Verfahren zur Herstellung neuer Steroidverbindungen Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 6-Fluor-17a-oxy- progesteronen der Formel

in welcher R' den Acylrest einer Carbonsäure be deutet, das dadurch gekennzeichnet ist, dass man ein 6-Fluor-5a,17a-dioxypregnan-3,20-dion-17-acylat mit einer Säure oder Base dehydratisiert.
Die Herstellung der Ausgangsmaterialien des erfindungsgemässen Verfahrens erfolgt, indem zu nächst ein 17a-Oxypregnenolon-3,17-diacylat in Lö sung in einem inerten organischen Lösungsmittel mit einer organischen Persäure umgesetzt wird. Die Re aktion wird im allgemeinen bei niedrigen Tempera turen, z. B. bei -10 bis 4-1011C durchgeführt. Es können jedoch auch höhere oder niedere Tempe raturen verwendet werden, wie z. B. -30 bis + 40 C.
Die Persäure wird üblicherweise in einer Menge von 10-20% über der fünffachen theoretisch erfor- derlichen Menge verwendet.
Nach beendigter Reak tion wird das Gemisch neutralisiert, mit Wässer ge waschen und das derart erhaltene Gemisch von 5a,6a-Oxydo-3ss,17a-dioxypregnan 20-on-3,17-di- acylat sowie das entsprechende 5ss,6ss-Epimere durch Verdampfen des Lösungsmittels gewonnen. Bei Ver wendung von Peressigsäure zur Bildung der 5,6- OxydoVerbindung wird dem Reaktionsgemisch vor zugsweise ein trockenes Alkaliacylat, beispielsweise Natriumacetat, zugesetzt:
Durch Chromatographieren des Gemisches und nachfolgende Umkristallisation erhält man 5a,6a- Oxydo-3ss,17a-dioxypregnan-20-on-3,17-diacylat und sein Epimer 5ss,6ss-Oxydo-3ss,17a-dioxypregnan-20- on-3,17-diacylat.
Das derart erhaltene 5a,6a-Oxydo-3ss,17a-dioxy- pregnan-20-on-3,17-diacylat, in Lösung in einem ge eigneten organischen Lösungsmittel, wird mit Fluor wasserstoff zum 6ss-Fluor-3ss,5a,17a-trioxypregnan- 20-on-3,17-diacylat umgesetzt. Der Fluorwasserstoff kann gasförmig oder in wässriger Lösung von bei spielsweise 4811/o oder in Form eines bei Behandlung mit einer Säure Fluorwasserstoff abgebenden Metall- fluorids verwendet werden.
Die Reaktion kann zwischen etwa -70 und + 50 C durchgeführt werden. Bei Verwendung von wasserfreiem Fluorwasserstoff werden üblicherweise tiefere Temperaturen, z. B. zwischen -70 und + 10 C bevorzugt. Bei Verwendung von wässrigem Fluor wasserstoff kann die Reaktion zwischen 0 C und Raumtemperatur durchgeführt werden. Ebenso kön nen bei der Herstellung des Fluorwasserstoffes in situ durch Reaktion eines Metallfluörids, wie Ka- liumbifluorid, und einer Säure, Reaktionstempera turen zwischen 0 C und Raumtemperatur oder darüber und bis zu 90 C angewendet werden.
Bei tiefen Temperaturen werden Lösungsmittel wie Chloroform, Methylenchlorid und insbesondere Tetra- hydrofuran verwendet. Bei der Umsetzung des Epoxyds mit Kaliumbifluorid und einer Säure wer den organische Säuren bevorzugt, z. B. Essigsäure, Propionsäure, Ameisensäure oder dergleichen. Es können jedoch auch andere Lösungsmittel, z. B. Neopentylalkohol, Isopropanol und dergleichen, mit Mineralsäuren wie Schwefelsäure, Perchlorsäure oder dergleichen verwendet werden.
Die Reaktionsdauer beträgt üblicherweise zwi schen 15 Minuten und 4 Stunden, wobei für wasser freien Fluorwasserstoff 2-3 Stunden üblicherweise genügen. Bei Verwendung von Kaliumbifluorid werden Reaktionszeiten von 12 Stunden bis 5 Tagen ver wendet. Nach Beendigung der Reaktion wird das Produkt auf bekannte Weise .isoliert, z. B. durch Neutralisieren des überschüssigen Fluorwasserstoffes mit einer Base und Extraktion des Produktes mit einem mit Wasser nicht mischbaren Lösungsmittel.
Durch Verdampfen der Lösungsmittel erhält man das rohe Material, welches durch Umkristallisation aus organischen Lösungsmitteln gereinigt wird und reines 6ss -Fluor - 3ss,5a, 7,u - trioxypregnan-20-on-3,17-di- acylat ergibt.
Das rohe oder gereinigte 6ss-Fluor-3#,5a,17a-tri- oxypregnan-20-on-3,17-diacylat wird sodann in einem mit Wasser mischbaren Lösungsmittel, vor zugsweise in einem wässrigen oder Aceton-Säure- Medium hydrolysiert. Als Lösungsmittel eignen sich besonders Alkanole, wobei Methanol und Äthanol bevorzugt werden;
doch lassen sich . auch andere geeignete, mit Wasser mischbare Lösungsmittel ver wenden, wie beispielsweise tert. Butylalkohol, Pro pylalkohol, Isopropylalkohol, Dioxan, Aceton, Essig säure oder dergleichen.
Die Hydrolyse des 6ss-Fluor-3,B,5a,17a-trioxy- pregnan - 20 - an - 3,17 - diacylats ergibt 6ss-Fluor- 3ss,5a,17.a-trioxypregnan-20-on=17-acylat; die nach folgende Oxydation dieser Verbindung mit Natrium- bichromat in Essigsäure führt zu 6ss-Fluor-5a,17 , dioxypregnan-3,20-dion-17-acylat. Das rohe Produkt kann auf bekannte Weise gereinigt werden, z. B.
durch Umkristallisieren aus organischen Lösungsmit teln, wie Aceton, Äthylacetat, Skellysolve B-Hexanen, Methanol, tert. Butylalkohol, Äther oder dergleichen oder Gemischen solcher Verbindungen; man erhält auf diese Weise reines, als Ausgangsmaterial für vorliegendes Verfahren geeignetes 6ssFluor-5a,17a- dioxypregnan-3,20-dion-17-acylat.
Das derart erhaltene 6f Fluor-5a,17a-dioxy- pregnan-3,20-dion-17-acylat wird daraufhin chemisch dehydratisiert. Die Dehydratisierung kann in alka lischer oder saurer Lösung erfolgen. In der bevor zugten Ausführungsform der Erfindung wird die saure Dehydratisierung gewählt.
Das Ausgangsmate rial wird in Lösungsmitteln, die mit der verwendeten Säure nicht reagieren, wie Methylenchlorid, Chloro form, Dioxan, Tetrachlorkohlenstoff und dergleichen gelöst oder suspendiert und die gewünschte Säure die ser Lösung oder Suspension zugesetzt. Für diese Reak tion besonders geeignete Säuren sind starke Säuren wie gasförmiger Chlorwasserstoff oder Bromwasser stoff, Schwefelsäure und - dergleichen, wobei gas- förmiger Chlorwasserstoff bevorzugt wird.
Zur De- hydratisierung mittels Alkali wird das Ausgangs material in Methanol, Äthanol, Dioxan oder einem andern geeigneten, mit der verwendeten Base nicht reagierenden Lösungsmittel gelöst. Die Lösung wird durch Einleiten von Stickstoff vom Sauerstoff befreit und anschliessend mit einer sauerstofffreien Alkalilauge umgesetzt.
Natrium- oder Kaliumhydroxyd sind die bevorzugten Laugen, doch können auch Alkalialko- holate, Bariumhydroxyd, Calciumhydroxyd oder der gleichen verwendet werden.
Je nach der verwendeten Menge der Säure oder Base erhält man die 6a- und 6f Isomeren. Die De- hydratisierung führt zunächst zum 6f-Isomeren, das sich, da es in starken Säuren oder Basen weniger stabil ist, in das 6a-Isomere umlagert.
