Verfahren zur Herstellung von 19-Nor-45-androsten-3ss,17ss-diolen Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen 17a-Nieder- alkyl- und 17a-Niederalkinyl-Derivaten von 19-Nor-45- androsten-3/3,17ss-diol. Diese erfindungsgemäss her gestellten 17a-Niederalkyl- und 17a-Niederalkinyl- Derivate (inbegriffen deren Ester, wie später aus geführt wird) von 19-Nor-45-androsten-3ss,
17ss-diol sind androgene Stoffe mit einem hohen anabolisch- androgenen Verhältnis sowie einer deutlichen anti- östrogenen Wirkung, insbesondere bei oraler Ver abreichung. Sie sind deshalb von Nutzen zur Erzie lung einer Milchdrüsenschrumpfung, zur Verbesse rung von dehibilitiertem Gewebe sowie zur Erzielung anderer bekannter antiöstrogener Wirkungen und Stoffwechselwirkungen, indem sie auch eine minimale androgene Wirkung besitzen.
Es wurde gefunden, dass diese neuen 17a-Nieder- alkyl- und 17a-Niederalkinyl-Derivate von 19-Nor- .J5-androsten-3ss,17/3-diol hergestellt werden können durch Behandeln von 17a-Niederalkyl- oder 17a- Niederalkinyl-Derivaten von 19-Nortestosteron zur Bildung der entsprechenden 17a-substituierten 3,17- Diacetate von 19-Nor-43,5-androstadien-3,
17ss-diol. Durch Behandeln dieser Verbindungen unter selekti ver Hydrolyse der 3-Enolestergruppe und Sättigung der 3,4-Doppelbindung, z. B. mit Reduktionsmitteln, erhält man die entsprechenden 17a-substituierten 17- Monoester von 19-Nor-d5-androsten-3ss,17ss-diolen. Durch Verseifung oder Hydrolyse erhält man das freie Diol, und durch übliche Veresterung dieser freien Alkohole gewinnt man die 3-Mono- oder 3,17-Diester davon.
So erhält man durch Behandeln der 3,17-Di- ester der 17a-substituierten 19-Nor-,13.s_androstadien- 3ss,17/3-diole mit Natriumborhydrid die 17-Monoester von 17a-substituierten 19-Nor-45-androsten-3ss,17ss- diol-Verbindungen und weiterhin durch Reaktion mit Lithiumaluminiumhydrid die freien Diole. Es wurde ferner gefunden, dass durch partielle Hydrierung der 17a-Alkinyl-19-nor-45-androsten-3ss,
17ss-diol-Verbin- dungen oder von deren Estern die entsprechenden Alkenyl-Derivate und durch vollständigere Hydrie rung die entsprechenden 17a-Alkyl-Derivate gebildet werden können.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen der Formel

worin R eine Niederalkylgruppe mit weniger als 7 C-Atomen, wie Methyl, Äthyl, Propyl oder Butyl, eine Niederalkinylgruppe mit 2-6 C-Atomen, wie Äthinyl, Propinyl-(1) oder Butinyl-(1), bedeutet, ist dadurch gekennzeichnet,
dass man ein entsprechendes 17a-Alkyl- oder 17a-Alkinylderivat des 19-Nor-ds- androsten-17ss-ol-3-ons mit Essigsäureanhydrid zum 3-Enolacetat-17-acetat oder zum 17-Acetat und letz teres mit Isopropenylacetat in Anwesenheit eines sau ren Katalysators zum 3-Enolacetat-17-acetat umsetzt und das erhaltene 3,17-Diacetat des entsprechenden 19-Nor-43,
5-androstadiens mit Natriumborhydrid in 3/4-Stellung selektiv hydriert und die Enolacetatgruppe hydrolysiert, worauf man das 17-Monoacetat des in 17a-Stellung entsprechend substituierten 19-Nor-dS- androsten-3ss,17ss-diols mit Lithiumaluminiumhydrid verseift.
Das erfindungsgemässe Verfahren wird anhand des folgenden Schemas näher erläutert:

Zur Durchführung des angegebenen Verfahrens behandelt man ein 17a.-Niederalkyl- oder 17a-Nieder- alkinyl-Derivat von 19-Nor-A4-androsten-3ss-ol-3-on, beispielsweise ein 17a-Methyl- oder 17a-Äthinyl- Derivat, in Gegenwart von Pyridin mit Acetanhydrid. Das Reaktionsgemisch wird während längerer Zeit grössenordnungsmässig 16 Stunden - auf Rückfluss er hitzt und dann in Wasser gegossen.
Durch Extrahie ren mit Äther und Eindampfen zur Trockne erhält man einen öligen Rückstand, bestehend aus dem rohen 17-Acetat des 17a-Niederalkyl- oder 17a-Nie- deralkinyl-Derivates von 19-Nor-J4-androsten-17ss-ol- 3-on. Dieses kann in üblicher Weise, z.
B. durch Chromatogrraphie, gereinigt oder direkt mit Isoprope- nylacetat und einer geringen Menge Säure behandelt werden zur Bildung des entsprechenden 3,17-Diace- tats eines niedrigen 17a-Alkyl- oder 17a-Alkinyl-19- nor-.A'.5-androstadien-3,17ss-diols. Diese letztgenannte Verbindung wird dann unter Sättigung der 3-Doppel- bindung und Hydrolyse der 3-Enolacetatgruppe wei terbehandelt.
