Verfahren zur Darstellung von Behydr ocortlcoster on. Es wurde gefunden, dass man zu Dehydro- corticosteron gelangen kann, wenn man ein Carbonsäurehalogenid eines Demivates der Atiocholansäure,
das im Ring A in 3-Stellung eine durch .Hydrolyse in Hydroxyl umwan delbare Gruppe und im Ring C in 11-Stellung eine Ketogruppe aufweist, mit Diazomethan zum Diazok-eton umsetzt,
im erhaltenen Pro dukt die geschützte Hydroxylgruppe in 3- Stellung in Freiheit ;setzt, in beliebiger Reihenfolge diese Hydroxylgruppe mit Oxy dationsmitteln in eine Ketogruppe und die 17ständige Gruppierung mit Säuren in eine veresterte KetoIgnippe umwandelt,
in a-Stel- lung zur 3-Ketogruppe eine Doppelbindung durch Halogenierung und nachträgliche Halogenwaeserstoffabspaltung einführt und schliesslich mit hydrolysierenden Mitteln be handelt.
Beim Rest in < 3-Stellung des Ausgangs stoffes kann es sich z. B. um eine durch an organische oder organische Säuren, wie Car- bonsäuren, Sulfonsäuren oder Halogenwasser- stoffsäuren, veresterte .oder durch Alkohole oder Phenole, wie Triarylmethyl- oder Ben- zylalkoho.e,
enolisierte oder aoetalisierte Car bonylverbindungen, verätherte Hydroxyl- gruppe handeln.
Die Umsetzung das Carbonsäurehalogenids mit Diazomethan erfolgt unter Halogenwas- serstoffabspa-Itung und Bildung eines Diazo- ketons -CO-CHN2, wenn ständig ein Über schuss von Diazoverbindung zugegen ist.
Das erhaltene Diazoketon kann in rohem Zustande oder nach Abtrennung und Reini- gung weiterverarbeitet werden. Die freie Hydroxylbaruppe in. 3-Stellung kann mit hydrolysierenden Mitteln erzielt werden.
Die freie 3-Hydroxylgruppe wird durch Oxydationsmittel, z. B. mit Chromsäure in Eisessig oder durch Erhitzen mit Kupfer pulver, Einwirkung von Metallalkoholaten bezw. -phenolaten in Gegenwart von Ketonen, wie Aceton oder Cyclohexanon, in eine Keto- gruppe übergeführt.
Nach .oder auch vor Oxydation der 3- Hydroxylgruppe wird die vorhandene Diazo- ketongruppierung durch Einwirkung von Säu- ren, z.
B. wasserfreien oder verdünnten orga nischen oder anorganischen Säuren, beispie-Is:- weis-e Essigsäure, Propionsäure, Buttersäuren, Crotonsäuren, Palmitinsäure, Benzoesäure, Phenylessigsäure, Schwefelsäure, Methan sulfosäure, Toluolsulfosäure, Halogenwasser stoffsäure, Phosphorsäure oder Borsäure,
in eine veresterte Ketolgruppe übergeführt. Es können so z. B. Acylate, Halogenide oder Sulfonsäureester erhalten werden.
Die Doppelbindung in a-Stellung zur 3- Ketogruppe wird durch Halogenierung und nachträgliche Halogenwasserstoffabspaltung gebildet.
Schliesslich wird die veresterte Ketol- gruppe mit hy dxoly sierenden Mitteln verseift. Dabei wird man auf die relative Alkali empfindlichkeit der a-Oxy-keton-Gruppierung Rücksicht nehmen, so dass man vorteilhaft mit Säuren oder mit ganz schwachen Alka- lien, wie Biearbonaten, arbeitet.
Das Verfahrensprodukt, das d 4-Preg nen- 3,11,20-trio:n-21.-ol (Dehydrocorticosteron), ist bekannt. Es soll therapeutische Verwen dung finden oder als Zwischenprodukt zur Herstellung therapeutisch wertvoller Verbin dungen dienen.
<I>Beispiel:</I> 23 Teile 3B Acetoxy-ll-keto-ätiocholan- säure vom F. 110-112 (erhältlich z. B. aus dem Methylester der 3a,12ss-Dioxy-ätio-cho- lans2ure durch Umwandlung in das entspre chende 12-Monoacetat über das 3,12-Diacetat, Oxydation mit nachträglicher Verseifung zur 3-Keto-12ss-oxy-ätiocholansäure, Überfüh rung in 3-Keto-12ss-benzoxy-ätio-cholansäure- methylester,
thermische Spaltung zum d 11,1z_ 3-K$to-ätio-cholensäuremethylester, Hydrie rung der 3=Ketogruppe zur Hydroxylgruppe, Umsetzung mit unterbromiger Säure, Oxyda tion des entstandenen Bromhydrins mit Chromsäure zum Bromketon und anschlie- Pende Entbromung mit Zinkstaub)
weiden bei 0 in 140 Teilen reinstem Thionylchlorid gelöst und unter Feuchfgkeitsausschluss: zu nächst 1j2 Stunde bei 0 , dann noch 16 Stun den bei 18 stehen gelassen.
Hierauf wird im Vakuum bei 40 Badtemperatur :eingedampft, der Rückstand in 500 Teilen abs. Benzol ge löst und in eine aus 100 Teilen Nitroso- inethylharn3toff bereitete, über Kaliumhy- droxydgetrocknete und frisch destillierte äthe rische Lösung von Diazomethen bei 0 einge tragen, wobei sofortige Gasentwicklung zu beachten ist.
Die Mischung bleibt unter Feuchtigkeitsa.usschluss (KOH) zunächst 2 Stunden bei 0 , dann noch 16 Stunden bei 18 stehen, wird hierauf bei 50 Badtempera- tur stark eingeengt und zuletzt im Vakuum völlig getrocknet. Der Rückstand wiegt 25 Teile.
Das so erhaltene rohe 21-Diazo-pregnan- 3ss-ol-11,20-dionacetat wird in 300 Teilen Methanol gelöst, mit der Lösung von 15 Tei len Kaliumhydroxyd in 20 Teilen Wasser und 700 Teilen Methanol versetzt und 8 Stun den bei 20 stehen gelassen.
Hierauf wird die Lösung von 32 Teilen Kaliumbicarbonat in 1000 Teilen Wasser zugegeben., das Methanol im Vakuum entfernt und die verbleibende Suspension mit viel Äther ausgeschüttelt. Die mit Wasser gewaschene und über Natrium sulfat getrocknete Ätherlösung wird bei 50 Ba.dtemperatur stark eingeengt und hierauf im Vakuum völlig eingedampft, wobei. 24 Teile rohes 21-Diazo-pregnan-3ss-ol-11,20-dion als hellbraunes Harz zurückbleiben.
Diese werden mit 300 Teilen, reinstem wasserfreiem Eisessig 1/ Stunde auf 95-100 erhitzt und dass dabei abgespaltene Gas in einem Messzylinder über Wasser aufgefangen. Nach Beendigung der Stickstoffentwicklung wird im Vakuum eingedampft. Es verbleiben 24 Teile Rückstand, der in an sich bekannter Weise über Aluminiumoxyd chromato:gra- phisch gereinigt wird.
Die Benzol-Äther=Eluate geben beim Um- kristallisieren aus Äther-Petroläther das Pregnan-3ss-21-diol-1.11?0-dion-21-momoacetat in farblosen Blättchen vom F. 178-181 .
(Acetylie:rung mit Essigsäure-anhydrid und Pyridin würde zum entsprechenden Diacetat vom F. 169-171 führen.) 8,5 Teile dies beschriebenen M.onoacetates werden in 100 Teilen Eisessig gelöst, mit 100 Teilen 2 % iger Chromtiioxyd-Eisessiglösung (= 2 Teile Cr03) versetzt und 16 Stunden bei. 20 stehen,
gelassen. Hierauf wird im Vakuum bei 30 Badtemperatur stark einge engt, mit -'@Tass,er versetzt und mit Äther aus geschüttelt. Die mit verdünnter Schwefel säure, Sodalösung und Wasser gewaschene und über Natr¯i.umslüfat getrocknete Äther lösung wird stark eingeengt und die bald ein setzende Kristallisation durch Zusatz von etwas Petroläther vervollständigt. Man erhält auf dies,
e Weise das Pregnan-3,11,20-trion-21- el-acetat in farblosen Nadeln vom F. 153 bis 155 und der spezifischen Drehung [a] D = -J- 107,2 4 (c = 0,783 in Aceton).
Zu demselben Produkt gelangt man auch, wenn das 21-Diazo-pregnan-3f-ol-11,20-dion zunächst z. B. bei Zimmertemperatur mit A.luminium-isopropylat und Aceton behan delt und die erhaltene Verbindung anschlie ssend mit Eisessig erhitzt wird.
Durch Ve27nischen von 10 Teilen Brom mit 384 Teilen Eisessig wird zunächst eine n-Bromlösung bereitet. 9,6 Teile des Produk tes vom F.
153-155 werden in 50 Teilen Eisessig gelöst und hierauf mit 2 Tropfen der obigen Bromlösung versetzt. Nach einigen Minuten tritt plötzliche Entfärbung ein., worauf man unter Kühlung und Umschwen ken langsam noch 48 Teile der Bromlösung zugibt,
die fast momentan entfärbt wird. Nach Eindampfen im Vakuum bei 25 Bad- temp.eratur tritt Krztstallisation ein. Die mit Äther gewaschenen farblosen Kristalle schmel zen bei 180--185 . Sie stellen das 4-Brom- pregnan-3,11,20-trion-21-ol-acetat dar.
Dieses Bromid wird mit 100 Teilen abso lutem Pyridin 5 Stunden unter Rückfluss ge kocht. Nach Eindampfen im Vakuum wird in viel Äther aufgenommen, mit wenig ver- dünnter Salzsäure, Sodalösung und Wasser gewaschen,
über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird über eine Säule von Aluminiumoxyd chromatogra- phisch gereinigt, wobei die ersten mit Benzol eluierten Anteile tief und unscharf schmel zende Kristalle geben.
Die weiteren mit Benzol sowie mit Benzol-Äther eluierten Fraktionen g<B>W</B> eben nach zweimaligem Umkris.tallisi@eren aus Aceton-Äther farblose Nadeln vom F. 175-178 und der spezifischen Drehung [a]v =+ 210,7' 3 (c=0,676 in Aceton).
Es handelt sich um das 44-Pregnen-3,11,20- trion - 21- o.1 - acetat (Dehy dro - corticosteron- a.oetat). Eine Probe reinstes natürliches De- hydro-corticosteron-acetat schmilzt unter den selben Bedingungen bei 177-179 ,
zeigt die obige spezifische Drehung und gibt keine Schmelzpunktdepression im Gemisch mit dem Synthesenprodukt. Durch Verseifung mit methylalkoholischer Salzsäure oder wässerig- methylalkoh-olischem Kaliumbioarbonat wird das freie Dehydro-corticosteron vom F. 174 bis 180 hergestellt.
An Stelle dem 3,ss-Acetoxy-11-keto-ätio- cholanusäure läss.t sich als Ausgangsstoff auch ein in 3-Stellung stereoisomerer Ester ver wenden.

which has a group which can be converted into hydroxyl by hydrolysis in the 3-position and a keto group in the 11-position in the C ring, reacts with diazomethane to form the diazoketone,
In the product obtained, the protected hydroxyl group in the 3-position is set free; in any order, this hydroxyl group is converted into a keto group with oxidizing agents and the grouping with acids into an esterified keto group,
introduces a double bond in the a position to the 3-keto group by halogenation and subsequent elimination of hydrogen halide and finally treats it with hydrolyzing agents.
When the rest in the <3 position of the starting material it can be, for. B. by an organic or organic acids, such as carboxylic acids, sulfonic acids or hydrogen halide acids, esterified. Or by alcohols or phenols, such as triarylmethyl or benzyl alcohol.

The reaction of the carboxylic acid halide with diazomethane takes place with elimination of hydrogen halide and formation of a diazo ketone -CO-CHN2 if there is always an excess of diazo compound.
The diazoketone obtained can be further processed in the raw state or after separation and purification. The free hydroxyl group in the 3-position can be achieved with hydrolyzing agents.
The free 3-hydroxyl group is oxidized by oxidizing agents, e.g. B. with chromic acid in glacial acetic acid or by heating with copper powder, exposure to metal alcoholates or. -phenolates in the presence of ketones, such as acetone or cyclohexanone, converted into a keto group.
After or before oxidation of the 3-hydroxyl group, the diazo ketone grouping present is removed by the action of acids, e.g.
B. anhydrous or dilute organic or inorganic acids, for example: white acetic acid, propionic acid, butyric acid, crotonic acid, palmitic acid, benzoic acid, phenylacetic acid, sulfuric acid, methanesulfonic acid, toluenesulfonic acid, hydrogen halide acid, phosphoric acid or boric acid,
converted into an esterified ketol group. It can be B. acylates, halides or sulfonic acid esters can be obtained.
The double bond in the a-position to the 3-keto group is formed by halogenation and subsequent elimination of hydrogen halide.
Finally, the esterified ketol group is saponified with hy dxolizing agents. In doing so, the relative alkali sensitivity of the a-oxy-ketone group will be taken into account, so that it is advantageous to work with acids or with very weak alkalies such as carbonates.

thermal cleavage to d 11,1z_ 3-K $ to-etio-cholenic acid methyl ester, hydrogenation of the 3 = keto group to the hydroxyl group, reaction with hypobromous acid, oxidation of the bromohydrin formed with chromic acid to give bromine ketone and subsequent debromination with zinc dust)
Weiden at 0 in 140 parts of the purest thionyl chloride and with exclusion of moisture: left to stand at 0 for the next 1j2 hour, then at 18 for 16 hours.
Then in a vacuum at 40 bath temperature: evaporated, the residue in 500 parts abs. Benzene dissolved and added to an ethereal solution of diazomethene prepared from 100 parts of nitrosoethyl urea, dried over potassium hydroxide and freshly distilled, whereby the immediate evolution of gas must be observed.
The mixture remains under the exclusion of moisture (KOH) initially for 2 hours at 0, then for a further 16 hours at 18, is then strongly concentrated at a bath temperature and finally completely dried in vacuo. The residue weighs 25 parts.
The crude 21-diazo-pregnan-3ss-ol-11,20-dione acetate obtained in this way is dissolved in 300 parts of methanol, the solution of 15 parts of potassium hydroxide in 20 parts of water and 700 parts of methanol is added and the mixture is kept at 20 for 8 hours calmly.
The solution of 32 parts of potassium bicarbonate in 1000 parts of water is then added, the methanol is removed in vacuo and the remaining suspension is extracted by shaking with plenty of ether. The ether solution, washed with water and dried over sodium sulfate, is strongly concentrated at 50 Ba.demperature and then completely evaporated in vacuo, with. 24 parts of crude 21-diazo-pregnan-3ss-ol-11,20-dione remain as a light brown resin.
These are heated to 95-100 with 300 parts of the purest anhydrous glacial acetic acid 1 / hour and the gas that is split off is collected in a measuring cylinder over water. An n-bromine solution is first prepared by mixing 10 parts of bromine with 384 parts of glacial acetic acid. 9.6 parts of the product from F.
153-155 are dissolved in 50 parts of glacial acetic acid, and 2 drops of the above bromine solution are then added. After a few minutes there is sudden discoloration, whereupon 48 parts of the bromine solution are slowly added while cooling and swirling,
which is almost instantly discolored. After evaporation in a vacuum at a bath temperature of 25, crystallization occurs. The colorless crystals, washed with ether, melt at 180--185. They represent 4-bromo-pregnane-3,11,20-trione-21-ol-acetate.
This bromide is refluxed with 100 parts of absolute pyridine for 5 hours. After evaporation in vacuo, it is taken up in a lot of ether, washed with a little dilute hydrochloric acid, soda solution and water,
dried over sodium sulfate and evaporated. The residue is chromatographically purified over a column of aluminum oxide, the first fractions eluted with benzene giving deep and indistinct melting crystals.
The other fractions g <B> W </B> eluted with benzene as well as with benzene ether just after two umkris.tallisi@eren from acetone-ether colorless needles of F. 175-178 and the specific rotation [a] v = + 210.7 '3 (c = 0.676 in acetone).
It is the 44-pregnen-3,11,20-trione-21- or 1-acetate (dehydro-corticosterone-a.oetat). A sample of the purest natural dehydro-corticosterone acetate melts under the same conditions at 177-179,
shows the above specific rotation and gives no melting point depression in admixture with the synthesis product. The free dehydrocorticosterone with a mp of 174 to 180 is produced by saponification with methyl alcoholic hydrochloric acid or aqueous methyl alcoholic potassium bicarbonate.
Instead of the 3, ß-acetoxy-11-keto-ethio-cholanic acid, an ester stereoisomeric in the 3-position can also be used as the starting material.
