Verfahren zur Herstellung von l-Keto-4-oxy-polyhydrophenanthrenen
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 1-Keto-4-oxy-poly- hydrophenanthrenen, welches dadurch gekennzeichnet ist, dass 1, SDioxy-polyhvdro- phenanthrene in 1-. Stellung oxydliert werden.
In einer besonderen Ausführungsform bezielit sieh das erfindungsgemässe Verfahren auf die Herstellung von 4b-Methyl-l,2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10,10a-dodecahydrophenanthren-4-ol 1, 7-dion der Formel

Diese Substanz sowie die aus ihr leicht ab leitbaren 7-EnolÏther- oder-Ketalderivate und 4-Aeylderivate sind wertvolle Zwischenprodukte für die Synthese von Steroidver- bindungen und ändern Polyhydrophenanthren- verbindungen.
Diese neuen Verbindungen besitzen drei kondensierte Kohlenstoffringe, welche den Ringen A, B und C der Steroid verbindungenentsprechen,und eine anguläre Methylgruppe in Stellung 4b, welche der bei vielen Steroidverbindungen vorhandenen angulären Methylgruppe in 10-Stellung entspricht.
Nach einer Ausführungform des erfin dungsgemässen Verfahrens kann das neue 4b Methyl- dodecahydrophenanthren-4-ol-l,7-dion unter Verwendung von 4b-Methyl-1,'2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 110a-dodeeahydrophenanthren-1, 4- diol-7-on als Ausgangsmaterial gemäss folgendem Reaktionssehema hergestellt werden :

Die Verbindung I wird zum Beispiel zuerst mit einem geeigneten Mittel behandelt, um den 7-Keto-Substituenten zu blockieren oder zu sehützen, was zum Beispiel durch Bildung eines Enoläther-oder Ketal-derivates geschehen kann.
Dieses Derivat (II) wird dann oxydiert und so die 1-Oxygruppe in eine Ketogruppe verwandelt. Das Oxydationspro- dukt (III) wird zur 4-Oxy-l, 7-diketo-Verbin- dung (IV) hydrolysiert, welche durch Acy lierung in die entsprechende 4-Acyloxy-Verbindung übergeführt werden kann.
Das Blockieren der 7-Ketogruppe kann, wie oben gesagt, durch Bildung eines Enol äther oder Ketal-derivates geschehen. Die zweite Methode, vorzugsweise die Umwandlung in eine cyclisehe Ketalgruppe, etwa eine Athylendioxygruppe, erwies sich aber als zweckmässiger. Bei der Herstellung des Ketals verschiebt sich die Doppelbindung von der 8, 8a- in di 8a, 9-Stellung.
Demgemäss wird Xb- Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahy- drophenanthren-4-ol-1, 7-dion (IV) wie folgt hergestellt :

Im ersten Schritt dieses Verfahrens wird Verbindung I in das entsprechende 7-Athylendioxy-Derivat, nÏmlich 4b-Methyl-7-Ïthylen dioxy 1t, ^', 3, 4, 4a, 4b, 5., 6,,'7, 8, 10, l0a-dodecahydrophenanthren-l, 4-diol (IIA) umgewandelt. Dies kann leicht so geschehen, da. die Verbindung I mit Glykol in Gegenwart eines sauren Katalysators, wie p-Toluolsulfonsäure, er hitzt wird.
Die Reaktion wird mit Vorteil in einem inerten Lösungsmittel vorgenommen, etwa in Äthylendichlorid. Dazu wird eine Mischung von Verbindung I, Glykol und Äthylendichlorid, erhitzt und das bei der Reaktion gebildete Wasser fortlaufend als azeotrope Mischung mit Äthylendichlorid abdestilliert Naeh beendeter Reaktion wird das gebildete Athylendioxyderivat durch Ein dampfen der Lösung und Kristallisieren des Rückstandes aus einem geeigneten Lösungsmittel, etwa Aceton, gewonnen.
