Aus dem erhaltenen Ge- misch können das genannte Methyloxazolina-steroid und die gleichzeitig entstandenen beiden enantiomeren 17-Ketos'teroide auf übliche Weise, zweckmässig auf chromatographischem Wege, abgetrennt und isoliert werden.

die ihrerseits durch Umsetzung entspre chender 20-Oxosteroide, z. B. von 3ss-Acetoxy-16- methylpregnädien-20-on, mit Hydroxylamin erhalten werden können.
Die erfindungsgemäss erhältlichen Steroid-Deri- vate sollen für therapeutische Zwecke oder als Aus gangsmaterialien für die Herstellung pharmazeutischer Präparate dienen.
<I>Beispiel</I> a) Zur Herstellung des Ausgangsstoffes, 3ss-Acet- oxy-17-acetylamino -16-methyl-5,16-pregnadien (1I), werden 29 g 3ss-Acetoxy-16-methyl-5,16-pregnadien- 20-on (Wettstein, Helv. Chim. Acta 27, 1802)
in <B>175</B> mI Pyridin gelöst und mit 8 g pulverisiertem Hy- droxylämin-Hydrochlorid versetzt. Beim Erwärmen entsteht zunächst eine klare Lösung, die sich bald In folge der Abscheidung eines las trübt.
Nach fünf tägigem Stehen wird zunächst Wasser, dann 2n Schwe felsäure zugesetzt und mehrmals mit Äther ausge- schüttelt. Nach dem Trocknen über Na2S04 und Ab dampfen des Lösungsmittels hinterbleiben 29 g festes rohes 3ss-Acetoxy-16-methyl-5,16-pregnadien-20-on- oxim (I),
das durch fraktionierte Umkristallisation aus abs. Äthanol gereinigt werden kann. Man erhält vor wiegend ein Oxim vom Schmelzpunkt 181-182 C;
[a] D = - 62 (c = 1 in CHHC13) neben einer kleinen Menge eines höher schmelzenden Isomeren vom Schmelzpunkt 216-217 C; [a] D = - 90 (c = 1 in CHC13). Die Gesamtausbeute an gereinigtem Produkt beträgt etwa 26 g.
Die Trennung der isomeren Oxime der Summen formel C24Hs503N kann chromatograpnisch an AIU- miniumoxyd (neutral, Aktivität IH) erfolgen. Das niedriger schmelzende Isomere ist leichter, das höher schmelzende, das mit Schmelzpunkten bis 223 bis 224 C ([a] D =-98,6 ) erhalten werden kann, schwerer eluierbar.
Die Infrarotspektren der beiden Oxime in KBr- Presslingen unterscheiden sich in charakteristischer Weise durch folgende Banden:

Im Fingersprint- gebiet ist noch eine Anzahl von Nebenbanden vor handen, deren Lage bei den beiden isomeren Oximen nicht genau übereinstimmt.
Das p-Toluolsulfonylderivat des niedrig schmel zenden Oxims (C31H41O5NS) schmilzt bei 151 bis 154 C. Das niedrig schmelzende Oxim kann in das hoch schmelzende Oxim umgewandelt werden, wenn man z. B. 300 mg davon in 10 ml Eisessig löst und trop fenweise mit einer Lösung von 110 mg Bortrifluorid- ätherat in 2 ml Eisessig versetzt.
Nach 16stündigem Stehen bei Zimmertemperatur wird auf Eis gegossen, mit 30 % iger Natronlauge alkalisch gemacht und in Äther aufgenommen. Ausbeute: 240 mg hochschmel- zendes Oxim. Gelegentlich erhält man auch kleine Mengen einer instabilen Oximform vom Schmelzpunkt 155-l58 C,
die sich nach einigen Tagen in das niedrig schmel zende Isomere (F. = 179-180 C) umwandelt. 3,5 g des niedrig schmelzenden Oxims (F. =<B>179</B> bis 182 C) werden in Anteilen in eine mit Eis und Wasser gekühlte Lösung von 3,5 g Phosphorpenta- chl'orid und 3,5 g Phosphoroxychlorid in 21 ml Pyri- din eingetragen. Bei der Zugabe fällt ein rotorange farbene Komplexverbindung aus.
Nach einstündigem Rühren wird das Reaktionsgemisch auf Eis gegossen und mit 80 ml Salzsäure (10%ig) anges'äuer't. Die ausgefallene Substanz wird abgenutscht, in Methanol aufgenommen und das Lösungsmittel abgedampft.
