4,4,14 - trimethyl -11,20 - diketo-pregnan der Bruttoformel. C2,tH3803 schmilzt. bei 241 bis 217 (unter Zersetzung). Es soll als Zwi- sehenprodukt zur Herstellung von Heilmit teln dienen.
Für die Oxydation des Choladiens verwen- ((ei man insbesondere eine Verbindung des 6-wertigen Chroms, wie Chromsäure, ferner Permanganat., Ozon, Peroxyde, wie Benzoper- säure, Phthalmonopersäure oder Wasserst.off- superoxyd,
vorteilhaft in Gegenwart von Os- miumtetroxyd. Je nach den verwendeten Oxy dationsmitteln wird die Dien-Struktur direkt aufgespalten, oder man erhält primär Ozonicle, Glykole oder Oxyde. Die Ozonide lassen sich hydrolytiseh, reduktiv oder oxyda- tiv aufspalten, die Oxyde durch Hydrolyse in Glykole überführen. Die Spaltung dieser Glykole wird z.
B, mittels Chromsäure, Blei tetraacetat oder Perjodsäure vorgenommen.
Für die Hydrolyse der Estergruppe in 3-Stelling verwendet man z. B. alkalische oder saure Mittel, wie Alkalimetall-hydroxyde, -karbonate oder -bikarbonate, Erdalkalime- tall-hydroxyde oder Mineralsäuren., vorteil haft in wasserhaltigen Lösungsmitteln, wie @rä.ssrigen niedrigen Alkoholen oder wässrigem Dioxan, Tetrahydrofuran oder mit Wasser mischbaren niederen Ketonen, z. B. Aceton.
Der Ausgangsstoff ist in 3:StelIung z. B. mit einer aliphatisehen oder aromatischen Säure, wie mit Essigsäure, Propionsäure oder Benzoesäure, verestert. und trägt in 24-Stel- lung z. B. 2 H-Atome oder zwei Kohlenwas serstoffreste, insbesondere aromatische Reste, wie Phenylgruppen.
Die als Ausgangsstoffe in Betracht fallen den Ester lassen sich aus Derivaten von 27 Kohlenstoffatorne zählenden Säuren, die durch oxydativen Abbau von tetracyclischen Triterpenen um 3 Kohlenstoffatome erhält lich sind, insbesondere ihren Estern, Nitrilen; Halogeniden oder Anhydriden herstellen. Solche Säuren sind z.
B. die den mehrfach ungesättigten tetracyclischen Triterpenen, wie Lanosta-dienol, Agnosterin, Euphol und Tirucallol entsprechenden Säuren mit 27 Kohlenstoffatomen, die in 7,8,9,11-Stellung die Gruppierung

