Die Einführung von Halogenatomen in die 17a- Stellung erschien von besonderem Interesse, da Ha logen-Substitutionen in unmittelbarer Nachbarschaft von wichtigen Keton- oder Carbinolgruppen zu einem biologisch hochaktiven Produkt führen kön nen. So sind beispielsweise zahlreiche 9a- oder 12a halogenierte, 11-oxydierte Steroide wirksame Corti- coide. 21-Fluorprogesteron ist ein sehr aktives Luteoid und einige 21-fluorierte Corticoide besitzen ebenfalls starke Aktivität.
Eine Reihe repräsentativer Reaktionen, welche zu 17-halogenierten Hormon-Analogen der Proge- steron-Corticoidgruppe und Ausgangsmaterialien des vorliegenden Verfahrens führen, können, zusammen mit dem erfindungsgemässen Verfahren, wie folgt dargestellt werden:

Ausgangsmaterialien der Formel Va können wie folgt hergestellt werden: Ein 3 - Acyloxy - 5 - pregnen - 20- on-Derivat der Formel I wird in an sich bekannter Weise in eine Mischung der entsprechenden 17-En-20-ol-acetate II und<B>111</B> oder zu analogen Enolestern umgewandelt durch Einwirkung von Mitteln wie Essigsäureanhydrid in Gegenwart von Katalysatoren, wie z.
B. Para- toluolsulfonsäure. Die Mischung wird dann zu einem 5,6,17-Trihalogenid der allgemeinen Formel Va halogeniert. Das Progesteronderivat 1 kann auch direkt zum Trihalogenid Va halogeniert werden.
Gemäss vorliegender Erfindung wird sodann die Acyloxygruppe des Trihalogenids vorzugsweise in saurem Medium hydrolysiert; die resultierende 3 - Oxy - 5,6,17 - trihalogen - verbindung V kann mit Chromsäure in einem geeigneten Lösungsmittel wie Essigsäure, Aceton, Chloroform, Tetrachlorkohlen- stoff, Methylenchlorid usw. oxydiert werden unter Bildung des Trihalogenderivates der Formel IV.
Die Doppelbindung in 5,6-Stellung wird durch partielle Dehalogenierung vorzugsweise mit einem Alkali- jodid, beispielsweise mit Natriumjodid, hergestellt, vorzugsweise z.
B. durch eine kurze Behandlung bei erhöhter Temperatur in einem geeigneten Lösungs mittel, beispielsweise durch kurzes Kochen der Mi schung in trockenem Aceton am Rückfluss, vorzugs weise unter Sauerstoffausschluss. Das resultierende 17-Monohalogenid der Formel VII wird durch Be handlung mit Säure in das d4-17-halogenierte Produkt der Formel VIII umgewandelt.
Die Reaktionsfolge kann auch umgekehrt werden, indem die Acyloxy-trihal'ogen-Verbindung der For mel Va unter Einwirkung eines Alkalijodids teilweise dehalogeniert wird und das resultierende Acyloxy- monohalogenid der Formel VIa in saurem Medium hydrolysiert wird zum entsprechenden freien Alkohol der Formel VI. Der letztere kann mit Chromsäure oxydiert werden zum 45-3-Oxo-halogenid der For mel VII, welches, wie oben beschrieben,
zum 17-ha- logenierten Endprodukt der Formel VIII umgelagert wird.
Als besonders wirksames Therapeutikum hat sich das so erhältliche 17a-Brom-progesteron erwiesen, dessen Herstellung folgendem Schema folgt:

Bei der Herstellung dieser besonderen Verbin dung kann 5a,6ss,17a-Tribrom-3ss-acetoxy-5-allo- pregnan (XIIIa) als Ausgangsmaterial verwendet werden. Es wurde aus d5-3ss-Acetoxy 20-oxopregnan (1X) über die Mischung der isomeren Enolacetate X und XI hergestellt.
Die Bromierung wurde in Ge- genwart von Kaliumacetat durchgeführt, um Neben reaktionen zu vermeiden.
Die Acetylgruppe der Tribrom -Verbindung (XIIIa) wurde mit Salzsäure in einer Mischung von absolutem Methanol und Methylenehlorid hydroly- siert und das resultierende unstabile Oxy-tribromid (XIII) mit Chromsäure in Essigsäure oder in Aceton in Gegenwart von Schwefelsäure zum Tribromketon (XII) oxydiert.
Dieses unstabile Produkt wurde nicht in reinem Zustand isoliert, sondern selektiv in 5- und 6-Stellung durch die Wirkung von Natriumjodid in siedendem Aceton entbromiert. Ein Teil des Reak tionsproduktes (vgl. XV) hatte sich dabei während der Reaktion in das d4-3-Keto-derivat (XVI) umge lagert. Um die Verschiebung der Doppelbindung zu vervollständigen, wurde das rohe Produkt während kurzer Zeit mit Salzsäure in siedendem Äthanol be handelt.
Das resultierende rohe 17a-Brom-progesteron (XVI) wurde gereinigt durch Chromatographieren an Silikagel und durch Umkristallisierung aus Äther- Hexan.
17a-Brom-progesteron kann ebenfalls auf andere Weise erhalten werden, und zwar in einer gegenüber der eben beschriebenen Art wesentlich vorteilhafteren Weise, indem alle Zwischenprodukte stabil sind und kristalline Produkte darstellen. Das Tribromketon (XIIIa) wird selektiv zum Acetoxy-monoketon (XIVa) debromiert mit Natriumjodid in siedendem Aceton. Dieses Vorgehen ergibt bessere Resultate als das langdauernde Behandeln bei Zimmertemperatur.
Die AcetyIgruppe der Verbindung (XIVa) wird mit Salzsäure in Methanol oder mit Perchlorsäure in Methanol hydrolysiert und das erhaltene Oxybrom- keton (XIV) wird mit Chromsäure in Aceton in Ge genwart von Schwefelsäure oxydiert. Das rohe Reak tionsprodukt (vgl. XV) wird direkt zu 17a-Brom- progesteron umgelagert.
<I>Beispiel 1</I> ce) Herstellung des Ausgangsmaterials 5a,61i,17a- Tribrom-3r-acetoxy-5-allopregnan-20-on (XlIIa): Einer Lösung von 19,758 g 3/3 - Acetoxy- 5 - pregnen-20-on (IX) in 1 Liter Essigsäureanhydrid wurden 5,56 g p-Toluolsulfonsäure zugesetzt. Die Lösung wurde durch Destillation langsam auf ein Volumen von 500 cm3 im Verlaufe von 8 Stunden reduziert.
Anschliessend wurde der grösste Teil des Lösungsmittels im Vakuum entfernt und der Rück stand mit Äther extrahiert. Die ätherische Lösung wurde mit Wasser, eisgekühlter, gesättigter Bicarbo- natlösung und Wasser gewaschen und getrocknet. Das Lösungsmittel wurde entfernt und das resultie rende Produkt in einer kleinen Menge Benzol auf gelöst und an 200g Aluminiumoxyd (pH 6) absor biert. Die Eluierung mit 1,5 1 Benzol ergab 19,477 g einer gelblichen, kristallinen Mischung der Enol- acetate X und XI.
Diese Mischung wurde in 250 cm- Essigsäure gelöst und tropfenweise unter Rühren innerhalb von 10 Minuten und bei Raumtemperatur mit einer Lö sung von 17,09 g Brom in 103,6 cm3 Essigsäure, welche mit einer Lösung von 4,762 g Kaliumacetat in 3 em3 Wasser vermischt worden war, versetzt. Am Ende der Beimischung dieser Bromlösung kristalli- sierte das Tribromid (XIIIa) aus dem Reaktions gemisch.
Die Filtration ergab 20,665 g farbloser Blättchen, welche nach einer Umkristallisation aus Methylenchlorid -Methanol 19,148 g Tribromid (XIIIa), Fp. 156-157 ergaben. Die Mutterlaugen wurden mit dem Filtrat der ursprünglichen Kristalli sation vereint und 70 cm3 Wasser zugefügt. Die Mi schung wurde auf 85 erwärmt und in üblicher Weise während 30 Minuten mit 10 g Zinkstaub behandelt.
Die übliche Aufarbeitung ergab ein amorphes Pro dukt, welches nach Chromatographieren 4,424 g des Ausgangsmaterials I, Fp. 136-138 ergab (totale Ausbeute an Tribromid XIIIa aus IX im Hinblick auf die Rückgewinnung des Ausgangsmaterials, 90%). b) 5a,6,d,17a-Tribrom-3/3-oxy-5-allopregnan- 20-on (XIII):
Eine Lösung von 2 g Tribromacetat XIIIa, Fp. 156-157 wurde in 15 cm?, Methylenehlorid ge- löst und mit 30 cm3 einer 1% igen Lösung von Chlorwasserstoff in absolutem Methanol behandelt.
Die Lösung wurde bei Raumtemperatur während 24 Stunden dunkel gehalten. Hierauf wurde Äther zu gefügt und die Lösung mit Wasser, eisgekühlter Na- tri.umbicarbonatlösung und Wasser gewaschen und hierauf getrocknet. Die Lösungsmittel wurden ent fernt, zuerst bei atmosphärischem Druck, anschlie- ssend im Vakuum. Nach Beifügung von Methanol kristallisierte das Produkt aus.
Die Filtration ergab <B>1</B> 57 g Oxytribromid XIII, Fp. 121-123 (Zers.), der nach Zufügung von Tribromacetat XIIIa eine Depression erfuhr. Das Produkt ist nicht stabil. Der Schmelzpunkt wurde durch Umkristallisation aus Me- thylenchlorid-Methanol auf 93-96 gesenkt. Das Addukt wurde hierauf ohne weitere Reinigung in der folgenden Reaktion verwendet.
5a,6,ss,17a-Tribrom-5-allopregnan-3,20-dion (XII): a) Einer Lösung von 1 g Oxy-tribromid XIII, Fp. 92-93 (Zers.) in 10 cm3 Essigsäure wurden bei Raumtemperatur 135 mg Chromsäure in 2 cm3 Essigsäure und 0,5 cm3 Wasser zugefügt. Nach 18 Stunden wurde die Mischung in Wasser geschüttet und mit Äther extrahiert. Die ätherische Lösung wurde mit Wasser, eisgekühlter Natriumbicarbonat- lösung und Wasser gewaschen und getrocknet.
Das Lösungsmittel wurde im Vakuum entfernt. Im Ver lauf dieser Operation kristallisierte ein Teil des Pro duktes aus. Die Mutterlaugen wurden getrocknet und aus Methanol umkristallisiert. Es wurden 663 mg (66%-) kristallines Tribromdiketon XII, Fp. 92-93 (Zers.) erhalten. Das Produkt war sehr unstabil.
b) Eine Lösung von 1,4 g Oxytribromid XIII, Fp. 95-96 , in 20 cm3 trockenem Aceton wurde mit 2 cms einer Lösung behandelt, welche aus 100 g Chromsäure, 160 g konzentrierter Schwefelsäure und 500 em3 Wasser bereitet worden war.
Die Reak tionsmischung wurde während 24 Stunden bei 0 ge halten und anschliessend mit Äther extrahiert. Die ätherische Lösung wurde mit Wasser, eisgekühlter Natriumbisulfitlösung, eisgekühlter Natriumbicarbo- natlösung und Wasser gewaschen und getrocknet. Die Entfernung des Lösungsmittels im Vakuum er gab 1,4 g eines teilweise kristallinen, nicht stabilen Reaktionsproduktes (vgl. XII), welches in der Folge ohne weitere Reinigung verwendet wurde.
Teilweise Entbromierung des Tribromids IV Rohes Tribromidketon XII (1,4 g) wurde in 50 cm3 absolutem Aceton gelöst und während 10 Minuten mit 750 mg Natriumjodid am Rückfluss ge kocht. Die Lösung wurde auf Raumtemperatur ab gekühlt und das freie Jodid durch Zusatz von 36 cm3 einer 0,1n Thiosulfatlösung titriert. Hierauf wurde die Mischung mit Äther extrahiert und die ätherische Lösung mit Wasser gewaschen und getrocknet.
Die Entfernung des Lösungsmittels im Vakuum ergab 985 mg eines rohen Monobromids XV, das etwa -1-1-3-Keton enthielt, welches durch ein Absorptions maximum geringer Intensität bei 240 ma angezeigt wurde.
17a-Bromprogesteron (XVI) aus Tribromid XIIIa über die Zwischenprodukte XIII, XII und XV: Das obenerwähnte rohe amorphe d5-3,20-Dioxo- bromid (XV) 985 mg) wurde in 40 cm3 Methanol ge löst und während 5 Minuten mit 0,5 em3 Wasser, das einen Tropfen konzentrierter Salzsäure enthielt, am Rückfluss gekocht.
Das Produkt wurde mit Äther extrahiert und die ätherische Lösung mit Wasser, eisgekühlter Natriumbicarbonatlösung und mit Wasser gewaschen und getrocknet. Die Lösungsmittel wurden im Vakuum entfernt und das resultierende Produkt mit Benzol getrocknet. Es wurden 979 mg Nadeln erhalten, welche rohes 17a-Brom-progesteron (XVI) darstellten.
Das Produkt wurde in Benzol gelöst und über 30 g Silikagel chromatographiert. Benzol, das 2 % Äthylacetat enthielt, eluierte 780 mg kristallines Material, das nach einer Umkristallisation aus Me- thylenchlorid-Hexan bei 162-163 schmolz. Eine Probe wurde zu Analysenzwecken zweimal umkristal lisiert; Fp. 162-163 .

C 64,24 H 7,39 Br 20,51 <I>Beispiel 2</I> 17a-Brom-3p-acetoxy-5-pregnen-20-on (XIVa) Eine Lösung von 2,27g Tribromacetat XIIIa, Fp. 156-157 in 40 cm3 absolutem Aceton wurde während 10 Minuten in Stickstoffatmosphäre mit einer Lösung von 2 g Natriumjodid in 10 cm:' abso lutem Aceton am Rückfluss gekocht.
Nach Abkühlung wurde das freigewordene Jod mit 75 cm3 einer 0,1 n Natriumthiosulfatlösung titriert. Hierauf wurde das Produkt mit Wasser ausgefällt. Die Filtration ergab 1,315g Monobromid XIVa, das in Blättchen kristal lisierte und bei 140,5-141 schmolz.
Die Mutter laugen ergaben eine weitere Portion von 225 mg XIVa, Fp. 136-139 (totale Ausbeute 92%). Eine Probe wurde zu Analysezwecken zweimal aus Me- thylenchlorid-Methanol umkristallisiert; Fp. 146 bis 147 .
Analyse: Berechnet für C,3H3.303Br: C 63,51 H 7,61 Br 18,27 Gefunden: C 63,30 H 7,63 Br 18,17 17a-Brom-3/3-hydroxy-5-pregnen-20-on (XIV): a) Acetat XIVa (370 mg, Fp. 144,5-145,5 ) wurde in 15 cm3 absolutem Methanol, welches 150 mg Chlorwasserstoff enthielt, gelöst. Die Lösung wurde während 24 Stunden bei Raumtemperatur ge halten. Durch Zusatz von Wasser wurden 282 mg des Oxybromids XIV, Fp. 133-135 , ausgefällt (Ausbeute 84 l0).
Das Produkt zeigte zeitweilig einen doppelten Schmelzpunkt von 100j133-135 . Eine Probe wurde zu Analysezwecken zweimal aus wässri- gem Methanol umkristallisiert und ergab Blättchen vom Fp. 143,5-144,5 .
Analyse: Berechnet für C..,1H310,Br: C 63,79 H 7,90 Br 20,21 Gefunden: C 63,52 H 7,73 Br 20,36 b) Eine Lösung von 391 mg Acetat (XIVa) in 15 cm3 Methanol, das 0,38 cm3 72 /o iger Perchlor- säure enthielt, wurde während 24 Stunden bei Raum temperatur gehalten. Die Ausfällung mit Wasser er gab 286 mg des Oxybromids XIV, Fp. 139,5-l40,5 , das identisch ist mit dem unter a) beschriebenen Produkt.
Oxydation des 17a-Brom-3/3-oxy-5-pregnen-20-ons (17("-Brom-5-pregnen-3,20-dion [XV]) Einer Lösung von 286 mg des Oxybromids XIV in 40 cm3 absolutem Aceton wurden 0,24 cm3 der oben beschriebenen oxydierenden Lösung (hergestellt aus<B>100</B> g Chromsäure, 160 g konzentrierter Schwe felsäure und 500 cm3 Wasser) tropfenweise unter Stickstoffatmosphäre beigefügt. Nach 5 Minuten wurde das Reaktionsprodukt mit Wasser ausgefällt.
Es wurden 239 mg rohes Diketon XV erhalten, das in Nadeln kristallisierte und bei 100-101 schmolz (Ausbeute 84%). Da das Ultraviolettspektrum des Produktes ergab, dass die Doppelbindung sich teil weise verschoben hatte, wurde das rohe Produkt ohne Reinigung in der folgenden Reaktion ver wendet.
17a-Brom-progesteron (XVI) über die Zwischen produkte XIVa, XIV und XV Das oben beschriebene kristalline Diketon XV wurde, wie oben beschrieben, umgelagert. Durch Kristallisation aus wässrigem Äthanol wurden 158 mg reines 17a-Brom-progesteron, Fp. 157-159 , erhal ten. Die Identität des Produktes mit dem vorgängig beschriebenen Produkt wurde durch Infrarot-Analyse erwiesen.

In the preparation of this particular compound, 5a, 6ss, 17a-tribromo-3ss-acetoxy-5-allo-pregnane (XIIIa) can be used as starting material. It was prepared from d5-3ss-acetoxy 20-oxopregnane (1X) via the mixture of the isomeric enol acetates X and XI.
The bromination was carried out in the presence of potassium acetate in order to avoid side reactions.
The acetyl group of the tribromo compound (XIIIa) was hydrolyzed with hydrochloric acid in a mixture of absolute methanol and methylene chloride and the resulting unstable oxy-tribromide (XIII) was oxidized with chromic acid in acetic acid or in acetone in the presence of sulfuric acid to give the tribromometone (XII) .
This unstable product was not isolated in the pure state, but selectively debrominated in the 5- and 6-positions by the action of sodium iodide in boiling acetone. Part of the reaction product (cf. XV) had been rearranged during the reaction into the d4-3-keto derivative (XVI). To complete the shift in the double bond, the crude product was treated for a short time with hydrochloric acid in boiling ethanol.
The resulting crude 17a-bromo-progesterone (XVI) was purified by chromatography on silica gel and by recrystallization from ether-hexane.
17a-bromo-progesterone can also be obtained in another way, in a way that is considerably more advantageous than that just described, in that all intermediate products are stable and are crystalline products. The tribromoketone (XIIIa) is selectively debrominated to acetoxy-monoketone (XIVa) with sodium iodide in boiling acetone. This procedure gives better results than long-term treatment at room temperature.
The acetyl group of the compound (XIVa) is hydrolyzed with hydrochloric acid in methanol or with perchloric acid in methanol and the oxybromoketone (XIV) obtained is oxidized with chromic acid in acetone in the presence of sulfuric acid. The crude reaction product (cf. XV) is rearranged directly to 17a-bromogesterone.
<I> Example 1 </I> ce) Preparation of the starting material 5a, 61i, 17a- tribromo-3r-acetoxy-5-allopregnan-20-one (XlIIa): A solution of 19.758 g of 3/3 - acetoxy- 5 - pregnen-20-one (IX) in 1 liter of acetic anhydride was added to 5.56 g of p-toluenesulfonic acid. The solution was slowly reduced to a volume of 500 cm3 over the course of 8 hours by distillation.
Most of the solvent was then removed in vacuo and the residue was extracted with ether. The ethereal solution was washed with water, ice-cold, saturated bicarbonate solution and water and dried. The solvent was removed and the resulting product was dissolved in a small amount of benzene and absorbed in 200g aluminum oxide (pH 6). Elution with 1.5 l of benzene gave 19.477 g of a yellowish, crystalline mixture of the enol acetates X and XI.
This mixture was dissolved in 250 cm acetic acid and added dropwise with stirring over a period of 10 minutes and at room temperature with a solution of 17.09 g of bromine in 103.6 cm 3 of acetic acid, which was mixed with a solution of 4.762 g of potassium acetate in 3 cm 3 of water had been transferred. At the end of the addition of this bromine solution, the tribromide (XIIIa) crystallized from the reaction mixture.
Filtration gave 20.665 g of colorless flakes which, after recrystallization from methylene chloride-methanol, gave 19.148 g of tribromide (XIIIa), melting point 156-157. The mother liquors were combined with the filtrate from the original crystallization and 70 cm3 of water were added. The mixture was heated to 85 and treated in the usual way for 30 minutes with 10 g of zinc dust.
The usual work-up gave an amorphous product which, after chromatography, gave 4.424 g of the starting material I, melting point 136-138 (total yield of tribromide XIIIa from IX with regard to the recovery of the starting material, 90%). b) 5a, 6, d, 17a-tribromo-3/3-oxy-5-allopregnan-20-one (XIII):
A solution of 2 g of tribromoacetate XIIIa, melting point 156-157, was dissolved in 15 cm 3 of methylene chloride and treated with 30 cm 3 of a 1% strength solution of hydrogen chloride in absolute methanol.
The solution was kept dark at room temperature for 24 hours. Ether was then added and the solution was washed with water, ice-cold sodium bicarbonate solution and water and then dried. The solvents were removed, first at atmospheric pressure, then in vacuo. After adding methanol, the product crystallized out.
Filtration gave 1 57 g of oxytribromide XIII, m.p. 121-123 (decomposition), which experienced depression after the addition of tribromoacetate XIIIa. The product is not stable. The melting point was lowered to 93-96 by recrystallization from methylene chloride-methanol. The adduct was then used in the following reaction without further purification.
5a, 6, ss, 17a-tribromo-5-allopregnan-3,20-dione (XII): a) A solution of 1 g of oxy-tribromide XIII, melting point 92-93 (decomp.) In 10 cm3 of acetic acid was added Room temperature 135 mg chromic acid in 2 cm3 acetic acid and 0.5 cm3 water were added. After 18 hours the mixture was poured into water and extracted with ether. The ethereal solution was washed with water, ice-cold sodium bicarbonate solution and water and dried.
The solvent was removed in vacuo. During the course of this operation, part of the product crystallized out. The mother liquors were dried and recrystallized from methanol. 663 mg (66%) of crystalline tribromodiketone XII, melting point 92-93 (decomp.) Were obtained. The product was very unstable.
b) A solution of 1.4 g of oxytribromide XIII, melting point 95-96, in 20 cm3 of dry acetone was treated with 2 cms of a solution which had been prepared from 100 g of chromic acid, 160 g of concentrated sulfuric acid and 500 cubic meters of water.
The reaction mixture was kept at 0 for 24 hours and then extracted with ether. The ethereal solution was washed with water, ice-cold sodium bisulfite solution, ice-cold sodium bicarbonate solution and water and dried. Removal of the solvent in vacuo gave 1.4 g of a partially crystalline, unstable reaction product (cf. XII) which was subsequently used without further purification.
Partial debromination of the tribromide IV. Crude tribromide ketone XII (1.4 g) was dissolved in 50 cm3 of absolute acetone and refluxed with 750 mg of sodium iodide for 10 minutes. The solution was cooled to room temperature and the free iodide was titrated by adding 36 cm3 of a 0.1N thiosulfate solution. The mixture was then extracted with ether and the ethereal solution washed with water and dried.
Removal of the solvent in vacuo gave 985 mg of a crude monobromide XV, which contained about -1-1-3-ketone, which was indicated by an absorption maximum of low intensity at 240 ma.
17a-bromoprogesterone (XVI) from tribromide XIIIa via the intermediates XIII, XII and XV: The above-mentioned crude amorphous d5-3,20-dioxobromide (XV) 985 mg) was dissolved in 40 cm3 of methanol and with 0 for 5 minutes .5 em3 of water containing a drop of concentrated hydrochloric acid was refluxed.
The product was extracted with ether and the ethereal solution washed with water, ice-cold sodium bicarbonate solution and with water and dried. The solvents were removed in vacuo and the resulting product dried with benzene. 979 mg of needles were obtained, which were crude 17a-bromo-progesterone (XVI).
The product was dissolved in benzene and chromatographed over 30 g of silica gel. Benzene containing 2% ethyl acetate eluted 780 mg of crystalline material which, after recrystallization from methylene chloride-hexane, melted at 162-163. A sample was recrystallized twice for analysis; M.p. 162-163.

C 64.24 H 7.39 Br 20.51 <I> Example 2 </I> 17a-Bromo-3p-acetoxy-5-pregnen-20-one (XIVa) A solution of 2.27 g of tribromoacetate XIIIa, m.p. 156-157 in 40 cm 3 of absolute acetone was refluxed for 10 minutes in a nitrogen atmosphere with a solution of 2 g of sodium iodide in 10 cm: of absolute acetone.
After cooling, the released iodine was titrated with 75 cm3 of a 0.1 N sodium thiosulphate solution. The product was then precipitated with water. Filtration yielded 1.315 g of monobromide XIVa which crystallized into leaflets and melted at 140.5-141.
The mother liquors gave a further portion of 225 mg of XIVa, melting point 136-139 (total yield 92%). For analysis purposes, a sample was recrystallized twice from methylene chloride-methanol; Mp. 146 to 147.
Analysis: Calculated for C, 3H3.303Br: C 63.51 H 7.61 Br 18.27 Found: C 63.30 H 7.63 Br 18.17 17a-Bromo-3/3-hydroxy-5-pregnen- 20-one (XIV): a) Acetate XIVa (370 mg, m.p. 144.5-145.5) was dissolved in 15 cm3 of absolute methanol which contained 150 mg of hydrogen chloride. The solution was kept at room temperature for 24 hours. By adding water, 282 mg of oxybromide XIV, melting point 133-135, were precipitated (yield 84 l0).
The product showed a double melting point of 100j133-135 at times. A sample was recrystallized twice from aqueous methanol for analysis purposes and gave flakes with a melting point of 143.5-144.5.
Analysis: Calculated for C .., 1H310, Br: C 63.79 H 7.90 Br 20.21 Found: C 63.52 H 7.73 Br 20.36 b) A solution of 391 mg acetate (XIVa) in 15 cm3 of methanol containing 0.38 cm3 of 72% perchloric acid was kept at room temperature for 24 hours. Precipitation with water gave 286 mg of oxybromide XIV, melting point 139.5-140.5, which is identical to the product described under a).
Oxidation of 17a-bromo-3/3-oxy-5-pregnen-20-one (17 ("- Bromo-5-pregnen-3,20-dione [XV]) in a solution of 286 mg of oxybromide XIV in 40 cm3 0.24 cm3 of the oxidizing solution described above (prepared from 100 g chromic acid, 160 g concentrated sulfuric acid and 500 cm3 water) were added dropwise to absolute acetone under a nitrogen atmosphere. After 5 minutes, the reaction product was precipitated with water .
239 mg of crude diketone XV were obtained, which crystallized in needles and melted at 100-101 (yield 84%). Since the ultraviolet spectrum of the product indicated that the double bond had partially shifted, the crude product was used in the following reaction without purification.
17a-Bromo-progesterone (XVI) via the intermediates XIVa, XIV and XV The crystalline diketone XV described above was rearranged as described above. By crystallization from aqueous ethanol, 158 mg of pure 17a-bromo-progesterone, melting point 157-159, were obtained. The identity of the product with the product described above was proven by infrared analysis.
