Verfahren zur Herstellung eines Acyloxyderivates des

Produkt mit Chromsäure in Eisessig oxydiert, das Oxydationsprodukt mit Kalilauge und Methanollösung hydrolysiert, danach chromatographiert und durch Acetylierung des 3-ständigen Hydroxyls als Pregnadienolonacetat gewonnen. Über die Ausbeute wurde nichts angegeben, sie ist jedoch nach eigener Erfahrung niedrig.
Es wurde gefunden, dass man die Ausbeute des acylierten Produktes erhöhen'kann, wenn man zur Acylierung anstatt Essigsäureanhydrid Propionsäureanhydrid verwendet. Auf diese Weise kann man das A5, 16-Pregnadien-3ss-ol-20-on-propionat mit einer Ausbeute von etwa 507o, bezogen auf Solasodin, erhalten. Abgesehen von der erhöhten Ausbeute, liegt ein weiterer Vorteil des Verfahrens darin, dass das neue Acylderivat in Methanol 2, 5-mal schwerer löslich ist, als das Acetat, so dass in der Mutterlauge nur sehr wenig Endprodukt zurückbleibt.
Von der neuen Propionsäureverbindung kann man zu den wertvollen Steroidhormonen auf ähnliche Weise gelangen, wie das bei der Acetylverbindung bekannt ist.
Um festzustellen, ob nicht bei der Isomerisation mittels Eisessig eine Umesterung stattfindet, wurden Versuche durchgeführt, bei welchen bei der Acylierung mittels Propionsäureanhydrid die Isomerisation auch mit Propionsäure durchgeführt und bei der folgenden Oxydation als Lösungsmittel auch Propionsäure benutzt wurde. In diesem Falle wurde das gewünschte Produkt mit ebenso guter Ausbeute erhalten, was darauf schliessen lässt, dass eine Umesterung nicht stattgefunden hat. Nach Abtrennung des kristallinen Produktes wird die Mutterlauge zur Trockne verdampft, das erhaltene ölige Produkt wird in gleicher Menge Methanol gelöst, mit dreifacher Menge Äther im Schütteltrichter gemischt und die neutralen und basischen Stoffe voneinander mittels einer 8%igen Salzsäurelösung getrennt.
Die Ätherlösung wird neutral gewaschen, getrocknet, der Äther abdestilliert und der Rückstand aus Methanol umkristallisiert. Auf diese Weise kann man auf Solasodin bezogen noch weitere 2% Propionat gewinnen.
Beispiel 1 : 50 g Solasodin (Schmelzpunkt 191-194 C) wird in einem Gemisch von 700 ml Pyridin und 200 ml Propionsäureanhydrid gelöst und 3 Stunden am Ölbad zum Sieden erhitzt. Danach wird Pyridin und überschüssiges Propionsäureanhydrid im Vakuum abdestilliert und der Rückstand an einem Öl- bad, dessen Temperatur nicht mehr als 110 C beträgt, vollständig getrocknet. Man erhält. auf diese Weise 68 g Rohprodukt. Dieses Dipropionylsolasodin wird in 500 ml Eisessig gelöst und die Lösung 2 1/4 Stunden zum Sieden erhitzt. Die die isomerisierte Dipropionatverbindung enthaltende Lösung wird dann zusammen mit einer Lösung von 20 g CrO in 300 ml Eisessig während 15 Minuten zu 300 ml Eisessig langsam hinzugefügt.
Die überschüssige Chromsäure wird durch Hinzufügung von 10 g Natriumbisulfit zersetzt und danach zu der Lösung die gleiche Menge Wasser hinzugefügt. Die Lösung wird danach mit insgesamt 8 1 Benzol in vier Portionen extrahiert. Die Benzollösung wird mit Natriumbicarbonat und danach mit Wasser neutral gewaschen und über Natriumsulfat getrocknet. Die wasserfreie Lösung wird filtriert und das Benzol abdestilliert. Man erhält 60 g Rückstand. Aus diesem Oxydationsprodukt wird der Propionylaminoisocapronsäure-Rest durch Wärmespaltung wie folgt entfernt :

Das Oxydationsprodukt wird am Ölbad auf 1600C erhitzt und 3/4 Stunden gerührt. Zu dem noch warmen und flüssigen Öl werden 100 ml Methanol rasch hinzugefügt.
Die homogene Lösung wird 1 Stunde bei Zimmertemperatur und danach 1 Stunde bei OOC stehen gelassen. Das kristalline Produkt wird abgenutscht und viermal mit 10 ml eisgekühltem Methanol gewaschen und bei 60-70 c bis Gewichtskonstanz getrocknet. Man erhält auf diese Weise 21, 5 g A5, 16-Pregnadien-3ss-ol-20-on-propionat (F = 172-174 C), Ausbeute, bezogen auf Solasodin, 48%.

Aus der Mutterlauge kann man noch weitere Mengen dieser Substanz erhalten, wenn man die Mutterlauge einengt, in gleicher Menge Methanol aufnimmt und danach die dreifache Menge Äther hinzufügt und fünfmal je mit der gleichen Menge 8% iger wasseriger Salzsäure ausschüttelt.
Die Ätherlösung wird dann mit Natriumbicarbonatlösung und danach mit Wasser neutral gewaschen und über Natriumsulfat getrocknet. Die Lösung wird filtriert, das Lösungsmittel abdestilliert, der Rückstand in der gleichen Gew.-Menge Methanol gelöst und bei 00C stehen gelassen. Auf diese Weise erhält man 870 mg Propionat mit einem Schmelzpunkt von 161-168 C. Durch Umkristallisieren aus Methanol erhält man 550 mg Propionat (F = 172-1740C). Die Gesamtausbeute ist somit 49. 3'10. berechnet auf den Ausgangsstoff.
Beispiel 2 : Eine Lösung von 5 g Solasodin in 70 ml Pyridin- (F = 191-194 C) - wird mit 20 ml Propionsäureanhydrid versetzt, 3 Stunden am Ölbad zum Sieden erhitzt und danach im Vakuum zur Trockne verdampft. Der sirupartige Rückstand (5, 4 g) wird in 50 ml Propionsäure kalt gelöst und danach 2 1/4 Stunden am Ölbad zum Sieden erhitzt. Die abgekühlte Lösung wird zusammen mit einer Lösung von 2 g croc in 30 ml Propionsäure zu 30 ml Propionsäure während 15 Minuten tropfenweise hinzugefügt.
Nach einstündigem Stehen wird das Reaktionsgemisch gemäss Beispiel 1 weiter verarbeitet, wobei man ebenfalls mit guter Ausbeute das #5, 16-Pregnadien-3ss-ol-20-on-propionat erhält.

The product is oxidized with chromic acid in glacial acetic acid, the oxidation product is hydrolyzed with potassium hydroxide and methanol solution, then chromatographed and obtained as pregnadienolone acetate by acetylating the 3-position hydroxyl. Nothing was given about the yield, but our experience has shown that it is low.
It has been found that the yield of the acylated product can be increased if propionic anhydride is used for the acylation instead of acetic anhydride. In this way, the A5, 16-pregnadien-3ss-ol-20-one propionate can be obtained with a yield of about 5070, based on solasodine. Apart from the increased yield, another advantage of the process is that the new acyl derivative is 2.5 times more soluble in methanol than the acetate, so that very little end product remains in the mother liquor.
From the new propionic acid compound one can get to the valuable steroid hormones in a similar way as is known with the acetyl compound.
In order to determine whether transesterification does not take place in the isomerization using glacial acetic acid, tests were carried out in which the isomerization was also carried out with propionic acid during the acylation using propionic anhydride and propionic acid was also used as solvent in the subsequent oxidation. In this case, the desired product was obtained with an equally good yield, which suggests that transesterification did not take place. After the crystalline product has been separated off, the mother liquor is evaporated to dryness, the oily product obtained is dissolved in the same amount of methanol, mixed with three times the amount of ether in a separating funnel and the neutral and basic substances are separated from one another using an 8% hydrochloric acid solution.