Wie aus dem Schema hervorgeht, eignet sich zu diesem Zweck Natriumborhydrid als Reduk tionsmittel, und als Produkt erhält man das 17-Acetat des 17a-Niederalkyl- oder Alkinyl-Derivates von 19- Nor-.i5-androsten-3ss,17ss-diol. Vorzugsweise löst man das Ausgangsmaterial für diesen letzten Schritt in einem inerten organischen Lösungsmittel, wie Metha nol, Äthanol oder Tetrahydrofuran oder einem Ge misch davon,
und behandelt mit dem Natriumbor- hydrid in einer geringen Wassermenge bei Zimmer temperatur während etwa 20 Stunden. Das nach den üblichen Isolierungsverfahren erhaltene 17-Acetat wird dann zum freien Diol verseift oder hydrolysiert. Zu diesem Zweck verwendet man vorzugsweise Li- thiumaluminiumhydrid unter Rückflussbedingungen in Gegenwart eines inerten organischen Lösungsmittels, wie Äther.
Wenn das so gebildete Diol bei Zimmer temperatur in Pyridinlösung in üblicher Weise mit dem Anhydrid oder Säurechlorid einer Kohlenwasser- stoffcarbonsäure mit 2-12 C-Atomen behandelt wird, so erhält man den entsprechenden 3-Monoester. An derseits gewinnt man durch Behandeln mit einem Säureanhydrid in Pyridin unter Rückflussbedingungen während längerer Zeit, z. B. 16-20 Stunden, das ent sprechende 3,17-Diacylat.
Zur Bildung der entsprechenden 17a-Alkenyl- Derivate von 19-Nor-45-androsten-3/i,17ss-diol oder dessen Estern wurden die entsprechenden 17a-Alki- nyl-Verbindungen partiell hydriert.
In dieser Weise erhielt man durch Behandlung mit etwa 1 Mol Was serstoff in Gegenwart von 2 % Palladium auf Calcium- carbonat als Katalysator sowie von Pyridin die ent sprechenden Alkenyl-Derivate. Durch weitere Hydrie rung unter Verwendung von etwa 1 Mol Wasserstoff und 5,Q,', Palladium auf Kohle als Katalysator in Di- oxan wurde die Doppelbindung des 17-Alkenyl-Deri- vates gesättigt,
wobei man die entsprechende 17- Alkyl-Verbindung erhielt. Die 17-Alkyl-Verbindun- gen wurden auch direkt aus den entsprechenden 17- Alkinyl-Verbindungen erhalten unter Verwendung von 2 Mol Wasserstoff mit 5 % Palladium auf Kohle als Katalysator und mit Dioxan als Reaktionsmedium.
Wie aus dem Schema hervorgeht, können die 3,17- Diester der 17a-Niederalkyl- oder 17a-Niederalkinyl- Derivate von 19-Nor-43,5-androstadien-3,17ss-diol auch direkt aus den Ausgangsverbindungen her gestellt werden, indem man diese in Gegenwart eines Säurekatalysators, vorzugsweise p-Toluolsulfonsäure, mit einem überschuss des entsprechenden Säurean- hydrids erhitzt.
Durch Behandeln dieses 3,17-Diesters mit Natriumborhydrid unter den oben angegebenen Bedingungen erhält man dann den 17-Monoester des 17-Niederalkyl- oder 17-Alkinyl-Derivates von 19- Nor-45-androsten-3ss,17ss-diol. Durch Hydrierung des 17-Niederalkinyl-17-monoesters unter den oben an gegebenen Bedingungen gelangt man zum entspre chenden 17-Niederalkenyl- bzw.
17-Niederalkyl-Deri- vat, durch Verseifung oder Hydrolyse zum freien Diol und durch passende Veresterung zum 3,17-Diester, welcher verschiedene Estergruppen aufweisen kann.
<I>Beispiel 1</I> Ein Gemisch aus 4 g 17a-Methyl-19-nor-testoste- ron, 15 cm3 Pyridin und 20 cm3 Acetanhydrid wurde 16 Stunden lang auf Rückfluss erhitzt, in Wasser ge gossen und mit Äther extrahiert. Der Auszug wurde mit verdünnter Salzsäure, Wasser, Natriumbicarbo- natlösung und nochmals mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft.
Der ölige Rückstand bestand aus dem Acetat von 17a-Methyl-19-nor-testosteron und zeigte bei der Infrarot-Untersuchung die charak teristischen Banden der 17ss-Acetoxygruppe (1730 cm-') und der d4-3-Keto-Gruppierung (1680 cm-i).
Dieses Acetat von 17a-Methyl-19-nor-testosteron wurde in 40 cm3 Isopropenylacetat gelöst und mit einer Lösung von 0,2 cm- konzentrierter Schwefel säure in 10 cm' Isopropenylacetat vermischt. Durch langsame Destillation innert 2 Stunden wurde die Lösung auf ein Endvolumen von 15 cm3 konzentriert.
Darauf wurden weitere 40 cm3 Isopropenylacetat so wie 0,8 cm3 Schwefelsäure in 40 cm3 Isopropenyl- acetät zugesetzt, und das Gemisch wurde nochmals langsam innert 2 Stunden auf ein Volumen von 20 cm3 konzentriert. Darauf goss man in Wasser, ex trahierte mit Äther, wusch den Ätherauszug mit Was ser bis zur Neutralität, trocknete über wasserfreiem Natriumsulfat und dampfte zur Trockne ein.
Der ölige Rückstand bestand aus dem Diacetat von 17a- Methyl -19 - nor-d3,5-androstadien-3ss,17ss-diol, wel ches ohne weitere Reinigung für die nächste Stufe ver wendet wurde.
Eine Lösung des Diacetats von 17a-Methyl-19- nor-d5-androsten-3ss,17ss-diol in 150 cm3 Äthanol wurde im Laufe einer Stunde langsam zu einem ge rührten Gemisch aus 10 g Natriumborhydrid in 20 cm3 Wasser und 200 cm-' Äthanol eingeführt, welches zuvor auf eine Temperatur zwischen 0 und 5 C gekühlt worden war, wobei dafür gesorgt wurde, dass die Temperatur des Reaktionsgemisches in die- sein Bereiche blieb. Nachdem das Gemisch über Nacht bei Zimmertemperatur gehalten worden war, wurde es unter Vakuum auf ein kleines Volumen kon zentriert, mit 2n Salzsäure angesäuert und mit Chloro form extrahiert.