aufweisen. Man gelangt durch folgende Umwandlungen zu den Ausgangsstoffen: Reduktion der 8,9- Doppelbindung, z. B. mittels Zink und Eis essig, Eliminierung der 7-Ketogruppe, z. B.
nach .der Methode von Wolff-Kishner oder über Äthylen=dithioketale und anschliessende Entschwefelung mit Raney-Nickel, schliesslich Überführung der in 3-Stellung veresterten 3 - Ox@# -..4,4,14 - trimetliy l -11-ket.o-eholansäure- derivate in die entsprechenden 'zo,z3- Choladiene.
Zu letzterem Zweck werden die 3-Oxy- 4,4,14-trimethyl-11-keto-cholansäure-derivate mit metallorganischen Verbindungen, wie Nagnesiumverbindungen, insbesondere Aryl- magnesiumhalogeniden, wie Phenylmagne- siumhalogeniden,umgesetzt. Auf diese Weise erhält man tertiäre Carbinole oder Ketone,
welch letztere sich durch weitere Behandlung mit metallorganischen Verbindungen oder mit Wasserstoff in Carbinole überführen lassen. Aus den gebildeten Carbinolen spaltet. man direkt: Wasser ab oder ersetzt die IIy- droxylgruppe zunächst durch Halogen oder eine substituierte Oxygruppe. Anschliessend lässt sich die so umgewandelte Hydroxyl- gruppe beispielsweise durch thermisehe Zer setzung oder durch Behandlung mit Säure bzw. alkoholabspaltenden Mitteln entfernen.
Die Bildung der Doppelbindung kann auch durch Einwirkung der metallorganischen Verbindung bei erhöhter Temperatur direkt erfolgen. Zur Halogenierung des erhaltenen @1ca- Cholens in 22-Stellung wird dieses mit Halo gen, wie Brom, oder ain Stickstoff z. B.
durch Brom oder Chlor halogenierten Säure- imiden oder -amiden, besonders N-halogenier- ten Suceinimiden oder Phtlialimiden, ferner auch X-halogenierten Aeetamiden, Propion- amiden, Diacetamiden, oder kernhalogenierten oder -nitrierten Acetaniliden oder Benz- aniliden umgesetzt.
Die Halogenierung wird vorteilhaft unter Belichten und/oder Zusatz eines Katalysators, z. B. Benzoylperoxyd, in einem indifferenten l.ösnn-smit-tel, wie Tetra ehlorkohlenstoff, oder aliphatischen oder ali- eyelisehen Kohlenwasserstoffen, gegebenen falls unter Erwärmen der Reaktionslösung, durchgeführt-. Zur Belichtung kann künst liches oder starkes natürliches Licht verwen det- werden.
Durch Abspaltung von Halogenwasser stoff durch Erhitzen oder insbesondere durch Behandlung mit basischen -Mitteln, z. B. mit tertiären Aminen, wie Pvridin, Collidin, Chinolin, Dimethylanilin oder Cyelohexyl- amin, gewinnt man das in. 3-Stellung ver- esterte dzo,z3-3-Oxy-4,4,14-trimethyl-11-keto- eholadien.
Im. folgenden Beispiel besteht zwischen Gewielitsteil und Volumteil die Bleiehe Be ziehung wie zwischen. (,'ramm und Kubik zentimeter. Die Temperaturen sind in Celsius graden angegeben. Beispiel:
Eine Lösung von 10,9 CTewielitsteilen ,J 20,z3 - 3 _ Aeetoxy - 4,4,14 - triniethyl -11- keto- '?4,?4-diphenyl-eholadien in einer 'Mischung von 340 Volumteilen. 1Ietlii-lenehlorid, 2-10 Volumteilen,
Eisessig und 48 Volumteilen Wasser wird unter Rühren bei 0 bis 3 wäh rend 1 Stunde mit einer Lösung von 9 Ce- wiclitsteilen Clironiti-ioxyd in 144 Volumteilen 80-proz. Essigsäure versetzt. Das Oxydations- gemisch wird dann ?34 Stunden bei 0 be lassen, dann innerhalb von 30 Minuten. auf 18 erwärmt und dann noch 21h Stunden bei dieser Temperatur stehen gelassen.
Die über schüssige Chromsäure wird dureb vorsichtige Zugabe von Methanol zerstört, das Reaktions gemisch in Wasser eingerührt und der Nie dersehla- in einem (,'emiseh von Äther und Methylenehlorid aufgenommen.

Man erhält das 3 Aeetoxy-4,4,14-trimethyl-11,
20-diketo-preg- nan der Bruttoformel C26H4004 vom F. =232-233 ; [a] D=+109 in Chloroform.
Durch alkalische Verseifung und nach Umkristallisieren aus einer Mischung von Methylenehlorid und Methanol erhält man das unscharf unter Zersetzung bei 241 bis 247 schmelzende, ab 231 leicht sinternde 3-Oxy -4,4,14-trimethyl-11,20-diketo-pregnan der Bruttoformel C24H3S03.
Das als Ausgangsstoff versendete 420.23- 3- Aeetox@--4,4,14 -t.rimethyl -11-keto - 24,24-di- phenyl-eholadien kann folgendermassen her gestellt werden:
8 Gewichtsteile Trisnor-acetoxy-lanostan- dion-säure-methylester werden in 12 Volum- teilen absolutem Chloroform und 8 Volum- teilen Äthylendithioglycol gelöst. und durch die Lösung ein kräftiger Strom von trocke nem Chlorwasserstoffgas geleitet. Nach dem Entfernen der Salzsäure und des Lösungs mittels im Vakuum in der Kälte wird der Rückstand aus Methanol und einer Mischung von Chloroform und Methanol kristallisiert.
Man erhält 8,2 Gewichtsteile des Mono-äthy- lendithioketals der Bruttoformel C32H5o05S2 von) F.206-208 . [a] D= +22 in Chloroform. 1)ureh erneute Behandlung der Alutt.erlaugen mit Äthylendithioglykol und Chlorwasser stoffgas kann eine weitere Menge des Mono- ätliylentlithioketals gewonnen werden.