Ist das Me dium zu Beginn der Dehydratisierung stark sauer, so erhält man nur 6a-Fluor-17a-oxyprogesteron-17- acylat. Ein schwach saures oder alkalisches Medium ergibt 6ss-Fluor-17a-oxyprogesteron-17-acylat, wäh rend ein stark basisches Medium den freien Alkohol als a-Epimeres, das heisst 6a@-Fluor-17a-oxyproge- steron, ergibt. Das 6f-Isomere kann nachträglich in das 6a-Isomere umgewandelt werden durch Behand lung mit einer starken Säure oder Base.
Die derart erhaltenen 6-Fluor-17a-oxyproge- steron-17-acylate oder der freie Alkohol 6-Fluor- 17a-oxyprogesteron, der gebildet wird, wenn eine konzentrierte Lösung der starken Lauge für die De- hydratisierung verwendet wird, kann aus dem Re aktionsgemisch isoliert und nach üblichen Methoden gereinigt werden. Solche Methoden sind z. B. das Verdünnen mit Wasser und entweder Filtrieren oder Extraktion der Mischung mit einem mit Wasser nicht mischbaren Lösungsmittel, z.
B. Methylenchlorid, Chloroform, Hexanen, Benzol, Äther und dergleichen und anschliessendes Verdampfen des Lösungsmittels. Die derart erhaltenen Feststoffe können auf übliche Art weiter gereinigt werden, z.
B. durch Umkristalli- sieren aus einem organischen Lösungsmittel, wie Me thanol, Äthanol, Skellysolve B-Hexanen, Äthylacetat, Benzol oder dergleichen, um das reine 6-Fluor-17a- oxyprogesteron oder dessen 17-Acylat zu erhalten.
In einer gesonderten Verseifungsstufe lässt sich 6-Fluor-17a-oxyprogesteron aus einem 6-Fluor-17a- oxyprogesteron-17-acylat herstellen.
Zu diesem Zwecke lässt man 6-Fluor-17a-progesteron-17-acylat mit einem Alkalihydroxyd wie Natrium- oder Kalium hydroxyd oder einem Karbonat wie Natrium- oder Kaliumkarbonat, Bikarbonat oder dergleichen oder einem Erdalkalimetallhydroxyd wie Barium- oder Calciumhydroxyd in alkoholischer Lösung bei Raum temperatur während 1-24 Stunden reagieren, um 6-Fluor-17a oxyprogesterön zu erhalten.
Um das Produkt aus dem Reaktionsgemisch zu isolieren, wird das Reaktionsgemisch zuerst mit wässriger Säure wie Essigsäure, Salzsäure oder Schwefelsäure neutra lisiert und anschliessend mit Wasser verdünnt und mit einem mit Wasser nicht mischbaren organischen Lösungsmittel, wie Methylenchlorid, Chloroform, Benzol, Äther oder dergleichen, extrahiert und die Extrakte getrocknet und eingedampft. Auf diese Weise erhält man das rohe Material.
Dieses rohe Material kann sodann aus organischen Lösungsmit teln wie Methanol, Athanol, Aceton, Skellysolve B- Hexanen, Äthylacetat, Methylenchlorid oder der gleichen umkristallisiert werden und ergibt reines 6-Fluor-17a-oxyprogesteron. Aus dem derart erhal tenen 6-Fluor-17a-oxyprogesteron können wiederum Ester hergestellt werden,
indem man nach bekann ten Methoden zur Veresterung tertiärer Oxygruppen in der 17a-Stellung eines Steroids der Pregnanreihe verestert. So beschreiben beispielsweise Huang- Minlon et a1., J. Am. Chem. Soc. 74, 5394 (l952) sowohl eine kalte Methode wie eine heisse Me thode , welche beide gleichermassen zur Veresterung von 6a-Fluor-17a-oxyprogesteron geeignet sind.
Sind bei der Veresterung des 6ss-Isomeren wiederum die 6ss-Isomeren erwünscht, so sind milde Bedingungen erforderlich (kalte Methode, geringe Menge saurer Katalysator), da sonst das 6g3 Isomere in das 6a-Iso- mere übergeführt wird.