Der Auszug wurde mit Wasser neu tralgewaschen, über wasserfreiem Natriumsulfat ge trocknet und eingedampft. Der Rückstand wurde in 200 cm3 Äther gelöst, und die Lösung wurde einer Suspension von 3 g Lithiumaluminiumhydrid in 150 cms Äther tropfweise und unter ständigem Rüh ren zugesetzt. Man erhitzte das Gemisch 30 Minuten lang unter Rückfluss, zerstörte überschüssiges Reagens durch Zugabe von Äthylacetat, goss das Gemisch in Wasser und trennte die organische Schicht ab. Diese wurde mit Wasser gewaschen, über wasserfreiem Na triumsulfat getrocknet und eingedampft.
Der ölige Rückstand wurde in einem Gemisch aus gleichen Tei len Benzol und Hexan gelöst, und die Lösung wurde in einer Säule von 100 g gewaschener Tonerde chro- matographiert. Durch Eluieren der Säule mit 750 cm3 eines Benzol-Äther-Gemisches (9 :1) erhielt man 17a - Methyl - 19 - nor-,1 5-androsten-3ss,17ss-diol mit einem Schmelzpunkt von 177-186 C.
Durch Um kristallisieren aus Aceton-Hexan erhielt man die reine Verbindung, Schmelzpunkt 185-187 C, [a]D -18 (Chloroform). Die Verbindung zeigte im Ultraviolett-Spektrum kein Absorptionsmaximum.
Zur Veresterung wurde ein Gemisch aus 1 g 17a- Methyl-19-nor-45-androsten-3ss,17ss-diol, 20 cms Pyridin und 1 cm?- Acetanhydrid über Nacht bei Zim mertemperatur stehengelassen und dann in Wasser gegossen. Das Produkt wurde mit Äthylacetat extra hiert, mit 2n Salzsäure, Wasser, Natriumbicarbonat- Lösung und nochmals mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und einge dampft.
Durch Kristallisieren des Rückstandes aus Aceton-Hexan erhielt man das 3-Acetat von 17a- Methyl-19-nor-45-androsten-3ss,17ss-diol.
Eine Lösung von 1 g 17a-Methyl-19-nor-d5-an- drosten-3/:,17ss-diol in 12,5 cms Pyridin wurde mit 5 cm3 Acetanhydrid vermischt und 16 Stunden lang auf Rückfluss erhitzt.
Nach Aufarbeiten gemäss oben angeführtem Beispiel 1 zur Veresterung des 17a-Me- thyl-19-nor-testosterons gelangt man zum Diacetat von 17a-Methyl-19-nor-45-androsten-3ss,17ss-diol. <I>Beispiel 2</I> 1 g des 17-Acetats von 17a-Methyl-19-nor-45- androsten-3ss,17ss-diol wurde in 20 cm3 Pyridin ge löst, mit 1 cms Propionsäureanhydrid vermischt und auf dem Dampfbad 1 Stunde lang erhitzt.
Man goss das Gemisch in Wasser und extrahierte mit Äthyl- acetat. Das Reaktionsprodukt wurde dann nach dem gleichen Verfahren wie in Beispiel 2 isoliert. Man er hielt so das 3-Propionat-17-acetat von 17a-Methyl- 19-nor-45-androsten-3ss,17ss-diol.
<I>Beispiel 3</I> Ein Gemisch aus 4 g 17a-Äthinyl-19-nor-testoste- ron, 50 cm3 Pyridin und 20 cms Acetanhydrid wurde 16 Stunden lang auf Rückfluss erhitzt und dann in Wasser gegossen. Man extrahierte das Produkt mit Äther, wusch mit verdünnter Salzsäure, Wasser, Na- triumbicarbonat-Lösung und nochmals mit Wasser, trocknete über wasserfreiem Natriumsulfat und dampfte zur Trockne ein.
Der Rückstand bestand zur Hauptsache aus dem 17-Acetat von 17a-Athinyl-19- nor-testosteron und zeigte bei der Infrarot-Unter- suchung die charakteristischen Banden der 17ss-Acet- oxygruppe (1730 cm-') und der J4-3-Keto-Gruppie- rung (1680 cm-'). Dieses rohe 17-Acetat von 17a- Äthinyl-19-nor-testosteron wurde chromatographisch gereinigt, und die reine Substanz wurde isoliert;
sie hatte folgende Eigenschaften: Schmelzpunkt 161 bis 162 C, [a]D -33 (Chloroform), d",a, 240 m,u, log E 4,2.
Man löste den wie im obigen Absatz beschrieben durch Abdampfen des Äthers erhaltenen Rückstand in 40 cm3 Isopropenylacetat, vermischte mit einer Lö sung von 0,2 cm3 Schwefelsäure in 10 cm3 Isoprope- nylacetat und konzentrierte durch langsame Destilla tion innert 2 Stunden auf ein Schlussvolumen von 15 cm3. Man setzte 40 cm3 Isopropenylacetat und 0,
8 cm3 Schwefelsäure in 40 cm3 Isopropenylacetat zu .und konzentrierte das Gemisch nochmals innert 2 Stunden auf ein Volumen von 20 cm3. Man goss darauf in Wasser, extrahierte mit Äther, wusch mit Wasser neutral, trocknete über wasserfreiem Natrium sulfat und dampfte zur Trockne ein. Der Rückstand bestand aus dem Diacetat von 17a-Äthinyl-19-nor- A3,5_androstadien-3,17ss-diol in roher Form.
Durch Umkristallisieren aus Methanol erhielt man die reine Substanz mit einem Schmelzpunkt von 166-169 C, [a]D -203 (Chloroform), a",a, 234-236 m,u., log E 4,27.
