Verfahren zur Herstellung von 1, 4-Diketopolyhydrophenanthrenen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 1, 4-Diketo- polyhydrophenanthrenen, das dadurch gekenn- zeichnet ist, dass entsprechende 1, 4-Dioxy- polyhydrophenathrene oxydiert werden. Das Verfahren kommt vor allem für die Herstel- lung des 4b-Methyl-1, 2, 3, 4, 4a,, 4b, ! 5,. 6, 7, 9, 10, 10adodecahydrophenathren-1,4, 7-trions und seiner Derivate in Betracvht.

8 9 und seine Derivate sind wertvolle Zwischenprodukte für die Synthese von Steroidverbin- dungen und anderer Polyhydrophenanthrene.
Sie besitzen die den Ringen A, B undi C der Steroidverbindungen entsprechenden, kondensierten Kohlenstoffringe und eine anguläre Methylgruppe in 4b-Stellung, die der in zahlreichen Sterinen vorhandenen anguläre Me thylgmvxppe in 10-Stellung entspricht.
Für die Herstellung der genannten Triketo- verbindungen wird am besten 4b-Methyl 1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahydrophenan- thren-1, 4-diol-7-on (I) als Ausgangsstoff verwendet, wobei eine der steroisomeren Formen des 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5,, 6,7,9,10,10a-dode cahydrophenanthren-1, 4, 7-trions (XI) durch die folgenden Reaktionen erhalten wird :

R bedeutet einen Substituenen, der sich durch Hydrolyse in die Ketogruppe umwandeln lässt.
Bei diesem Vorgehen wird die 7-Keto- gruppe der als Ausgangsmaterial dienenden Verbindung I zuerst durch Bildvng eines geeigneten Derivates blockiert, zum Beispiel eines Enoläthers oder vorzugsweise eines Ketals, welches sich leicht zum Ausgangsmaterial hydrolysieren lässt. Dieses Derivat der Verbindung I wird dann zur Umwandlung der Hydroxygruppen in Ketogruppen oxydiert, worauf das oxydierte Produkt zur Verbindung XI hydrolysiert wird. Bei der Herstellung des Ketalderivates verschiebt sich die Doppelbindung von der 8, 8a-Stellung nach 8a, 9. Bei der Hydrolyse des Ketalderivates findet eine Rückverschiebung dler Doppelbindung in die 8, 8a-Stellung statt.
Bei der bevorzugten Ausführungsform dieses Verfahrens wird die Verbindung I zuerst in das entsprechende 7-Äthylendioxyderivat verwandelt, aus welchem durch Oxydation und anschliessende Hydrolyse die Verbindung XI entsteht. Diese Reaktionsreihe lässt sich wie folgt darstellen :

Das 4b-Met. hyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 1% 10a-dodecahydrophenanthren-1, 4- diol (VIII) kann leicht erhalten werden, indem man die Verbindung I mit Glykol in Gegenwart eines sauren Katalysators, wie p- Toluolsulfonsäure, erhitzt.
Diese Reaktion wird am besten in einem inerten Losungsmit- tel, zum Beispiel inÄthylendichlorid,durch- geführt. Das geschieht etwa durci Erhitzen einer Mischung von Verbindung I, Glykol, Äthylendichlorid und p-'Toluolsulfonsäure und Abdestillieren des gebildeten Wassers als azeotrope Mischung mit dem Äthylendiehlorid.
Nach beendeter Beaktion wird das Äthylen- dioxyderivat, die Verbindung VIII, durch Eindampfen der Lösung und Umkristallisie- ren des Produktes aus einem geeigneten Losungsmittel, wie Aeeton,gewonnen.