Die Herstellung der Ausgangsverbindungen für das erfindungsgemässe Verfahren ist in den nachfol genden Präparationen erläutert: Präparation <I>1 A:</I> 17a-Oxypregnenolon-3,17-diacetat. 10 g p-Toluolsulfonsäure wurde innerhalb 10 Minuten (unter Rühren in Stickstoffatmosphäre) einem Gemisch von 25 g 17a-Oxypregnenolon in 400 ml Essigsäure und 100 ml Essigsäureanhydrid zugesetzt. Die Lösung wurde nach etwa 30 Minuten klar. Nach zwei weiteren Stunden wurde das Reak tionsgemisch in Eiswasser gegossen und der gebildete Niederschlag gesammelt, gut mit Wasser gewaschen und getrocknet.
Man erhielt 30 g eines bei 166 bis 178" C schmelzenden Produktes. Die Umkristallisie- rung aus einem Gemisch von Aceton und Skellysolve B-Hexanen ergab 24,2 g reines 17u Oxypregnenolon- 3,17-diacetat mit einem Schmelzpunkt von 179 bis 181 C und einer Drehung [a]L, von -65 in Chloro form.
Auf dieselbe Art können 17a-Oxypregnenolon-3,17-dicaproat, 17a-Oxypregnenolon-3,17-diphenylacetat, 17a-Oxypregnenolon-3,17-di(ss-cyclopentyl- propionat), 17a-Oxypregnenolon-3,17-diacrylat, 17a-Oxypregnenolon-3,17-divalerat, 17a-Oxypregnenolon-3,17-ditrimethylacetat, 17a-Oxypregnenolon-3,17-di-t-butylacetat, 17a-Oxypregnenolon-3,17-dicyclopentyl- carboxylat, 17a-Oxypregnenolon-3,17-diäthylbutyrat, 17a-Oxypregnenolon-3,17-dicyclohexylacetat,
17a-Oxypregnenolon-3,17-di-o-toluat, 17a-Oxypregnenolon-3,17-dimonoglutarat, 17a-Oxypregnenolon-3,17=dimonodiglycolat, 17a-Oxypregnenolon-3,17-dimono-@-methyl- glutarät, 17a-Oxypregnenolon-3,17-dimono-ss,ss-dimethyl- glutarat, 17a-Oxypregnenolon-3,17-diäthoxyacetat, 17a-Oxypregnenolon-3,17-dilaurat, 17a Oxypregnenolon-3,17-dibutyrat, 17a-Oxypregnenolon-3,17-dipropionat, 17a-Oxypregnenolon-3,17-diisovalerat,
17a-Oxypregnenolon-3,17-diönanthat und 17a-Oxypregnenolon-3,17-dicaprylat hergestellt werden, indem man 17a-Oxypregnenolon in einer Lösung, welche ein Gemisch der entspre chenden Säure und ihres Anhydrids enthält, zusam men mit p-Toluolsulfonsäure auflöst. Der rohe Di- ester wird wie oben beschrieben isoliert.
Falls die entsprechende Säure oder ihr Anhydric? fest sind, kann ein inertes Lösungsmittel wie Ben,ol, Chloro form oder Dioxan verwendet werden, um die Lö sung zu bewerkstelligen und ein flüssiges Vereste- rungsmedium zu ergeben. Präparation <I>1 B:</I> 17a-Oxypregnenolon-17-acetat. 0,4 g 17a-Oxypregnenolon-3,17-diacetat in 15 ml Methanol und 0,13 ml konzentrierter Salzsäure wurde während 1 Stunde am Rückfluss gekocht.
Dann wurde etwa die Hälfte des Methanols unter Einwirkung eines Stickstoffstroms abdestilliert; 0,35 g des ProduAtes wurde mit Wasser ausgelaugt, gesam melt, getrocknet und aus Aceton umkristallisiert. Es ergab 17a - Oxypregnenolon - 17 - acetat mit einem Schmelzpunkt von 224-228 C und einer Drehung [a]D von -69 in Chloroform.