Man löste 4 g des Diacetats von 17a-Äthinyl-19- nor-43.5-androstadien-3,17ss-diol in 250 cm3 Metha nol und 120 cm3 Tetrahydrofuran, kühlte auf 0 C und vermischte mit einer Lösung von 5 g Natrium- borhydrid in 10 cm3 Wasser. Das Gemisch wurde 20 Stunden lang bei 0-5 C gehalten, mit Eisessig an gesäuert, unter vermindertem Druck auf ein kleines Volumen konzentriert, gekühlt und mit Wasser ver dünnt.
Der Niederschlag wurde gesammelt, mit Was ser gewaschen, getrocknet und aus Äther umkristalli siert, wobei man das 17-Acetat von 17a-Äthinyl-19- nor-45-androsten-3ss,17ss-diol erhielt, Schmelzpunkt 105 C, [a]D -62 (Chloroform). Die Verbindung zeigte keine selektive Absorption im Ultraviolett- Spektrum.
Ein Gemisch aus 1 g des obigen Acetats, 100 cm3 wasserfreiem Tetrahydroxyfuran und 1 g Lithium- aluminiumhydrid wurde während 1 Stunde auf Rück fluss erhitzt und dann gekühlt. Man zerstörte über schüssiges Hydrid durch Zugabe von Äthylacetat und setzte dann gesättigte Natriumsulfat-Lösung und was serfreies Natriumsulfat zu. Man filtrierte den Nieder schlag von anorganischen Salzen ab und wusch mit Tetrahydrofuran. Filtrat und Waschflüssigkeit wurden vereinigt und unter vermindertem Druck zur Trockne eingedampft.
Durch Kristallisieren des Rückstandes aus Methanol erhielt man freies 17a-Äthinyl-19-nor- _15-androsten-3ss,17ss-diol, Schmelzpunkt 215-217 C, [a]@ -52 (Chloroform), keine selektive Absorption im Ultraviolett-Spektrum.
<I>Beispiel 4</I> 100 mg eines 2%igen Palladium-auf-Calcium- Katalysators, suspendiert in 20 cm3 reinem Pyridin, wurden bei Atmosphärendruck und Zimmertempera tur vorhydriert, worauf man eine Lösung von 1 g 17a- Äthinyl-19-nor-45-androsten-3ss,17ss-diol in 20 cm3 reinem Pyridin zusetzte und die Hydrierung fortsetzte, bis etwa 1 molares Äquivalent Wasserstoff absorbiert war und die Wasserstoffaufnahme praktisch aufhörte,
was ungefähr 30 Minuten erforderte. Der Katalysator wurde abfiltriert und mit Äthylacetat gewaschen. Das mit den Waschflüssigkeiten vereinigte Filtrat wurde mit weiterem Athylacetat verdünnt und mit verdünn ter Salzsäure gerührt.
Die Äthylacetat-Lösung wurde abgetrennt, mit wässeriger Natriumbicarbonat-Lösung und Wasser gewaschen, über wasserfreiem Natrium sulfat getrocknet, filtriert und unter vermindertem Druck zur Trockne eingedampft. Durch Kristallisie ren des Rückstandes aus Aceton-Hexan erhielt man 17a-Vinyl-19-nor-45-androsten-3ss,17ss-diol.
<I>Beispiel 5</I> Man gab 100 mg 17a-Vinyl-19-nor-45-androsten- 3ss,17ss-diol zu einem vorreduzierten Gemisch aus 20 cm3 Dioxan und 40 mg 5 % igem Palladium-auf- Kohle-Katalysator und hydrierte das Gemisch bei At mosphärendruck und Zimmertemperatur, bis ein mo- lares Äquivalent Wasserstoff absorbiert war.
Man fil trierte dann durch Celite und wusch den Rückstand mit ein wenig Dioxan. Das Filtrat wurde mit der Waschflüssigkeit vereinigt und unter reduziertem Druck zur Trockne eingedampft, und der Rückstand wurde aus Aceton-Hexan kristallisiert, wobei man 17a-Äthyl -19-nor-45-androsten-3ss,17ss-diol erhielt.
<I>Beispiel 6</I> Unter den gleichen Hydrierungsbedingungen, wie sie in den Beispielen 6 und 7 beschrieben wurden, hy drierte man 1 g des 17-Acetats von 17a-Äthinyl-19- nor-45-androsten-3ss,17ss-diol zum 17-Acetat von 17a-Vinyl-19-nor-45-androsten-3ss,17ss-diol und re duzierte diese Verbindung weiter zum 17-Acetat von 17a-Äthyl-19-nor-45-androsten-3ss,17ss-diol.
<I>Beispiel 7</I> Nach dem Verfahren von Beispiel 5 hydrierte man die Athinylgruppe des 17-Acetats von 17a-Äthinyl- 19-nor-A5-androsten-3ss,17ss-diol oder des freien Diols, unter Absorption von 2 Äquivalenten Wasser stoff, zum 17-Acetat von 17a-Athyl-19-nor-45-andro- sten-3ss,17ss-diol bzw.
dem freien Diol. <I>Beispiel 8</I> Ein Gemisch aus 5 g 17a-Äthinyl-19-nor-testoste- ron, 50 cm3 Acetanhydrid und 500 mg p-Toluolsul- fonsäure wurde 1 Stunde lang auf dem Dampfbad er hitzt; hierauf goss man in Eiswasser und erhitzte auf dem Dampfbad 1/2 Stunde lang bis zur völligen Hy drolyse des Anhydridüberschusses. Nach dem Abküh len extrahierte man das Produkt mit Äther, wusch mit Wasser, Natriumbicarbonat-Lösung und noch mals mit Wasser, trocknete über wasserfreiem Na triumsulfat und dampfte zur Trockne ein.