Nach alkalischer Verseifung erhält man die Trisnor-lanostanolon-säure der Bruttofor mel C27H4404, die nach dem Umlösen aus Essigester bei 155-157 schmilzt. [a]D=+58 in Chloroform. Durch Behandlung mit.
Diazo- methan entsteht der entsprechende Trisnor- lanostanolonsäLire-methylester der Brutto formel C2gH4604 vom F.160-161 ([a] =-I-56 in Chloroform), der aus einer Mischung von Methylenchlorid und Hexan umkristallisiert werden kann.
Durch Acetylierimg der Tris- nor-lanostanolon-säLire mit Pyridin und Ace- tanhydrid gelangt man zur Acetoxy-säure der Bruttoformel C'291-4605 vom F.
171-172 ,[a]D=+53 , Zur Herstellung des Acetoxy-diphenyl- äthen-Derivates (423-3-Acetoxy-4,4,14-trime- thyl-11-keto-24,24-diphenyl-cholen) werden 15 Gewichtsteile Trisnor - acetoxy - lanostanon- säuremethylester 3 Stunden mit 1500 Volum- teilen ln-äthanolischer Kalilauge am Rück fluss erhitzt,
die Verseifungslösung in ver dünnte Schwefelsäure gegossen und die aus gefallene Oxy-säure mit Chloroform extra hiert. Die mit Wasser neutralgewaschene Chloroformlösung wird mit überschüssiger ätherischer Diazomethanlösung versetzt, der nach dem Abdampfen des Lösungsmittels erhaltene,
rohe Oxy-ester in 450 Vollanteilen absolutem Benzol gelöst und die Lösung durch Abdestillieren von 150 Volumteilen des Lösungsmittels vollständig von Wasser be freit. Die noch warme Lösung wird dann innerhalb von 15 Minuten zu einer siedenden Grignard-Lösung, hergestellt aus 15 Gewichts teilen Magnesium, 2ss0 Volumteilen absolutem Äther und 68 Volumteilen Brombenzol, zu fliessen gelassen.
Darnach werden etwa 200 Volumteile Äther abdestilliert, das zurück bleibende Reaktionsgemisch unter Rühren noch 7 Stunden am Rüekfluss weiter gekocht und dann auf ein Gemisch von 800 Gewichts teilen Eis und 45 Gewichtsteilen Ammonium chlorid gegossen.
Das bei der Reaktion gebil dete Diphenyl lässt sich durch Wasserdampf- clestillation entfernen. Die zurückbleibenden, schwerflüchtigen, kristallisierten Reaktions produkte werden abgenutscht, in 400 Volum- tei.len Benzol -elöst, die Lösung filtriert und eingedampft.
Der kristalline Rückstand wird 3 Stunden auf dem Wasserbade mit. 125 Vo- lumteilen Acetanhydrid und 125 Volumtei- len Py ridin acetcliert, danach das überschüs- sige Acetanh@#drid mit Wasser zerstört, das ausgefallene Acetylierungsprodukt in Chloro form aufgenommen und die Chloroform- lösung mit werd.
Salzsäure, ges. Natrium- bicarbonat-Lösung und schliesslich mit Was ser gewaschen. Nach dem Eindampfen des Chloroforms löst man den Rückstand in -):)0 Volumteilen Benzol und erhitzt.

Die noch heisse, tiefgefärbte Lösung wird dann mit 450 Vo- lumteilen Petroläther und wenig Aktivkohle versetzt, weitere 45 Minuten am Rüekfluss gekocht und dann durch eine Säule aus 150 Gewichtsteilen Aluminiumoxyd filtriert.
Nach der Entwieklun- des Chromatogramms mit 1000 Volumteilen Petroläther-Benzol (1:1)-Cemiseh, 250 Volumteilen Benzol. und 350 Volumteilen Benzol-Äther (1:1.)-Gemisch werden insgesamt 16,8 Gewichtsteile schwach gelbe Kristalle gewonnen, die zwischen 160 bis 180 schmelzen.

Zur Reini gung wird das Rohprodukt. an einer Säule von 150 Gewichtsteilen Aluminiumoxyd chromatographiert. Mit 900 Volumteilen Pe- troläther-Benzol (1:
