Verfahren zur Darstellung von J','; 11,1z -21-Oxy-pregnadien-3,20-dion. Es wurde gefunden, dass man zu d4,sa1,>>- "l-Oxy-pregnadien-3,20-dion gelangen kann, wenn man ein Carbonsäureha.logenid eines Derivates der Äthiocholansäure, .das im Ring A in 3-Stellung eine durch Hydrolyse in Hydroxyl umwandelbare Gruppe und im Ring C in 11,12-Stellung eine Doppelbin dung aufweist,
mit Diazomethan zum Diazo- heton umsetzt, im erhaltenen Produkt die ge- #iehützte Hydroxylgruppe in 3-Stellung in Freiheit setzt, in beliebiger Reihenfolge diese HydroxylgTuppe mit Oxydationsmitteln in eine Ketogruppe und die 17ständige Grup pierung mit Säuren in eine veresterte Ketol- gruppe umwandelt, ,
in a-Stellung zur 3-Keto- gruppe eine Doppelbindung durch Halogenie- rung und nachträgliche Halogenwasserstoff- abspaltung einführt und schliesslich mit hy- drolysierenden Mitteln behandelt.
Beim Rest in 3-Stellung des Ausgangs- 21 kann es sich z. B. um eine durch anorganische oder organische Säuren, wie Carbonsäuren, Sul'fonsäuren oder Ha.logen- wasserstoffsäuren, veresterte oder durch Al kohole oder Phenole, wie Tizarylmethyl- oder Benzylalkohole, enolisierte oder acetalisierte Carbonylverbindungen,
verätherte 11yd,roxyl- gruppe handeln.
Die Umsetzung des. Carbonsäurehalogenids mit Diazomethan erfolgt unter Halogen wasserstoffabspaltung und Bildung eines Di- azoketons -CO-CHN. insbesondere dann, wenn ständig ein Übe@rschuss von Diazo- methan zugegen ist. Das erhaltene Diazoketon kann in rohem Zustande oder nach Abtrennung und Reini gung weiter verarbeitet werden.
Die freie Hydroxylgruppe in 3-Stellung wird mit Ily- drol'y sierenden Mitteln hergestellt.
Die freie 3-Hydroxylgruppe wird durch Oxydationsmittel, z. B. mit Chromsäure in Eisessig, oder durch Erhitzen mit Kupfer pulver, Einwirkung von Metallalkoholaten bzw. -phenolaten in Gegenwart von Ketonen, wie Aceton oder Cyclohexanon, in eine Keto- gruppe übergeführt.
Nach oder auch vor der Oxydation der 3-Hydroxylgruppe wird die vorhandene Di- azoketongruppierung durch Einwirkung von Säuren, z. B. wasserfreien oder verdünnten, organischen oder anorganischen Säuren, bei spielsweise Essigsäure, Propionsäure, Butter säuren, Crotonsäuren, Palmitinsäure, Benzoe- säure, Phenyless .igsäure, Schwefelsäure, Me- thansulfosäure,
Toluolsulfosäure, Halogen wasserstoffsäure, Phosphorsäure oder Bor säure, in eine veresterte Ketolgruppe über geführt. Es können so z. B. Acylate, Halo genide oder Sulfonsäureester erhalten werden.
Die Doppelbindung in a-Stellung zur 3-Ketogruppe wird durch Halogenierung und nachträgliehe Halogenwasserstoffabspaltung gebildet.
Schliesslich wird die veresterte Ketol- gruppe mit hydrolysierenden Mitteln ver seift. Dabei wird man auf die relative AlkaIi- empfindl.ichkeit der a-Oxy-keton-Grruppie- rung Rüeksicht nehmen, so dass man vorteil- haft mit Säuren oder mit ganz schwachen Al- kalien, wie Bicarbonaten, arbeitet.
Das neue Verfahrensprodukt, das d4,5:11,12- 21-Oxy-pregnadien-3,20-dion, bildet ein Acetat vom F. 142-143 . Es soll therapeu tische Verwendung finden oder als Zwischen produkt zur Herstellung therapeutisch wert voller Verbindung dienen.
<I>Beispiel:</I> 1,1 Teile d11,12- 3a - Acetoxy - ätiocholen- säure (erhalten z. B. gemäss Patent Nr. 242994 [Gase 1996/m]) werden in 6 Teilen reinsten Thionylchlorid gelöst und unter Feuchtig- keitsausschluss 16 Stunden bei Zimmertempe ratur stehen gelassen.