Durch Um kristallisieren des Rückstandes aus Methanol erhielt man das Diacetat von 17a-Äthinyl-19-nor-d3,5-an- drostadien-3,17ss-diol, welches mit der in Beispiel 5 beschriebenen Verbindung identisch war. <I>Beispiel 9</I> Ersetzt man im Verfahren des vorangehenden Beispiels das Acetanhydrid durch Propionsäurean- hydrid, so erhielt man das Dipropionat von 17a- Äthinyl-19-nor-43,5-androstadien-3,17ss-diol.
Auf dem in Beispiel 5 beschriebenen Wege wurde diese Verbindung in das 17-Propionat von 17a- Äthinyl-19-nor-45-androsten-3ss,17ss-diol übergeführt, welches, wie das 17-Acetat, weiter in das freie Diol umgewandelt wurde.
<I>Beispiel 10</I> Durch Anwendung der Verfahren der Beispiele 6 und 7 auf das 17-Propionat von 17a-Äthinyl-19-nor- 45-androsten-3ss,17ss-diol erhielt man die 17-Propio- nate von 17a-Vinyl-19-nor-45-androsten-3ss,17ss-diol und von 17a-Äthyl -19 - nor - 45 - androsten-3ss,17ss- diol.
Diese 17-Propionate und 17-Acetate der 17a- Vinyl- und 17a-Athyl-Derivate von 19-Nor-45-an- drosten-3ss,17ss-diol wurden durch Reaktion mit Li- thiumaluminiumhydrid wie in Beispiel 5 beschrieben in die entsprechenden freien Diole übergeführt.
<I>Beispiel 11</I> Nach dem Verfahren der vorausgehenden Bei spiele, jedoch ausgehend von andern 19-Nor-testoste- ron-Derivaten, welche in 17a-Stellung durch andere niedrige Alkinylgruppen substituiert waren, enthielt man die 17-Ester der entsprechenden 17a-Alkinyl-19- nor-45-androsten-3ss,17ss-diole, deren Dreifachbin- dung dann partiell oder vollständig hydriert wurde.
Beispielsweise wurden folgende Reaktionen durch geführt: Gemäss dem Verfahren von Beispiel 5 wurde 17a-Propinyl-(1)-19-nor-testosteron in das 17-Acetat von 17a-Propinyl-(1)-19-nor-45-androsten-3,17ss-diol übergeführt; nach dem Verfahren von Beispiel 11 wurde 17a-Butinyl-(1)-19-nor-testosteron in das 17- Propionat von 17a-Butinyl-(1)-19-nor-45-androsten- 3ss,17ss-diol übergeführt;
durch Anwendung der Re aktion mit Lithiumaluminiumhydrid von Beispiel 5 erhielt-man die entsprechenden freien Diole der letz teren -Verbindungen; durch Hydrieren derselben ge mäss Beispiel 6 erhielt man 17a-Propenyl-(1)-19-nor- i5-androsten-3ss,17ss-diol bzw. 17a-Butenyl-(1)-19- nor-45-androsten-3ss,17ss-diol;
durch vollständige Sät tigung der Dreifachbindung gemäss Beispiel 7 entstan den 17a - Propyl - 19 - nor - 95-androsten-3ss,17ss-diol bzw. 17ra <I>-</I> Butyl - 19 - nor - d 5-androsten-3ss,17ss-diol. Durch Hydrierung der 17-Ester der entsprechenden Diole erhielt man die hydrierten Verbindungen in Form der entsprechenden Ester.
<I>Beispiel 12</I> Ersetzte man in den Verfahren der Beispiele 3 und 8 17a-Äthinyl-19-nor-testosteron durch ein 19-Nor- testosteron mit einer Methyl-, Äthyl-, Propyl- oder Butylgruppe in 17a-Stellung, so erhielt man die 17a- Methyl-, -Äthyl-, -Propyl- und -Butyl-Homologen der Diester von 19-Nor-43,5-androstadien-3,17ss-diol,
der entsprechenden 17-Ester von 19-Nor-d5-andro- sten-3ss,17ss-diolen bzw. der entsprechenden freien Diole.
<I>Beispiel 13</I> Ersetzte man in den Beispielen 8 und 11 das Acetanhydrid bzw. Propionsäureanhydrid durch ein anderes Anhydrid einer gesättigten oder ungesättig ten, geraden oder verzweigten, cyclischen oder ge mischt cyclisch-aliphatischen, unsubstituierten oder durch Methoxy-, Halogen- oder andere Gruppen sub- stituierten Carbonsäure mit bis zu 12 C-Atomen,
so erhielt man alle die in den früheren Beispielen be schriebenen Verbindungen in Form der betreffen den Ester. Insbesondere wurden die Butyrate, Hämi- succinate, Caproate, Benzoate, Trimethylacetate, Phenoxyacetate, Phenoxypropionate, Phenylpropio- nate, Cyclopentylpropionate, Dichlorphenoxyacetate und ss-Chlorpropionate hergestellt.
<I>Beispiel 14</I> Man löste 1 g 17a-Äthinyl-19-nor-45-androsten- 3ss,17ss-diol, das nach dem in Beispiel 5 beschriebe nen Verfahren erhalten worden war, in 20 cm3 Pyri- din, vermischte mit 1 cm3 Acetanhydrid und liess über Nacht bei Zimmertemperatur stehen. Das Gemisch wurde in Wasser gegossen und mit Äthylacetat extra hiert.
Der Auszug wurde mit 2n Salzsäure, Wasser, Natriumbicarbonat-Lösung und nochmals mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrock net und zur Trockne eingedampft. Durch Kristallisa tion des Rückstandes aus Aceton-Hexan erhielt man das 3-Acetat von 17a-Äthinyl-19-nor-45-androsten- 3ss,17ss-diol.
<I>Beispiel<B>15</B></I> Ein Gemisch aus 1 g 17a-Äthinyl-19-nor-Q5-an- drosten-3ss,17ss-diol und 10 cm3 Acetanhydrid wurde 16 Stunden lang auf Rückfluss erhitzt und dann wie im vorhergehenden Beispiel beschrieben aufgearbeitet. Man erhielt so das Diacetat von 17a-Äthinyl-19-nor- 45-androsten-3ss,17ss-diol.
<I>Beispiel 16</I> Ein Gemisch aus 1 g des 17-Acetats von 17a- Äthinyl-19-nor-45-androsten-3ss,17ss-diol, 20 cm3 Py- ridin und 1 cm?, Propionsäureanhydrid wurde 1 Stunde lang auf dem Dampfbad erhitzt und dann in Wasser gegossen. Das Produkt wurde mit Äthylacetat extra hiert und dann in gleicher Weise isoliert, wie es in Beispiel 16 beschrieben ist. Man erhielt so das 3-Pro- pionat-17-acetat von 17a-Äthinyl-19-nor-.15-andro- sten-3ss,17ss-diol.
<I>Beispiel 17</I> Nach den in den Beispielen 14 bis 16 beschriebe nen Veresterungsverfahren veresterte man die freien 17a-substituierten 19-Nor-45-androsten-3ss,17/3-diole zu 3-Monoestern bzw. 3,17-Diestern, und die 3- Monoester und 17-Monoester dieser Diole wurden in die 3,17-Diester mit gleichen oder verschiedenen Ra dikalen übergeführt.
Die verwendeten Anhydride leiteten sich von den in Beispiel 12 beschriebenen Säuren her.
Process for the preparation of 19-nor-45-androstene-3ss, 17ss-diols The present invention relates to a process for the preparation of new 17a-lower alkyl and 17a-lower alkynyl derivatives of 19-nor-45-androstene 3 / 3.17ss-diol. These 17a-lower alkyl and 17a-lower alkynyl derivatives (including their esters, as will be explained later) of 19-nor-45-androstene-3ss,
17ss-diol are androgenic substances with a high anabolic-androgenic ratio and a significant anti-estrogenic effect, especially when administered orally. They are therefore useful for achieving milk gland shrinkage, for improving dehibilitized tissue and for achieving other known anti-estrogenic effects and metabolic effects by also having minimal androgenic effects.
It has been found that these new 17a-lower alkyl and 17a-lower alkynyl derivatives of 19-nor- .J5-androstene-3ss, 17/3-diol can be prepared by treating 17a-lower alkyl- or 17a-lower alkynyl -Derivatives of 19-nortestosterone to form the corresponding 17a-substituted 3,17-diacetates of 19-nor-43,5-androstadien-3,
17ss-diol. By treating these compounds with selective hydrolysis of the 3-enol ester group and saturation of the 3,4 double bond, e.g. B. with reducing agents, the corresponding 17α-substituted 17-monoesters of 19-nor-d5-androstene-3ss, 17ss-diols are obtained. The free diol is obtained by saponification or hydrolysis, and the 3-mono- or 3,17-diesters thereof are obtained by conventional esterification of these free alcohols.
Thus, treating the 3,17-diesters of the 17a-substituted 19-nor-, 13.s_androstadien-3ss, 17/3-diols with sodium borohydride gives the 17-monoesters of 17a-substituted 19-nor-45-androstene -3ss, 17ss- diol compounds and furthermore the free diols by reaction with lithium aluminum hydride. It was also found that by partial hydrogenation of the 17a-alkynyl-19-nor-45-androstene-3ss,
17ss-diol compounds or their esters the corresponding alkenyl derivatives and by more complete hydrogenation the corresponding 17a-alkyl derivatives can be formed.

To carry out the specified process, a 17a-lower alkyl or 17a-lower alkynyl derivative of 19-nor-A4-androsten-3ss-ol-3-one, for example a 17a-methyl or 17a-ethynyl derivative, is treated , in the presence of pyridine with acetic anhydride. The reaction mixture is heated to reflux for an extended period of the order of 16 hours and then poured into water.
Extracting with ether and evaporation to dryness gives an oily residue consisting of the crude 17-acetate of the 17a-lower alkyl or 17a-lower alkynyl derivative of 19-nor-J4-androsten-17ss-ol- 3- on. This can be done in the usual way, for.
B. by chromatography, purified or treated directly with isopropenyl acetate and a small amount of acid to form the corresponding 3,17-diacetate of a lower 17a-alkyl- or 17a-alkynyl-19-nor-.A'.5 -androstadien-3,17ss-diols. This last-mentioned compound is then treated further with saturation of the 3-double bond and hydrolysis of the 3-enol acetate group.
As can be seen from the scheme, sodium borohydride is a suitable reducing agent for this purpose, and the product obtained is the 17-acetate of the 17a-lower alkyl or alkynyl derivative of 19-nor-.i5-androstene-3ss, 17ss-diol. The starting material for this last step is preferably dissolved in an inert organic solvent, such as methanol, ethanol or tetrahydrofuran or a mixture thereof,
and treated with the sodium borohydride in a small amount of water at room temperature for about 20 hours. The 17-acetate obtained by the usual isolation processes is then saponified or hydrolyzed to give the free diol. For this purpose, lithium aluminum hydride is preferably used under reflux conditions in the presence of an inert organic solvent such as ether.
If the diol thus formed is treated in the customary manner in pyridine solution at room temperature with the anhydride or acid chloride of a hydrocarboxylic acid having 2-12 carbon atoms, the corresponding 3-monoester is obtained. On the other hand, by treating with an acid anhydride in pyridine under reflux conditions for a long time, e.g. B. 16-20 hours, the corresponding 3,17 diacylate.
To form the corresponding 17a-alkenyl derivatives of 19-nor-45-androstene-3 / i, 17ss-diol or its esters, the corresponding 17a-alkynyl compounds were partially hydrogenated.

