Nach einer bevorzugten Ausführungsart des erfindungsgemässen Verfahrens verwendet man als Ausgangsstoff der Formel n ein 3-Acyloxy-AS-10-formylsteroid und setzt dieses in einer ersten Verfahrens- ; rufe nach Wittig mit einem Triaryl-alkylidenphosphoran zur entsprechenden 19-Alkylidenverbindung ier Formel III um. Die Herstellung der Phosphoran-Komponente, wie beispielsweise des Triphenyl- - äthyliden-phosphorans der Formel (C H) P = CHCH, sowie deren Umsetzung mit dem 10-Formyl- ; teroid (Wittig-Reaktion) können nach an sich bekannten Methoden erfolgen (vgl. z. B. Angewandte Chemie 71, 260 [1959]).
Man geht dabei zweckmässig so vor, dass man das Phosphoran unmittelbar 1ach seiner Bereitung in situ mit dem 10-Formylsteroid bei Temperaturen zwischen etwa 35 und 1350 C, vorzugsweise im Bereich von etwa 70 bis 800 C, umsetzt. Das resultierende 19-Alkylidensteroid der mormel ni kann beispielsweise dadurch isoliert werden, dass man die Reaktionslösung eindampft und ien Eindampfrückstand chromatographiert. Verbindungen mit hydrolysierbaren Gruppen, wie beispielsweise Acyloxygruppen in 3- und 17-Stellung, werden bei der Aufarbeitung zum Teil verseift.
Die erhaltene 19-Alkylidenverbindung der Formel III wird hierauf durch Hydrierung der im Mole-

kül vorhandenen Äthylenbindungen und durch Hydrolyse von allenfalls vorhandenen veresterten oder verätherten Hydroxygruppen in das entsprechende 19-Alkylsteroid der Formel IV übergeführt. Hydrie- rung und allfällige Hydrolyse können dabei, wie erwähnt, in beliebiger Reihenfolge vorgenommen wer- den. Die Hydrierung erfolgt zweckmässig mit katalytisch aktiviertem Wasserstoff bei Raumtemperatur und Atmosphärendruck. Als Hydrierungskatalysatoren kommen die üblichen Metallkatalysatoren in Be- tracht, wie Nickel-, Palladium- oder Platinkatalysatoren. Die Hydrierung kann in saurem Medium (z. B. in Eisessig) oder in neutralem Medium (z.
B. in Äthanol, Benzol, Toluol usw. ) vorgenommen werden. Bei Verwendung saurer Medien führt die Hydrierung von 5, 6-ungesättigten Verbindungen der
Formel III bevorzugt zu 5a-Isomeren.
Die Dehydrierung mit Dichlordicyanobenzochinon erfolgt vorzugsweise in Benzol oder Dioxan, die
Dehydrierung mit Chloranil vorzugsweise in Amylalkohol.
Die als Ausgangsstoffe verwendeten 10-Formylsteroide, können soweit es sich um noch nicht be- kannte Verbindungen handelt, nach an sich bekannten Methoden gewonnen werden.
Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen der Formel I sind hormona], z. B. anabolisch, androgen, östrogen und/oder antiprogestativ wirksam. Sie besitzen ferner positiv ino- trope Herzaktivität. Sie können dementsprechend als Heilmittel verwendet werden.
Beispiel l : Zu einer Suspension von 9, 6 g Triphenyläthylphosphoniumbromid in 1000 ml Te- trahydrofuran gibt man bei Raumtemperatur unter Rühren auf einmal eine ätherische Lösung von 2, 2 g
Phenyllithium, wobei Rotfärbung eintritt. Nun setzt man eine Lösung von 1 g 19-Oxo-A5-androsten-38,
17ss-diol-diacetat in 40 ml Tetrahydrofuran auf einmal zu und rührt das Gemisch zwei Tage am Rückfluss. Dabei hellt sich die Farbe des Gemisches allmählich auf. Die gelbe Reaktionslösung wird dann unter reduziertem Druck zur Trockene eingedampft und der Rückstand mit Pyridin-Acetanhydrid (200 ml Pyridin, 100 ml Acetanhydrid) behandelt zwecks Wiedereinführung der teilweise abgespaltenen Acetylgruppen.
Man erhält so 5 g eines orange gefärbten, teilweise kristallinen Produktes, welches durch Chromatographie an der 45fachen Menge Silicagel und anschliessendes Umkristallisieren aus tiefsiedendem Petroläther gereinigt wird. Das so erhaltene 19-Äthyliden-AS-androsten-3ss, 17ss-diol- - diacetat (600 mg ; 600/0) schmilzt nach Umkristallisation bei 1170 C.
400 mg der erhaltenen 19-Äthylidenverbindung werden in 15 ml Eisessig in Gegenwart eines Pla- tinkatalysators (vorhydriertes PtO) unter Normalbedingungen hydriert. Die Hydrierungskurve steigt steil an, um sich nach der Aufnahme von 2 Moläquivalenten Wasserstoff brüsk abzuflachen. Nach Filtration vom Katalysator und Eindampfen des Filtrats erhält man 435 mg eines gelben Öls, welches in Äther gelöst und unter Rühren bei Zimmertemperatur zwecks Verseifung der Acetoxygruppen mit 500 mg Lithiumaluminiumhydrid behandelt wird. Nach Zugabe von verdünnter Salzsäure extrahiert man das Gemisch mit Äther. Aus der Ätherlösung erhält man 342 mg rohes 19-Äthyl-5a-androstan-3B, 17ss-diolin Form eines hellgelben Schaums.
Zu einer Lösung dieses Diols in 15 ml Eisessig gibt man 1, 37 ml einer Lösung von 400 mg Chromtrioxyd in 0, 4 ml Wasser und 1, 6 ml Eisessig und hält das Gemisch 11/4 h bei 50 C. Zur braungrünen Lösung gibt man 15 ml Äthanol zwecks Zerstörung des überschüssigen Chromtrioxyds. Nach der Aufarbeitung erhält man 316 mg 19-Äthyl-5a-androstan-3, 17-dion in Form eines gelben Öls, welches durch Chromatographie an der 80fachen Menge Silicagel und anschliessendes Umkristallisieren aus Äther-Petroläther gereinigt wird. Schmelzpunkt : 1230C.
Das alsAusgangsmaterial verwendete19-0xo-A5-andro- sten-3 6, 17ss-diol-diacetat kann wie folgt erhalten werden :
In eine Lösung von 8 g des Oxims von 19-0xo-AS-androsten-3B, 176-diol-diacetat in 800 ml Eisessig tropft man unter Rühren 400 ml einer wässerigen, 5'7eigen Natriumnitritlösung und lässt das Gemisch nach 1 h Rühren 1 bis 3 Tage bei Raumtemperatur stehen. Anschliessend giesst man das Gemisch auf 3000 ml Eiswasser und extrahiert erschöpfend mit Äther.
Nach der üblichen Aufarbeitung erhält man 10, 1g eines gelben Öls, aus welchem durch Chromatographie an Silicagel und anschliessende Kristallsation aus Äther-Petroläther die genannte 10-Oxo-verbindung in einer Ausbeute von 450/0 erhalten werden kann. Schmelzpunkt : 150-1520 C.
Beispiel 2 : 2, 4 g 19-Äthyliden-A-androsten-3ss, 17ss-diol-diacetat (hergestellt gemäss Beispiel piel1, erster Absatz) werden in 150 ml Methanol gelöst. Nach Zugabe von 2 g Natriumhydroxyd wird die Lösung 5h bei Raumtemperatur gerührt und hierauf im Vacuum zur Trockene eingedampft. Das nach Aufarbeitung des Rückstandes erhaltene kristalline 19-Äthyliden-A5-androsten-3ss, 17ss-diol sin- tert ab 900 C, wird dann wieder fest und schmilzt schliesslich bei 1650 C (unkorrigiert). Die Ausbeute ist praktisch quantitativ.
4, 3g von so erhaltenem 19-Äthyliden-A -androsten-3ss, 17ss-diol werden mit 1, 5 g vorhydrier-

tem Platinoxyd in 250ml Eisessig unter Schütteln hydriert. Nach 11/2 h beträgt die Wasserstoffaufnahme 570 ml (Theorie 600 ml). Das Reaktionsgemisch wird filtriert und das Filtrat zur Trockene eingedampft. Aus dem Rückstand lassen sich 4, 25 g rohes 19-Äthyl-5a-androstan-3ss, 17ss-diol isolieren, das bei 177 bis 1780 unvollständig schmilzt. Es enthält als Nebenprodukt geringe Mengen an 5ss-Isomeren.
4, 25 g des so erhaltenen rohen 19-Äthyl-5et-androstan-38 -178 -diol werden in 695 ml Aceton gelöst. Die Lösung wird auf 20 C abgekühlt. Bei dieser Temperatur werden 17, 4 ml Jones-Reagens zugetropft. (Es wurde ein Jones-Reagens folgender Zusammensetzung verwendet : 66, 8 g Chromtrioxyd, 57. 5 mol konzentrierte Schwefelsäure, Wasser ad 250 ml.) Nach 5 min werden weitere 17, 4 ml des Jones-Reagens'zugetropft. Nach 5 min wird das Reaktionsgemisch in 2, 5 I Eiswasser (30 g Natriumbicarbonat enthaltend) gegossen. Das resultierende trübe Gemisch wird 5mal mit je 600 ml Äther ausgezogen. Die Extrakte werden vereinigt, mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft.
Als Rückstand verbleiben 3, 92 g eines Öls, in dem auf Grund dünnschichtchromatographischer Analyse das 5ct-Isomere, d. h. das 19-Äthyl-5a-androstan-3, 17-dion stark überwiegt. Nach chromatographischer Reinigung (Kieselgel, Korngrösse 0, 05 bis 0, 2 mm ; Elution mit Ben-

Die erhaltene dunkelbraune Lösung wird mitWasser vermischt und die Mischung mit Äther mehrfach extrahiert. Die Ätherextrakte werden mit Natrium-thiosulfat-und Natriumbicarbonatlösung und zuletzt noch mit Wasser neutral gewaschen.
Nach Trocknen über Natriumsulfat und Einengen zur Trockene erhält man 180 mg rohes 2, 4-Di- brom-19-äthyl-5α-androstan-3,17-dion, das sofort wie folgt weiter verarbeitet wird :
Die erhaltene rohe Dibromverbindung (180 mg) wird im Gemisch mit 1, 0 ml Dimethylformamid, 85 mg Lithiumcarbonat und 65 mg Lithiumbromid 24 h auf 1100 C erhitzt, wobei die Suspension in Lösung geht. Die erhaltene Lösung wird mit 15 ml Wasser und einigen Tropfen Eisessig versetzt, 10 min lang bei Raumtemperatur gerührt und hierauf mit Methylenchlorid ausgezogen. Die Extrakte werden mit Natriumbicarbonatlösung und Wasser gewaschen, dann getrocknet und eingedampft.
Man erhält 120 mg eines Öls, aus welchem durch Säulenchromatographie an der dreissigfachen Menge Alox III mit einem Benzol/Petroläther-Gemisch (1:1) 19-Äthyl-#1,4-androstadien-3,17-dion vom Schmelzpunkt 1800 C isoliert werden kann.
