Das Brückenglied Y bedeutet vorzugsweise die -eH 2 -eH 2 -Gruppe. X und X I können gleich oder verschieden sein. Verbindungen, in denen X und Xl Wasserstoff oder einer der beiden Substituenten Wasserstoff und der andere Halogen, vorzugsweise Chlor oder Methyl darstellt, und Verbindungen, worin einer der beiden Substituenten Chlor und der andere Methyl bedeutet, sind besonders bevorzugt. Falls R l Alkyl bedeutet, so hat der Alkylrest vorzugsweise 4 bis 8 Kohlenstoffatome.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Sulfonylharnstoffe ist dadurch gekennzeichnet, dass man entsprechende Benzolsulfinyl- oder -sulfenylharnstoffe oxydiert und die Reaktionsprodukte gegebenenfalls zur Salzbildung mit alkalischen Mitteln behandelt.

Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinen hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise können die Umsetzungen in Abwesenheit oder Anwesenheit von Lösungsmitteln, bei Raumtemperatur oder bei erhöhter Temperatur durchgeführt werden.
Je nach dem Charakter der Ausgangsstoffe kann die eine oder andere der beschriebenen Verfahrensweisen in einzelnen Fällen einen gewünschten individuellen Benzolsulfonylharnstoff nur in geringen Ausbeuten liefern oder zu dessen Synthese nicht geeignet sein. In solchen verhältnismässig selten auftretenden Fällen macht es dem Fachmann keine Schwierigkeiten, das gewünschte Produkt auf einem andern der beschriebenen Verfahrenswege zu synthetisieren. Die erfindungsgemäss erhältlichen Benzolsulfonylharnstoffe können nach allgemein bekannten Verfahren in physiologisch unbedenkliche Salze überführt werden, z. B. durch Umsetzung mit Basen, Carbonaten oder Aminen. Die erfindungsgemäss erhältlichen Benzolsulfonylharnstoffe haben wertvolle therapeutische Eigenschaften und zeichnen sich insbesondere durch eine blutzuckersenkende Wirkung aus.
Sie dienen daher vorzugsweise zur Herstellung von Arzneimitteln.
Die blutzuckersenkende Wirkung der neuen Benzolsulfonylharnstoffe kann dadurch festgestellt werden, dass man sie als freie Verbindungen oder in Form der Natriumsalze in Dosen von 10 mg/kg

an normal ernährte Kaninchen verfüttert und den Blutzuckerwert nach der bekannten Methode von Hagedorn-Jensen, mit einem Autoanalyzer oder nach der Hexokinasemethode über eine längere Zeitdauer ermittelt.
In der nachfolgenden Tabelle sind die bei dieser Versuchsanordnung beobachteten Blutzuckersenkungen in % nach oraler Verabreichung von einigen erfindungsgemäss erhältlichen Benzolsulfonylharnstoffen zusammengestellt :

Acylaminoalkylbenzolsulfonylharnstoffe, die im Acylrest einen 2-Methoxy-oder 2-Äthoxy-nicotinsäurerest haben, sind zwar auch schon beschrieben worden (DE-OS 2419198), doch zeigten diese Verbindungen keine so starke oder lang anhaltende blutzuckersenkende Wirkung wie die erfindungsgemäss erhältlichen neuen Benzolsulfonylharnstoffe.
Die erfindungsgemäss erhältlichen Benzolsulfonylharnstoffe sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirksamkeit zur Behandlung des Diabetes mellitus dienen. Sie können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise

alkalische Mittel wie Alkali-oder Erdalkalihydroxyde,-carbonate oder-bicarbonate herangezogen werden.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Träger- und Hilfsstoffe, wie Talkum, Stärke, Milchzucker, Tragant oder Magnesiumstearat, enthalten.
Ein Präparat, das die neuen Benzolsulfonylharnstoffe als Wirkstoff enthält, z. B. eine Tablette, eine Kapsel oder ein Pulver mit oder ohne Zusätze, wird zweckmässig in eine geeignet dosierte Form gebracht. Als Dosis ist dabei eine solche zu wählen, die der Wirksamkeit des verwendeten Benzolsulfonylharnstoffs und dem gewünschten Effekt angepasst ist. Zweckmässig beträgt die Dosierung je Einheit etwa 1 bis 100 mg, vorzugsweise 5 bis 20 mg, jedoch können auch darüber oder darunterliegende Dosierungseinheiten verwendet werden, die gegebenenfalls vor Applikation zu teilen bzw. zu vervielfachen sind.
Die erfindungsgemäss erhältlichen Sulfonylharnstoffe können sowohl allein für die Behandlung des Diabetes mellitus eingesetzt werden als auch in Kombination mit andern oralen Antidiabetika.
Als solche kommen nicht nur blutzuckersenkende Sulfonylharnstoffe in Betracht, sondern auch Verbindungen von unterschiedlichem chemischen Aufbau, wie beispielsweise Biguanide, insbesondere das Phenyläthyl-biguanid oder das Dimethyl-biguanid.
Das nachfolgende Beispiel zeigt eine der zahlreichen Verfahrensvarianten, die zur Synthese der erfindungsgemäss herstellbaren Sulfonylharnstoffe verwendet werden können. Es soll jedoch nicht eine Einschränkung des Erfindungsgegenstandes darstellen.
Beispiel : N- {4- [2- (5-Chlor-2-ixobutyloxy-6-methyl-nicotinamido) -äthyl] -benzolsulfonyl} - - N'-butyl-harnstoff
2, 1 g 4- [2- (5-Chlor-2-isobutyloxy-6-methyl-nicotinamido)-äthylj-benzolsulfinsäure (hergestellt durch Reduktion des entsprechenden Sulfochlorids vom Fp. 84 bis 87 C mit Natriumsulfit) werden mit 1, 8 ml Thionylchlorid übergossen und über Nacht bei Raumtemperatur belassen. Nach dem Eindampfen im Vakuum trägt man den Rückstand in eine vorher bereitete Suspension von 0, 55 g N-Butyl-harnstoff in 10 ml Pyridin ein und rührt 15 min nach. Danach giesst man auf Eis und säuert mit verdünnter Essigsäure an. Nach dem Abfiltrieren des Niederschlags behandelt man diesen mit l% iger Ammoniak-Lösung und saugt erneut ab.
Der so erhaltene Sulfinylharnstoff wird in 20 ml Dimethylformamid gelöst und bei Raumtemperatur mit wässeriger Kaliumpermanganatlösung so lange versetzt, bis die Permanganatfarbe bestehen bleibt. Nachdem man die Suspension mit wenig Natriumsulfit entfärbt hat, saugt man vom Braunstein ab und säuert das klare Filtrat mit ver-

The benzenesulfonylureas obtainable according to the invention are preferably used for the production of orally administrable preparations with blood sugar-lowering activity for treating diabetes mellitus. They can be applied as such or in the form of their salts or in the presence of substances which lead to salt formation. For example, salt formation

alkaline agents such as alkali or alkaline earth metal hydroxides, carbonates or bicarbonates are used.
Suitable medical preparations are preferably tablets which, in addition to the process products, contain the usual carriers and auxiliaries, such as talc, starch, lactose, tragacanth or magnesium stearate.
A preparation that contains the new benzenesulfonylureas as an active ingredient, e.g. B. a tablet, a capsule or a powder with or without additives is suitably brought into a suitably dosed form. The dose to be selected is one which is adapted to the effectiveness of the benzenesulfonylurea used and the desired effect. The dosage per unit is expediently about 1 to 100 mg, preferably 5 to 20 mg, but it is also possible to use dosage units above or below, which may have to be divided or multiplied before application.
The sulfonylureas obtainable according to the invention can be used both for the treatment of diabetes mellitus alone and in combination with other oral antidiabetic agents.
As such, not only blood sugar-lowering sulfonylureas come into consideration, but also compounds of different chemical structures, such as biguanides, in particular phenylethyl biguanide or dimethyl biguanide.
The following example shows one of the numerous process variants which can be used for the synthesis of the sulfonylureas which can be prepared according to the invention. However, it is not intended to be a limitation of the subject matter of the invention.
Example: N- {4- [2- (5-chloro-2-ixobutyloxy-6-methyl-nicotinamido) ethyl] benzenesulfonyl} - - N'-butylurea
2.1 g of 4- [2- (5-chloro-2-isobutyloxy-6-methyl-nicotinamido) ethylj-benzenesulfinic acid (prepared by reduction of the corresponding sulfochloride of mp 84 to 87 C with sodium sulfite) are treated with 1.8 Pour ml of thionyl chloride and leave at room temperature overnight.