As can be seen from the scheme, the 3,17-diesters of the 17a-lower alkyl or 17a-lower alkynyl derivatives of 19-nor-43,5-androstadiene-3,17ss-diol can also be prepared directly from the starting compounds by this is heated in the presence of an acid catalyst, preferably p-toluenesulfonic acid, with an excess of the corresponding acid anhydride.
By treating this 3,17-diester with sodium borohydride under the conditions given above, the 17-monoester of the 17-lower alkyl or 17-alkynyl derivative of 19-nor-45-androstene-3ss, 17ss-diol is then obtained. Hydrogenation of the 17-lower alkynyl-17-monoester under the conditions given above leads to the corresponding 17-lower alkenyl or
17-lower alkyl derivative, by saponification or hydrolysis to the free diol and by suitable esterification to give the 3,17-diester, which can have various ester groups.
<I> Example 1 </I> A mixture of 4 g of 17a-methyl-19-nor-testosterone, 15 cm3 of pyridine and 20 cm3 of acetic anhydride was heated to reflux for 16 hours, poured into water and extracted with ether. The extract was washed with dilute hydrochloric acid, water, sodium bicarbonate solution and again with water, dried over anhydrous sodium sulfate and evaporated to dryness.
The oily residue consisted of the acetate of 17a-methyl-19-nor-testosterone and on infrared examination showed the characteristic bands of the 17ss-acetoxy group (1730 cm- ') and the d4-3-keto group (1680 cm -i).
This acetate of 17a-methyl-19-nor-testosterone was dissolved in 40 cm 3 of isopropenyl acetate and mixed with a solution of 0.2 cm concentrated sulfuric acid in 10 cm 'of isopropenyl acetate. The solution was concentrated to a final volume of 15 cm3 by slow distillation within 2 hours.
A further 40 cm3 of isopropenyl acetate and 0.8 cm3 of sulfuric acid in 40 cm3 of isopropenyl acetate were then added, and the mixture was again slowly concentrated to a volume of 20 cm3 over a period of 2 hours. It was then poured into water, extracted with ether, the ether extract was washed with water until neutral, dried over anhydrous sodium sulfate and evaporated to dryness.
The oily residue consisted of the diacetate of 17a-methyl-19-nor-d3,5-androstadien-3ss, 17ss-diol, which was used for the next stage without further purification.
A solution of the diacetate of 17a-methyl-19- nor-d5-androstene-3ss, 17ss-diol in 150 cm3 of ethanol was slowly transformed into a stirred mixture of 10 g of sodium borohydride in 20 cm3 of water and 200 cm3 in the course of one hour. Introduced ethanol, which had previously been cooled to a temperature between 0 and 5 C, it was ensured that the temperature of the reaction mixture remained in this range. After the mixture was kept at room temperature overnight, it was concentrated to a small volume under vacuum, acidified with 2N hydrochloric acid and extracted with chloroform.
The extract was neutral washed with water, dried over anhydrous sodium sulfate and evaporated. The residue was dissolved in 200 cm 3 of ether and the solution was added dropwise to a suspension of 3 g of lithium aluminum hydride in 150 cm 3 of ether, with constant stirring. The mixture was refluxed for 30 minutes, excess reagent was destroyed by adding ethyl acetate, the mixture was poured into water and the organic layer was separated. This was washed with water, dried over anhydrous sodium sulfate and evaporated.
The oily residue was dissolved in a mixture of equal parts of benzene and hexane, and the solution was chromatographed in a column of 100 g of washed alumina. Eluting the column with 750 cm3 of a benzene-ether mixture (9: 1) gave 17a - methyl - 19 - nor-, 15-androstene-3ss, 17ss-diol with a melting point of 177-186 C.
Recrystallization from acetone-hexane gave the pure compound, melting point 185-187 ° C., [a] D -18 (chloroform). The compound showed no absorption maximum in the ultraviolet spectrum.
For esterification, a mixture of 1 g of 17a-methyl-19-nor-45-androstene-3ss, 17ss-diol, 20 cms pyridine and 1 cm? Acetic anhydride was left to stand overnight at room temperature and then poured into water. The product was extracted with ethyl acetate, washed with 2N hydrochloric acid, water, sodium bicarbonate solution and again with water, dried over anhydrous sodium sulfate and evaporated.
Crystallization of the residue from acetone-hexane gave the 3-acetate of 17a-methyl-19-nor-45-androstene-3ss, 17ss-diol.
A solution of 1 g of 17a-methyl-19-nor-d5-anrosten-3 / :, 17ss-diol in 12.5 cm 3 of pyridine was mixed with 5 cm 3 of acetic anhydride and heated to reflux for 16 hours.
After working up as described in Example 1 above to esterify 17α-methyl-19-nor-testosterone, the diacetate of 17α-methyl-19-nor-45-androstene-3ss, 17ss-diol is obtained. <I> Example 2 </I> 1 g of the 17-acetate of 17a-methyl-19-nor-45-androsten-3ss, 17ss-diol was dissolved in 20 cm3 of pyridine, mixed with 1 cms of propionic anhydride and placed on the steam bath Heated for 1 hour.
The mixture was poured into water and extracted with ethyl acetate. The reaction product was then isolated by the same procedure as in Example 2. So he kept the 3-propionate-17-acetate of 17a-methyl-19-nor-45-androstene-3ss, 17ss-diol.

The residue consisted mainly of the 17-acetate of 17a-atynyl-19-nor-testosterone and showed the characteristic bands of the 17ss-acetoxy group (1730 cm- ') and the J4-3- in the infrared examination. 8 cm3 of sulfuric acid in 40 cm3 of isopropenyl acetate and concentrated the mixture again within 2 hours to a volume of 20 cm3. It was then poured into water, extracted with ether, washed neutral with water, dried over anhydrous sodium sulfate and evaporated to dryness. The residue consisted of the diacetate of 17a-ethynyl-19-nor-A3,5_androstadien-3,17ss-diol in crude form.
