Verfahren zur Herstellung von 17-Acetyl-steroiden Die Herstellung von Steroidverbindungen durch Totalsynthese, welche den Aufbau des Vierringsystems, die Einführung angulä.rer Methylgl@ippen in die Stellungen 10 und 13 und die Anlagerung der gewünschten Substi- tuenten an das Ringsystem .erfordert, stellt eine äusserst schwierige Aufgabe dar, die < furch die komplizierte Stereochemie der Steroidverbindungen noch weiter erschwert wird.
So können gesättigte Steroide mit min destens sechs As@=letrizentren in wenigstens vierundsechzig Modifikationen existieren. Dennoch wurden infolge des therapeutischen Wertes von Steroiden wie Cortison und der gleichen und wegen der Seltenheit der für die Cortisonherstellung verwendbaren Aus gangsstoffe Versuche zur Herstellung dieser Verbindungen durch Synthese unternommen.
Die vorliegende Erfindung hat ein neues Verfahren zur Umwandlung von Poly hydro- plienanthrenen in 1,2-Zyklopentanpolyhydro- phenanthrene zum Gegenstand, welches die Herstellung von Steroiden durch Total synthese ermöglicht.
So lassen sich unter An wendung des neuen Verfahrens unter andern d1-11-Keto-progesteron und d1-11-Hydroxy- progesteron Lind weitere Dodecahydrophen- anthrene herstellen, welche nützliche Zwi schenprodukte für die Gewinnung der ge nannten Progesterone darstellen.
Es wurde nämlich gefunden, dass Poly- hydrophenanthrene, welche im Ring C eine Metha.llylgruppe in Stellung 2 und eine Ca.rboxymethyl- oder veresterte Carboxy- methylgruppe in Stellung 1 aufweisen, durch geeignete Behandlung zum Ringschluss ver- a.nlasst werden können, wobei der D-Ring des Steroidskelettes gebildet wird, welcher in Stellung 17 eine Acetylgruppe trägt.
Das erfindungsgemässe Verfahren zur Herstellung von 17-Acetylsteroiden ist da: durch gekennzeichnet, dass man ein 2-Meth- a,llyl-polyhydrophenanthren, welches in 1- Stellung durch einen Rest der Formel -CH2COOR substituiert ist, worin R Wasserstoff, Alkyl, Aryl oder Ara.lkyl bedeutet, mit einem Re- duktionsmittel in das entsprechende 1-(2-Hy- droxy-äthyl)
- 2 - methallyl - polyhydrophen- a.nthren umwandelt und hierauf einerseits die Hydroxyäthylgruppe mit einem Alkyl-, Aryl- oder Aralkylsulfonylhalogenid in Gegenwart eines tertiären Amins in den Rest der Formel -CH2CH20 - S02R1 überführt, worin R1 Alkyl, Aryl oder Ara:
lkyl bedeutet., und anderseits die Methallylgruppe durch Oxydation und nachfolgende Spaltung in den Aeetonylrest umwandelt, worauf man das erhaltene 2-Acetonyl-l-sttlfonyloxyäthyl- polyhydrophenanthren mit einer Base cycli- siert. Die Reduktion der Verbindungen I zu den Verbindungen II kann z. B. mit Alkali- oder Erdalkalimetallen oder auch mit Alkali- oder Erdalkadi,,Aluminium-Hydriden erfolgen.
Es ist auch möglich, die Reihenfolge der Operationen gegenüber dem Schema in der Weise zu ändern, da.ss die Überführung der Methallylgruppe in die Acetonylgruppe vor der Herstellung des Sulfonatderiva,tes erfolgt. Die im Reaktionsschema dargestellte Reihen folge erwies sich aber als günstiger.
Die als Ausgangsstoff dienenden Poly- hydrophenanthrene können ausserdem in den Ringen A, B und C noch weitere Substituen- ten aufweisen, etwa.
Ketogruppen oder ge schützte Hydroxylgruppen, Zur Herstellung von Steroidverbindungen mit angulären 111e- thylgruppen in den Stellungen 10 und 13 werden in den Stellungen 2 und 4b durch 3Ie- thyl substituierte Polyhydrophenanthrene als Ausgangsstoffe verwendet.
