Sulfonyl-thioharnstoffen durch ein Sauerstoffatom kann in bekannter Weise, z. B. mitHilfe von Oxyden oder Salzen von Schwermetallen oder auch durch Anwendung von Oxydationsmitteln, wie Wasserstoffperoxyd, Natriumperoxyd, salpetriger Säure oder Permanganaten ausgeführt werden.
Die Thioharnstoffe können auch durch Behandlung mit Phosgen oder Phosphorpentachlorid entschwefelt werden. Als Zwischenstufe erhaltene Chlorameisensäureamidine bzw. Carbodiimide können durch geeignete Massnahmen, wie Verseifen oder Anlagerung von Wasser, in die Sulfonylharnstoffe überführt werden.
Gegenüber einigen Oxydationsmitteln, wie beispielsweise Wasserstoffperoxyd, verhalten sich die Isothioharnstoffäther wie die entsprechenden Thioharnstoffe. Sie können demgemäss ebenso wie diese für eine oxydative Entschwefelung eingesetzt werden.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinenhinsichtlichderRe- aktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise

können die Umsetzungen in Abwesenheit oder Anwesenheit von Lösungsmitteln, bei Raumtemperatur oder bei erhöhter Temperatur durchgeführt werden.
Je nach dem Charakter der Ausgangsstoffe kann die eine oder andere der beschriebenen Verfahrens- weisen in einzelnen Fällen einen gewünschten individuellen Sulfonylharnstoff nur in geringen Ausbeuten liefern oder zu dessen Synthese nicht geeignet sein. In solchen verhältnismässig selten auftretenden Fällen macht es dem Fachmann keine Schwierigkeiten, das gewünschte Produkt auf einem andern der beschriebe- nen Verfahrenswege zu synthetisieren.
Die blutzuckersenkende Wirkung der beschriebenen Sulfonylharnstoffe kann dadurch festgestellt werden, dass man sie in Form der Natriumsalze in Dosen von 10 mg/kg an normal ernährte Kaninchen verfüttert und den Blutzuckerwert nach der bekannten Methode von Hagedorn-Jensen oder mit einem Autoanalyzer über eine längere Zeitdauer ermittelt.
Beispielsweise zeigen der N--J4- [ss- (6-Chlorchinolin-8-carboxamido)-äthyl]-benzolsulfonyl)-N' -cyclo- pentylmethylharnstoff, der N- :- [ss- (2-Methoxy-5-methylbenzamido)-äthyl]-benzolsuIfonyl)-N'-cyclopentyl- methylharnstoff und der N-{4-[ss-(3-Chlorbenzamido)-äthyl]-benzolsulfonyl}-N'-cyclopentylmethylharnstoof in der oben angegebenen Dosis eine maximale Blutzuckersenkung von 50% und darüber.
Die beschriebenen Sulfonylharnstoffe sollen vorzugsweise zur Herstellung von oral verabreichbaren
Präparaten mit blutzuckersenkender Wirksamkeit zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, appli- ziert werden. Zur Salzbildung können beispielsweise alkalische Mittel, wie Alkali- oder Erdalkalihydroxyde, - carbonate oder-bicarbonate herangezogen werden.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Träger-und Hilfsstoffe, wie Talkum, Stärke, Milchzucker, Tragant oder Magnesiumstearat, enthalten.
Ein Präparat, das die beschriebenen Sulfonylharnstoffe als Wirkstoff enthält, z. B. eine Tablette oder ein Pulver mit oder ohne Zusätze, ist zweckmässig in eine geeignet dosierte Form gebracht. Als Dosis ist dabei eine solche zu wählen, die der Wirksamkeit des verwendeten Sulfonylharnstoffes und dem gewünschten Effekt angepasst ist. Zweckmässig beträgt die Dosierung je Einheit etwa 0,5 bis 100 mg, vorzugsweise 2 bis 10 mg, jedoch können auch darüber oder darunterliegende Dosierungseinheiten verwendet werden, die gegebenenfalls vor Applikation zu teilen bzw. zu vervielfachen sind.
Die erfindungsgemäss erhältlichen Sulfonylharnstoffe können sowohl allein für die Behandlung des Diabetes mellitus eingesetzt werden als auch in Kombination mit andern oralen Antidiabetika. Als solche kommen nicht nur blutzuckersenkende Sulfonylharnstoffe in Betracht, sondern auch Verbindungen von unterschiedlichem chemischen Aufbau, wie beispielsweise Biguanide, Insbesondere das Phenyläthyl-biguanid oder das Dimethyl-biguanid.
Die nachfolgenden Beispiele zeigen eine der zahlreichen Verfahrensvarianten, die zur Synthese der erfindungsgemäss hergestellten Sulfonylharnstoffe verwendet werden können. Sie sollen jedoch nicht eine Einschränkung des Erfindungsgegenstandes darstellen.
Beispiel l : N- {4- [ss- (2-Methoxy-5-chlorbenzamido)-äthyl]-benzolsulfonyl}-N'-cyclopentyl- methylharnstoff
7, 1 g Cyclopentylmethylharnstoff werden in 100 ml abs. Benzol suspendiert und mit 2, 4 g 50%igem Natriumhydrid (in Weissöl) versetzt. Man erhitzt 3 h bei 50 C, gibt 9,7 g 4- [ss- (2-Methoxy-5-chlorbenzami- do)-äthyl]-benzolsulfochlorid in 100 ml abs. Benzol zu und rührt 3 h bei 800C. Nach dem Erkalten schüttelt man das Reaktionsgemisch mehrfach mit Wasser aus und säuert nach Filtrieren die vereinigten wässerigen Extrakte mit Salzsäure an.
Das ausgefällte Reaktionsprodukt wird aus l% igem Ammoniak umgefällt und

Nach 10 min wird das Gemisch in 100 ml Eiswasser, das mit 20 ml 2n-Salzsäure versetzt wurde, gegeben. Der abgeschiedene Niederschlag wird mit l% igem Ammoniak behandelt, der Rückstand in Dimethylformamid gelöst und die Lösung unter Rühren mit einer wässerigen Lösung von Kaliumpermanganat versetzt, bis die Permanganatfarbe bestehen bleibt. Man entfärbt mit wenig Bisulfit, saugt den Braunstein ab und versetzt das Filtrat mit Wasser und Salzsäure.
Der erhaltene N-- [ss- (2-Methoxy-5-chlorbenzamido)-äthyl]-benzolsulfonyl}-N'-cyclopentylmethylharnstoff wird aus Methanol umkristallisiert und schmilzt bei 173 bis 1740C.