Recrystallization from methanol gave the pure substance with a melting point of 166-169 ° C., [a] D -203 (chloroform), a ", a, 234-236 m, u., Log E 4.27.
4 g of the diacetate of 17a-ethynyl-19-nor-43.5-androstadiene-3,17ss-diol were dissolved in 250 cm3 of methanol and 120 cm3 of tetrahydrofuran, cooled to 0 C and mixed with a solution of 5 g of sodium borohydride 10 cm3 of water. The mixture was kept at 0-5 C for 20 hours, acidified with glacial acetic acid, concentrated to a small volume under reduced pressure, cooled and diluted with water.
The precipitate was collected, washed with water, dried and recrystallized from ether, the 17-acetate of 17a-ethynyl-19-nor-45-androstene-3ss, 17ss-diol, melting point 105 C, [a] D -62 (chloroform). The compound showed no selective absorption in the ultraviolet spectrum.
A mixture of 1 g of the above acetate, 100 cm3 of anhydrous tetrahydroxyfuran and 1 g of lithium aluminum hydride was heated to reflux for 1 hour and then cooled. You destroyed excess hydride by adding ethyl acetate and then sat saturated sodium sulfate solution and what anhydrous sodium sulfate. The ethyl acetate solution was separated off, washed with aqueous sodium bicarbonate solution and water, dried over anhydrous sodium sulfate, filtered and evaporated to dryness under reduced pressure. Crystallizing the residue from acetone-hexane gave 17a-vinyl-19-nor-45-androstene-3ss, 17ss-diol.
<I> Example 5 </I> 100 mg of 17a-vinyl-19-nor-45-androsten- 3ss, 17ss-diol were added to a prereduced mixture of 20 cm3 of dioxane and 40 mg of 5% palladium-on-carbon Catalyst and hydrogenated the mixture at atmospheric pressure and room temperature until one molar equivalent of hydrogen was absorbed.

In the route described in Example 5, this compound was converted into the 17-propionate of 17a-ethynyl-19-nor-45-androstene-3ss, 17ss-diol, which, like 17-acetate, was further converted into the free diol .

by using the reaction with lithium aluminum hydride from Example 5, the corresponding free diols of the latter compounds were obtained; by hydrogenating them according to Example 6, 17a-propenyl- (1) -19-nor- i5-androstene-3ss, 17ss-diol or 17a-butenyl- (1) -19- nor-45-androstene-3ss, 17ss-diol;
through complete saturation of the triple bond according to Example 7, 17a - propyl - 19 - nor - 95-androstene-3ss, 17ss-diol or 17ra <I> - </I> butyl - 19 - nor - d 5-androsten- 3ss, 17ss-diol. Hydrogenation of the 17-esters of the corresponding diols gave the hydrogenated compounds in the form of the corresponding esters.
<I> Example 12 </I> In the procedures of Examples 3 and 8, 17a-ethynyl-19-nor-testosterone was replaced by a 19-nor-testosterone with a methyl, ethyl, propyl or butyl group in 17a Position, the 17a-methyl-, -ethyl-, -propyl- and -butyl homologues of the diesters of 19-nor-43,5-androstadiene-3,17ss-diol were obtained,
the corresponding 17-esters of 19-nor-d5-androsten-3ss, 17ss-diols or the corresponding free diols.
<I> Example 13 </I> If the acetic anhydride or propionic anhydride in Examples 8 and 11 is replaced by another anhydride of a saturated or unsaturated, straight or branched, cyclic or mixed cyclic-aliphatic, unsubstituted or methoxy, Halogen or other groups-substituted carboxylic acid with up to 12 carbon atoms,
all of the compounds described in the earlier examples were obtained in the form of the esters concerned. In particular, the butyrates, hemisuccinates, caproates, benzoates, trimethylacetates, phenoxyacetates, phenoxypropionates, phenylpropionates, cyclopentylpropionates, dichlorophenoxyacetates and ß-chloropropionates were produced.
Example 14 1 g of 17a-ethynyl-19-nor-45-androstene-3ss, 17ss-diol, which had been obtained by the method described in example 5, was dissolved in 20 cm 3 of pyridine , mixed with 1 cm3 acetic anhydride and left to stand overnight at room temperature. The mixture was poured into water and extracted with ethyl acetate.
The extract was washed with 2N hydrochloric acid, water, sodium bicarbonate solution and again with water, getrocked over anhydrous sodium sulfate and evaporated to dryness. Crystallization of the residue from acetone-hexane gave the 3-acetate of 17a-ethynyl-19-nor-45-androstene-3ss, 17ss-diol.
<I> Example<B>15</B> </I> A mixture of 1 g of 17a-ethynyl-19-nor-Q5-anrosten-3ss, 17ss-diol and 10 cm 3 of acetic anhydride was refluxed for 16 hours heated and then worked up as described in the previous example. The diacetate of 17α-ethynyl-19-nor-45-androstene-3ss, 17ss-diol was obtained in this way.
<I> Example 16 </I> A mixture of 1 g of the 17-acetate of 17a-ethynyl-19-nor-45-androstene-3ss, 17ss-diol, 20 cm3 pyridine and 1 cm3 propionic anhydride was 1 Heated on the steam bath for an hour, then poured into water. The product was extracted with ethyl acetate and then isolated in the same way as described in Example 16. The 3-propionate-17-acetate of 17α-ethynyl-19-nor-.15-androsten-3ss, 17ss-diol was obtained in this way.
<I> Example 17 </I> According to the esterification process described in Examples 14 to 16, the free 17a-substituted 19-nor-45-androstene-3ss, 17/3-diols were esterified to 3-monoesters or 3, 17-diesters, and the 3-monoesters and 17-monoesters of these diols were converted into the 3,17-diesters with the same or different radicals.
The anhydrides used were derived from the acids described in Example 12.