So kann das erfindungsgemässe Verfahren zur Herstellung von dl-11-Keto-progesteroit aus 2,4b-Dimethyl-l-carboxymethyl-2-metha,l lyl-7-äthylendioxyr1,2,3,4,4a,4b,5,6,'7,8,10,10a- dodecahydrophenanthren-4-ol oder aus der entsprechenden, statt der Carboxylgruppe eine veresterte Carboxymethylgruppe aufwei senden Verbindung verwendet. werden. Diese Reaktionen können durch das folgende Schema dargestellt werden:

In diesen Formeln bedeutet R. entweder Wasserstoff oder ein Alkyl-, Aryl- oder Aral- ky lradikal und R1 ein AllZ 1-, Aryl- oder Aralkylradikal.
Nach diesem Schema wird das als Aus- gangsstoff dienende 2,4b@Dimethyl-1-(ca.rboxy- methyl) - 2 methallyl-7-äthy lendioxy-1,2,3,4,4a" 4b@5,6,7,8,10"10a-dodecahydrophenanthren-4: o1 oder die entsprechende Verbindung mit ver- esterter Carboxymethvlgruppe VI zuerst zum entsprechenden primären Carbinol VII redu ziert.
Diese Reduktion gelingt leicht durch Einwirkung von Alkali- oder Erda.lkalimetall- Aluminium-Hydrid oder freiem Metall in einem inerten Reaktionsmedium (unter an derem in Dioxan, Tetrahydropyran, Tetra- hydrofuran, niedrigen Alkoholen und dergle:
i- chen). (Tewöhnlich wird die Reduktion mit Lithiuin- Aluminium-Hy drid in Tetrahy dro- furan bei Zimmertemperatur durchgeführt. Eine kürzere Reduktionszeit ergibt das Arbei ten bei 150 C. Nach beendeter Reaktion kann das gebildete Produkt leicht isoliert werden, indem die mit Wasser versetzte Reaktions mischung filtriert und das Filtrat unter ver mindertem Druck eingedampft. wird.
In gleicher Weise können mich andere in 1-Stellung eine Carboxymethyl- oder ver esterte Cai boxymethylgnippe und in 2-Stel- lung eine hlethallylgruppe aufweisende Do- decahydrophenanthrene zu den entsprechen den 2-Oxyäthylverbindungen reduziert wer den.
Zu dieser Reduktion besonders geeignet sind die niederen Alkylester, wie die Carbo- methoz-yrniethyl-, Ca.rbäthoxymethyl- und Ca.rbopropoxymethylverbindungen.
Im zweiten Schritt werden die Verbin dungen V II mit einem Alky 1-, Ary 1- oder Aralkyl-siilfonylhalogenid in die entsprechen den Sulfonsäurederivate VIII verwandelt.
Gewöhnlich wird die Bildung .des Toluiolsulfo- nylderivates durch Reaktion von 2,4b-Dime- thy1 -1- (2-hydroxy,-äthyl)-2-metha.llyl-7-ä,thy- lendioxy -1,2;3,4,4a,4b;5;6,7,8,10,;
lOa-dodecahy. drophenanthren-4-ol V II mit p-Toluolsulfo- ny lchlorid in Gegenwart von Pyridin bevor zugt., weil dabei unter optimalen BedingiLn- gen maximale Ausbeuten an Sulfonylderiv at erhalten werden können. Es können aber auch andere Sulfonylhalogenide zur Herstel lung der entsprechenden Sulfonsäurederivate verwendet werden, z.
B. 1Tethansulfony-1- halogenid oder Benzolsulfoliylhalogenid. Als Reaktionsmedium können statt des Pyridins auch andere tertiäre Amine, wie Picolin, Tri- methylamin und dergleichen, verwendet wer den. Nach beendeter Reaktion kann der ge bildete Sulfonsäureester isoliert werden, indem man die Reaktionsmischung mit Wasser verdünnt und die wässeAge Lösung mit einem wasseiiinlöslielien Lösungsmittel, z. B. Äther, extrahiert.
Der H-"- droxylsubst.ituent in 4-Stellung kann, wenn gewünscht, auf dieser Stufe des Verfahrens oxydiert werden, wobei die ent sprechende 11-Ketoverbindung XII entsteht. Es wurde gefunden, dass, sich diese Oxyda tion leicht mit. Hilfe des Chromsäurea.nliy- drid-Pyridin-Komplexes bewirken lässt. Das oxydierte Produkt. kann aus der mit Wasser verdünnten Reaktionsmischung durch Extrak tion mit .einem wasseiai.nlösliehen Lösungsmit tel, etwa. Äther, und Eindampfen des Ex traktes unter vermindertem Druck isoliert.
werden. Der erhaltene Rückstand kann durch Chromatographie an mit Säure gewaschenem Aluminiumoxyd weiter gereinigt werden.
Im nächsten Schritt wird die Methallyl- seitenkette bei C-2 der Verbindungen VIII und XII durch Oxydation und anschliessende Spaltung in ein Acetonylradikal übergeführt und so die Verbindunoen IX bzw. XIII er halten. Diese Oxydation kann auf verschie dene Arten geschehen, z.
B. indem die Ver bindung VIII oder XIII mit Osmium- tetroxy d zur Reaktion gebracht und der ge bildete Osmiatester unte=r Bildung des ent sprechenden Glykols zersetzt. wird, woraus sich durch Behandlung mit einem glykolspal- tenden Mittel, wie Perjodsäure oder Bleitetra aeeta.t die Acetonylverbindung gewinnen lässt.
Eine andere Möglichkeit zur Herstellung der acetonylsubstituierten Dodecahydrophena.n- threnverbindung bestellt im Ein rirkenlassen von Ozon auf die Verbindungen VIII oder XII und Zersetzung der gebildeten. Ozonide mit Hilfe eines Reduktionsmittels.
Anderseits ist es auch möglich, die Ver bindung VII bzw. die entsprechende 4-Keto- verbindung zuerst in die Acetonylverbindun- gen überzuführen und diese dann mit Sulfo- nyllialogenid in die Verbindungen IX bzw. XIII zu verwandeln.
Der Ringschluss der Verbindungen IX und XIII wird bewirkt, indem man diese Verbindungen mit einer Base behandelt. Als solche kommen in Frage: Triäthylamin, Al- kalimeta.lle, Alkali- und Erdalkalialkoxyde und Alkalihydride (z. B. Natrium-Ka.lium, Natriummethyla.t, Kaliiun-tert.-butylat, Na triumisopropy lat, l@?atiiumhydrid und der gleichen).
Die Reaktion wird üblicherweise in einem Lösungsmittel, wie einem niederen Alkohol, Benzol, Toliiol oder Äther durch geführt. Eine bevorzugte Form dieser Ring- schlussreaktion besteht in der Behandlung der Verbindung IX oder XIII mit Natrium- oder Kaliumalkoholat in einem niederen Al kohol, da. diese Reagentien leicht erhältlich sind Lind deren Verwendung eine gute Aus beute an gewünschter Steroidverbindung er gibt.
Gewöhnlich wird der R.ingsehluss durch- geführt, indem die Verbindungen IX oder XIII mit Natriummethylat in methanolischer Lösung bei Zimmertemperatur zur Reaktion gebracht werden.
Die gebildete Steroidverbin- dung kann isoliert werden, indem die Reak- tionsmisehung mit Wasser verdünnt und hier auf das gewünschte Produkt mit einem wasseiainlöslichen Lösungsmittel, wie Chloro form, extrahiert und der Extrakt. unter ver mindertem Druck eingedampft wird. Das so erhaltene Produkt kann durch Chroma.t.ogra- phie an säuregewaschenem Aluminiumoxyd weiter gereinigt werden.
Bei dieser zum Ringsehluss und zur Bil dung der gewünschten Steroidverbindung führenden Reaktion werden anscheinend pri mär die Isopregnenverbindungen XA oder XIV A- gebildet. Durch weitere Behandlung mit.
Alkali lassen sieh diese leicht zur ge wünschten normalen Pregnenverbindung epi- nierisieren.

Die so erhaltenen Pregnenvembindungen XIVB und XB können zur Abspaltung der blockierenden Äthylendioxygruppe und Re generierung der 3-Ketogruppe mit Säure hydrolysiert werden, wobei die Verbindungen XV (d1-11-Keto-progesteron) und XI (d1-11 Hydroxy-progesteron) entstehen, in welchen die Doppelbindung in die 4,
5-Stellung ver schoben ist. Natürlich kann man auch die Zwischenprodukte auf gleiche Art hydroly- si.eren und in die entsprechenden Ketoverbin- dungen umwandeln. Diese Hydrolyse kann z. B. mit Salzsäure, Perchlorsäure, p-Toluol- sulfonsäure und .dergleichen erfolgen.
Weiter kann die Verbindung XI (d1-11-Hydroxy- progesteron) durch Oxydation, beispielsweise durch Behandlung mit Chromsäureanhydi4d- Pyridin-Komplex, in d1-11-Keto-progesteron XV umgewandelt werden.
Die erfindungsgemäss hergestellten Ver bindungen sind auf dem Gebiete der Pharma- ceutica von Nutzen und dienen als Zwischen produkte bei der Herstellung von Nebennie- renrindenhormonen, wie 11-Dehydrocortico- steron (Kendalls Verbindung A) Cortison, Hydrocortison und dergleichen.
Man kann natürlich auch Verbindungen verwenden, die an Stelle der Äthylendioxyd- grupp.e andere leicht durch Hydrolyse in die Ketogruppe umwandelbare Substituenten aufweisen, also etwa die Enoläther oder an dere Ketale.
IndenfolgendenBeispielen sindjeweilsnur ,einzelne Stufen des Verfahrens beschrieben. Beispiel <I>1</I> 2,4b-Dimethyl-1-(2-hydroxy-äthyl)-2-meth- allyl- 7- äthylendioxy-1, 2, 3,.4,4a,4b,5,6,7,8,10, 10a-dodecahydrophenanthren-4-ol VII. Eine Lösung von 2,58 g der bei 157 bis l58 C schmelzenden stereoisomeren Form von 2,4b-Dimethyl-1- (1-carboxymethyl)
-2-methal lyl@7-äthylendioxy-1,2,3,4,4a,4b,5;6,7,8,p,10a- dodecahydrophenanthren-4-ol in 200 em3 ab solutem Tetra.hydrofuran wurde mz einer Lö sung von 0,80g Lithium-Aluminium-Hydrid in 8 em3 Tetrahydrofuran gegossen. Die Mi- sehung wurde während 20 Stunden gerührt und dann der Überschuss an Hydrid mit Wasser versetzt.
Die Reaktionsmischung wurde filtriert, getrocknet und im Vakuum eingedampft, wobei ein kristalliner Rückstand von 2,4b-Dimethyl-1-(2-hydroxy-äthyl)-2-meth- allyl- 7-äthylendioxy -1,2h 3,4,,4a.,4b,,5,6,7,8,10, 10a - dodecahydrophenanthren - 4 - ol zurück blieb. Das Produkt wurde aus Benzol umkri stallisiert, wobei es sich zeigte, da.ss es in zwei kristallisierten Modifikationen existiert, die bei 200-201 C und bei 210-211 C schmel zen.
Eine Lösung von 10 mg der bei 210 bis 211 C schmelzenden Modifikation in 1 em3 Tetrahydrofuran und 0,5 cm3 3-molarer Per- ehloraäure wurde 4 Stunden bei Zimmertem peratur stehengelassen und dann mit wässe riger Kaliumbicarbonatlösung neutralisiert.
Das Produkt, 2,4b-Dimethyl-1-(2-hy droxy- äthyl) i,2s-methallyl-7@keto-1,2,3,4,4a.,4b,5,6,7,8, 10,10a-dodecahydrophenanthren-4-ol mit dem Smp. 153-155 C wurde durch Extraktion mit Chloroform, Eindampfen des Extraktes und Umkristallisation des Rückstandes aus Äthylaeetat erhalten.
