Verfahren zur Herstellung von Steroiden der Östranreihe
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung biologisch-aktiver Steroide. Besonders in den Pregnanreihen sind in 16-Stellung alkylierte Steroide bekannt, in welchen sowohl die 1 6a-, wie die 1 6ss-Alkylverbindungen bedeutende biologische Wirkungen haben. Gewisse Vertreter dieser Gruppen, wie das 1 6a-Methyl- und das 1 6fl-Methyl-9a-fiuor-prednisolon, haben sich als von grossem therapeutischem Wert erwiesen, und zwar auf Grund ihrer Eigenschaften als Glucocorticoide und Antiphlogistica. In anderen Reihen, wie beispielsweise in der Östranreihe, wurden bisher derartig wertvolle 16-alkylierte Verbindungen nicht aufgefunden.
Es ist besonders überraschend, dass die in 1 613-Stel- lung alkylierten Derivate wesentlich weniger aktiv sind als die korrespondierenden 1 6a-Alkylverbindungen, die anabolische, androgene, progestative, die Ovulation verhindernde und die Schwangerschaft schützende Wirkungen haben.
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung der erwähnten neuen 16a-Alkylsteroide der Östranreihe der Formel:

worin R1 = H oder Acyl = = H oder eine Alkyl-, Alkenyl- oder Alkinylgruppe mit 1 bis 4 Kohlenstoffatomen und P und Q Wasserstoff oder die Methyl- oder Äthyl- gruppe sind.
Diese Verbindungen werden nach der Erfindung hergestellt, indem von einem Gemisch aus den 1 6u- und 1 6p-Alkylisomeren einer Verbindung der Formel

ausgegangen wird, worin X für einen niedrigeren aliphatischen Kohlenwasserstoffrest steht, und dass man einerseits aus dem Isomerengemisch das entsprechende 1 6a-Alkyhsomere abtrennt und anderseits eine Reduktion des aromatischen Ringes A mit einem Alkalimetall in flüssigem Ammoniak und anschliessend eine Säurebehandlung durchführt.
Die Erfindung bezieht sich im weiteren auch auf die Verwendung erhaltener in 1 7a-Stellung unsubstituierter 174-Hydroxy-steroide zur Herstellung von in 17a Stellung substituierten Derivaten davon, indem man die 1 7ss-Hydroxygruppe in den Verfahrensprodukten zur 17-Oxogruppe oxydiert und diese 17-Oxoverbindungen mit einer Metallverbindung eines Alkans, Alkens oder Alkins mit 1 bis 4 Kohlenstoffatomen umsetzt.
Das Gemisch aus dem 1 6a- und dem 1 6ss-Alkyl- isomeren kann auf verschiedene Weise erhalten werden.
Eine dieser Methoden besteht darin, dass man Östron selbst oder einen seiner 3-Ester oder 3-Äther in Anwesenheit einer Base mit einem Keton umsetzt, so dass man die entsprechende 1 6-Alkylidenverbindung erhält, die daraufhin in ein Gemisch aus der entsprechenden 1 6a- und 1 6ssAlkylverbindung übergeführt wird, indem man zuerst die 17-Ketogruppe und dann die 16-Alkylidengruppe reduziert.
Eine andere Methode, die sich insbesondere für die Herstellung der 16-Methylverbindungen eignet, besteht darin, dass man Östron oder eines seiner funktionellen Derivate mit Hilfe einer Mannich-Reaktion mit Formaldehyd und einem Amin, z. B. Dimethylamin oder Morpholin umsetzt, worauf man die Aminogruppe durch Wasserdampfdestillation oder durch Behandlung mit Silikagel abspaltet und die so erhaltene 1 6-Methy- len-17-Ketoverbindung wie oben zu den 16-Alkylidenverbindungen reduziert.
Die 16-Alkylidengruppe kann reduziert werden durch Behandlung mit Wasserstoff in Anwesenheit eines Metallkatalysators wie Platin oder Palladium.
Man kann die 1 6-Alkylverbindungen auch so herstellen, dass man auf das entsprechende 17-Ketosteroid eine Oxalylesterkondensation anwendet und das so erhaltene Produkt mit einem Alkylhalogenid behandelt.
Schliesslich kann man auch 16ssMethyl-östron oder eines seiner funktionellen Derivate einer Isomerisation unterwerfen.
Das durch eine der beschriebenen Methoden erhaltene Isomerengemisch wird nun in der Regel vor der Oxydation getrennt, um das gewünschte 1 6-Alkyl- isomere zu isolieren. Die Trennung kann durchgeführt werden mit Hilfe einer der üblichen Methoden, z. B.