Bei der zweiten Verfahrensstufe wird das Äthylendioxyderivat der Verbindung I zur Umwandlung der 1-Oxygruppe in eine Ketogruppe selektiv oxydiert. Das kann zwar auf verschiedene Weise geschehen, doch empfiehlt es sich, die Reaktion mit. Hilfe eines ZIetallalkoholates in Gegenwart eines Ketons a. uszuführen. Ganz allgemein zeigte es sich, dass die Reaktion besonders gut in Gegenwart eines inerten organischen Lösungsmittels verläuft, etwa in Benzol, Toluol oder Dioxan. So wird denn die Reaktion durchgeführt, indem die Verbindung IIA in einer Mischung von Keton und organischem Losungsmittel gelost, ein Metallalkoholat. zugesetzt, und die Mischung erwÏrmt wird. Geeignete Ketone sind neben andern Aceton, Met. hyläthylketon, Cyelo- hexanon, Benzol und p-Benzoehinon.
Beste Ergebnisse werden bei der Verwendung von Cyelohexanon als Keton, Benzol als inertem Losungsmittel und Aluminiumisopropylat als Metallalkoholat erhalten.
Es ist überraschend und unerwartet, dass sich auf diese Weise 4b-Methyl-7-äthylendi- oxy - 1,2,3,4,4a,4b, 5, ; 6,, 7, 8, 10,10a - dodecahydro yhenanthren-4, ol-l-on (IIIA) mit guter Ausheute aus der Verbindung IIA herstellen l, if') t, da bei diesem Vorgehen eher eine vollstÏndige Oxydation zur Diketoverbindung zu erwarten gewesen wÏre. ¯berraschenderweise wurde auch festgestellt, dass bei der Oxydation der Verbindung IIA eine Inversion der Konfiguration bei C l () a stattfindket, wodureh Verbindung IIIA entsteht, deren B/C-R-ing konfiguration von derjenigen der Verbindung IIA abweicht.
Die Verbindung IIIA kann auch auf an dere Weise hergestellt werden, nämlich indem eine pyridinisehe Lösung der Verbin- dung IIA mit Chromtrioxyd-Pyridin-Kom plex oxydiert und das erhaltene Produkt mit einer Base behandelt wird. Diese Oxydation wird am besten durchgeführt, indem eine lIisehung von IIA, Chromtrioxyd-Pyridvin- Komplex und Pyridin bis zur vollständigen Oxydation bei Zimmertemperatur stehenge- lassen wird.
Dann wird die Reaktionsmischung mit Wasser verdünnt und mit einem Lösungs- mittel, wie Benzol-Ather-Misehung, extra hiert. Beim Eindampfen dieses Extraktes wird eine Mischung von Oxydationsprodukten erhalten, welche durch Adsorption an Alumi- niumoxyd chromatographisch getrennt werden kann. Die Verbindung IIIA und andere Oxydationsprodukte können durch Eluieren mit Lösungsmitteln aus der chromatogra- phischen Säule gewonnen werden.
In der letzten Stufe des Verfahrens wird die Verbindung IIIA in die entsprechende 7 Ketoverbindung, Verbindung IV, umgewandelt, zum Beispiel indem sie mit Säuren, etwa Salz-oder Schwefelsäure, oder mit säurebildenden Substanzen, etwa p-Toluolsulfonyl ehlorid, behandelt wird.
Es wurde gefunden, dass die Verbindung IV auch durch Reaktion der Verbindung I mit N-Brom-acetamid und Behandlung des entstehenden Produktes mit einer Base erhalten werden kann, beispielsweise indem die Verbindung I in Pyridin gelöst, das N-Brom- acetamid zugesetzt und die Mischung mehrere Stunden bei Zimmertemperatur stehengelassen wird. Dann wird die Reaktionsmischung gekühlt und in einen Überschuss von Salzsäure gegossen. Diese Mischung wird mit einem Losungsmittel, etwa Chloroform, extrahiert und der Extrakt zur Trockne einge- dampft. Das Produkt wird aus dem Verdampfungsrüekstand gewonnen, indem man diesen an alkalischem Aluminiumoxyd chromatographiert und das Adsorbat mit einer ¯ther-Chloroform-Mischung eluiert.
Nach einer andern Ausführungsform der Erfindung kann die Verbindung IV aus Verbindung I auf folgende Weise hergestellt werden :

In den obenstehenden Formeln bedeutet Bi eine Acyloxygruppe.
Dazu wird Verbindung I mit einem, Acy lierungsmittel in das Triacyloxyderivat V verwandelt. Dieses wird selektiv zur 1-Oxy4-aeyloxy-7-keto-Verbindung VI hydrolysiert, welche zur entspreehenden 4-Aeyloxy-1, 7-diketo-Verbindung VII oxydiert wird. Daraus wird durch Hydrolyse Verbindung IV erhalten.
Von den vielen in Frage kommenden Aeylierungsmitteln eignen sich zur Umwandlung der Verbindung I in das entsprechende Tri aeylaxyderivat besonders niedere aliphatische Säureehloride, etwa Acetylchlorid. Die Verbindungen der Formel V, VIund VII mit R1 = Acetoxy werden im folgenden mit VA, VIA und VIIA bezeiehnet.
In der ersten Stufe wird durch Erhitzen der Verbindung I mit Acetylehlorid 4b Methyl - 1,2,3,4,4a,4b, 5, 6, 10, 10a - decahydrophenanthren-1, 4, 7-triol-1, 4, 7-triacetat (VA) gebildet. Naeh beendeter Reaktion wird die Reaktionsmisehung im Vakuum getrocknet und der Rüekstand an mit Säure gewa- schenem Aluminiumoxyd chromatographiert.
Das gewünschte Produkt, die Verbindung VA, wird durch Eluierung mit einer Mi- schung von Äther und Petroläther erhalten.
Ausserdem wurde aus dem Eluat eine kleine Menge 4-Methyl - 1,2,3,4,4a,4b,5,6, 10, 10a.- deeahydrophenant. hren-1, O, 7-triol-1 7-diaeetat. gewonnen. Als nächster Schritt wird Verbindung VA durch Behandlung mit Alkali selektiv zum 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a dodecahydrophenanthren-l, 4-diol-7-on-4-a@etat (VIA) hydrolysiert. Dies gesehieht leicht durch kurzes Erhitzen der VerbindungVA mit einer methanolisehen Lösung von Ka liumearbona. t. Das Methanol wird bei einer unterhalb Zimmertemperatur liegenden Temperatur aus der Reaktionsmischung abdestilliert.
Der so erhaltene, wässerige R ckstand wird dann mit Äther extrahiert, und aus dem Extrakt durch Eindampfen Verbindung VIA erhalten.
Die folgende Stufe besteht darin, Verbindung VIA zu 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5,6, 7, 9, 10, 10a-dodeeahydrophenanthren-4-ol-1, 7 dion-4-acetat (VIIA) zu oxydieren. Durch Reaktion einer Losung der Verbindung VIA in Essigsäure mit Chromtrioxyd geht dies ohne Sehwierigkeiten. Nach beendeter Reaktion wird Wasser zugesetzt und die entstehende Lösung mit Chloroform extrahiert.
Das Produkt, die Verbindung VIIA, ka. nn gewonnen werden, indem, die Chloroformlosung zur Trockne eingedampft und der Rüekstand mit einer milden Base behandelt wird, etwa. so, dass das Produkt an Alumi niumoxyd adsorbiert wird. Dadurch findet eine Inversion bei C-l0a statt, und es entsteht eine Verbindung, welche an der Verei nigmngsstelle des B-und C-Ringes eine andere Konfiguration aufweist
Im letzten Schritt wird Verbindung VIIA zur Verbindung IV hydrolysiert. Dies gesehieht zum Beispiel, indem eine Lösung von VIIA mit einem Alkali, etwa mit methanoli- sehem Kaliumhydroxyd, am Rüekfluss erhitzt t wird.