<I>Beispiel 2</I> 2,4b-Dimethyl-l-(2-hydroxy-ä:thyl)-2-meth- allyl-7--äthylendi;oxy-1,2, 3, 4, 4a.,4b, 5,6,7,8,10, 10a-dodecaliydrophenanthren-4-ol VII.
Eine Lösung von 2,54 g 2,4b-Dimethyl-l- carboxymethyl-2'-methallyl-7-äthylendioxy-1,2, 3,4,4a,4b,5,6,7,8,10,10a - dodeeahydrophenan- thren-4-ol, Smp. 250-251 C, in 500 ein absolutem Tetrahydrofuran wurde zu einer Lösung von 2,5g Lithium-Aliuninium-Hydiid in 25 em3 Tetrahydrofuran gegeben und die Mischung unter Rühren 30 Minuten am Räek- fluss erhitzt.
Dann wurde die Reaktions- mischuni,- mit Wasser versetzt und das Pro dukt auf die im Beispiel 1 angegebene Weise isoliert.
<I>Beispiel 3</I> 2,4b-Dimethyl-l-(2-tosyloxy-äthyl) 2-meth- allyl- 7 - äthylendioxy-1, 2,3,4,4a, 4b,5,6,7,8,10, 10a-dodecahydrophenanthren-4-ol VIII.
Zu einer wasserfreien Lösung von 302 ing 2,4b-Dimethy 1- l- (2-h.#-droV-äthy l) -2-metIial- lytr- 7 -äthy lendioxy-1,2,3,4,4a.,4b,5"6,7,8,10,10a, dodecahydrophenanthren-4-ol VII in Pyridin wurden 168 mg reines p-Toluol-sulfonylchlo- rid zugesetzt. Die Mischung blieb etwa- 20 Stunden bei Zimmertemperatur stehen,
worauf der Übersehuss an Tosylchlorid durch Zusatz einiger Tropfen wässeriger Natrium- bica.rbonatlösimg zersetzt wurde. Die Lösung -wurde hierauf mit Wasser verdünnt und mit Äther extrahiert. Der Extrakt wurde im Va-. kuum zur Trockne eingedampft.
Aus dein kristallinen Rückstand wurde reines, bei 157 bis 158" C schmelzendes 2,4b-Dimethyl-1-(2- tosy loxy- äthyl) - 2 - methallyl-7-äthylendioxy- 1,2,3,4,4a,4b,5, 6, 7, 8,10,10a - dodecahy drophen- anthren-4-ol VIII durch fraktionierte Kri stallisation aus einem Benzol-Petrolät.her- Gemisch erhalten.
Ein anderer Versuch unter Verwendung von 2,47 g Verbindung VII und 1,48 g p-To- lu.ol-sulfonylehlorid wurde bei 0 C und einer Reaktionsdauer von etwa. 20 Stunden erfolg reich durchgeführt.
<I>Beispiel 4</I> 2,4b-Dimethyl-1- (2-tosy loxy-äthyl) -2-meth- ally 1-.7 -äthy lendioxy-1, 2, 3, 4, 4a,4b, 5,6, 7,8,10, lüa-dodecahydropheilanthren-4-on XII.
160 mg 2,4b - Dimethyl -1- (2 - tosyloxy- äthyl) s-2,methallyl-7-ütliy lendioxy-1,2,3,4,4a, 4b,5,6,7,8,10,10a-dodeeahy dr opheilanthren-4-ol in 1 em3 wasserfreiem Pyridin wurden zu einer Lösung von 160 mg Chromsätireanhy- drid in 1 em3 Pyridin gebracht.