Extraktion, Kristallisation oder auf chromatographischem Wege. Besonders die letzte Trennmethode hat sich als wirkungsvoll erwiesen. Als Absorbens benutzt man dabei gewöhnlich Aluminiumoxyd oder Silikagel und als Eluans und anderen einen aromatischen Kohlenwasserstoff, wie Benzol, Xylol oder Toluol oder einen aliphatischen Äther, wie Dimethyläther, Dioxan oder Tetrahydrofuran oder einen halogenierten aliphatischen Kohlenwasserstoff, wie Chloroform, oder auch Petroläther oder Äthylacetat oder Gemische aus diesen Lösungsmitteln.
Das Mengenverhältnis zwischen dem rl- und dem 3-Isomeren in dem Gemisch hängt davon ab, auf welche Art dieses hergestellt wurde, jedoch auch von der Natur des 16-Alkylsubstituenten. In den 1 6-Methyl- verbindungen sind die beiden Isomeren in praktisch gleicher Menge anwesend, jedoch herrscht bei längeren 1 6-Alkylketten das 1 6a-Alkylisomere vor.
Es wurde gefunden, dass es bei chromatographischer Trennung des 1 6c und des 16ss-Alkylisomeren zweckmässig ist, zunächst die 1 7-Ketogruppe zu reduzieren oder die Trennung mit einem 17ss-Hydroxy-17a-alkyl- derivat durchzuführen.
Die resultierenden 16a-Alkylöstradiolderivate, die in 17a-Stellung noch eine Alkyl- oder Alkenyl- bzw.
Alkinylgruppe aufweisen können, werden nun vorzugsweise durch Birch-Reduktion? > in die entsprechenden J4 - 3 - Ke.o -1 6a - alkylöstrenverbindungen übergeführt.
Diese Verbindungen sind an sich schon wertvoll aufgrund ihrer biologischen Eigenschaften, können jedoch auch als Ausgangsprodukte für die Herstellung der entsprechenden 3-Desoxosteroide benutzt werden.
Man kann, wie gesagt, eine 1 7a-Alkylgruppe, falls eine solche noch nicht vorhanden ist, in die Endprodukte einführen, und zwar durch Oxydation der erhaltenen 1 7ss-Hydroxyverbindung und nachfolgende Alkylle- rung.
Im Falle einer 17a-Alkylierung einer J4-3,17-Di- keto-16ct-alkylverbindung ist es zweckmässig, die 3-Ketogruppe zunächst vorübergehend zu schützen, beispielsweise durch einen Enoläther oder einen Enolester.
Die in 17a-Stellung einzuführende Alkylgruppe ist ein gesättigter oder ungesättigter aliphatischer Kohlenwasserstoffrest mit 1 bis 4 Kohlenstoffatomen. Als Beispiele seien die folgenden Gruppen erwähnt: Methyl, Äthyl, Propyl, Isopropyl, Butyl, Vinyl, Propenyl, Allyl, Methallyl, Äthinyl, Propinyl, Propargyl oder Butinyl.
Die Alkylierungsreaktion kann so durchgeführt werden, dass man an die 17-Ketogruppe der betreffenden 1 6a-Alkylöstrenverbindung ein Metallderivat eines ge sättigten oder ungesättigten aliphatischen Kohlenwasserstoffes addiert. Das Metallderivat kann ein Magnesiumhalogenid, z. B. das Magnesiumbromid, des betreffenden Kohlenwasserstoffes sein.
Die 1 7fl-Hydroxy-17a-alkinylverbindungen können auch dargestellt werden durch eine Additionsreaktion zwischen dem entsprechenden 17-Ketosteroid und einem 3fach ungesättigten Kohlenwasserstoff in Anwesenheit eines Alkalimetalles oder eine Alkaliverbindung, wie eines Alkalimetallalkoholates, oder durch Addition einer Metallverbindung eines 3fach ungesättigten Kohlenwasserstoffes an die 1 7-Ketogruppe des Ausgangsproduktes; diese Metallverbindung kann eine Alkalioder Erdalkalimetallverbindung sein.
Zur Herstellung einer gesättigten 17a-Alkylverbindung kann man auch ein Metallderivat eines gesättigten Kohlenwasserstoffs, insbesondere ein Alkaliderivat, wie Methyllithium oder Äthyllithium verwenden.
Die so erhaltenen L-14-3-0xo-16a-alkyl-17P-hydroxy- östren-verbindungen, die in 17a-Stellung alkyliert sein können, können gegebenenfalls an der 17ss-Hydroxyl- gruppe verestert werden. Zur Esterbildung wird vorzugsweise eine gesättigte oder ungesättigte organische Carbonsäure mit 1-18 Kohlenstoffatomen benutzt.
Als Beispiele für geeignete Säuren seien die folgenden erwähnt:
Essigsäure, Propionsäure, Valeriansäure, Önanthsäure, Caprylsäure, Caprinsäure, Laurinsäure, Decylensäure, Cyclohexylbuttersäure, Phenylpropionsäure, Palmitinsäure, Stearinsäure, Malonsäure und Bernsteinsäure.