Das Produkt, Verbindung IV, wird gewonnen, indem die Reaktionsmisehung mit einem Lösungsmittel, zum Beispiel Benzol, extrahiert und das L¯sungsmittel vollständig abgedampft wird.
Wenn man die Verbindung IIIA mit einem geeigneten Reduktionsmittel reduziert, so entsteht die Verbindung IIA', eine stereoisomere Form von IIA. Diese neue Verbindung, nÏmlich 4b-Methyl-7-athylendioxy-1, ?', 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodeeah.-drophenantllren-1, 4-diol (IIA') besitzt an der Verbindungsstelle des B-und C-Ringes eine andere Konfiguration als Verbindung IIA. Die Verbindung IIA'hat einen Schmelzpunkt von etwa 175 bis 175 C, währenddem Verbindung IIA bei etwa 189 C schmilzt.
In analoger Weise entsteht bei der Reduktion von 4b-Methyl-7-äthylendioxy- 1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophelian- thren-1, 4-dion (IX) eine andere stereoisomere Modifikation von IV und IIA', nämlieh Verbindung X, welche einen Schmelzpunkt von 7. 98 bis 198, 5¯ C besitzt. Es wurde gefunden, dass sich die Verbindungen IIA'und X am besten durch Reaktion der Verbindungen IIIA und IX mit Lithium-Aluminium-Hydrid in einem geeigneten Lösungsmittel, etwa in Tetra- hydrofuran, n, herstellen lassen.
Die Verbindung I ist neu. Sie kann durch Reaktion von 5-Methyl-6-keto-perhydronaphthalin-1, 4-diol mit Methylvinylketon in Ge genwart eines alkalischen Kondensationsmit- tels, etwa einer starken Base (eines Alkali metallhydroxydes und dergleichen) hergestellt werden.
Das a-AIethyl-6-keto-perhydronaphthalin- 1, 4-viol seinerseits kann wie folgt hergestellt werden : ¯-¯thoxy-propionaldehyd wird mit ¯thylmagnesiumbromid zur Reaktion gebracht, wobei 1-tithoxy-3-oxy-pentan entsteht, das mit Chromsäure zu l-Athoxy-3-keto-pentan oxy- diert wird. Dieses Oxydationsprodukt wird in Gegenwart von Chlorwasserstoff mit Athyl- o-formiat behandelt, das entstandene 1, 3, 3 Triäthoxy-pentan dureh Reaktion mit heissem Kaliumbisulfat in 3-¯thoxy-1,3-pentadien verwandelt und dieses nach Diels-Alder mit Bemochinon kondensiert, wodurch 5-Methyl-
6-Ïthoxy - 1,4,4a,5,8,8a - hexahydronaphthalin1, 4-dion entsteht.
Daraus wird durch Reduktion mit Wasserstoff in Gegenwa. rt von Raney- Nickel 5-Methyl-6-athoxy-1, 2, 3, 4, 4a, 5, 8, 8a-oeta- hydronaphthalin-1, 4-dion erhalten, welches durch Behandlung mit Lithiumaluminium- hydrid in das entsprechende Diol, 5-Methyl- 6-Ïthoxy-l,2,3,4,4a,5, 8, 8a-octa. hydronaphthalin- 1, 4-diol übergeht. Aus diesem entsteht schliess- lieh durch Hydrolyse 5-Methyl-6-keto-per hydronaphthalin-l, 4-diol.
Beispiel 1 il : Herstellung von 4b-Methyl-7-äthylen- dioxy-l, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 110i 10a@dodecahydro- phenanthren-l,4-diol (IIA).
In eine mit einem Rührer, einem Einlitertropftrichter und einem Kühler in Destillier- stellnng versehene Fünfliterflasche wurden 38, 9 g (0, 155 Mol) 4b-Methyl-l,2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahydrophenanthren-l, 4-diol- 7-on (I), 40 cm3 (0, 645 Mol) Glykol, 2500 cm3 Äthylendichlorid und 0, 4 g (0, 002 Mol) p- Toluolsulfonsäure gebracht.
Bei eingeschalteter Rührung wurde genügend erwärmt, um innert dreier Stunden 3 Liter eines aus dem Reaktionswasser und Äthylendichlorid beste henden azeotropen Gemisches abzudestillieren.
Um die Reaktionsteilnehmer in Lösung zu halten, wurden während, dieser Zeit 1500 em3 Lösungsmittel zugesetzt. Nach drei Stunden wurde die Reaktionsmischung abgekühlt und mit 50 em3 einer wässerigen, normalen Kalium- bicarbonatlösung kräftig durchgesch ttelt.
Die wässerige Schicht wurde abgetrennt und zweimal mit Äthylendichlorid extrahiert. Alle drei Äthylendichloridextrakte wurden verei nigt, mit wasserfreiem Magnesiumsulfat getrocknet und eingedampft. Das Konzentrat wurde in einem Liter Aceton aufgenommen und bis zur beginnenden Kristallisation eingeengt. Durch Filtration der kalten Mischung wurde rohes, kristallines 4b-Methyl-7-äthylen- dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydro- phenanthren-1, 4-diol erhalten, das durch Umkristallisieren aus Aceton weiter gereinigt werden kann.
Schmelzpunkt des reinen Produktes 189 C job : Herstellung von 4b-Methyl-7-äthylen- dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-l-on-4-ol (IIIA) neben 4b-Me thyl-7-ä. thylendioxy-1, 2, 3, 4., 4a, 4b, l5, 6, 7, 8, 10, 10a dodecahydrophenanthren-1, 4-dion (IX).
86, 5 g (0, 294 Mol) Ob-Methyl-7-äthylen- dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1, 4-diol wurden in einer Mischung von 2250 em3 Benzol und 2 ; 130 g = 2250 em3 (2. 1, 7 Mol) Cyclohexanon gelöst.
Der Lösung wurden 86, 5 g (0, 424 Mol) Aluminiumisopropylat zugesetzt und die Mischung am R ckflu¯ 12 Stunden erhitzt. Dann wurden 25 em3 Wasser zugesetzt und das dadurch gebildete, zusammengeballte Aluminiumhydroxyd abfiltriert. Das Filtrat wurde im Vakuum vollständig eingedampft und der Rüekstand mit Petroläther ausgezogen, wodurch rohes, kristallines 4b-Methyl-7-äthylen- dioxy-1, 2., 3, 4, 4a, 4b,5,6,7,8,10,10a-dodecahydrophenanthren-l-on-4-ol erhalten wurde. Ein durch Umkristallisieren gereinigtes Produkt schmolz bei 219-220 C.
Im Petrolätherauszug befand sich 4b-Me- thyl-7-Ïthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a dodecahydrophenanthren-1, 4-dion, das nach Umkristalisation bei 120 C schmolz.
C: Herstellung von 4b-Methy1-1,2, 3, 4, 4a, 4b, 5, 6,7, 8, 10', l0a - dodecahydrophenanthren - 4-ol1, 7-dion (IV).
Eine Lösung von 0, 05 g (0, 001 Mol) 4b- Methyl-7-äthylendioxy-l, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, l0a - dodecahydrophenanthren - 4 - ol-l-on und einigen Milligrammen p-Toluolsulfonylchlorid in 10 cm3 Alethanol wurde am Rüekfluss anderthalb Stunden erhitzt und dann eingeengt.
Der troekene Rückstand wurde mit Ather aufgenommen und mit einer wässerigen Lösung von Kaliumbiearbonat, welche einen Tropfen Pyridin enthielt, kräftig geschüttelt und dann getrennt, über wasserfreiem Natriumsulfat getrocknet. und zu einem dünnen Íl eingedampft. Das rohe Produkt, 4b-Methy1-1, 2, 3, 4, 4a, 4b, 5,6, 7, 8, 10, 10a,-dodecahydrophenan- thren-4-ol-1, 7-dion, kristallisierte bald aus diesem ¯l aus. Es kann durch Kristallisation aus Athylacetat gereinigt, werden und schmilzt dann bei 201 bis 203"C.