Dann wird im Vakuum eingedampft und der Rückstand, das rohe 411,12-3-Acetoxy-ätiocholensäure-chlorid, in 10 Teilen absofutem Benzol gelöst.
Die benzolische Lösung des so erhaltenen Ausgangsstoffes wird nun bei 0 in eine trockene, aus 5 Teilen Nitroso-m@ethylharn- stoff bereitete ätherische Lösung von Diazo- methan eingetragen. Die Mischung bleibt zu nächst 2 Stunden bei 0 , dann noch 12 Stun den bei Zimmertemperatur stehen, wird hierauf bei 50 Badtemperatur eingedampft und im Vakuum getrocknet.
Der gelbe Rück stand (rohes Diazoketon) wird in 15 Teilen Methanol gelöst, mit der Lösung von 0,75 Teil galiumhydroxyd in 1 Teil Wasser und 35 Teilen Methanol versetzt und 8 Stunden bei Zimmertemperatur stehengel'assen. Dann wird eine Lösung von 1,6 Teilen Kalium karbonat in 50 Teilen Wasser zugegeben, das Methanol im Vakuum entfernt und das Pro dukt durch Abdampfen mit absolutem Benzol völlig getrocknet. Nun wird mit 200 Teilen absolutem Benzol,
80 Teilen trockenem Ace ton und 5 Teilen Aluminium-phenolat 20 Tage in einer Ampulle eingeschmolzen stehen gelassen. Hierauf wird stark eingeengt, mit Äther aufgenommen, die Lösung mit ganz verdünnter Salzsäure, Sodalösung und Wasser gewaschen, über Natriumsulfat getrocknet, eingedampft und der Rückstand im Vakuum bis zur völligen Entfernung des freien Phenols -sehr gut getrocknet.
Dieses Roh- Produkt wird mit 15 Teilen reinstem, wasser- freiem Eisessig 1/2 Stunde auf 100 erhitzt, wobei zuerst reichlich Stickstoff entweicht. Eindampfen im Vakuum gibt das rohe J11.12_ 21-Acetoxy-pregnen-3,20-dion. Es wird nach der Durchlaufmethode über A1203 chromato- graphisch gereinigt und schmilzt dann, aus Äther-Petroläther umkristallisiert, bei 151 bis 152 .
Dieses Diketon wird in Eisessig mit 2 Mol Brom bromiert, und aus dem rohen Bromierungsprodukt wird durch 5stündiges Kochen mit absolutem Pyridin oder durch 2stündiges Erwärmen mit Dimethylanilill auf 150 Bromwasserstoff abgespalten. Das so erhaltene Rohprodukt wird noch 15 31i- nuten mit 0,5 Teil Zinkstaub und 10 Teilen Eisessig unter ständigem Umschwenken auf 80 erwärmt.
Nach Filtration wird im Va kuum eingedampft, mit Äther aufgenommen, die Lösung mit verdünnter Salzsäure, Soda lösung und Wasser gewaschen, über Na triumsulfat getrocknet und eingedampft. Der Rückstand wird chromatographiseh gereinigt. Man erhält auf diese Weise das J4,1;11;12-21- Acetoxy-pregnadien-3,20-dion (Anhydro-cor- ticostcron-acetat) vom F. 142-143 , das durch Verseifung mit wässerig-methylalho- holischexn Kaliumbikarbanat in das freie 44,5;11,12-21-Oxy-pregnadien-3,20-dion über führt wird.
Zum gleichen Produkt gelangt. man in analoger Weise ausgehend von einer _"j11,12_ 3ss- Acyloxyätiocholensäure.

It has been found that d4, sa1, >> - "l-oxy-pregnadiene-3,20-dione can be obtained by using a carboxylic acid halide of a derivative of ethiocholanic acid, the one in ring A in the 3-position group which can be converted into hydroxyl by hydrolysis and has a double bond in ring C in position 11,12,
reacts with diazomethane to form diazohetone, in the product obtained releases the protected hydroxyl group in the 3-position, in any order converts this hydroxyl group into a keto group with oxidizing agents and converts the 17-position group into an esterified ketol group with acids, ,
in the a-position to the 3-keto group introduces a double bond by halogenation and subsequent elimination of hydrogen halide and finally treated with hydrolyzing agents.

The reaction of the carboxylic acid halide with diazomethane takes place with elimination of hydrogen and formation of a diazoketone —CO — CHN. especially when there is always an excess of diazomethane. The diazoketone obtained can be processed further in the raw state or after separation and purification.
The free hydroxyl group in the 3-position is produced with Ilydrol'y-sizing agents.
The free 3-hydroxyl group is oxidized by oxidizing agents, e.g. B. with chromic acid in glacial acetic acid, or by heating with copper powder, the action of metal alcoholates or phenolates in the presence of ketones, such as acetone or cyclohexanone, converted into a keto group.
After or before the oxidation of the 3-hydroxyl group, the diazo ketone grouping present is removed by the action of acids, e.g. B. anhydrous or dilute, organic or inorganic acids, for example acetic acid, propionic acid, butyric acids, crotonic acids, palmitic acid, benzoic acid, phenylacetic acid, sulfuric acid, methanesulfonic acid,
Toluenesulfonic acid, hydrohalic acid, phosphoric acid or boric acid, passed into an esterified ketol group. It can be B. acylates, halides or sulfonic acid esters can be obtained.
The double bond in the a-position to the 3-keto group is formed by halogenation and subsequent elimination of hydrogen halide.
Finally, the esterified ketol group is saponified with hydrolyzing agents. The relative alkali sensitivity of the α-oxy-ketone group will be taken into account, so that it is advantageous to work with acids or with very weak alkalis such as bicarbonates.
The new product of the process, d4,5: 11,12-21-oxy-pregnadiene-3,20-dione, forms an acetate of F. 142-143. It should find therapeutic use or serve as an intermediate product for the creation of a therapeutically valuable connection.
<I> Example: </I> 1.1 parts of d11, 12-3a - acetoxy - etiocholenic acid (obtained for example according to patent no. 242994 [Gase 1996 / m]) are dissolved in 6 parts of the purest thionyl chloride and left to stand under exclusion of moisture for 16 hours at room temperature.
It is then evaporated in vacuo and the residue, the crude 411,12-3-acetoxy-etiocholenic acid chloride, dissolved in 10 parts of absolute benzene.
The benzene solution of the starting material obtained in this way is now introduced at 0 into a dry, ethereal solution of diazomethane prepared from 5 parts of nitroso-m @ ethylurea. The mixture remains at 0 for the next 2 hours and then at room temperature for another 12 hours, is then evaporated at 50 bath temperature and dried in vacuo.
The yellow residue (crude diazoketone) is dissolved in 15 parts of methanol, treated with a solution of 0.75 part of galium hydroxide in 1 part of water and 35 parts of methanol and left to stand for 8 hours at room temperature. Then a solution of 1.6 parts of potassium carbonate in 50 parts of water is added, the methanol is removed in vacuo and the product is completely dried by evaporation with absolute benzene. Now with 200 parts of absolute benzene,
80 parts of dry acetone and 5 parts of aluminum phenolate were allowed to stand melted in an ampoule for 20 days. It is then strongly concentrated, taken up with ether, the solution washed with very dilute hydrochloric acid, soda solution and water, dried over sodium sulphate, evaporated and the residue dried very well in vacuo until the free phenol has been completely removed.
This crude product is heated to 100 for 1/2 hour with 15 parts of the purest, anhydrous glacial acetic acid, with abundant nitrogen escaping first. Evaporation in vacuo gives the crude J11.12_ 21-acetoxy-pregnen-3,20-dione. It is purified chromatographically using the continuous flow method over A1203 and then, recrystallized from ether-petroleum ether, melts at 151 to 152.
This diketone is brominated in glacial acetic acid with 2 moles of bromine, and the crude bromination product is split off by boiling with absolute pyridine for 5 hours or by heating with dimethylanilil to 150% hydrogen bromide for 2 hours. The crude product obtained in this way is heated to 80 for a further 15 31 minutes with 0.5 part zinc dust and 10 parts glacial acetic acid with constant swirling.
After filtration, it is evaporated in vacuo, taken up with ether, the solution washed with dilute hydrochloric acid, soda solution and water, dried over sodium sulfate and evaporated. The residue is purified by chromatography. In this way the I4,1; 11; 12-21-acetoxy-pregnadiene-3,20-dione (anhydro-corticostcron-acetate) of F. 142-143, which is obtained by saponification with aqueous methylalcoholicxn Potassium bicarbanate is converted into the free 44.5; 11.12-21-oxy-pregnadiene-3,20-dione.