Eine Lösung von 20 g 17a-Oxypregnenolon- 3,17-diacetat und 2,5 g Natriumacetat in 250 ml Chloroform wurde auf 5 C gekühlt und unter Rühren mit 25 ml 40 % iger Peressigsäure versetzt. Nach 3 Stunden wurde die Lösung zuerst mit Wasser,
dann mit verdünnter Natronlauge und noch mals mit Wasser gewaschen, getrocknet und vom Lösungsmittel befreit. Der Rückstand liess sich nicht kristallisieren und wurde über eine 400 g Florisil (ein synthetisches Magnesiumsilikat) aufweisende Kolonne chromatographiert. Das ss-Oxyd wurde mit Skellysolve B plus 6 % Aceton eluiert und aus Me- thanol umkristallisiert.
Man erhält 3,7 g 5ss,6ss- Oxydo-3ss,17a-dioxypregnan-20-on-3,17-diacetat, das einen Schmelzpunkt von 168-17311C und eine Drehung [ ]D von -45 in Chloroform aufwies.

Präparation <I>3:</I> 5,6-Oxydo-3,8,17a-dioxypregnan 20-on-17-acetat. Eine Lösung von 150 g rohes 17a-Oxypregnen- olon-17-acetat (erhalten gemäss Präparation 1 A) in etwa 6 Liter Benzol wurde auf 15 C gekühlt und unter Rühren 17g wasserfreies Natriumacetat zu gesetzt,
gefolgt von 170 ml 40o/oiger Peressigsäure. Die Temperatur der Reaktionsteilnehmer wurde wäh rend 2i/4 Stunden auf 15 C gehalten und die Lö sung anschliessend zweimal mit Wasser unter leichtem Schwenken gewaschen. Starkes Schütteln während des dritten Waschens bewirkte eine Anfällung des a-Oxydes (II). Es wurde gesammelt, mit Wasser und mit Benzol gewaschen und getrocknet und ergab 95 g eines bei 248-249 C schmelzenden Produktes.
Die Umkristallisation aus Aceton führte zu reinem 5a,6a-Oxydo-3ss,17a-dioxypregnan 20-on-17-acetat mit einem Schmelzpunkt von 251-252 C und einer Drehung [ ]D von<B>-710</B> in Chloroform.

10 ml gekühltes Methylenchlorid wurde zu 1,5 g Fluorwasserstoff, der in einem Trockeneis-Acetonbad gekühlt worden war, zugesetzt. 4,3 g 5a,6a-Oxydo- 3,8,17ä <I>-</I> dioxypregnan - 20 - on- 3,17 - diacetat (II)
in 20 ml gekühltes Methylenchlorid wurden dem Fluor- wasserstoff-Methylenchlorid zugesetzt. Die entstan dene Lösung wurde während 4 Stunden bei Raum temperatur stehengelassen und alsdann mit 2 ml Pyridin versetzt. Das Methylenehlorid wurde ent fernt und Wasser zugesetzt. Das gummiartige un lösliche Material wurde abgetrennt und gut mit Äther gewaschen, worauf saubere Kristalle erhalten wurden.
Das Produkt wurde aus Aceton umkristalli siert und ergibt 1,2 g 6ss-Fluor-3ss,5a,17a-trioxy- pregnan-20-on-3,17-diacetat mit einem Schmelzpunkt von 253 C und einer optischen Drehung [ab von -33 in Chloroform.

39 ml vorgekühltes Chloroform und 98 ml vor gekühltes Tetrahydrofuran wurden unter Schwenken zu 53g Fluorwasserstoff in einer Polyäthylenflasche, die in einem Trockeneis-Acetonbad gekühlt wurde, zugesetzt. Diese Lösung wurde einer, in einem Trockeneis-Acetonbad gekühlten Lösung von 45 g 5a,6a-Oxydo <I>-</I> 3ss,17 <I>-</I> dioxypregnan-20-on-17-acetat (1I) in 200 ml Chloroform zugesetzt.
Nach Auf bewahrung während 21/4 Stunden bei 15 C wurde das Reaktionsgemisch in 1 Liter Eiswasser, das <B>100</B> ml Pyridin enthielt, gegossen. Die obere wässrige Schicht wurde nun zweimal mit Methylenchlorid extrahiert. Diese Extrakte wurden mit der untern Chloroformschicht vereint und mit verdünnter Salz säure, Wasser, verdünntem Natriumkarbonat und wieder mit Wasser gewaschen. Jede Wasserphase wurde mit derselben Methylenchloridlösung zurück extrahiert, und die vereinten Extrakte wurden über Magnesiumsulfat getrocknet, filtriert und zur Trockne verdampft.
Der Rückstand war ein glasartiger Fest Stoff. Präparation <I>7:</I> 6ss-Fluor-5a,17a-dioxypregnan-3,20-dion- 17-acetat. 40 g 6ss Fluor-3ss,5a,17a-trioxypregnan-20-on- 17-acetat wurden in 500 ml Essigsäure gelöst und dieser Steroidlösung sofort 45g Natriumbichromat- Dihydrat in 450 ml Essigsäure unter Schwenken und Kühlen in einem Wasserbad während etwa 5 Minuten zugesetzt.
Das Reaktionsgemisch wurde bei Raum temperatur während etwa 11/2 Stunden stehengelas sen. Dann wurde die Lösung auf 15 C gekühlt und das Produkt abfiltriert und zweimal mit kleinen Portionen Methanol gewaschen. Nach dem Trocknen wog das Produkt 34,7 g und wies einen Schmelz punkt von 246-250 C auf. Die- Umkristallisierung aus siedendem Methanol ergab 6ss-Fluor-5a,17a- dioxypregnan-3,20-dion-17-acetat mit einem Schmelz punkt von 265-267,5 C.
<I>Beispiel</I> 6-Fluor-17a-oxyprogesteron-17-acetat Eine Aufschlämmung von 10 g 6ssi-Fluor-5a,17,c- dioxypregnan 3,20-dion-17-acetat in 100 ml Chloro form und 1 ml absolutem Äthanol wurde auf -5 C in einem Eis-Kochsalz-Bad gekühlt und sodann mit Chlorwasserstoff gesättigt. Nach 4 Stunden wurde Stickstoff durch die Reaktionsmischung geleitet und anschliessend zweimal mit Wasser, einmal mit Na triumbikarbonat und wiederum mit Wasser gewa schen.
Nach dem Trocknen und' Filtrieren wurde das Lösungsmittel entfernt und-der Rückstand aus einem Gemisch von Aceton und Skellysolve B-Hexanen umkristallisiert. Eine Ausbeute von 6,1 g 6a-Fluor- 17a-oxyprogesteron-17-acetat mit einem Schmelz- punkt von 252-256 C (Zers.) wurde erhalten. Die Umkristallisation einer zweiten Portion des Pro duktes ergab 0,7 g vom Schmelzpunkt 248-255 C.
Behandelt man im obigen Verfahren andere 6ss-Fluor- 5a,17a-dioxypregnan-3,20-dion-17-acylate in Chloro- formlösung mit Chlorwasserstoff, so erhält man die entsprechenden 6a -Fluor -17a - oxyprogesteron-17- acylate.
Das erhaltene Steroid kann folgendermassen zum 6a-Fluor-17a-oxyprogesteron verseift werden: Eine Lösung von 1,9 g 6a-Fluor-17a-oxyproge- steron-17-acetat in 50 ml 90 %. igem Methanol plus l "/u Natronlauge und 20 ml Methylenchlorid wurde mit Stickstoff gespült.
Nach dem Stehen über Nacht bei Raumtemperatur wurde die Lösung mit Methy- lenchlorid verdünnt und dreimal mit Wasser gewa schen. Die wässrigen Schichten wurden mit Methy- lenchlorid zurückextrahiert. Die Extrakte wurden vereint, getrocknet und auf etwa 20 ml konzentriert und über eine Kolonne von 150 g Florisil (synthe tisches Magnesiumsilikat)
chromatographiert. Nach dem Waschen der Kolonne mit zunehmenden Men gen Aceton in Skellysolve B wurde das Produkt mit 2-80/e Aceton in 1 : 1-Gemischen von Skellysolve B und Methylenchlorid eluiert. The steroid obtained can be saponified to give 6a-fluoro-17a-oxyprogesterone as follows: A solution of 1.9 g of 6a-fluoro-17a-oxyprogesterone-17-acetate in 50 ml of 90%. Strong methanol plus 1 "/ u sodium hydroxide solution and 20 ml methylene chloride were flushed with nitrogen.