Beispiel 2 il @ Herstellung von 4b-Methyl-l,2, 3, 4, 4a, 4b, 5, 6, 10, l0a-decahydrophenanthren-l, 4, 7-triol 1, 4, 7-triaeetat (VA).
In einem gegen Feuehtigkeitszutritt gesehützten Reaktionsgefäss wurden 0, 480 g 4b Methy] -1,2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahy- drophenanthren-1, 4-diol-7-on in 7 cm3 frisch destilliertem Aeetylchlorid vier Stunden am Rüekfluss erhitzt. Schon nach einer Stunde war alles klistalline Ausgangsmaterial in Losung gegangen, und nach vier Stunden konnte keine Chlorwasserstoffentwicklung mehr festgestellt werden. Alles flüchtige Material wurde im Vakuum entfernt, zuletzt durch ErwÏrmen im Hochvakuum. Der Bückstand wurde an 15 g mit SÏure gewaschenem Aluminiumoxyd chromatographiert.
Mit einer Mischung aus 3 Teilen Ather und 7 Teilen Petroläther (Siedepunkt 30 bis 60 C) liel sich kristallines Triaeetat mit dem Schmelz- punkt 132 bis 1316 C eluieren. Nach dreimaligem Umkristallisieren schmolz das reine Produkt VA bei 137, 5 bis 139 C.
Mit einer Ather-PetrolÏther-Mischung im Verhältnis 7 : 3 wurde b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 10, 10a-dodecahydrophenanthren-l,4,7-triol1, 7-diacetat eluiert, dessen Schmelzpunkt bei 1. 17 bis 150¯ C lag. Durch zweimaliges Umkristallisieren aus Äther stieg er auf 150 bis 151 C.
Durch Verk rzung der Reaktionsdauer liens sich der Anteil an Diacetat erhöhen.
B : Herstellung von 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a - dodecahydrophenanthren -1,4 diol-7-on-4-acetat (VIA).
Eine Lösung von 0, 455 g Verbindung VA in 25 cm3 einer 4prozentigen Kaliumcar- bonatlösung in 715prozentigem Methanol wurde 10 Minuten am Rückfluss erhitzt. Das Me- thanol wurde, im Vakuum der Wasserstrahl- luftpumpe aus der unter Zimmertemperatur abgekühlten Mischung abgedampft. Der wÏsserige Rückstand wurde fünfmal mit Äther extrahiert, der Ätherextrakt getrocknet und eingedampft. Es kristallisierte Verbindung VIA mit dem Sehmelzpunkt 156 bis 165 C und sehied sich aus. Eine aus Äther umkristallisierte Probe schmolz bei 168 bis 170 C.
C : Herstellung von 4b-Methyl-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 9, 10, 10a-dodecahydrophenanthren-4 ol-1, 7-dion-4-acetat (VIIA).
Eine eiskalte Lösung von 0, 180 g Verbindung VIA in 5 em3 90prozentiger EssigsÏure wurde allmählich mit einer L¯sung von 0, 18 g Chromtrioxyd in 1, 8 cm3 90prozenti- ger Essigsäure versetzt. Die Reaktionsmischung blieb ohne K hlung eine Stunde ste hen, wobei sie sich auf Zimmertemperatur er wärmte. Nun wurden drei Volumen Wasser zugesetzt und die erhaltene Lösung sechsmal mit Chloroform extrahiert. Nach dem Wasehen mit wässeriger Natriumbicarbonatlosung und Trocknen wurde das Chloroform abgedampft, wobei ein harzartiges Material zurüekblieb, das nieht zur Kristallisation gebracht werden konnte.

Eine Losung-von40mgVerbindungVIIA in 1 em3 Benzol wurde mit 1 cm3 methanoli sehem, normalem Kaliumhydroxyd vermischt und die Mischung am Rüekfluss eine Stunde gekocht. Nun wurde Benzol und dann 1 cm3 10 /o Salzsäure zugesetzt. Die organische Schicht wurde abgetrennt, getrocknet und eingedampft, wobei ein Produkt zurüekblieb, das nach Kristallisation aus Aceton bei 194 : bis 199 C schmolz. Durch Umkristallisieren aus Aceton wurde reine Verbindung IV mit dem Schmelzpunkt 201 bis 202¯ C erhalten.
Beispiel 3
Herstellung von Verbindung IV durch die Oxydation von Verbindung I mit N-Brom- acetamid.
Eine Lösung von 0, 877 (0, 0035 Mol) Verbindung I in 9 en, Pyridin wurde hergestellt, indem die Mischung erwärmt und die erhal tene Losung vorsichtig auf Zimmertempera tur abgekühlt wurde, um ein Wiederauskristallisieren zu verhindern. Mehrere Umkristallisationen dieses Produktes aus Methanol führten zum. Ausgangsstoff, Verbindung I, mit dem Schmelzpunkt 241 bis 2441, C. Der Aeetonauszug entliielt ein rohes Produkt mit dem Schmelzpunkt 155 bis 185¯ C. Durch viermaliges Umkrista@i- sieren aus Aeeton konnte eine kleine Menge Verbindung IV.
Schmelzpunkt 197 bis 199 C, erhalt : en werden. Wenn das rohe Material (Schmelzpunkt 155 bis 185¯ C) an basischem Aluminiumoxyd chromatographiert wurde, enthielt das Eluat mit einer 7 : 31 Äther-Chloro form-Xischung die Ha. uptfraktion mit dem Schmelzpunkt 195 bis 200¯C. Daraus konnte durch einmaliges Umkristallisieren reine Verbindung IV erhalten werden, deren Schmelz- punkt und Mischschmelzpunkt mit einer andern Probe von Verbindung IV bei 201 bis 202 C lag.

A : Herstellung von 4bMethyl-7-äthlyen dioxy-1, 2, 3, 4, 4a, 4b, 6, ! 7,8,10,10a-dodecahydrophenanthren-1, 4-diol (IIA), einer stereoisomeren Modifikation von Verbindung IIA.
Zu 1 g Lithiumaluminiumhydrid in 25 cm3 trockenem Tetrahydrofuran wurde unter R hren langsam eine Losung von 0, 884 g Verbindung IIIA in 25 em3 trockenem Tetra hydrofuran gegeben. Die Mischung wurde eine Stunde bei Zimmertemperatur gerührt, dann wurden tropfenweise 4, 0 cm3 Wasser zugegeben und die entstandene, anorganische Ausscheidung abfiltriert. Das Tetrahydrofuran wurde abdestilliert und der zurückbleibende, kristalline Rückstand aus Aceton umkristallisiert, wodurch eine erste Menge von Verbindung IIA', Schmelzpunkt 175 bis 176 C, erhalten wurde. Preparation of compound IV by the oxidation of compound I with N-bromoacetamide.
A solution of 0.877 (0.0035 mol) of compound I in 9 en, pyridine was prepared by heating the mixture and carefully cooling the resulting solution to room temperature to prevent recrystallization. A: Production of 4bMethyl-7-ethylene dioxy-1, 2, 3, 4, 4a, 4b, 6,! 7,8,10,10a-dodecahydrophenanthrene-1,4-diol (IIA), a stereoisomeric modification of compound IIA.
To 1 g of lithium aluminum hydride in 25 cm3 of dry tetrahydrofuran was slowly added a solution of 0.884 g of compound IIIA in 25 cm3 of dry tetrahydrofuran with stirring. The mixture was stirred for one hour at room temperature, then 4.0 cm 3 of water were added dropwise and the inorganic precipitate formed was filtered off. The tetrahydrofuran was distilled off and the remaining crystalline residue was recrystallized from acetone, whereby a first amount of compound IIA ', melting point 175 to 176 ° C., was obtained. A second amount with a melting point of 160 to 173 ° C could be obtained from the end.
By recrystallizing the substance which had first crystallized out from acetone, a product with a melting point of 175 to 176 ° C. was obtained.