Die Veresterung kann in üblicher Weise durchgeführt werden durch Umsetzung der 17ss-Hydroxyver- bindung mit der entsprechenden Säure oder einem ihrer Derivate, wie dem Anhydrid oder einem Halogenid, z. B. dem Säurechlorid.
Bei der oben beschriebenen Methode wurde aus praktischen Gründen zunächst eine Trennung durchgeführt zwischen dem 16p-Alkylisomeren und dem gewünschten 16a-Alkylisomeren, worauf das resultierende 16a-Alkylisomere den obgenannten Umsetzungen unterworfen wurde. Man kann natürlich die Trennung auch in einer anderen Phase der Synthese durchführen, jedoch ist dies umständlicher.
Beispiel 1
Zu einer siedenden Suspension von 10 g LiAlH4 in 1 Liter Tetrahydrofuran wurde eine Lösung von 10 g 3 -Methoxy- 17 -keto-16-methylen-A18510)-östratn.en in 800 ml Tetrahydrofuran hinzugefügt und das Reaktionsgemisch 2 Stunden im Sieden gehalten. Nach Abkühlen wurden 100 ml Wasser zugeführt und das Gemisch über Hyflo abgesaugt. Der Rückstand wurde mit Chloroform gewaschen und das Filtrat im Vakuum zur Trockene eingedampft, und der Rückstand aus Aceton/Äther umkristallisiert. Man erhielt 8,20 g
3-Methoxy-1 7fi-hydroxy-1 o-methylen-
A1'8,5(10)-östratrien.

5 g dieser Verbindung wurden in 500 ml absolutem Äthanol gelöst und 1,2 g Platinoxyd zugefügt. Nun wurde das Reaktionsgemisch hydriert, wobei die Reaktion abgebrochen wurde, nachdem 0,6 Liter H2 aufgenommen worden waren. Beim Aufarbeiten wurde eine Mischung aus 3-Methoxy-1 6a- und 3-Methoxy-16,ll'- methyl-17ss-hydroxyw 335't0)-östratrien erhalten.
5 g dieser Mischung wurden über 150 g Silikagel in Petroläther/Benzol (1:1) chromatographiert und ergaben nach dem Eluieren drei Fraktionen. Die dritte Fraktion (1,5) enthielt das reine 3-Methoxy-17,8- hydroxy-16a-methyl-zlt,35(t )-östratrien.
Zu 300 ml flüssigem Ammoniak wurde eine Lösung von 1 g dieser Verbindung in 100 ml Tetrahydrofuran hinzugefügt. Bei -40 C wurden dem Gemisch 3 g Lithium zugefügt und diese dann bei dieser Temperatur noch 3 Stunden weitergerührt, worauf tropfenweise 30 ml absolutes Äthanol zugegeben wurden. Nach Abdampfen des Ammoniak wurde das Gemisch in Wasser ausgegossen und einer Extraktionsbehandlung in 60 ml Methanol unterworfen, worauf 35 ml 3n HC1 zugefügt werden. Das Gemisch wurde nun 20 Minuten unter Rückfluss gehalten, in Wasser ausgegossen und mit Methylenchlorid extrahiert. Nach Umkristallisieren wurde das 43 -Keto-1 6a-methyl- 1 7P-hydroxy-östren erhalten.
Zu einer Lösung von 2,25 g dieser Verbindung in 16 ml Pyridin wurden 3,8 Essigsäureanhydrid zugefügt.
Die Lösung wurde 7 Stunden bei 350 C gerührt, worauf 30 ml Wasser zugegeben wurden. Nun wurde das Gemisch weitere 2 Stunden gerührt und dann nach Zugabe von 100 ml Wasser mit Äther extrahiert. Der Ätherextrakt wurde mit 2n HC1 und ln Natriumhydroxyd gewaschen, über Natriumsulfat vorgetrocknet und dann zur Trockene eingedampft. Das Produkt erwies sich nach Umkristallisieren aus Methanol als das 17-Acetat des #4-3-Keto-16α-methyl-17ss-hydroxy-östrens.
Auf die gleiche Weise wurden die 17-Ester der Trimethylessigsäure, der Capronsäure und der ss-Phenyl- propionsäure hergestellt.
Zu einer Lösung von 1,16 g d4-3-Keto-16a-methyl- 174-hydroxy-östren in 10 ml Pyridin wurden unter Rühren 2 ml Caprinsäurechlorid zugegeben. Das Reaktionsgemisch wurde 15 Stunden bei Raumtemperatur gehalten, worauf 20 ml Wasser zugegeben wurden. Nun wurde die Lösung noch 2 Stunden gerührt und wiederum 100 ml Wasser zugefügt, worauf das Gemisch mit Äther extrahiert wurde. Der Extrakt wurde wie oben, bei der Herstellung des 17-Acetats, weiterbehandelt und ergab das 17-Decanoat des J4-3-Keto-16a- methyl- 1 7fl-hydroxy-östrens.
Auf die gleiche Weise wurden das 17-Onanthat und das 17-Palmitat hergestellt.
Beispiel 2
Zu einer Lösung von 10,3 g zl4-3-Keto-16a-methyl- 17ss-hydroxy-östren in 1 Liter Aceton wurden tropfenweise bei 100 C 10 ml einer 8n CrO3-Lösung zugegeben.
Das Gemisch wurde bei dieser Temperatur 10 Minuten gerührt und dann mit Methylenchlorid extrahiert. Der anfallende Rückstand wurde über 50 g Silikagel in Benzol filtriert. Nach Umkristallisierung des Rückstandes aus einem Gemisch aus Äther und Petroläther erhielt man das #4-3,17-Diketo-16α-methyl-östren.
5 g dieser Verbindung wurden gelöst in 30 ml Dioxan und 6 ml Äthylorthoformiat und eine Lösung von 60 mg p-Toluolsulfonsäure in 6 ml Dioxan und 2 ml absolutem Äthanol zugegeben. Das Gemisch wurde 1 Stunde gerührt, worauf noch 2 ml Pyridin zugefügt wurden. Nun wurde das Gemisch im Vakuum zur Trockene eingedampft und der Rückstand aus Äthanol umkristallisiert. Man erhielt den 3-Enoläthyläther des #4-3,17-Diketo-16α-methyl-östrens.
In Stickstoffatmosphäre bei 0 wurden 5,5 g 3-Enol äther von J4-3,17-Diketo-16a-methyl-östren zu einer Lösung von Allylmagnesiumbromid hinzugefügt, welch letztere hergestellt worden war durch Zugabe von 16 g Magnesium und 300 ml Äther zu einer Lösung von 24 ml Allylbromid in 24 ml Äther. Das Reaktionsgemisch wurde 5 Stunden bei 0 gerührt und dann über Nacht bei Raumtemperatur gehalten. Am nächsten Morgen wurde dem Gemisch 10 %ige Schwefelsäure bei 0 zugefügt, die Ätherschicht abgetrennt, mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum zur Trockene eingedampft.
Der Rückstand wurde einer chromatographischen Behandlung über 300 g Silikagel in Benzol/Äther (4:1) unterworfen und ergab das d4-3-Keto- 16a-methyl- 1 7fl-hydroxy- 1 7o-allyl-östren.
Die Veresterung dieser Verbindung gemäss Beispiel 1 ergab das 17-Butyrat bzw. 17-Caprylat bzw.
17p-Phenylpropionat.
Auf die oben beschriebene Weise wurde der 3-Enol äther des A4-3,17-Diketo-16a-methyl-östrens überführt in die entsprechenden n4-3-Keto- 1 6a - methyl - 17ss - hydroxy-17a-methyl- bzw. -17a-äthyl- oder -17a-butylverbindungen.
Beispiel 3
10 g östron-3-äthyläther wurden gelöst in 200 ml Benzol, worauf bei 300 C 10 ml Diäthyloxalat und 6 g einer 50 siegen Suspension von NaH in Ö1 zugefügt wurden. Das Gemisch wurde 5 Stunden bei 450 gerührt und auf 0 abgekühlt, worauf ihm 15 ml Methanol und 500 ml Wasser zugefügt wurden. Die entstehenden zwei Schichten wurden getrennt und die Wasserschicht mit Petroläther ausgewaschen, mit 2n HC1 angesäuert und weiter extrahiert. Der Rückstand aus dem Extrakt wurde in 300 ml Aceton gelöst, worauf 40 ml Äthyljodid und 12 g Kaliumcarbonat zugefügt wurden. Dann wurde das Gemisch 24 Stunden unter Rühren im Sieden gehalten, abgekühlt und über Hyflos abgesaugt.
Das Filtrat wurde auf ein kleines Volumen eingeengt, in Wasser ausgegossen und das Gemisch mit Methylenchlorid extrahiert. Der Rückstand aus dem Extrakt wurde gelöst in einer Lösung aus 2 g Natrium und 200 ml Äthanol, 24 Stunden bei Raumtemperatur gehalten, in Wasser ausgegossen und das Gemisch mit Methylenchlorid extrahiert. Eine Lösung des Extraktrückstandes in einem Gemisch aus Petroläther und Benzol (2:1) wurde über 110 g Aluminiumoxyd filtriert, worauf aus dem Eluat eine Mischung aus 1 6a-Äthyl- und 16ss-Äthyl-östren-3-äthylenäther erhalten wurde.
Diese Mischung wurde mit Hilfe der in den vorigen Beispielen beschriebenen Methoden über "4-3-Keto-16α- äthyl-17ss-hydroxy-östren in -1 A4-16a-Äthyl-17ss-hydroxy- östren überführt.

Die Veresterung dieser Verbindung nach der in Beispiel 1 beschriebenen Methode führte zu den 1 7-Estern der Essigsäure, der Önanthsäure, der Palmitinsäure und der ss-Phenylpropionsäure.
Durch Oxydation der 1 7ss-Hydroxygruppe mit Chromsäure und nachfolgende Alkylierung wurde die obige Verbindung übergeführt in die entsprechenden 17ss-Hydroxy-17a-äthyl-, 17ss-Hydroxy-17a-allyl- und 17ss-Hydroxy- 7a-äthinyl-verbindungen.
Auf analoge Weise wurden das 24-3-Keto-16a-(l- methyl-propyl)-l 7/S-hydroxy-östren, das z14-3-Keto-16a- (1-äthyl-propyl)- 1 7ss-hydroxy-östren und die entspre chenden 3-Desoxo-Verbindungen hergestellt.
Beispiel 4
Gemäss Beispiel 1 wurde das 3-Methoxy-17-keto 16-isopropyliden-#1,3,5(10)-östratrien mit LiAlH4 zu der entsprechenden 17ss - Hydroxy - Verbindung reduziert.
(F. 183-1850 C; [a]D = +340 [CHC13]), worauf diese Verbindung übergeführt wurde in das d33s5(l0)-3-Meth- oxy- 16a-isopropyl- 17ss - hydroxy - östratrien, und zwar durch Reduktion mit Sauerstoff in Anwesenheit von PtO2. Das erhaltene Produkt war noch verunreinigt mit einer kleinen Menge der entsprechenden 1 6ss-Isopropyl- verbindungen.
Dieses Gemisch aus der 16a- und der 1 6ss-Isopro- pylverbindung wurde auch erhalten durch Behandeln von 3-Methoxy- 1 6ss-Isopropyl- 1 7-keto-A 5,5(10)-östratrien mit KOH in Methanol (oder mit HC1 in Essigsäure) und nachfolgende Reduktion mit LiAlH4 und chromatographische Trennung.
10 g des oben erwähnten Gemisches wurden gelöst in 800 ml Tetrahydrofuran und auf -400 gekühlt, worauf 3 Liter flüssiges Ammoniak und dann langsam 20 Lithium zugegeben wurden. Das Reaktionsgemisch wurde nun 2 Stunden bei M00 gerührt, worauf das überschüssige Lithium mit 200 ml Äthanol entfernt wurde. Nach Abdampfen des Ammoniaks wurde das Gemisch behandelt wie in Beispiel 1, worauf der erhaltene Rückstand in 1 Liter Methanol, dem 350 ml 3n HCl zugefügt worden waren, gelöst wurde. Die Lösung wurde 20 Minuten unter Rückfluss gehalten, in Wasser ausgegossen und das wässrige Gemisch mit Methylenchlorid extrahiert.
Nach Umkristallisieren wurde das #4-3-Keto-16a-isopropyl-17ss-hydroxy-östren (F. 89 bis 910 C; [ajr = +40 [CHCl3]) erhalten.
Die Veresterung dieser Verbindung ergab die 17-Ester der Essigsäure, der Trimethylessigsäure, der ssPhenylpropionsäure bzw. der Caprinsäure.
Beispiel 5
Gemäss Beispiel 2 wurde das A4-3-Keto-16a-iso propyl-17ssöstren in die entsprechenden 1 7Hydroxy- 17a-äthinyl- bzw. 1 7fl-Hydroxy- 1 7a-allylverbindungen übergeführt.

UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man das Gemisch durch Extraktion, Kristallisation und/oder Chromatographie trennt.
2. Verfahren nach Patentanspruch I und Unteranspruch 1, dadurch gekennzeichnet, dass die Isolierung der gewünschten 16a-Alkyl-17ss-Hydroxyverbindung durch chromatographische Trennung über Aluminiumoxyd oder Silikagel erfolgt.
3. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine erhaltene 17-Hydroxyverbindung zur entsprechenden 17-Acyloxyverbindung verestert.
PATENTANSPRUCH II
Verwendung einer nach dem Verfahren gemäss Patentanspruch I erhaltenen 1 7-Hydroxy-Steroidverbin- dung zur Herstellung der entsprechenden 1 7fl-Hydroxy- 17a-alkyl- bzw. -alkenyl- bzw. -alkinyl-verbindung, dadurch gekennzeichnet, dass man die erhaltene 17 Hydroxyverbindung durch Oxydation in die 17-Ketoverbindung umwandelt und diese mit einer Metallverbindung eines Alkans oder Alkens bzw. Alkins mit 1 bis 4 Kohlenstoffatomen umsetzt.

The invention relates to a process for the production of biologically active steroids. Particularly in the Pregnan series, steroids alkylated in the 16-position are known, in which both the 16a and 16ss-alkyl compounds have significant biological effects. Certain representatives of these groups, such as 16a-methyl- and 16fl-methyl-9a-fluoro-prednisolone, have proven to be of great therapeutic value because of their properties as glucocorticoids and anti-inflammatory drugs. Such valuable 16-alkylated compounds have not been found in other series, such as the estran series.
It is particularly surprising that the derivatives alkylated in the 1613 position are significantly less active than the corresponding 16a-alkyl compounds which have anabolic, androgenic, progestative, ovulation-preventing and pregnancy-protecting effects.
The invention relates to a process for the preparation of the mentioned new 16a-alkyl steroids of the estran series of the formula:

The invention further relates to the use of 174-hydroxy-steroids obtained unsubstituted in the 17a-position for the preparation of derivatives thereof substituted in the 17a-position by oxidizing the 17ss-hydroxy group in the process products to the 17-oxo group and this 17-oxo group -Oxo compounds with a metal compound of an alkane, alkene or alkyne having 1 to 4 carbon atoms.
The mixture of the 16a and 16ss-alkyl isomers can be obtained in various ways.
One of these methods consists in reacting oestrone itself or one of its 3-esters or 3-ethers in the presence of a base with a ketone, so that the corresponding 16-alkylidene compound is obtained, which is then converted into a mixture of the corresponding 16a - and 16ss-alkyl compound is converted by first reducing the 17-keto group and then the 16-alkylidene group.
Another method, which is particularly suitable for the preparation of the 16-methyl compounds, is that one oestrone or one of its functional derivatives using a Mannich reaction with formaldehyde and an amine, e.g. B. dimethylamine or morpholine is reacted, whereupon the amino group is split off by steam distillation or by treatment with silica gel and the 16-methylene-17-keto compound thus obtained is reduced to the 16-alkylidene compounds as above.
The 16-alkylidene group can be reduced by treatment with hydrogen in the presence of a metal catalyst such as platinum or palladium.
The 16-alkyl compounds can also be prepared by applying an oxalyl ester condensation to the corresponding 17-ketosteroid and treating the product thus obtained with an alkyl halide.
Finally, 16ss-methyl-oestrone or one of its functional derivatives can also be subjected to isomerization.
The isomer mixture obtained by one of the methods described is now generally separated before the oxidation in order to isolate the desired 16-alkyl isomer. The separation can be carried out using any of the usual methods, e.g. B.
Extraction, crystallization or by chromatography. The last separation method in particular has been found to be effective. The absorbent used is usually aluminum oxide or silica gel and the eluant and others an aromatic hydrocarbon such as benzene, xylene or toluene or an aliphatic ether such as dimethyl ether, dioxane or tetrahydrofuran or a halogenated aliphatic hydrocarbon such as chloroform, or petroleum ether or ethyl acetate or Mixtures of these solvents.
The quantitative ratio between the rl- and the 3-isomer in the mixture depends on how this was prepared, but also on the nature of the 16-alkyl substituent. It has been found that, in the case of chromatographic separation of the 16c and 16ss-alkyl isomers, it is advantageous to first reduce the 17-keto group or to carry out the separation with a 17ss-hydroxy-17a-alkyl derivative.

These compounds are valuable in and of themselves because of their biological properties, but can also be used as starting materials for the production of the corresponding 3-deoxosteroids.
As already mentioned, a 17α-alkyl group, if one is not already present, can be introduced into the end products by oxidation of the 17ss-hydroxy compound obtained and subsequent alkylation.
In the case of a 17α-alkylation of a J4-3,17-diketo-16ct-alkyl compound, it is expedient to initially protect the 3-keto group temporarily, for example with an enol ether or an enol ester.
The alkyl group to be introduced in the 17a position is a saturated or unsaturated aliphatic hydrocarbon radical having 1 to 4 carbon atoms. The following groups may be mentioned as examples: methyl, ethyl, propyl, isopropyl, butyl, vinyl, propenyl, allyl, methallyl, ethynyl, propynyl, propargyl or butynyl.
The alkylation reaction can be carried out in such a way that a metal derivative of a saturated or unsaturated aliphatic hydrocarbon is added to the 17-keto group of the 16α-alkyl estrogen compound in question. The metal derivative can be a magnesium halide, e.g. B. the magnesium bromide, the hydrocarbon in question.
The 17fl-hydroxy-17a-alkynyl compounds can also be prepared by an addition reaction between the corresponding 17-ketosteroid and a triple unsaturated hydrocarbon in the presence of an alkali metal or an alkali compound, such as an alkali metal alcoholate, or by adding a metal compound of a triple unsaturated hydrocarbon to the 1 7-keto group of the starting product; this metal compound can be an alkali or alkaline earth metal compound.
A metal derivative of a saturated hydrocarbon, in particular an alkali derivative such as methyl lithium or ethyl lithium, can also be used to produce a saturated 17α-alkyl compound.
The L-14-3-0xo-16a-alkyl-17P-hydroxy-oestrene compounds thus obtained, which can be alkylated in the 17a-position, can optionally be esterified on the 17ss-hydroxyl group. A saturated or unsaturated organic carboxylic acid having 1-18 carbon atoms is preferably used for ester formation.
The following are examples of suitable acids:
Acetic acid, propionic acid, valeric acid, enanthic acid, caprylic acid, capric acid, lauric acid, decylenic acid, cyclohexylbutyric acid, phenylpropionic acid, palmitic acid, stearic acid, malonic acid and succinic acid.
The esterification can be carried out in the usual way by reacting the 17ss-hydroxy compound with the corresponding acid or one of its derivatives, such as the anhydride or a halide, eg. B. the acid chloride.
In the method described above, for practical reasons, a separation was first carried out between the 16p-alkyl isomer and the desired 16a-alkyl isomer, after which the resulting 16a-alkyl isomer was subjected to the above-mentioned reactions. The separation can of course also be carried out in a different phase of the synthesis, but this is more complicated.

A solution of 10 g of 3-methoxy-17-keto-16-methylene-A18510) -estratn.en in 800 ml of tetrahydrofuran was added to a boiling suspension of 10 g of LiAlH4 in 1 liter of tetrahydrofuran and the reaction mixture was kept boiling for 2 hours. After cooling, 100 ml of water were added and the mixture was filtered off with suction through Hyflo. The residue was washed with chloroform and the filtrate was evaporated to dryness in vacuo, and the residue was recrystallized from acetone / ether. 8.20 g were obtained

5 g of this compound were dissolved in 500 ml of absolute ethanol and 1.2 g of platinum oxide were added. The reaction mixture was then hydrogenated, the reaction being terminated after 0.6 liters of H2 had been absorbed. During work-up, a mixture of 3-methoxy-16a and 3-methoxy-16,11'-methyl-17ss-hydroxy (335't0) estratriene was obtained.
5 g of this mixture were chromatographed over 150 g of silica gel in petroleum ether / benzene (1: 1) and gave three fractions after elution. The third fraction (1.5) contained the pure 3-methoxy-17,8-hydroxy-16a-methyl-zlt, 35 (t) -estratriene.
A solution of 1 g of this compound in 100 ml of tetrahydrofuran was added to 300 ml of liquid ammonia. 3 g of lithium were added to the mixture at -40 ° C. and the mixture was then stirred for a further 3 hours at this temperature, after which 30 ml of absolute ethanol were added dropwise. After the ammonia was evaporated, the mixture was poured into water and subjected to an extraction treatment in 60 ml of methanol, followed by the addition of 35 ml of 3N HCl. The mixture was then refluxed for 20 minutes, poured into water and extracted with methylene chloride. After recrystallization, the 43-keto-16a-methyl-17P-hydroxy-estrogen was obtained.
To a solution of 2.25 g of this compound in 16 ml of pyridine, 3.8 acetic anhydride was added.
The solution was stirred at 350 ° C. for 7 hours, after which 30 ml of water were added. The mixture was then stirred for a further 2 hours and then extracted with ether after adding 100 ml of water. The ether extract was washed with 2N HCl and 1N sodium hydroxide, predried over sodium sulfate and then evaporated to dryness. The product, after recrystallization from methanol, was found to be the 17-acetate of # 4-3-keto-16α-methyl-17ss-hydroxy-oestrene.
The 17-esters of trimethyl acetic acid, caproic acid and ß-phenylpropionic acid were prepared in the same way.
To a solution of 1.16 g of d4-3-keto-16a-methyl-174-hydroxy-oestrene in 10 ml of pyridine, 2 ml of capric acid chloride were added with stirring. The reaction mixture was kept at room temperature for 15 hours, after which 20 ml of water were added. The solution was then stirred for a further 2 hours and 100 ml of water were again added, whereupon the mixture was extracted with ether. The extract was treated as above for the preparation of the 17-acetate, and gave the 17-decanoate of J4-3-keto-16a-methyl-17fl-hydroxy-estren.
The 17-onanthate and the 17-palmitate were prepared in the same way.
Example 2
10 ml of an 8N CrO3 solution were added dropwise at 100 ° C. to a solution of 10.3 g of z14-3-keto-16a-methyl-17ss-hydroxy-oestrene in 1 liter of acetone.
The mixture was stirred at this temperature for 10 minutes and then extracted with methylene chloride. The resulting residue was filtered through 50 g of silica gel in benzene. After recrystallization of the residue from a mixture of ether and petroleum ether, the # 4-3,17-diketo-16α-methyl-oestrene was obtained.
5 g of this compound were dissolved in 30 ml of dioxane and 6 ml of ethyl orthoformate and a solution of 60 mg of p-toluenesulfonic acid in 6 ml of dioxane and 2 ml of absolute ethanol were added. The mixture was stirred for 1 hour after which another 2 ml of pyridine was added. The mixture was then evaporated to dryness in vacuo and the residue was recrystallized from ethanol. The 3-enolethyl ether of # 4-3,17-diketo-16α-methyl-oestrene was obtained.
In a nitrogen atmosphere at 0, 5.5 g of 3-enol ether of J4-3,17-diketo-16a-methyl-oestrene were added to a solution of allyl magnesium bromide, the latter having been prepared by adding 16 g of magnesium and 300 ml of ether a solution of 24 ml of allyl bromide in 24 ml of ether. The reaction mixture was stirred at 0 for 5 hours and then kept at room temperature overnight. The next morning, 10% sulfuric acid was added to the mixture at 0, the ether layer was separated off, washed with water, dried over sodium sulfate and evaporated to dryness in vacuo.
The residue was subjected to chromatographic treatment over 300 g of silica gel in benzene / ether (4: 1) and gave the d4-3-keto-16a-methyl-17fl-hydroxy-17o-allyl-oestrene.
The esterification of this compound according to Example 1 gave 17-butyrate or 17-caprylate or
17p-phenylpropionate.
In the manner described above, the 3-enol ether of A4-3,17-diketo-16a-methyl-oestrene was converted into the corresponding n4-3-keto-16a-methyl-17ss-hydroxy-17a-methyl or -17a-ethyl or -17a-butyl compounds.
Example 3
10 g of estrone-3-ethyl ether were dissolved in 200 ml of benzene, whereupon 10 ml of diethyloxalate and 6 g of a 50% suspension of NaH in oil were added at 300 ° C. The mixture was stirred at 450 for 5 hours and cooled to 0, whereupon 15 ml of methanol and 500 ml of water were added. The resulting two layers were separated and the water layer was washed out with petroleum ether, acidified with 2N HCl and further extracted. The residue from the extract was dissolved in 300 ml of acetone, whereupon 40 ml of ethyl iodide and 12 g of potassium carbonate were added. The mixture was then kept at the boil for 24 hours with stirring, cooled and filtered off with suction through Hyflos.
The filtrate was concentrated to a small volume, poured into water and the mixture extracted with methylene chloride. The residue from the extract was dissolved in a solution of 2 g of sodium and 200 ml of ethanol, kept at room temperature for 24 hours, poured into water and the mixture extracted with methylene chloride. A solution of the extract residue in a mixture of petroleum ether and benzene (2: 1) was filtered through 110 g of aluminum oxide, whereupon a mixture of 16 alpha-ethyl and 16 p-ethyl-estren-3-ethylene ether was obtained from the eluate.
This mixture was converted into -1 A4-16a-ethyl-17ss-hydroxy-oestrene via "4-3-keto-16α-ethyl-17ss-hydroxy-oestrene using the methods described in the previous examples.

The esterification of this compound according to the method described in Example 1 led to the 17-esters of acetic acid, enanthic acid, palmitic acid and β-phenylpropionic acid.
By oxidation of the 17ss-hydroxy group with chromic acid and subsequent alkylation, the above compound was converted into the corresponding 17ss-hydroxy-17a-ethyl, 17ss-hydroxy-17a-allyl and 17ss-hydroxy-7a-ethinyl compounds.
The 24-3-keto-16a- (1-methyl-propyl) -l 7 / S-hydroxy-oestrene, the z14-3-keto-16a- (1-ethyl-propyl) -1 7ss- hydroxy-esters and the corre sponding 3-deoxo compounds produced.
Example 4
According to Example 1, the 3-methoxy-17-keto 16-isopropylidene- # 1,3,5 (10) -estratriene was reduced with LiAlH4 to the corresponding 17ss-hydroxy-compound.
(M.p. 183-1850 C; [a] D = +340 [CHC13]), whereupon this compound was converted into the d33s5 (10) -3-methoxy-16a-isopropyl-17ss-hydroxy-estratriene, namely by reduction with oxygen in the presence of PtO2. The product obtained was still contaminated with a small amount of the corresponding 16 p-isopropyl compounds.
This mixture of the 16a and the 16ss-isopropyl compound was also obtained by treating 3-methoxy-16ss-isopropyl-17-keto-A 5,5 (10) -estratriene with KOH in methanol (or with HC1 in acetic acid) and subsequent reduction with LiAlH4 and chromatographic separation.
10 g of the above-mentioned mixture was dissolved in 800 ml of tetrahydrofuran and cooled to -400, whereupon 3 liters of liquid ammonia and then slowly 20 lithium were added. The reaction mixture was then stirred for 2 hours at M00, after which the excess lithium was removed with 200 ml of ethanol. After evaporating off the ammonia, the mixture was treated as in Example 1, and the residue obtained was dissolved in 1 liter of methanol to which 350 ml of 3N HCl had been added. The solution was refluxed for 20 minutes, poured into water and the aqueous mixture extracted with methylene chloride.
After recrystallization, the # 4-3-keto-16a-isopropyl-17ss-hydroxy-oestrene (mp 89 to 910 C; [ajr = +40 [CHCl3]) was obtained.
The esterification of this compound gave the 17-esters of acetic acid, trimethyl acetic acid, ß-phenylpropionic acid and capric acid.
Example 5
According to Example 2, the A4-3-keto-16a-isopropyl-17ssosten was converted into the corresponding 17-hydroxy-17a-ethinyl or 17fl-hydroxy-17a-allyl compounds.
