umsetzt und das so erhaltene 3-Enamin des 7a Methyl-17cn- aJkinylsteroids hydrolysiert.
Die erfindungsgemäss erhaltenen Verbindungen können durch Umsetzung mit geeigneten Anhydriden oder Halogeniden von Kohlenwasserstoff-Carbon- säuren in die entsprechenden 17-Acylate überführt werden.
Zu den organischen Carbonsäuren, aus denen erfindungsgemässe Acylate erhalten werden, gehören gesättigte und ungesättigte aliphatische und aroma tische Säuren, wie:
Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, tert. Butylessigsäure, Valeriansäum, Isovaleriansäuze, Capronsäure, C.aprinsäure, Decansäure, Dodecans-äure, Acrylsäure, Crotonsäure, Hexins-äure, Heptinsäure, Octinsäure, Cyclobutancarbonsäure, Cyclopentancarbonsäure,
Cyalohexancarbonsäure, Dimethylcyclohexancarbonsäure, Benzoes-äure, Tolylsäure, Naphthoesäure, Äthylbenzoesäure, Phenylessigsiäure, Naphthylessigsäure, Phenylvaleriansäure, Zimtsäure, Phenylpropiolsäure, Phenylpropionsäure, p-Hexyloxyphenylpropionsäure,
p-Butyloxyphenylpropionsäure und ähnliche p-Alkoxyphenylalkansäuren. Die erfindungsgemäss erhältlichen Verbindungen besitzen anabolische und androgene Wirksamkeit bei verbessertem Verhältnis von anabolischer zu andro- gener Wirksamkeit.
Sie weisen ferner antiöstrogene, gonadotropin-inhibierende, progestationale, wachs tumsfördernde, die Fruchtbarkeit herabsetzende und das Zentralnervensystem beruhigende Wirkung auf, die sie zur Verwendung in der Human- und Veteri- närmedizin geeignet machen.
Die Verbindungen sind ferner wertvoll als Zwi schenprodukte zur Herstellung von 7aMethyl-17a- alkyl testostezonen und deren 17-Acylaten. Die er findungsgemäss hergestellten Verbindungen können z. B. in Gegenwart eines Hydrierungskatalysators wie Palladium/Kohle unter Überführung der 17-Alkinyys- gruppe in die 17 Alkylgruppe hydriert werden.
Die so erhaltenen 7a-Methy117-alkyl-testosterone und deren 17-Acylabe .sind selbst anabolisch und andro- gen wirksam bei verbessertem Verhältnis von ana- bolischer zu androgener Wirkung. Sie besitzen ferner antiöstrogene, gonadotropin-inhibierende,
progesta- tionale, wachstumsfördernde, die Fruchtbarkeit her absetzende und das Zentralnervensystem beruhigende Wirkung auf, die sie zur Verwendung in der Human- und Veterinärmedizin geeignet machen.
Die-1Ierstellung der Ausgangsmaterialien des er findungsgemässen Verfahrens kann. wie folgt gesche- hen: Man oxydiert 7a Methyl testosteron (siehe Campbell und Babcock, J.
Am. Chem. Soc.-81, 4069 [1959]) mit Oxydationsmitteln wie Chromsäure, Na- triumbichromat, Kaliumbichromat oder dergleichen in bekannter Weise unter Bildung von 7a-Methyl androsten-3,17-dion. Diese Verbindung wird, z. B.
durch Umsetzung mit einem sekundären zyklischen Alkylenamin mit 5 bis 7 Ringatomen wie: Pyrrolidin, Piperidin, Morpholin, C-alkylsubstituierten Pyrrolidinen, Piperidinen oder Morpholinen wie 2,4-Dimethyl pyrrolidin, 3-Propyl piperidin, 2-Methyl pyrrolidin, 3,
4-Dimethylpyrrolidin, 3 Äthylpyrrolidin, 3-Isopropyl pyrrolidin, 3,3-Dimethyl-pyrrohdin, 2-Methyl-morpholin, 3-Methylmorpholin oder dergleichen, in das entsprechende 3-Enamin übergeführt. Man bevorzugt zu diesem Zweck die Verwendung von Pyrrolidm.
Das 3-Enamin wird vorteilhafterweise hergestellt, indem man das 7a-Metihyl-androsten-3,17-dien und das zyklische sekundäre Amin gemeinsam in Gegen- wart eines geeigneten Lösungsmittels, z.
B. in einem niedrigen Alkohol wie Methanol, Athanol oder der gleichen, erwärmt, Das Eiamin scheidet sich im allgemeinen aus diesem Gemisch ab und' kann nach der Isolierung und kurzem Trocknen ohne weitere Reinigung in der Reaktionsstufe verwen det
werden.
Das so erhaltene 3-Enamin des 7a-Methyl-an- drosten-3,17-dions wird sodann erfindungsgemäss z. B. mit einem Alkalimetallderivat wie z.
B. einem Natrium- oder Kaliumderivat, des geeigneten Alleins, vorzugsweise in Gegenwart eines inerten Lösungs- mittels wie Dimethylfrnmamid, Dimethylsulfoxyd oder dergleichen,
umgesetzt. Dasi sich bildende 3-Enamin des 7a-Methyl 17a-alkinyl-testosterons wird im all- gemeinen nicht aus dem Reaktionsgemisch isoliert, sondern in situ nach Zersetzung des Gemisches mit Wasser, Ammoniumchlorid oder dergleichen mit einer wässrigen Lösung einer Base wie Natriumhydroxyd,
Kaliumhydroxyd oder dergleichen hydrolysient.
Das gebildete 7a-Methyl-17a@alkinyd-üestosteron kann falls erwünscht, auf übliche Weise, z. B. durch Umkristallisieren, Chromatographioren oder derglei- chen, gereinigt werden.
Die 7a Methyl 17a-alkinyl testosterone können ferner durch Umsetzung des vorstehend beschriebe- nen 3 Eiamins des. 7a-Methyl-androsten-3,17-dions mit .einem Adkinylmagnesiumhalogenid erhalten wer den.
Man arbeitet hierbei vorzugsweise in Gegen wart eines Lösungsmittels wie Dimethyläüher, Tetra- hydrofuran oder dergleichen. Vorzugsweise wird das, Grignard-Reagens im überschuss von etwa 10 Mol pro Mol 3-Enamin verwendet.
Das 3-Enamin wird im allgemeinen nicht isoliert, sondern in situ wie vorstehend beschrieben, zum freien 7a-Methyl 17a- alkinyl-testosteron hydrolysiert.
Die 17-Acylate können nach an sich bekannten Methoden, z. B. durch Umsetzung des 7a-Methyl- 17a - alkinyl - üestosterons mit dem entsprechenden Säureanhydrid oder Säurehalogenid in Gegenwart einer. tertiären organischen Base wie Pyridin erhal ten werden.
Auf diese Weise können 7a-Methyl-17a- alkinyl-testosteron -17 - acylate organischer Carbon säure wie Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, tert.-Butylessigs.äure, Valeriansäure, Isovaleriansäure, Capronsäure, Carpryl.säuse, Decansäure, Acrylsäure, Crotonsäure, Hexinsäure,
Heptinsäure, Octinsäure, Cyclobutancarbonsäure, Cyclopentancarbonsäure, Cyclopentencarbonsäure, Cyclohexancarbons:
äure, Cyclopentylpropionsäure, p Butoxyphenylpropionsäure, p-Hexyloxyphenylpropionsäure, Benzoesäure, Toluylsäure, Naphtoesäure, Äthylbenzoesäure, Phenylessigs,äure, Naphthylessigsäure, Phenylvalerian9äure, Zimtsäure, Phenylpropiollsäure und dergleichen hergestellt werden.
Die erfindungsgemäss erhrältlichen Verbindungen können Säugetieren, Vögeln. und Menschen in einer Vielfalt von oralen und parenteralen Verabreichungs- formen einzeln oder im Gemisch mit anderen, gleich zeitigRTI ID="0002.0240"WI="17" HE="4" LX="1231" LY="1828"> wirksamen Verbindungen zugeführt werden.
Sie können zusammen mit einem festen oder flüssigen Träger, in dem sie gelöst, dispergiert oder suspen- diert vorliegen, angewendet werden.
Festen Zusam- mensetzungen kann die Form von Tabletten, Pul vern, Kapseln, Pillen oder dergleichen, zwecks ein facher Verabreichung und: genauer Dosierung vor zugsweise mit einheitlicher Steroidkonzentration, ge geben werden. An flüssigen Zusammensetzungen kommen Lösungen, Emulsionen, Suspensionen, Syrupe oder Elixiere in Frage.
Die in 17a-Stellung alykierten 17f-Hydroxyverbindungen werden vor zugsweise oral verabreicht, da sie hierbei eine be sonders günstige Wirkung entfalten.
<I>Präparat 1:</I> 7a-Methyl 19-norte9tosteron-acetat 30 ml Tetrahydrofuran (erhalten durch Percolie- ren von techn. Tetrahydrofuran über eine Säule aus neutralem Aluminiumoxyd unter Verwerfen der er sten 15 ml des Percolats)
wurden in einem Eisbad gekühlt und unter Rühren in einer Stickstoffatmo- sphäre mit 25 ml einer 3-molaren Methydmagner siumbromidlösung und 0,4g Kupfer (I)-bromid ver setzt.
Diesem Gemisch wurde eine Lösung von 3 g 6-Dehydro-19-nortestosteron 17-acetat und geringen Mengen CuBr in 50 ml wie vorstehend gereinigtem Tetrahydrofuran untrer Rühren zugesetzt.
Das. Gemisch wurde unter Kühlen noch 10 Mi nuten gerührt und sodann in ein. mit Stickstoff durchspültes Gemisch aus mit Eis verdünnter Salz säure, gesättigt mit Natriumchlorid-Äther, gegossen. Die Ätherschicht wurde abgetrennt und nacheinander mit Kochsalzlösung, verdünnter Natronlauge + Koch salz, und wiederum mit Kochsalzlösung gewaschen.
Die Ätherextrakte wurden über Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft. Der Rückstand wurde in. 5 ml Pyridin + 5 ml Acet- anhydrid gelöst. Nach 18stündigem Stehen bei Raum temperatur wurde Eis und Wasser zugegossen und das Produkt mit Äther extrahiert.
Der Extrakt wurde mit verdünnter Säure, verdünnter Natronlauge und Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wurde in Methylenchlorid gelöst und die Lösung an Magnesiumsilikat (Florisil) chromatogra- phiert. Die Säule wurde mit Skeuysolve B-Hexanen mit steigendem Gehalt an Aceton eluiert. Die Frak tionen,
in welchen durch IR-Analyse ein Gehalt an 7a-Methyl-19-nortestosteron-17-acetat ermittelt wurde, wurden. vereinigt und zur Trockne eingedampft.
Der ölige Rückstand wurde in Aceton gelöst und über eine mit Aceton getränkte Säule von 20 g Celite und 10 g Dareo chromatographiert. Die Säule wurde mit Aceton eluiert. Die ersten 750 ml wur den zur Trockne eingedampft,
der Rückstand aus 7a - Methyl-19-nortestos@teron -17 - acetat wurde aus Methanol umkristallisiert. Man erhielt 1 g kristalli- nes Material vom F. 111-114 ; [a]D + 48 (Chloro- form). Das UV-Spektrum dieser Verbindung (in Äthanol) besass ein Maximum bei 240 m,u (s=17,350). Analyse: Berechnet für C21H3602 C 76,32 H 9,15 Gefunden:
C 76,28 H 9,44 <I>Präparat 2:</I> 7a-Methyl-19-nortestosteron Eine Lösung von. 3 g 7a-Methyl-19-nortestosteron- 17-acetat in 40 ml einer 5 % igen Lösung von Ka- liumearbonat in. 80 % igem wässrigem Methanol wurde unter Stickstoff 2 Stunden am Rückfluss gekocht.
Das Gemisch wurde mit Äther extrahiert, der Äther extrakt mit Wasser gewaschen und über Magnesium- sulfat getrocknet, filtriert und zur Trockne einge dampft. Der Rückstand wurde mit Äther verrieben, der dabei resultierende Feststoff wurde isoliert. Man erhielt .so 7a-Methyl-19-norbestoseron als kristalli- neu Feststoff vom F. 1.40-145,5 . Eine analysen reine Probe schmolz bei 145-146 ;
[a]D + <I>55 </I> (Chloroform) (nach Umkrisiallisieren aus einem Ge- misch aus Aceton und Skellysolve B). Das UV- Spektrum (in Äthanol) besass ein Maximum bei 241 rag (E = 17,150).
Analyse: Berechnet für C19H2802 C 79,12 H 9,78 Gefunden: C 79,13 H 10,19 <I>Präparat 3:</I> 7a-Methyl-19-norandrosten 3,17-dion Eine Aufschlämmung von 1,4 g Chromtrioxyd in 15 ml Pyridin wurde unter Rühren und Kühlen eine Lösung von 1,4 g 7ca Methyl-19-nortestosteron in 15 ml Pyridin zugegeben.
Nach beendeter Zugabe wurde das Gemisch bei Raumtemperatur (etwa 20 ) 20 Stunden gerührt, sodann mit einem Gemisch. aus gleichen Teilen Benzol und Äther verdünnt und über eine Schicht aus Diatomeenerde (Celite) fil triert. Die Filterschicht wurde gut mit einem 1 : 1- Gemisch aus Äther und: Benzol, dann Wasser und wiederum mit dem Lösungsmittelgemsch gewaschen.
Das Filtrat und die organische Schicht der Wasch flüssigkeit wurden vereinigt und mehrmals mit Wasser gewaschen. Das- Waschwasser wurde nochmals. mit Äther + Benzol (1 :
1) extrahiert. Die vereinigten organichen Schichten wurden getrocknet und zur Trockne eingedampft. Der Rückstand wurde mit Äther verrieben, der dabei erhaltene Feststoff wurde abfiltriert. Man erhielt 1,4 g kristallines 7a-Methyl- 19-norandrosten-3,17-dion vom F. 195-198 .
Eine durch Umkristallisieren aus Aceton erhaltene An alysenprobe schmolz bei 201-204 . Das UV-Ab- sorptionsspektrum, dieser Verbindung (in Äthanol) zeigte ein. Maximum bei 239,5 mg (a = l7,000).
Analyse: Berechnet für C19H2602 C 79,69 H 9,15 Gefunden: C 79,66 H 8,87 <I>Präparat 4:</I> 7a-Methyl-19-norandrosten-3,17-dion-3- pyrrolidyl-enamin. Einer Lösung aus 10 mg 7a-Methyl-19-norandro- sten-3,17-dion in wenig siedendem Methanol wurde 1 Tropfen Pyrrolidin zugesetzt. Die resultierende Lösung wurde eingeengt und: abgekühlt.
Der sich ab scheidende kristalline Feststoff wurde abfiltriert, mit wenig Methanol gewaschen und getrocknet. Er be stand aus 7a-Methyl-19-norandrosten-3,17-dion-3- pyrrolidyl-enamin, F. 151-160 . Das UV-Spektrum (in Äther) besass ein Maximum bei 282 mu <I>(a =</I> 23,450). Das IR-Spektrum (in. Mineralöl) besass Maxima bei 1735, 1635, 1600, 1200, 1180, 1155 und 1035 cm-'.
<I>Beispiel 1:</I> 7a-Methyl-17a-räthinyl 19-nortestosteron 1 ml einer 20 gew. % igen Suspension von Na- triumacetylid in. Xylol wurde zentrifugiert. Der sich

Diese Lösung wurde mehrmals mit Methylenchlorid extrahiert. Die vereinigten Extrakte wurden mit verdünnter Salz säure, verdünnter Sodalösung und Wasser gewa sehen, über Natriumsulfat getrocknet, filtriert und zur Trockne eingeengt. Der Rückstand (3,9 g) wurde zusammen mit den vorstehend genannten 2 g, in Aceton gelöst, mit einem Gemisch aus.
Holzkohle (Darco), Diatomeenerde (Celite) und Magnesnum- silikat (Florisil) behandelt. Das Gemisch wurde fil- triert und das Filtrat zur Trockne eingedampft.
Der Rückstand wurde aus Aceton-Skellysolve B umÜai- sballisIert, wobei 3,9 g kristallines 7a-Methyl-17a- äthinyl-testosteron vom F. 190-192,5 erhalten wur den.
Aus Äthylacetat erhielt man eine analysenreine Probe vom F. 191-193 ; [a]D + 41 (Chloroform). Das UV-Spektrum (in Äthanol) zeigte ein Maximum bei 242 mg (s = 16,550).
Analyse: Berechnet für C22H3o02 C 80,93 H 9,26 Gefunden: C 80,86 H 9,33 <I>Beispiel 4:</I> 7a-Methyl-17a-(1-propinyl)-testosteron Ersetzt man im. Verfahren gemäss Beispiel 3 C das Natriumacetylid durch Nabriummethylacetylid, so wird das 7a-Methyl--17a-(1-propinyl)-testosteron als kristalliner Feststoff erhalten.
<I>Beispiel 5:</I> 7a-Methyl-17a-(1-butinyl)-testosteron Diese Verbindung erhält man gemäss dem Ver fahren des Beispiels: 3 C, indem man das dort ver wendete Natriumacetylid durch Natriumäthylacetylid ersetzt.
<I>Beispiel 6:</I> 7a4Methyl-17a-äthinyl-testosteron-17-propionat Einer Lösung von 5 g 7a-Meihyl 17a-äthinyl- testosberon in 20 ml Pyridin werden 5 ml Propion- säureanhydrid zugesetzt.
Die Lösung wird am Rück flur gekocht, bis im IR Spektrum keine Hydroxyl- gruppe mehr sichtbar ist.
Dann wird mit gestossenem Eis und Wasser verdünnt, der sich abscheidende Fest stoff abfiltriert und aus wässrigem Aceton umkristal- lisiert. Man erhält 7a-Methyl-17a-äthinyl-testosteron- 17-propionat als kristallinen Feststoff.
Ersetzt man im vorstehenden Verfahren das Pro- pionsäureanhydrid durch ein anderes geeignetes Kohlenwas,sers:toffearbonsäureanhydrid, so erhält man das entsprechende 17-Acylat des 7a-Methyl-17a-äthinyltestosterons wie z.

An analytically pure sample melted at 145-146;