Die Verbindung VIII wird dann mit einem Oxydationsmittel behandelt, um die Hydr oxygruppen zuKetogruppen zu oxydieren. Die Oxydation kann zum Beispiel mit einem Me tallalkoholat in'Gegenwart eines Ketons geschehen. Es zeigtesieh,dass diese Rea. ktion am besten in Gegenwart eines inerten, orga- nischen Losungsmittels (unter andern Benzol, Toluol, Dioxan) vor sich geht. Zur Durchführung der Reaktion wird am besten wie folgt vorgegangen : Die Verbindung VIII wird in einer Mischung von organischem Losungs- mittel und einem Keton gelöst, das Metallalkoholat zugefügt und die Mischung erwärmt.
Als Ketone eignen sich dabei unter andern Aceton, Methyläthylketon, Cyclohexanon, Benzil und p-Benzochinon. Am besten verläuft die Reaktion bei der Verwendung von Cyclohexanon als Keton, Benzol als Lösungsmittel und Aluminiumisopropylat als Metallalkoholat. Nach beendeter Reaktion wird der Re aktionsmischung Wasser zugesetzt, das aus geschiedene Metallhydroxyd abfiltriert, das Filtrat zur Trockne eingedampft, der Rückstand mit PetToläther ausgezogen und die Verbindung X aus dieser Lösung gewonnen.
Die Oxydation der Verbindung VIII kann auch mit Pyridin-Chromtrioxyd-Komplex in Pyridin erfolgen. In diesem Fall wird nach beendeter Oxydation die Verbindung X gewonnen, indem die Reaktionsmischung mit Wasser verdünnt und hierauf mit einer Benzol-Jither-Mischung ausgeschiittelt wird.
Die nicht wässerige Schicht wird eingedampft, der erhaltene Rückstand an alkalischem Alu- miniumoxyd chromatographiert und das Adsorbat mit Benzol eluiert.
Bei der Oxydation der Verbindung VIII nach dem oben angegebenen Verfahren entstehen ausser Verbindung X auch die unvoll- ständig oxydierten Verbindungen 4b-Methyl- 7-äthylendixoy -1, 2, 3, 4, 41,4b,5, 6, 7, 8, 10, 10a-do decahydrophenanthren-l-on-4-ol (IX) und 4b Methyl-7-äthylendioxy-1,2,3,4,4a,4b,5,6, 7, 8, 10, l0a-dodecahydrophenanthren-1-ol-4-on (XVII) mit den Strukturformeln

Diese Verbindungen können nach dem für die Oxydation von Verbindung VIII beschriebenen Verfahren weiter oxydiert werden. Sie können aber auch zuerst mit Säure hydTolysiert werden, um die 7-thylendioxygruppe in eine Ketogruppe zu verwandeln, worauf die erhaltene Diketoverbindung zur Verbindung XI oxydiert wird.
So kann die Verbindung XVII nach dem folgenden Verfahren in die Verbindung XI umgewandelt werden :

Dazu wird zum Beispiel diie Verbindung XVII zuerst durch Erwärmen mit Säure zu 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5,6,7,8,10, 10a-dodeca hydrophenanthren-1-ol-4, 7-dion (XXI) hydrolysiert. Dieses wird mit Chromtrioxyd zum 1, 4, 7-Trion (XXII) oxydiert, welches ein Stereo, isomer der Verbindung XI mit einem Schmelzpunkt von etwa o1164 bis 166 C darstellt. Durch Erhitzen mit einer Base, etwa mit alkalischem Aluniniumaxyd, wird daraus die stereoisomere Verbindung XI mit einem Schmelzpunkt von 117 bis 117, 5 C gebil- det.
Bei der oben beschriebenen Oxydation der Verbindung VIH mit einem Metallalkoholat in Gegenwart eines Ketons findet eine Inversion der B/C-Ring-Bindung unter Bildung der Verbindungen X und XI statt, welche eine andere B/C-Ring-Bindung aufweisen als Verbindung VIII. Die Verbindung IX kann nach dem folgenden Verfahren in die Verbindung XI umgewandelt werden :

Beispielsweise wird die Verbindung IX in Pyridin mit Pyridin-Chromtrioxydkomplex behandelt und die entstehende Verbindung X durch saure Hydrolyse in die Verbindung XI umgewandelt.