Der in Benzol lösliche Anteil wird über Aluminium oxyd (neutral, Aktivität HI) chromatographiert. Die Benzol - Äther (4: 1 und 2 : 1) - Fraktionen ergeben 3,3 g rötlich gefärbtes Amid, das nach mehrmaligem Umkristallisieren aus Methylenchlorid - Petroläther <B>1,8</B> g 3ss - Acetoxy -17-acetylamino-16-methyl.-5,16- androstadien (Il)
mit der Summenformel C24Hss0sN ergibt. F. = 217-218 C; [a]D = - 25,4 (c = 1 in CHCl3). Durch weitere Umkristallisation kann der Schmelzpunkt bis auf 220-222 C erhöht werden.
Aus den Kristallsätionsmutterlaugen können tie fer schmelzende Isomere (F. = z. B. 181-l85 C) gleicher Zusammensetzung isol'ier't werden.
Diese zeig gen ein ähnliches, aber nicht identisches IR-Spektrum. Zum Beispiel is't die Bande bei 11,05 ,u stärker aus gebildet;
dagegen fehlen die schwachen Bariden bei 3,11 und <I>3,22</I> ,u, und die Amid-Doppelbande, ist um 0,1 ,u nach längeren Wellen, -nämlich 6,05 und: 6,15,u verschoben.
Möglicherweise liegt hier das isomere Amid mit der Methylgruppe am Kohlenstöffatom 16 oder an den Kohl'enstoffatomen 16 und 17 vor, oder es handelt sich um das zweite mögliche Umlagerungs- produkt einer Beckmannschen Umlagerung mit der Gruppe -CONHCH3 am Kohlenstoffatom 17.
Zur Abspaltung der O-Acetylgruppe werden 150 mg Amid (II) vom Schmelzpunkt 217 C in, 6 ml Tetrahyd'rofuran gelöst und unter Rühren mit 4 ml in alkoholischer KOH sowie gegebenenfalls mit 4 ml 30 % igem H202 versetzt,
worauf ein weisser Nieder schlag ausfällt. Nach 20 Minuten langem Erhitzen auf dem Wasserbad wird abgesaugt und aus abs. Methanol. umkristallisiert. Zur weiteren Reinigung löst man in Chloroform, filtriert durch eine Säule aus Aluminiumoxyd (neutral, Aktivität III) und kristalli- siert nochmals aus CH30H; F. = 233-234 C.
Sum menformel: C22M3O2N. b) Zu einer Lösung von 1,8 g des Amids (1I) in 60 ml Eisessig gibt man tropfenweise 700 mg Bortri- fluoridätherat in 5 ml Eisessig und erhitzt 3 Stunden unter Rückfluss, worauf sich die Lösung über Braun violett dunkelgrün färbt. Nach dem Erkalten giesst man auf Eis und schüttelt zweimal mit Äther aus.
Dieser Extrakt enthält Etwa 1,2 g des Rohketons (11I), dem je nach Menge des zum Ausschütteln angewand ten Äthers wechsälnde Mengen des gebildeten Oxa- zolins (IV) beigemengt sind. Das Oxazolin erscheint bei der chromatoggraph ischen Reinigung des Roh ketons über .ein synthetisches Magnesiumsilikat, das unter dem Handelsnamen Florisil bekannt ist, in den Benzol-Äther (z.
B. 4 : 1)-Eluaten. c) Zur Isolierung des Oxazol'ins aus dem gemäss b erhaltenen Gemisch wird die bereits mit Äther ex trahierte wässrigsaure Lösung mit NaOH (30%ig) auf p11 9 gebracht und mehrmals mit Äther extra hiert.
Die Ätherauszüge dampft man ein, löst die er haltene basische Substanz in Benzol, filtriert die Lö sung durch eine Aluminiumoxyd-Säule (neutral, Akti vität III), dampft ein, kristallisiert aus Petroläther um und erhält in verdünnter Eisessigsäure unlösliche, in verdünnter Salzsäure dagegen lösliche Kristalle vom Schmelzpunkt 143-144 C.
Das 3ss - Acetoxy - (17, 16-d) - 2' - methyloxazolino - d5 - androsten (IV) zeigt ausser der (Acetyl)-Carbonylbande (5,76,u) ein ähn liches IR-Spektrum wie 2-Methyl-d2-oxazolin. Sum- menformel: 24113503N; [a] D = - 8 (c = 1 in OHCl3).
