Verfahren zur Herstellung von 11-Dehydro-corticosteron. Im Hauptpatent ist ein Verfahren be schrieben zur Darstellung des wertvollen 11- 1)ehydro-corticosterons (d4,5-Pregnen-3,11,20- trion-21-ol), welches dadurch gekennzeichnet ist, dass man ein Carbonsäurehalogenicl eines Derivates der rltiocholansäure,
das im Ring A iii 3-Stellung eine durch Hydrolyse in Hydro- xyl umwandelbare Gruppe und im Ring C in 11-Stellung eine Ketogruppe aufweist, mit Diazomethan umsetzt, in dem erhaltenen Diazo- keton die geschützte Hy droxylgruppe in 3- Stellung in Freiheit setzt,
in beliebiger Rei henfolge diese Ilydroxylgruppe mit Oxyda tionsmitteln in eine Ketogruppe überführt lind die Diazoketongruppierung mittels Säu ren in eine veresterte Ketolgruppe umwandelt, iu a-Stellung zur 3-Ketogruppe eine Doppel bindung einführt durch Halogenierung und naehträgliche Halogenwasserstoffabspaltung und sehliesslich mit hydrolysierenden Mitteln behandelt.
Das Verfahren des Hauptpatentes gestat tet die Herstellung des wertvollen 11-Dehydro- eorticoSterons (44,5-Pregnen-3,11,20-trion-21- ol). In einem diesbezüglichen Beispiel wird die Teilsynthese besehrieben des Dehydro- cortieosteronaeeta.tes, ausgehend von einem 3 ss-Acetoxy -11-keto-ätiocholansäure-halogenid. Die letzte Stufe besteht darin, dass im erhal tenen Pregnan-3,11,
20-trion-21-ol-acetat durch Bromierung und Bromwasserstoffabspaltung eine Doppelbindung in -1,5-Stellung einge führt wird. Diese Reaktion verläuft aber mit einer unbefriedigenden Ausbeute, möglieher- weise weil die Bromierung nicht ausschliess lich in 4-Stellung erfolgt.
Inzwischen wurde gefunden, dass man ebenfalls zu 11-Dehydro-corticosteron bzw. zu seinen Derivaten gelangen kann, wenn man von 45,6-3_Oxy-11-keto - ätiocholensäure - halo- geniden ausgeht, deren Hydroxy lgruppe ge schützt ist, und analog dem Verfahren des hauptpatentes die genannten-Ausgangsstoffe mit Diazomethan umsetzt,
in den erhaltenen Diazoketonen in beliebiger Reihenfolge einer seits die geschützte Hydroxylgruppe in 3-Stel- lung in Freiheit setzt und zur Ketogruppe oxydiert und anderseits die Diazoketongrup- pierung in eine freie oder veresterte Ketol- gruppierung umwandelt und gegebenenfalls mit hydrolysierenden und/oder veresternden :1Iitteln behandelt.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung von 11-De- liydro-corticosteron, welehes dadureh gekenn zeichnet ist, dass man ein 45,6-3-Oxy-11-keto- ätiocholensäurehalogenid, dessen Hy droxyl- gruppe geschützt, ist, mit Diazomethan um setzt,
in dem erhaltenen Diazoketon in be liebiger Reihenfolge einerseits die geschützte Hy droxylgruppe in 3-Stellung in Freiheit setzt und zur Ketogruppe oxydiert und an derseits die Diazoketongruppierung mit Säu ren in eine veresterte Ketolgruppierung um- ,_; andelt und diese mit hydrolysierenden Mit teln behandelt.
Die als Ausgangsmaterial benötigte 45.6- 3-Ox@--11-keto-ätiocholensäuren waren im Zeit- punkt der Anmeldung des Hauptpatentes noch nicht bekannt. Der erste gangbare Weg zu ihrer synthetischen Darstellung ist in dem Patent Nr.266363 beschrieben. Der Schutz der Hydroxylgruppe, die Bereitung der Säure halogenide sowie die Durchführung der übri gen beanspruchten Massnahmen kann analog der Angaben des Hauptpatentes Nr.244341 erfolgen.
Das vorliegende Verfahren hat gegenüber demjenigen, ausgehend von gesättigten 3-Oxy- 11-keto-ätiocholensäuren, den Vorteil, dass die schlecht verlaufende endstufige Einführung der Doppelbindung wegfällt, da bereits die neuen Ausgangsstoffe eine Doppelbindung enthalten. Auf diese Weise wird die Aus beute an dem gewünschten Endprodukt we sentlich verbessert.
Beispiel 13,5 Teile d5,6-3ss-Acetoxy-11-keto-ätiocho- lensäare vom Schmp. 214-219 C werden in 70 Teilen reinstem Thionylchlorid aufgelöst und die erhaltene Lösung während 16 Stun den bei 6 C und dann eine halbe Stunde bei Zimmertemperatur stehen gelassen.
Nach dem Eindampfen im Vakuum wird der Rückstand in 300 Teilen absolutem Benzol aufgelöst und die Lösung sodann bei 0 C in eine getrock nete ätherische Diazomethanlösung eingetra gen, die aus 50 Teilen Nitrosomethyl-Harn- stoff erhalten wurde. Das Ganze wird zwei Stunden stehen gelassen, sodann im Vakuum bei 30 C stark eingeengt, von wenig Floeken abfiltriert und das Filtrat ganz eingedampft.
Als Rückstand wird das d5,6-21-Diazo-3 ss- acetoa-y pregnen-11,20-dion erhalten. Dieses acetylierte Diazoketon wird mit 12 Teilen Kaliumhydroxyd in 600 Teilen Methanol versetzt und die Mischung einige Stunden bei Zimmertemperatur stehen ge lassen. Nach Zusatz von 300 Teilen Wasser wird das Methanol im Vakuum bei 30 C ab destilliert und der Rückstand mit viel Äther ausgeschüttelt. Die ätherischen Auszüge wer den mehrmals mit-Wasser ausgewaschen, über Natriumsulfat getrocknet und bei 20 C ein- gedampft.
Der Rückstand (13,2 Teile) liefert aus Chloroform und wenig Äther umkristalli siert 6,3 Teile d5,6-21-Diazo-3ss-oxy-pregnen- 1.1,20-dion vom Schmp. 161-163 C (Zerset zung). Die Hutterlaugen werden eingedampft. (6,9 Teile), worauf der Rückstand an Alu miniumoxyd chromatographiert weitere 5,2 Teile reine Kristalle in Form von blassgelben, zu Drusen vereinigten Blättchen liefert. Total ausbeute 11,5 Teile.
11,5 Teile d5,6 - 21- Diazo-3 ss-oxy-pregnen- 11,20-dion vom Schmp. 161-163 C wurden in 100 Teilen reinstem Eisessig aufgelöst und die Lösung auf 95 C erwärmt, darauf noch kurz auf 102 C gebracht, worauf die Lösung im Vakuum bei 30 C vollständig eingedampft wird. Der Rückstand (12,5 Teile) lässt sich bei 0,01 mm Druck und 180-190 C Bad temperatur destillieren.
Es wurde erhalten 9,2 Teile d5,6-Pregnen-3f 21-diol-11,20-dion-21- monoacetat. 9,2 Teile dieser Verbindung wurden mit 15 Teilen Aluminium-tert.butylat., 300 Teilen absolutem Benzol und 150 Teilen Aceton im evakuierten Rohr während 25 Stunden auf 100 C erhitzt. Das Reaktionsgemisch wurde in viel Äther aufgelöst, die Lösung mit ver dünnter Salzsäure, Natriumcarbonatlösung und Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft.
Der erhaltene Rückstand (8,5 Teile) wurde durch Chromato- gr aphie an Aluminiumoxyd gereinigt. Die mit Benzol-Äther und mit reinem Äther eluier- ten Anteile wurden aus Aceton-Äther um kristallisiert und ergaben etwa 6 Teile 11-De- hydro-corticosteron-acetat in Gestalt von farb losen Nadeln vom Schmp. 177-179 C und der spezifischen Drehung (a)D = -I- 208,8 30 und (a)
