Verfahren zur Darstellung von Dehydrocorticosteron. Es wurde gefunden, dass man zu Deliy dro- corticosteron gelangen kann, wenn man ein Carboiisäurehalogenid eines Derivates der :ltiocholansäure, das in 3- und 11-Stellung durch Hydrolyse in Hydroxyl um-,van.delbare Gruppen aufweist, mit Diazomethan zum Di- :
@zolietori umsetzt, im erhaltenen Produkt die Peschützten Hydroxylgruppen in 3- und 11- Siellung in Freiheit setzt,
in bzlizbigcr Reihenfolge diese Hy droxylgruppen mit Oay- da.tionsmitteln in Ketogruppen und die 1 7- stä ndige Gruppiei@ing mit Säuren in eine v er- esterte Ketolgruppe umwandelt,
in a-Stellung zur Ketob ruppe eine Doppelbindung durch I3alogenierung und nachträgliche Halogen- vassersi:offabspa.ltung einführt und schliess lich mit hydrolysierenden Mitteln behandelt.
Bei den Resten in 3- und 11-Stellung des .Iusbangsstoffes kann es sich z. B. um durch unorganische oder organische Säuren, wie Ca.rbonsäuren, Sulfonsäuren oder Halogen- wasserstoffsäuren, veresterte oder durch Al- holiole oder Phenole, wie Trianllmelityl- oder Benzylalkohole, enolisierte oder acetalisierte Carbonylvc@rbiridungen,
verätherte Hydroxyl- gruppen handeln.
Die L mgetzung des Carbonsäurelial'ogenids iit Diazomethan erfolgt unter Halogen- wasserstoffabspaltung und Bildung eines Di- azol:etons -CO-CHN" insbesondere dann, wenn ständig ein tberschuss von Diazo- rnetlian. zugegen ist.
Das erhaltene Diazolketon kann in rohem Zustande oder nach. Abtrennung und Reini- gung weiter verarbeitet werden. Die freien Hy droxylgruppen in 3- und 11-Stellung wer den mit hydroly sierenden Mitteln hergestellt.
Die freien 3- und 11-Hydroxylgruppen werden durch Oxydationsmittel, z. B. mit Chromsäure in Eisessig oder durch Erhitzen mit Kupferpulver, Einwirkung von Metall- alkoholaten bzw. -phenolaten in Gegenwart von Ketonen, wie Aceton oder Cyclohexanon, in Ketogruppen übergeführt.
Nach oder auch vor der Oxydation der S und 11-Hydroxylgruppen wird die gegebenen falls vorhandene Diazoketongruppierung durch Einwirkung wasserfreier oder ver dünnter organischer oder anorganischer Säu ren, beispielsweise Essigsäure, Propionsäure, Buttersäuren, Crotonsäuren, Palmitinsäure, Benzoesäure, Phenylessigsäure, Schwefel- sKure, l@lethansulfosäure, Toluolsulfos.äure, Halogenwa:
sserstoffsäure, Phosphorsäure oder Borsäure, in eine veresterte Ketolgruppe übergeführt. Es können so z. B. Acylate, Halogenide oder Sulfonsäureester erhalten werden.
Die Doppelbindung in a-Stellung zur 3- Ketogruppe wird durch Halogenierung und nachträgliche ITalogenwasserstoffabspa.ltung gebildet.
Schliesslich wird die veresterte Ketol- gruppe mit hydrolysierenden Mitteln ver seift. Dabei wird man auf die relative Alkali- empfindl'ichkeit der a-Oxy-keton-Gruppie- riAg Rücksicht nehmen, so dass man vorteil- haft mit Säuren oder mit ganz schwachen Alkalien, wie Bicarbonaten, arbeitet.
Das Verfahrensprodukt, das 44-Pregnen- 3,11,20-trion-21-ol (Dehydrocorticosteron), ist bekannt. Es soll therapeutische Verwendung finden oder als Zwischenprodukt zur Her stellung therapeutisch wertvoll-er Verbindun gen dienen.
<I>Beispiel:</I> 23 Teile 3a,llss-Diacetoxy-ätiocholansäure werden bei 0 in 140 Teilen reinstem Thionyl- chlorid gelöst und unter Feuchtigkeitsaus- schluss zunächst 1/2 Stunde bei 0 , dann noch 16 Stunden bei 18 stehengelassen.
