el eine durch Hydrolyse in Hydroxyl umwandelbare Gruppe und im Rinb C in 9,11-Stellung eine Doppelbindung aufweist, mit Diazo- methan zum Diazoketon umsetzt, im erhal tenen Produkt die geschützte Hydroxyl- gruppe in 3-Stellung in Freiheit setzt,
in he.- liebiger Reihenfolge diese Hydroxylgruppe mit Oxydationsmitteln in eine Ketogruppe und die 17stä.ndige Gruppierung mit Säu ren in eine veresterte Ketolgrizppe umwan delt., in a-Stellung zur 3-Ketogruppe eine T)
oppelbindung durch Halogenierung und irachträgliche Halogenwasserstoffabspaltung einführt und schliesslich mit hydrolysieren- den Mitteln behandelt.
Beim Rest. in 3-Stellung des Ausgangs- :toffes kann es sich z. B. um eine durch an- or-anische oder organische Säuren, wie Car- bonäuren, Sulfonsäuren oder Halogenwas- urstoffsä Uren, veresterte oder durch Alko hole oder Phen 1e, wie Triarylmethyl- oder Benzylalkohole,
eilolisierte oder acetali.sierte Carbonylverbindungen, verätherte Hydroxyl- @,gnippe handeln.
Die Umsetzung des Carboiit"ä.urehaloge- vids mit Diazomethan erfolgt unter Halogen- und Bildung eines f)iazol@etons - CO-CHN2, wenn ständig ein 1'bci@@ehuss von Diazoverbin dung zugegen ist. Das erhaltene Diazoketon kann in rohem Zustande oder nach Abtrennung und Reini gung weher verarbeitet werden.
Die Bildung der freien Hydroxylgruppe in 3-Stellung er folgt mit hydrolysierenden Mitteln.
Die freie 3,-Hydroxylgruppe wird durch Oxydationsmittel z. B. in an sich bekann ter Weise durch oxydierende Mittel wie Chromsäure in Eisessig oder durch dehydrie rende Mittel wie Erhitzen mit Kupferpulver, Einwirkung von Metallalkoholaten bzw. -phenolaten in Gegenwart von Ketonen, wie Aceton oder Cyclohexanon, in eine Keto- gruppe übergeführt.
Nach oder auch vor Oxydation der 3-Hydroxylgruppe wird die vorhandene Diazoketo-ngruppierung durch Einwirkung von Säuren z. B. wasserfreien oder verdünn ten, organischen oder anorganischen Säuren, beispielsweise Essigsäure, Propionsäure, But tersäuren, Crotonsäuren, Pa.lmitinsäure, Ben- zoesäure, Phenylessigsä,ure, Schwefelsäure, llethansulfosäure, Toluolsulfosäure, Halo genwasserstoffsäure, Phosphorsäure oder Borsäure,
in eine veresterte Ketolgruppe übergeführt. Es können so z. B. Acylate, Halogenide oder Sulfonsäureester erhalten werden.
Die Doppelbindung in a-Stellung zur 3-Ketogruppe wird in an sich bekannter Weise durch Ila.logenierunb und nachträg- liebe- Halogenwasserstoffabspaltung gebildet.
Schliesslich wird die veresterte Ketol- gruppe mit hydrolysierenden Mitteln ver- seift. Dabei wird man auf die relative Alkali empfindlichkeit .der a-Oxy-keton-Gruppie- rung Rücksicht nehmen, so dass man vorteil haft mit Säuren oder mit ganz schwachen Alkalien, wie Bicarbonaten, arbeitet.
Das neue Verfahrensprodukt, .das d 4,5: 9,11- 21-Oxy-pregnadien-3:,20-dion bildet ein Ace tat vom F. 158-159 . Es, soll therapeutische Verwendung finden oder als Zwischenpro dukt zur Herstellung therapeutisch wertvoller Verbindungen dienen.
<I>Beispiel:</I> Die Darstellung des in diesem Beispiel verwendeten Ausgangsstoffes kann auf fol gende Weise vorgenommen werden: 1 Teil 3,11- Diketo - ätio-cholansäurs - methylester (,Smp. 184 korr., erhältlich z.
B. aus dem Methylester der 3a, 12ss-Di- oxy-ätio-cholansäure durch Umwandlung in das entsprechende 12-Monoacetat 'über das 3,12-Diacetat, Oxydation mit nachträglicher Verseifung zur 3-Keto-12ss-oxy-ätiochola.n- säure, Überführung in 3-Feto-12ss-benzoxy- ätio-cholansäuremethylester, thermische Spal tung zum d 11,1a - 3,
- Keto - ätio - cholansöure- methylester, Umsetzung mit unterbramiger Säure, Oxydation des entstandenen Bro:m- hydrins mit Chro:m-säure zum Bromketon und anschliessende Entbromung mit Zinkstaub, werden in 20 Teilen reinstem Eisessig nach Zusatz von 0,4 'Teil Platinoxyd hydriert bis die Wasserstoffaufnahme völlig aufhört, was etwa 8 Stunden benötigt.
Nach Filtration wird eingedampft und der Rückstand 4 Stun den mit der Lösung von 2 Teilen Kalium hydroxyd in 2 Teilen Wasser und 20 Teilen Methanol unter Rückfluss gekocht. Nach Zu satz von 10 Teilen Wasser wird das Metha nol im Vakuum entfernt, der Rückstand unter Kühlung mit Salzsäure bis zur kongosauren. Reaktion versetzt und -die krystallin ausge fällte Säure abgenutscht, mit Wasser ge waschen und im Vakuum getrocknet. Das Produkt schmilzt unscharf und stellt ein Ge misch der raumisomeren 3a- und 3ss,11 Dioxy- ätio-cholansäuren dar.
Zur partiellen Acety- lierung wird es mit. 5 Teilen Eisessig und 1 Teil Essigsäureanhydrid 1 Stunde unter Rüekfluss gekocht, dann werden in .der Hitze tropfenweise 2 Teile Wasser zugegeben, wor auf man noch 1.5, Minuten weiter kocht. Nach Zusatz von mehr Wasser wird im Vakuum stark eingeengt und die Säure durch erneu ten Wasserzusatz völlig ausgefällt, abge- nutscht und im Vakuum getrocknet.
Das gelb liche krystalline Material besitzt die Zu sammensetzung C=2H340; und schmilzt un scharf, es stellt ein Gemisch der raumisome- ren 3u- und Siss-Acetoxy-11-oxy-ätio-cho:lan- säuren dar. Die Ausbeute beträgt 1.1 Teile.
Das gut getrocknete Produkt wird in 6 Tei len reinstem Thionylchlorid gelöst und unter Feuchtigkeitsausschluss 16 Stunden bei Zim mertemperatur stehen gelassen. Dann wird im Vakuum eingedampft und der Rückstand, das rohe d9.11_3-Acetoxy-ä.tio-chol'ensäure- chlorid, in 10 Teilen absolutem Benzol ge löst.
Die benzolisc.he Lösung des so erhaltenen Ausgangsstoffes wird nun bei 0 in eine trockene, aus 5 Teilen Nitroso-methylharll- stoff bereitete ätherische Lösung von Dia.zo- methaneingetragen. Die Mischung bleibt zu nächst 2 Stunden bei 0 , dann noch 12 St.un- .den bei Zimmertemperatur stehen, wird hier auf bei 509 Badtemsperatur eingedampft und im Vakuum getrocknet.
Der gelbe Rückstand (1.l5 Teile rohes Diazoketon) wird in 15 Teilen Methanol gelöst, mit der Lösung von 0,75 Teil Kaliumhydroxyd in 1. Teil Wasser und 35 Teilen Methanol versetzt und 8 Stun- .den bei Zimmertemperatur stehen gelassen.
Dann wird eine Lösung von 1.6 Teilen Ka- liumbicarbonat in 50 Teilen Wasser zuge geben, das Methanol im Vakuum entfernt und .das Produkt durch Abdampfen mit absolu tem Benzol völlig getrocknet. -Nun wird mit 2'00 Teilen absolutem Benzol, 80 Teilen trockenem Aceton und 5 Teilen Aluminium- phenolat 20 Tage in einer Ampulle einge schmolzen stehen gelassen. Hierauf wird stark eingeengt, mit Äther aufgenommen, die Lösung mit ganz verdünnter Salzsäure.