Das Pikrat des Oxazolins CsoH3sON4 wird in der üblichen Weise mit ätherischer Pikrinsäurelösung her- gestellt. Nach mehrmaligem Umkristallisieren aus Essigester schmilzt es bei 194-l95 C.
d) Aufspaltung des Oxazolinringes durch Ben- zoylierung 50 mg Oxazolin, 0,5 ml Dioxan, 2 m1 Wasser und 55 mg Natriumcarbonat werden in einem Erlenmeyer- kolben mit 0,1 ml Benzoylchlorid versetzt. Nach drei stündigem Rühren -lässt man über Nacht bei Zimmer- temperatur stehen.
Dann werden 20 ml Wasser zu gegeben, und es wird mit Chloroform extrahiert. Die organische Phase wäscht man gründlich mit NaHC03 und Wasser. Nach dem Abdampfen des Lösungsmit tels bleiben 88 mg 3,16-Diace'toxy-16-methyl-17-ben- zoylamino-45-androsten (V) zurück, welches nach dreimaligem Umkristallisieren aus Alkohol-Wasser bei 235-236 C schmilzt. Summenformel:
C31H4105N. (Journ. Am. Chem. Soc. 70, 3872) hergestellten 3,8- Acetoxy - 16 - methyl - 45 - androsten -17 - an keine Schmelzpunktserniedrigung.
Die Benzol-Äther-Eluate des Rohketons enthalten das Oxazolin (IV).
Die Verseifung der 3ständigen Acetylgruppe im Keton (III) kann durch zweistündiges Kochen in mit Kaliumcarbonat gesättigtem 90 % igem Methanol be wirkt werden. Die Isolierung des in Äther löslichen Produktes erfolgt in üblicher Weise. F. = 169-171' C (CH30H/H20) (Lit. Wert = 174-175 C).
Summen formel: C20113002# Aus den Mutterlaugen der Kristallisation können tiefer schmelzende Isomere (z. B. F. = 90-92 C; 111-113 C) der gleichen Zusammensetzung und mit ähnlichem IR-Spektrum wie das oben genannte Keton (11I) isoliert werden. Sie enthalten möglicherweise das mit dem Keton (III)
hinsichtlich der räumlichen Lage der Methyl'gruppe am Kohlenstoffatom 16 isomere Keton.

To split off the O-acetyl group, 150 mg of amide (II) with a melting point of 217 ° C. are dissolved in 6 ml of tetrahydrofuran and, while stirring, 4 ml of alcoholic KOH and, if necessary, 4 ml of 30% H 2 O 2 are added,
whereupon a white precipitate falls. After 20 minutes of heating on the water bath, suction is performed and abs. Methanol. recrystallized. For further purification, it is dissolved in chloroform, filtered through a column of aluminum oxide (neutral, activity III) and crystallized again from CH30H; F. = 233-234 C.

This extract contains about 1.2 g of the crude ketone (11I), to which varying amounts of the oxazoline (IV) formed are added depending on the amount of ether used to shake it out. The oxazoline appears in the chromatographic purification of the crude ketone via a synthetic magnesium silicate, which is known under the trade name Florisil, in the benzene ether (e.g.
B. 4: 1) eluates. c) To isolate the oxazol'ins from the mixture obtained according to b, the aqueous acidic solution already extracted with ether is brought to p119 with NaOH (30%) and extracted several times with ether.
The ether extracts are evaporated, the basic substance obtained is dissolved in benzene, the solution is filtered through an aluminum oxide column (neutral, activity III), evaporated, recrystallized from petroleum ether and obtained insoluble in dilute glacial acetic acid, but in dilute hydrochloric acid soluble crystals with a melting point of 143-144 C.
The 3ss - acetoxy - (17, 16-d) - 2 '- methyloxazolino - d5 - androstene (IV) shows, apart from the (acetyl) carbonyl band (5,76, u), a similar IR spectrum as 2-methyl- d2-oxazoline. Molecular formula: 24113503N; [a] D = -8 (c = 1 in OHCl3).
The picrate of the oxazoline CsoH3sON4 is prepared in the usual way with an ethereal picric acid solution. After repeated recrystallization from ethyl acetate it melts at 194-195 C.
d) Splitting of the oxazoline ring by benzoylation 50 mg of oxazoline, 0.5 ml of dioxane, 2 ml of water and 55 mg of sodium carbonate are mixed with 0.1 ml of benzoyl chloride in an Erlenmeyer flask. After stirring for three hours, it is left to stand overnight at room temperature.
Then 20 ml of water are added and it is extracted with chloroform. The organic phase is washed thoroughly with NaHCO3 and water. After evaporation of the solvent, 88 mg of 3,16-diacetoxy-16-methyl-17-benzoylamino-45-androstene (V) remain, which melts at 235-236 C after three recrystallization from alcohol-water. Sum formula:
