Verfahren zur Darstellung von d'-Pregnen-3,20-dion-llss,21-diol. Es wurde gefunden, dass man zu d4-Preg- nen -<B>3,20</B> - dien -11ss - 21- diel gelangen kann, wenn man ein Carbonsäurehalogenid einer 3,l 1ss - Dioxy - ätiocholanAure, dessen ]Ely- droxylgruppen in 3- und 11-Stellung ge schützt sind, mit Diazomethan zum Diazo- kcton umsetzt,
im erhaltenen Produkt die ge schützten Hydroxylgruppen in 3- und 11 Stellung in Freiheit setzt, in beliebiger Reihenfolge die Hydroxylgruppe in 3-Stel- lung mit Oxydationsmitteln in eine Keto- gruppe und die 17ständige Gruppierung mit Säuren in eine veresterte Ketolgruppe um wandelt,
in a-Stel'lung zur 3-Ketogruppe- eine Doppelbindung durch Halogenierung und nachträgliche Halogenwasserstoffabspaltung einführt und schliesslich mit Ilydrolysieren- den Mitteln behandelt.
Bei den Resten in 3- und 11-Stellung des Ausgangsstoffes kann es sich z. B. um durch anorganische oder organische Säuren, wie Carbonsäuren, Sulfonsäuren oder Ha,logen- wasserstoffsäuren, veresterte oder durch Al kohole oder Phenole, wie Triarylmethyl- oder Benzyl'alkohole, enolisierte oder acetalisierte Carbonylverbindungen,
verätherte Hydroxyl- gruppen handeln. Die geschützte Hydroxyl- W Cr uppe in 11 -Stellung des Ausgangsstoffes C, befindet sieh in anderer räumlicher Lage als diejenige des Corticosterons.
Die Umsetzung des Carbonsäurehalogenids mit Diazomethan erfolgt unter Halogen- wasserstoffabspa,ltung und Bildung eines Diazoketong-CO---CHNz insbesonders dann, wenn ständig ein Überschuss von Diazo- methan zugegen ist.
Das erhaltene Diazoketon kann in rohem Zustande oder nach Abtrennung und Reini gung weiter verarbeitet werden. Die freien Hydroxylgruppen in 3- und 11-Stellung wer den mit hydrolysierenden Mitteln hergestellt.
Die freie 3-Hydroxylgruppe wird z. B. durch Einwirkung von Metallalkoholaten bzw. -phenolaten in Gegenwart von Ketonen, wie Aceton oder Cyclobexanon, in eine Keto- gruppe übergeführt.
Nach oder auch vor der Oxydation der 3-Hydrogylgruppe wird die vorhandeneDiazo- ketongruppierung durch Einwirkung wasser freier oder verdünnter organischer oder an- organischer Säuren, beispielsweise Essig säure, Propionsäure, Buttersäuren, Croton- säuren, Palmitinsäure, Benzoesäure, Phenyl- essigsäure, Schwefelsäure,
Methansulfosäure, Toluolsulfosäure, Halogenwass-erstoffsäure, Phosphorsäure oder Borsäure, in eine ver esterte Ketolgruppe übergeführt. Es kön nen so z. B. Aeylate, Halogenide oder Sul- fonsäureester erhalten werden.
Die Dappelbindung in a-Stellung zur 3- Ketogruppe wird durch Halogenierung und nachträgliche Halogenwasserstoffabspaltung gebildet.
Schliesslich wird die veresterte Ketol- gruppe mit hydrolysierenden Mittelh verseift. Dabeiwird man auf dieTelativeAlkaliempfind- lichkeit :der a-Oxyketon-Gruppierung Rück sicht nehmen, so dass man, vorteilhaft mit Säuren oder mit ganz schwachen Alkalien, wie Bikarbonaten, arbeitet.
Das neue Verfahrensprodukt bildet farb lose Kristalle und soll therapeutische Verwen- dungfinden oder als Zwischenprodukt zur Her stellung therapeutisch wertvoller Verbindun gen dienen.
<I>Beispiel:</I> 23 Teile 3ss,llss-Diacetoxy-ätiochol'ansäure (hergestellt z. B. durch oxydativen Abbau der Lakton-Seitenkette des Sarmentogenin-di- acetates mit Kaliumpermanganat, Abspaltung der 14-Hydroxylgruppe mit Phosphoroxy- chlorid und Pyridin und Hydrierung der ent standenen Doppelbindung)
werden bei 0 in 140 Teilen reinstem Thionylchlorid gelöst und unter Feuchtigkeitsausschluss zunächst 1/2 Stunde bei 0 , dann noch 16 Stunden bei 18 stehengelassen. Hierauf wird im Vakuum bei 40 Badtemperatur eingedampft, der Rückstand in 500 Teilen absolutem Benzol gelöst und in eine aus 100 Teilen Nitroso- methylharnstoff bereitete,
über Kalium- hydroxyd getrocknete und frisch destillierte ätherische Lösung von Diazomethan bei 0 eingetragen, wobei sofortige Gasentwicklung zu beobachten ist. Die Mischung bleibt unter Feuehtigkeitsausschluss (KOH) zunächst 2 Stunden bei 0 , dann noch 16 Stunden bei 18 stehen,
wird hierauf bei 50 Badtempera- tur stark eingeengt und zuletzt im Vakuum völlig getrocknet.
Das so erhaltene rohe 21-Diazo-pregnen- 3ss,llss-diacetoxy-20-on wird in 300 Teilen Methanol' gelöst, mit der Lösung von 15 Teilen Kaliumhydroxyd in 20 Teilen Wasser und 700 Teilen Methanol versetzt und 8 Stunden bei 20 stehengelassen.
Hierauf wird die Lösung von 32 Teilen Kaliumbikarbonat in 1000 Teilen Wasser zugegeben, das Me thanol' im Vakuum entfernt und die verblei bende Suspension mit viel Äther ausges6hüt- telt. Die mit Wasser gewaschene und über Natriumsulfat getrocknete Atherlösung wird bei 50 Badtemperatur stark eingeengt und hierauf im Vakuum völlig eingedampft,
wo bei rohes 21-Diaza-pregnan-3f,ll@-diol-20-on zurückbleibt. Dieses wird nun. mit 2000 Teilen absolutem Benzol, 800 Teilen trocke nem Aceton und 50 Teilen Aluminium phenolat 20 Tage in einer Ampulle einge schmolzen stehengelassen. Hierauf wird stark eingeengt, mit Äther verdünnt, die Lösung mit ganz verdünnter Salzsäure, Sodalösung und Wasser gewaschen, über Natriumsulfat getrocknet, eingedampft und der Rückstand im Vakuum bis zur völligen Entfernung des freien Phenols sehr gut getrocknet.
Dieses Rohprodukt wird mit 300 Teilen reinstem wasserfreiem Eisessig 1/2 Stunde auf 95-100 erhitzt und das dabei abgespaltene Gas in einem Messzylinder über Wasser auf gefangen. Na-eh Beendigung der Stickstoff- entwicklung wird im Vakuum eingedampft. Der verbleibende Rückstand wird in an sich bekannter Weise. über Aluminiumoxyd chro- matographisch gereinigt.
Beim Umkristalli- sieren aus Äther-Petroläther erhält man so das Pregnan-llss,21-diol-3,20-dion-21-mono- acetat in farblosen Kristallen.
Durch Vermischen von 10 Teilen Brom mit 884 Teilen Eisessig wird nun zunächst eine n-Bromlösung bereitet. 9,6 Teile des Zwischenproduktes werden in 50 Teilen Eis- essig gelöst und hierauf mit 2 Tropfen der obigen Bromlösung versetzt.
Nach einigen Minuten tritt plötzliche Entfärbung ein., worauf man unter Kühlung und Umschwen ken langsam noch 48 Teile der Bromlösung zugibt, die fast momentan entfärbt wird. Nach Eindampfen im Vakuum bei 25 Bad- temperatur tritt Kristallisation ein.
Die mit Äther gewaschenen, farblosen Kristalle werden mit 100 Teilen absolutem Pyridin 5 Stunden unter Rückfluss gekocht. Nach Eindampfen im Vakuum wird in viel Äther aufgenommen, mit wenig verdünnter Salzsäure, Sadalösung und Wasser ge waschen, über Natriumsulfat getrocknet und.
eingedampft. Der Rückstand wird mit einer Säule von Aluminiumoxyd chromatographisch gereinigt, wobei die ersten mit Benzol eluier- ten Anteile tief und unscharf schmelzende Kristalle geben. Die weiteren mit Benzol so wie mit Benzol-Ather eluierten Fraktionen geben nach Umkristallisieren aus Aeeton- Äther farblose Nadeln.
Durch Verseifung derselben mit methyl'alkoholischer Salzsäure oder wässerig-methyl:alkoholischem Kalium bikarbonat erhält man das freie d4-Pregnen- 3.20-dion-llss,21-diol in Form farbloser Kri- aall'e.
An Stelle des Chlorids der 3ss,llss-Diacet- oxy-ätiocholansäure lässt sich als Ausgangs stoff z. B. auch ein Halogenid eines in 3-Stel- lung stereoisomeren Diesters, beispielsweise ein 3a,llss-Diacyloxy-ätiocholansäure-halo- benid verwenden.
Process for the preparation of d'-Pregnen-3,20-dione-llss, 21-diol. It has been found that d4-pregnen - <B> 3.20 </B> - diene -11ss - 21- can be obtained if a carboxylic acid halide of a 3.11ss - dioxy - ethiocholanic acid, whose] Ely- droxylgruppen in 3- and 11-positions are protected, reacts with diazomethane to diazo- octone,
in the product obtained sets the protected hydroxyl groups in the 3- and 11-position free, in any order converts the hydroxyl group in the 3-position into a keto group with oxidizing agents and the 17-position group into an esterified ketol group with acids,
introduces a double bond in a position to the 3-keto group by halogenation and subsequent elimination of hydrogen halide and finally treated with Ilydrolysieren- agents.
The residues in the 3- and 11-position of the starting material can be, for. B. to by inorganic or organic acids, such as carboxylic acids, sulfonic acids or Ha, logen- hydrogen acids, esterified or by alcohols or phenols, such as triarylmethyl or benzyl alcohol, enolized or acetalized carbonyl compounds,
act etherified hydroxyl groups. The protected hydroxyl W Cruppe in the 11 position of the starting material C is in a different spatial position than that of the corticosterone.
The reaction of the carboxylic acid halide with diazomethane takes place with elimination of hydrogen halide and formation of a diazoketone-CO --- CHN2, especially when there is always an excess of diazomethane.
The diazoketone obtained can be processed further in the raw state or after separation and purification. The free hydroxyl groups in the 3- and 11-positions who are produced with hydrolyzing agents.
The free 3-hydroxyl group is z. B. by the action of metal alcoholates or metal phenates in the presence of ketones, such as acetone or cyclobexanone, converted into a keto group.
After or before the oxidation of the 3-hydrogyl group, the diazo ketone grouping present is removed by the action of anhydrous or dilute organic or inorganic acids, for example acetic acid, propionic acid, butyric acids, crotonic acids, palmitic acid, benzoic acid, phenylacetic acid, sulfuric acid,
Methanesulfonic acid, toluenesulfonic acid, hydrohalic acid, phosphoric acid or boric acid, converted into an esterified ketol group. It can be so z. B. eylates, halides or sulfonic acid esters can be obtained.
The Dappel bond in the a-position to the 3-keto group is formed by halogenation and subsequent elimination of hydrogen halide.
Finally, the esterified ketol group is saponified with hydrolyzing agents. In doing so, one will take into account the relative sensitivity to alkali: the α-oxyketone group, so that it is advantageous to work with acids or with very weak alkalis such as bicarbonates.
The new process product forms colorless crystals and is intended to be used therapeutically or as an intermediate product for the production of therapeutically valuable compounds.
<I> Example: </I> 23 parts of 3ss, llss-diacetoxy-etiochol'anoic acid (produced e.g. by oxidative degradation of the lactone side chain of sarmentogenin di-acetate with potassium permanganate, cleavage of the 14-hydroxyl group with phosphoroxy- chloride and pyridine and hydrogenation of the double bond formed)
are dissolved at 0 in 140 parts of the purest thionyl chloride and left to stand initially at 0 for 1/2 hour and then at 18 for 16 hours with exclusion of moisture. It is then evaporated in vacuo at a bath temperature, the residue is dissolved in 500 parts of absolute benzene and prepared from 100 parts of nitrosomethylurea,
Ethereal solution of diazomethane, dried over potassium hydroxide and freshly distilled, entered at 0, with immediate evolution of gas being observed. The mixture remains with the exclusion of fire (KOH) initially for 2 hours at 0, then for 16 hours at 18,
is then strongly concentrated at 50 bath temperature and finally completely dried in vacuo.
The crude 21-diazo-pregnen-3ss, llss-diacetoxy-20-one obtained in this way is dissolved in 300 parts of methanol, mixed with a solution of 15 parts of potassium hydroxide in 20 parts of water and 700 parts of methanol and left to stand at 20 for 8 hours.
The solution of 32 parts of potassium bicarbonate in 1000 parts of water is then added, the methanol is removed in vacuo and the remaining suspension is shaken out with plenty of ether. The ether solution, washed with water and dried over sodium sulfate, is strongly concentrated at a bath temperature and then completely evaporated in vacuo,
where with raw 21-diaza-pregnan-3f, ll @ -diol-20-one remains. This will now. with 2000 parts of absolute benzene, 800 parts of dry acetone and 50 parts of aluminum phenolate for 20 days in an ampoule melted. It is then strongly concentrated, diluted with ether, the solution washed with very dilute hydrochloric acid, soda solution and water, dried over sodium sulphate, evaporated and the residue dried very well in vacuo until the free phenol has been completely removed.
This crude product is heated to 95-100 for 1/2 hour with 300 parts of the purest anhydrous glacial acetic acid and the gas which is split off is collected in a measuring cylinder over water. After the end of the nitrogen evolution, it is evaporated in vacuo. The remaining residue is in a manner known per se. Purified chromatographically over aluminum oxide.
When recrystallizing from ether-petroleum ether, the pregnane-llss, 21-diol-3,20-dione-21-monoacetate is obtained in colorless crystals.
By mixing 10 parts of bromine with 884 parts of glacial acetic acid, an n-bromine solution is now first prepared. 9.6 parts of the intermediate product are dissolved in 50 parts of glacial acetic acid, and 2 drops of the above bromine solution are then added.
After a few minutes there is sudden discoloration, whereupon 48 parts of the bromine solution are slowly added while cooling and swirling, which is almost instantaneously discolored. After evaporation in vacuo at a bath temperature, crystallization occurs.
The colorless crystals, washed with ether, are refluxed with 100 parts of absolute pyridine for 5 hours. After evaporation in vacuo, the mixture is taken up in a lot of ether, washed with a little dilute hydrochloric acid, Soda solution and water, dried over sodium sulfate and.
evaporated. The residue is purified by chromatography on a column of aluminum oxide, the first fractions eluted with benzene giving deep and indistinct melting crystals. The other fractions eluted with benzene as well as with benzene-ether give colorless needles after recrystallization from aeeton-ether.
By saponifying them with methyl alcoholic hydrochloric acid or aqueous methyl: alcoholic potassium bicarbonate, the free d4-pregnene-3.20-dione-llss, 21-diol is obtained in the form of colorless crystals.
Instead of the chloride of 3ss, llss-diacetoxy-ätiocholanäure can be used as a starting material z. For example, a halide of a diester which is stereoisomeric in the 3-position, for example a 3a, 11ss-diacyloxy-etiocholanic acid halide, can also be used.