Hierauf wird im Vakuum bei 40 Badtemperatur ein gedampft, der Rückstand in 500 Teilen abso lutem Benzol .gelöst und in eine aus 100 Teilen Nitrosomethylharnstoff bereitete, über Kaliumhydroxyd getrocknete und frisch de stillierte ätherische Lösung von Diazomethan bei 0 eingetragen, wobei sofortige Gas entwicklung zu beobachten ist. Die Mischung bleibt unter Feuchtigkeitsausschluss (KOH) zunächst 2 Stunden bei 0 ,
dann noch 16 i Stunden bei 18 stehen, wird hierauf bei 50 Badtemperatur stark eingeengt und 'zuletzt im Vakuum völlig getrocknet.
Das so erhaltene rohe 21-Diazo-pregnen- 3a,1lss-diacetoxy-20-on wird in 300 Teilen r Methanol gelöst, mit der Lösung von 15 Teilen Kaliumhydroxyd in 20 Teilen Wasser und 700 Teilen Methanol versetzt und 8 Stunden bei 20 stehengelassen. Hierauf wird die Lösung von 32 Teilen Kaliumbikarbonat in 1000 Teilen Wasser zugegeben, das Me thanol im Vakuum entfernt und die verblei bende Suspension mit viel Äther ausgeschüt telt.
Die mit Wasser gewaschene und über Natriumsulfat getrocknete Ätherlösung wird i bei 50 Badtemperatur stark eingeengt und hierauf im Vakuum völlig eingedampft, wo bei rohes 21-Diazo-pregnan-3a,llss-dioi-20-on zurückbleibt. Dieses wird mit 300 Teilen reinstem wasserfreiem Eisessig 1/2 Stunde auf 95 bis 1.00 erhitzt und das dabei abgespaltene Gas in einem Messzylinder über Wasser aufc-@- fangen. Nach Beendigung der Stiekstoff- entwickl'ung wird im Vakuum eingedampft.
Der verbleibende Rückstand wird in an sich bekannter Weise über Aluminiumoxyd chro- matographisch gereinigt,.
Die Benzol-Äther-Ei luate geben beim Uni- kristallisieren aus Äther - Petroläther das Pregnan-3a,11ss,21-triol-20-on-21-mono-acetat in farblosen Kristallen.
8,5 Teile des beschriebenen i)1onoacetatcs werden in 100 Teilen Eisessig gelöst, allmäh lich mit 200 Teilen 2 % iger Chromtrioxyd- Eisessiglösung (= 4 Teile Cr03) versetzt und 6 Stunden bei 20 stehengelassen.
Hier auf wird im Vakuum bei 30 Badtemperatur stark eingeengt, mit Wasser versetzt und mit Äther ausgeschüttelt. Die mit verdünnter Schwefelsäure, Sodalösung und Wasser ge waschene und über Natriumsulfat getrock nete Ätherlösung wird stark .eingeengt und die bald einsetzende Kristallisation durch Zusatz von etwas Petrolä-ther vervollständigt.
Man erhält auf diese Weise das Pregnan- 3a,llss-20-trion-21-ol-acetat in farblosen Na deln vom F. 153-155 und er spezifischen Drehung [a]" = -L 107,2 4 (c = 0,783 in Aceton).
Zu demselben Produkt gelangt man auch, wenn , das 21- Diazopregnan - 3a,llss - diol- 20-on zunächst z. B. bei Zimmertemperatur mit Aluminiumisopropylat und Aceton be handelt, die erhaltene Verbindung anschlie ssend mit Eisessig erhitzt und hierauf mit Chromsäure oxydiert wird.
Durch Vermischen von 10 Teilen Brom mit 384 Teilen Eisessig wird zunächst eine n-Bromlösung bereitet. 9,6 Teile des Pro duktes vom F. 153-155 werden in 50 Teilen Eisessig gelöst und hierauf mit 2 Tropfen der obigen Bromlösung versetzt. Nach eini gen Altnuten tritt plötzliche Entfärbung ein, worauf man unter Kühlung und ITmschwen- ken langsam noch 48 Teile der Bromlösung zugibt, die fast momentan entfärbt wird. Nach Eindampfen im Vakuum bei 25 Rad temperatur tritt Kristallisation ein. Die mit. Äther gewaschenen, farblosen Kristalle schmelzen bei 180-185 .

When unicrystallizing from ether - petroleum ether, the benzene-ether-eluates give the pregnane-3a, 11ss, 21-triol-20-one-21-mono-acetate in colorless crystals.
8.5 parts of the i) 1onoacetatcs described are dissolved in 100 parts of glacial acetic acid, 200 parts of 2% chromium trioxide glacial acetic acid solution (= 4 parts of Cr03) are gradually added and the mixture is left to stand at 20 for 6 hours.