Eine stereoisomere Modifikation der Verbindung X, das 4b-Methyl-7-athylendioxy- 1, 2, 3, 4, 4a, 4b, 5,6,7,8,10,10a - dodecahydropehnanthren-1, 4-dion XX kann wie folgt erhalten werden :

Dazu wird beispielsweise die Verbindung VIII zuerst selektiv aeyliert, zum Beispiel durch Reaktion ihrer Lösug in Pyridin mit Essigsäureanhydrid, wobei das 4b-Met. hyl-7- ähylendioxy-1,2,3,4,41,4b,5,6, 7, 8, 10, 10a - dode eahydrophenanthrenl, 4-diol-1-aeetat entsteht (XV).
Dieses wird mit einem Oxydationsmittel, etwa Pyridin-Chromtrioxyd, behandelt, wodurch das 4b-Methyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren I-ol-4-on-l-acetat (XVI) entsteht, das sich durch Hydrolyse mit einem Alkali, wie Kaliumearbonat, leicht in die entsprechende 1-Oxy-Ver- bindung überiühren lässt.
Es zeigte sich, dass bei dieser Hydrolyse der Verbindung XVI entweder die Verbindung XVII oder eine stereoisomere Verbindung XIX erhalten wird, je nach der Wahl der Reaktionsbedingungen. In mässig basisehem Milieu, unter Verwendung verhältnis- mässig geringer Kaliumcarbonatmengen entsteht Verbindung XVII, bei der Verwendung grösserer Waliumearbonatmengen und/oder verlängerter Reaktionsdauer die stereoisomere Verbindung XIX. Diese Verbindung XIX besitzt anscheinend eine B/C-Ringkonfiguration, die von derjenigen der Verbindung XVII abweicht.
Wenn die Verbindung XIX zum Beispiel durch Behandeln mit Pyridin-Chromitrozxydkomplex weiter oxydiert wird, entsteht 4b Methyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenathren-1, 4-dion XX, ein Stereoisomer von X. Gegenüber der bei 120 bis 121 Schmelzenden Verbidnung X zeigt die Verbindung XX einen Schmelzpunkt von 171 bis 172 Beim Hydrolysieren von XX mit Säure wird eine stereoisomere Form des 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahydrophenanthren-1, 4, 7-trions erhalten welche mit keiner der oben beschriebenen Verbindungen XI und XXII identisch ist.
Die als Ausgangsstoff verwendete Verbindung I ist neu. Sie kann hergestellt werden, indem 5-Methyl-perhydronaphthalin-1, 4-diol-6- on mit Methylvinylketon in Gegenwart eines alkalisehen Kondensierungsmittels, etwa einer starken Base wie Alkalihydroxyd, zur Reak- tion gebracht wird.
Das 5-Methyl-perhydronaphthalin-1, 4-diol6-on seinerseits wird wie folgt erhalten : -Äthoxy-propionaldehyd wird mit Äthylmagnesiumbromid umgesetzt, wobei 1-Äthoxy- 3-oxy-pentan entsteht. Dieses wird mit Chromsäure zum 1-Äthoxy-3-keto-pentan oxydiert, woraus durch Behandlung mit Äthyl-o-formiat undÄthanol in Gegenwart von Chlorwasser- stoff 1, 3, 3-Triäthoxy-pentan entsteht.
Dieses wird durch Reaktion mit heissem Kaliumbisul- fat in das 3-Äthoxy-1, 3-pentadien umgewan- delt, welches durch Diels-Alder-Kondensation mit Benzochinon 5-Methyl-6-iithoxy-1, 4, 4a, 5, 8, 8a-hexahydronaphthalin-1, 4-dion ergibt.
