Verfahren zur Darstellung von Corticosteron-ll-monoacetat. Es wurde befunden, dass man zu Cortieo- .-;leron-11-monoa.cetat gelangen kann, wenn inan ein Carbonsäurehalogenid der 11-a- Aeetoxy-ätiochola.iisä.ure, das in 3-Stellung eine durch Hydrolyse in Hydroxyl umwan- clelbare Gruppe aufweist, mit Diazomethan zum Diazoli#etoii umsetzt,
im erhaltenen Pro- (Ilikt die geschützte Hydroxylgruppe in 3-Stellung in Freiheit setzt, in beliebiger Reihenfolge diese :llydroxy lgruppe mit Oxy- d;
itionniitteln in eine Ketogruppe und die 17.tä.ndige Gruppierung mit Säuren in eine veresterte Ketolgruppe umwandelt, in a-Stel- hing zur 3-Ketogruppe eine Doppelbindung (hireli Halogenierung und nachträgliche 1-1a- Io#en-,vasserstuffabspaltung einführt und ;
rhliesslich mit. hydrolysierenden Mitteln die veresterte in eine freie Ketolgruppe über führt.
Beim Rest in 3-Stellung des Ausgangs =toffes:kann es sich z. B. um eine durch an- orgaiiiselie oder organische Säuren, wie C'ar1-)oiiaäuren, Sulfonsäuren oder lfalogen- wasserstoffsä,uren, veresterte oder durch Al kohole oder Phenole, wie Triarylmethyl- oder Benzylalkohole,
enolisierte oder aceta- lisierte Carbonylverbindungen,verä.therte1-Iy- droxylgruppe handeln. Die räumliche Lage der Acetoxygruppe in 11-Stellung des Aus entspricht derjenigen der 11- Hydroxylgruppe des Corticosterons.
Die Umsetzung des Carbonsäurehalo- genids mit Diazomethan erfolgt unter Ha.lo- genwasserstoffabspaltung und Bildung eines Diazoketons-CO-CHNz insbesondere dann, wenn ständig ein Überschuss von Diazo- methan zugegen ist.
Das erhaltene Diazoketon kann in rohem Zustande oder nach Abtrennung und Reini gung weiter verarbeitet werden. Die freie Hydroxylgruppe in 3-Stellung wird mit hy- drolysierenden Mitteln hergestellt.
Die freie 3-Hydroxylgruppe wird durch Oxydationsmittel, z. B. mit Chromsäure in Eisessig. oder durch Erhitzen mit Kupfer pulver, Einwirkung von Metallalkoholaten bzw. -phenolaten in Gegenwart von Betonen, wie Aceton oder Cyclohexanon, in eine Keto- guruppe übergeführt.
Nachoder auch vor der Oxydation der 3-Hy- droxylgruppe wird die vorhandene Diazo- ketongruppierung durch EinZvirkung von Säuren, z.
B. wasserfreien oder verdünnten, organischen oder anorganischen Säuren, bei spielsweite Essigsäure, Propionsäure, Butter säuren, Crotonsäuren, Palmitinsäure, Benzoe- säure, Phenylessigsäure, Schwefelsäure, Me- thansulfosä.ure, Toluolsulfosäure, Halogen wasserstoffsäure, Phosphorsäure oder Bor säure, in eine veresterte Ketolgruppe über- geführt. Es können so z.
B. Acylate, Ha- logenide oder Sulfonsäureester erhalten werden.
Die Doppelbindung in a-Stellung zur 3 Ketogruppe wird durch Halogenierung und nachträgliche Halogenwa.sserstoffabspaltung gebildet. Schliesslich wird die veresterte Ketol- gruppe mit hydrolysierenden Mitteln ver seift.
Dabei -v#4rd man auf die relative Alkali- ernpfindlichkeit der a-Oxyketon-Gruppierung Rücksicht nehmen, so dass man vorteilhaft mit Säuren oder mit ganz schwachen Alka- lien, wie Bikarbonaten, arbeitet.
Das neue Verfahrensprodukt, das Cortico- steron-11-monoacetat bildet farblose Kristalle und soll therapeutische Verwendung finden oder als Zwischenprodukt zur Herstellung therapeutisch wertvoller Verbindungen die nen.
<I>Beispiel:</I> 1,1 Teile 3a,lla-Diacetoxy-ätiocholan- säure werden in 6 Teilen reinstem Thionyl- chlorid gelöst und unter Feuchtigkeits- ausschluss 16 Stunden bei Zimmertemperatur stehengelassen. Dann wird im Vakuum ein gedampft und der Rückstand, das rohe 3a,lla-Diacetoxy-ätiocholansäure-chlorid, in 10 Teilen absolutem Benzol gelöst.
Die benzolische Lösung des so erhaltenen Ausgangsstoffes wird nun bei 0 in eine trockene, aus 5 Teilen Nitroso-methylharn- stoff bereitete ätherische Lösung von Diazo- methan eingetragen. Die Mischung 'bleibt zu nächst 2 Stunden bei 0 , dann noch 12 Stun den bei Zimmertemperatur stehen, wird hierauf bei 50 Badtemperatur eingedampft und im Vakuum getrocknet.
Der gelbe Rück stand (rohes Diazoketon) wird in 15 Teilen Methanol gelöst, mit der Lösung von 0,75 Teil Kaliumhydroxyd in 1 Teil Wasser und 35 Teilen Methanol versetzt und 8 Stunden bei Zimmertemperatur stehengelassen. Dann wird eine Lösung von 1,6 Teilen Kalium bikarbonat in 50 Teilen Wasser zugegeben, da.s Methanol im Vakuum entfernt und das Produkt durch Abdampfen mit absolutem Benzol völlig getrocknet.
Nun wird mit 200 Teilen absolutem Benzol, 80 Teilen trocke nem Aceton und 5 Teilen Aluminium- phenolat 20 Tage in einer Ampulle einge schmolzen stehengelassen. Hierauf wird stark eingeengt, mit Äther aufgenommen, die Lö sung mit ganz verdünnter Salzsäure, Sofia lösung und Wasser gewaschen, über Na- triumsulfat getrocknet, eingedampft und der Rückstand im Vakuum, bis zur völligen Ent fernung des freien Phenols sehr gut ge trocknet.
Einfacher lässt sich die freie 3-Hy- droxylgruppe mittels Chromsäure in Eisessig in an sich bekannter Weise zur Ketogruppe oxydieren, wobei man zweckmässig vorher die Diazoketon- in die veresterte Ketolgruppe umwandelt.
Das Rohprodukt wird nun mit 15 Teilen reinstem, wasserfreiem Eisessig 1/2 Stunde auf 100 erhitzt, wobei zuerst reichlich Stick stoff entweicht. Eindampfen im Vakuum gibt das rohe Pregnan-3,20-dion-lla,21- diacetat. Es kann, nach der Durchlauf methode über Al, , chromatographisch gerei nigt werden.
Dieses Diketon wird in Eisessig mit 1 Mol Brüm bromiert, und aus dem rohen Bromierungsprodukt durch 5stündiges Ko chen mit absolutem Pyridin oder durch #3'stündiges Erwärmen mit Dimethylanilin auf 150 Bromwasserstoff abgespalten.
Das so erhaltene Rohprodukt wird in Äther auf- genommen., die Lösung mit verdünnter Salz säure, Sodalösung und Wasser gewaschen, über Natriumsulfat getrocknet und einge dampft. Der Rückstand wird chromatogTa- phi.sch gereinigt.
Man erhält auf diese Weise das Corticosteron-11,22-diacetat, das durch Verseifung mit wässerig - methylalkoholi- achem Kaliumbikarbonat ins Corticosteron- 11-monoacetat überführt wird. Das Produkt stellt farblose Kristalle dar.
An Stelle des Halogenids der 3a,11a-Di- acetoxy-ätiocholansäure, lässt sich als Aus gangsstoff z. B. auch ein Halogenid eines in 3-Stellung stereoisomerer Diesters, das heisst ein 3ss-Acyloxy-lla-acetoxy-ätiocholansäure- halogenid verwenden.

enolized or acetalized carbonyl compounds, etherified 1-hydroxyl groups act. The spatial position of the acetoxy group in the 11-position of Aus corresponds to that of the 11-hydroxyl group of corticosterone.
The reaction of the carboxylic acid halide with diazomethane takes place with elimination of halogenated hydrogen and formation of a diazoketone — CO — CHN2, in particular when there is always an excess of diazomethane.
The diazoketone obtained can be processed further in the raw state or after separation and purification. The free hydroxyl group in the 3-position is produced with hydrolyzing agents.
The free 3-hydroxyl group is oxidized by oxidizing agents, e.g. B. with chromic acid in glacial acetic acid. or converted into a keto group by heating with copper powder, the action of metal alcoholates or metal phenolates in the presence of concretes such as acetone or cyclohexanone.
After or before the oxidation of the 3-hydroxyl group, the diazo ketone grouping present is removed by the action of acids, e.g.
B. anhydrous or dilute, organic or inorganic acids, for example acetic acid, propionic acid, butyric acids, crotonic acids, palmitic acid, benzoic acid, phenylacetic acid, sulfuric acid, methanesulphonic acid, toluenesulphonic acid, hydrohalic acid, phosphoric acid or boric acid in one esterified ketol group transferred. B. acylates, halides or sulfonic acid esters can be obtained.
The double bond in the a position to the 3 keto group is formed by halogenation and subsequent elimination of hydrogen halide. Finally, the esterified ketol group is saponified with hydrolyzing agents.

The new process product, corticosterone-11-monoacetate, forms colorless crystals and is intended to be used therapeutically or as an intermediate product for the production of therapeutically valuable compounds.
<I> Example: </I> 1.1 parts of 3a, IIIa-diacetoxy-etiocholanic acid are dissolved in 6 parts of the purest thionyl chloride and left to stand for 16 hours at room temperature with the exclusion of moisture. Then it is evaporated in vacuo and the residue, the crude 3a, IIIa-diacetoxy-ätiocholansäure-chloride, dissolved in 10 parts of absolute benzene.
The benzene solution of the starting material thus obtained is then introduced at 0 into a dry, ethereal solution of diazomethane prepared from 5 parts of nitrosomethylurea. The mixture remains at 0 for the next 2 hours and then at room temperature for a further 12 hours. It is then evaporated at a bath temperature and dried in vacuo.
The yellow residue (crude diazoketone) is dissolved in 15 parts of methanol, treated with a solution of 0.75 part of potassium hydroxide in 1 part of water and 35 parts of methanol and left to stand for 8 hours at room temperature. Then a solution of 1.6 parts of potassium bicarbonate in 50 parts of water is added, the methanol is removed in vacuo and the product is completely dried by evaporation with absolute benzene.
It is now allowed to stand melted in an ampoule for 20 days with 200 parts of absolute benzene, 80 parts of dry acetone and 5 parts of aluminum phenolate. It is then concentrated strongly, taken up with ether, the solution washed with very dilute hydrochloric acid, Sofia solution and water, dried over sodium sulfate, evaporated and the residue dried very well in vacuo until the free phenol has been completely removed.
The free 3-hydroxyl group can more easily be oxidized to the keto group by means of chromic acid in glacial acetic acid in a manner known per se, in which case the diazoketone group is advantageously converted into the esterified ketol group beforehand.
The crude product is then heated to 100 for 1/2 hour with 15 parts of the purest, anhydrous glacial acetic acid, with plenty of stick material escaping first. Evaporation in vacuo gives the crude pregnane-3,20-dione-IIIa, 21-diacetate. It can be purified chromatographically using the continuous flow method over Al.
This diketone is brominated in glacial acetic acid with 1 mol of bromine, and cleaved from the crude bromination product by boiling for 5 hours with absolute pyridine or by heating with dimethylaniline to 150% hydrogen bromide for 3 hours.
The crude product obtained in this way is taken up in ether. The solution is washed with dilute hydrochloric acid, soda solution and water, dried over sodium sulphate and evaporated. The residue is purified by chromatography.
In this way, corticosterone-11,22-diacetate is obtained, which is converted into corticosterone-11-monoacetate by saponification with aqueous - methyl alcoholic potassium bicarbonate. The product is colorless crystals.
Instead of the halide of 3a, 11a-di-acetoxy-ätiocholansäure, can be used as starting material z. B. also use a halide of a 3-position stereoisomeric diester, that is to say a 3ss-acyloxy-IIIa-acetoxy-ätiocholansäure- halide.