Durch Reduktion mit Wasserstoff und Raney Nickel erhält man daraus 5-Methyl-6-äthoxy- 1, 2, 3, 4, 4a, 5, 8, 8a-octahydronaphthalin-1, 4-dion, welches durch Behandlung mit Lithium-Alu minium-Hydrid in das entsprechende Diol, 5-Methyl-6-äthoxy-1, 2, 3, 4, 4a, 5, 8, 8a-octahydro- naphthalin-1, 4-diol übergeht, woraus schliess- lich druch Hydrolyse das 5-Methyl-6-keto-per hydronaphthalin-1, 4-diol erhalten wird.
In einem Teil der folgenden Beispiele sind jeweils nur einzelne Stufen der Herstellung der Triketopolyhydrophenanthrene beschrieben.
Beispiet 1
Herstellung von 4b-Methyl-7-äthylendioxy- 1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophen anthren-1, 4-diol (VIII).
In eine mit einem Rührer, einem 1-Liter- Tropftrichter und einem Tubus mit Kühler in Destillationsstellung ausgerüstete Fünfliter- flasche wurden 38, 9 g (0, 155 Mol) 4b-Methyl- 1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahydrophen anthren-1, 4-diol-7-on (I) 40 cm3 (0, 645 Mol) Glykol, 2500 cm3 Äthylendichlorid und 0, 4 g (0, 002 Mol) p-T'oluolsulfonsäure gebracht.
Nach Ingangsetzen der Rührung wurde genügend geheizt, um 3 Liter eines aus dem Losungsmittel und d'em gebildeten Wasser bestehenden aceotropen Gemisches innert dreier Stunden abzudestillieren. Um die Reaktionsteilnehmer in Lösung zu erhalten, wurden während dieser Zeit weitere 1i500 cm3 Äthy lendichlorid zugefügt. Nach 3 Stunden wurde die Reaktionsmischung abgekühlt und mit 50 cm3 wässeriger normaler Kaliumbicarbonatlösung kräftig durchgeschüttelt. Die wässe- rige Schicht wurde abgelassen und zweimal mit Athylendichlorid extrahiert. Alle drei Jithylendichloridextrakte wurden dann zusammengegeben, mit wasserfreiem Magnesium sulfat getrocknet und eingedampft.
Das Konzentrat wurde in einem Liter Aceton aufgenommen und die Lösung bis zur einsetzenden Kristallisation eingedampft. Durch Filtration des kalten Acetons wurden rohes, kristallines 4b-Methyl-7-äthylendioxt, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecalhydropehnathren-1,4-diol erhalten, das durch Umkristallisation aus Aceton weiter gereinigt werden kann. Schmelzpunkt des reinen Produktes 189\ C.
Beispiel 2
Herstellung von 4b-Methyl-7-athylendioxy 1, 2, 3., 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophen- anthren-1-on-4-ol (IX) und 4b-Methyl-7-äthy- lendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahy- drophenanthren-1, 4-dion (X).
86, 5 g (0, 294 Mol) 4b-methyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, l5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1, 4-diol wurden in einer Mischung von 2260 cm3 Benzol und 2130 g (= 2250 cm3) (21,7 Mole) Cyclhexanon gelöst, Dieser Losung wurden 86,5 g (0, 424 Mol) Aluminiumisopropylat zugesetzt und die Mi sehung am Rückfluss 12 Stunden erhitzt.
Dann wurden 25 cm3 Wasser zugesetzt und d'as dadureh gebildete, zusammengeballte Aluminiumhydroxyd abfiltriert. Das Filtrat wurde im Vakuum vollständig eingedampft und der Rückstand mit Petroläther ausgezo- gen, wodurch rohes, kristallines 4b-Methyl-7 äthylendioxy-1,2,3,4,41,5,6,7, 8, 10, 10a-dode cahydrophenanthren-l-on-4-ol erhalten wurde.
Ein durch Umkristallisieren gereinigtes Produkt schmolz bei 219 bis 220"C.
In dem zum Ausziehen verwendeten Pe troläther befand sich 4b-methyl-7-äthylen dioxy-1, 2, 3, 4, 4a, 4b,5, 6, 7, 8,10,10a-dodecahyrophenanthren-1, 4-dion, das nach Umkristallisa- tion bei 120 C schmolz.
Beispiel 3
Herstellung von b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahydrophenanthren-1, 4, 7 trion (XI).
Zu einer Lösung von 0,434 g (0, 0015 Mol) 4b-Methyl-7-äthylendioxy-1, 2', 3, 4, jazz 4b, 5,6,7,8,
10, 10a-dodecahydropehnanthren-1, 4-dion (X) in 10 cm3 Methanol wurde 1 cm3 2, 5 n
Salzsäure gegeben. Nachdem die angesäuerte
Mischung 21/2 Stunden am Rückfluss erhitzt worden war, wurden 8 em3 Wasser zugesetzt.
Das Methanol wurde vollständig abgedampft und die übrigbleibende Suspension dreimal mit Chloroform extrahiert. Die drei. Extrakte wurden vereinigt, mit wasserfreiem Magne siumsulfat getrocknet und eingedampft, wor auf das zurückbleibende rohe Produkt an
Aluminiumoxyd chromatographiert wurde.
Das Adsorbat wurde mit einer Äther-Chloro- form-Mischung eluiert. Das so erhaltene reine
4b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodeca hydrophenanthren-1, 4, 7-trion sehmilzt bei 117 C.
Beispiel 4
Herstellungvon4b-Methyl-7-äthylendioxyd-
1, 2,, 3, 4, 4a, 4b, 5,'6, 7, 8, 10, 10a-dodeeahydrophen- anthren-l, 4-dion (X) durch die Oxydation . von Verbindung IX mit Chromtrioxyd-Pyri din.
Eine Losung von 3, 12 g 4b-Methyl-7-äthy lendioxy-1, 2, 3, 4, 4a, 4b, 5, 16, 7, 8, 10, dodecahy drophenanthren-1-on-4-ol (IX) in 30 cm3
Pyridin wurde mit 3, 1 g Chromtrioxyd in 30 cm3 Pyridin vermischt. Das Reaktions gefäss wurde versehlossen, der Inhalt gut durchgemischt und die Mischung bei Zim mertemperatur stehengelassen. Dann wurde die Reaktionsmisehung in Wasser gegossen und dreimal mit Benzol-Äther (1 : 1,) extra hiert, wobei zum Brechen der Emulsion je weils über Diatomeenerde filtriert wurde.
Die vereinigten organischen Lösungen wur den mit Wasser gewaschen, mit entwässer- tem Magnesiumsulfat getrocknet, eingedampft und der Rückstand im Hochvakuum getrock net. Kristallisation aus Äther ergab Verbin dung X mit dem Schmelzpunkt 117 bis 121 C.
Beispiel 5
Herstellung von 4b-Methyl-7-äthylendi- oxy-1, 2, 3, 4, 4a, 5, 6, 7, 8, 10, 10a-dodecahydrophen athren-1,4-diol-1-acetat (XV).
Eine Lösung von 2,00 g Verbindung VIII in 20 em. trockenem Pyridin und 10 cm3 Essigsäureanhydrid wurde 10 Minuten auf 1@ C erhitzt. Der Oberschuss an Essigsäure- anhydrid wurde mit Eiswasser hydrolysiert und die Reaktionsmisehung auf 100 cm3 verdünnt. Die kristalline Ausscheidung wurde abfiltriert, gründlich mit Wasser gewaschen und getrocknet. Eine zweimal aus Aceton umkristallisierte Probe war reine Verbindung XV und schmolz bei 183, 5 bis 184 C.
Beispiel 6
Herstellung von 4b4Methyl-7-äthylendioxy- 1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophen anthren-1-ol-4-on-l-acetat (XVI).
2 g Chromtrioxyd wurden in 20 cm3 trockenem Pyridin unter Kühlung gelost.
Zur erhaltenen Suspension des orange ge färbten Komplexes wurde eine ! Losung von 2, 00 g Verbindung XV in 20 em3 trockenem Pyridin gegeben. Nach gutem Durchmischen des Inhaltes blieb das. verschlossene Reak- tionsgefäss über Nacht stehen. Dann wurde die Reaktionsmischung in 100 cm3 Wasser gegossen und driemal mit je 100 cm3 Benzol extrahiert. Zur Trennung der Extrakte war Filtration über Kieselgur nötig. Die verei nigten Benzollösungen wurden mit Wasser gewaschen, mit wasserfreiem Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wurde zum Entfernen des Pyridihs im Hoehvakuum getrocknet und dann in Äther kri stallisiert.
Nach zweimaliger Umkristallisation war reine Verbindung XVI vorhanden.

Herstellung von 4b-Methyl-7-ä. thylendioxy- 1, 2, 3, 4, 4a, 4b, 5,6,7,8,10,10a- dodecahydrophenanthren-1-ol-4-on (XVII)
0, 726 g Verbindung XVI wurden in 10 em3 0, 25 n Kaliumcarbonatlösung in 75- prozentigem Methanol gelöst und die Lösung 12 Minuten am Rüekfluss erhitzt. Dann wurde Wasser zugesetzt und das Methanol im Va kuum abdestilliert. Das organisehe Material wurde in Chloroform aufgenommen und die chloroformische Lösung getrocknet und ein gedampft, wodurch ein bei 137 bis 140 C schmelzendes Produkt erhalten wurde.
Durch Umkristallisation ausÄtherwurde reine Verbindung XVII mit dem Smp. 142 bis 143 C erhalten.
Eine Mischung mit dem Ausgangsacetat schmolz bei 130 C. Nach dem Durchgang über alkalisches Aluminiumoxyd wurde un- veränderte Verbindung XVII zuriickgewonnen.
Durch A. cetylierung mit Essigsäureanhydrid-Pyridin wurde das Ausgangsacetat Verbindung XVI erhalten.
BeispieZ 8
Herstellung von 4b-Methyl-7-äthylendi oxy-1, 2, 3, 4, 4a, 4b,5,6,7,8,10,10a-dodecahydrophenanthren-1, 4-dion (XVITI) und4b-Methyl- 7-äthylendioxy- 1,2,3, 4, 4a,4b,5,6,7, 8, 10, 10a-do deeahydrophenantliren-l, 4-dion (X), einer stereoisomeren Form von XVII'I.
Eine Lösung von 0, 182 : g Verbindung XVII in 2 em3 Pyridin wurde mit 0, 20 g Chromtrioxyd in 2 em3 UPyridin bei Zimmer- temperatur über Nacht oxydiert. Durch Verdünnen mit Wasser und Extrahieren mit Benzol wurde ein nichtkristallines Produkt erhalten. Eine Lösung dieses Materials in Ather schied beim Stehen Kristalle mit einem Schmelzpunkt von 135 bis 145 C aus.
Mehrere Umkristallisationen aus Xther ergaben Verbindung XVIII mit dem Schmelz- punkt 146 bis 148 C.
Die erste ätherische Mutterflüssigkeit wurde über alkalisches Aluminiumoxyd geschickt. Verbindung X mit dem : Schmelz- punkt 118 bis 119, 5 C wurde erhalten.
Mischsehmelzpunkt mit einer andern Probe von Verbindung X : 118, 5 bis 120 C.
Beispiel 9
Herstellung der Verbindungen X, IX, XVII und XVIII durch die Chromtrioxyd Pyridin-Oxydation von Verbindung VIII.