Die Misclning blieb 16 Stunden bei Zimmertemperatur stehen, wurde dann mit Wasser verdünnt und mit Äther extrahiert. Der Extrakt wurde mit Äther gewaschen, getrocknet und im Va kuum vollständig eingedampft. Der in einer Benzol-Petroläther-Mischung gelöste, kristal line Rückstand wurde an sauergewaschenem Aluminiumoxyd chroinatographiert. Eluation mit Äther ergab reines 2,4b-Dimethyl-1-(2- tosyl ox""(- äthyl )
-- 2 - methallyl - 7- ä.thylendioxy- 1,2,3,4,4a,:5,6,7,8,10,l0a - dodecahydrophenan - thren-4-on XII mit dem Schmelzpunkt 156 bis 158 C. Beispiel <I>5</I> 2,4b-Dimethy 1-1- (2-tosyloxy-äthyl)-2-ace- tony 1- 7 - ätliylendioxyl-1,2,3,4,4a,4b,5,6,7,8,10, 10a-dodecahy drophenanthren-4-oii XIII.
Zu 445 mg in 5 em3 Benzol gelöstem 2,4b- Diinetbyl,-l-('2,-tosyloxy-äthyl) 9-metha.llyl-7- ätliyIendiox-, -1,2,3,4,;4a,4b,5,6,7,8,10,10a@-dode- cahydrophenanthren - 4 - an wurden 208 mg Osmiumtetroxy d gegeben. Die Lösung blieb eine Stunde bei Zimmertemperatur stehen. Nun wurden 7 cm3 Äthanol und eine Lösung von 0,7g Natriumsulfit in 4 cm3 Wasser zu gesetzt und die Mischung 20 Minuten ge schüttelt.
Die obere, organische Schicht wurde dann dekantiert und filtriert, die untere Schicht mit Äthanol gewaschen. Der Alkohol wurde filtriert und mit der obern Schicht vereinigt. Die das gewünschte Produkt ent haltende Benzol-Alkohol@Lösung wurde im Vakuum auf einen Zehntel ihres Volumens eingedampft, das Konzentrat hierauf mit Wasser verdünnt und mit Chloroform ausge zogen.
Durch Eindampfen im Vakuum wurde aus dem gewaschenen und getrockneten Extrakt 2,4b-Dimethyl-1- (2-tosyjoxy-ät.hyl)- 2- (2,3-dihy droxy - 2 -,methyl-propyl) -7-äthy len- dioxy-'1,2,3,4,4a,4b;5,;6,7,8,10,10a: dodecahydro- phenanthren-4-on als fester, amorpher Rück stand erhalten.
Dieser wurde in 4 em3 Methanol und 1 cm3 Pyridin gelöst Lind der Lösung 250 mg Perjodsäure in 0,5 em3 Wasser zugesetzt. Nach 30minutigem Stehen bei Zimmertempe ratur wurde mit Wasser verdünnt und mit Chloroform extrahiert. Der organische Ex- ti-akt wurde gewaschen, getrocknet Lind im Vakuum zu einem 6I eingedampft..
Durch Chromatogra.phie an saii,ergewaschenem Alu miniumoxyd und Eluierung mit Äther wurde das gewünschte Produkt, 2,4b-Dimethyl-1-(2- tosyloxy-'äthyl;) , 9-" acetonyl,7-äthylendi!oxy- 1,2,3,;
4@4a,4b,5,6, 7,8,10,.10a - dodecahydrophen - anthren-4-on XIII mit dem Schmelzpunkt 104--108 C (unter Zersetzung) erhalten.
Die Behandlung mit Osmiumtetroxyd kann mit gutem Erfolg auch in einer 9 :1 Benzol-Tetrahydrofuran-Lösung mit 7,84 g Verbindung XII und 4,8 g Osmiumtetroxyd durchgeführt werden.
Beispiel <I>6</I> d5-3 - Äthylendioxy -11,20 - diketo-17-iso- pregnen RIVA.
Zu einer Lösung von 80 mg 2,4b-Dimethyl- 1-(2-t.osyloxy-äthyl) \2-a.cetonyl-7-äthylendioxy- 1,2,3,4,4a,4b,5,6,7,8,10,10a - dodecahydrophen- ant.hren-4-on in 1 cm3 Methanol wurden 0,08 em3 einer doppeltnormalen Lösung von Natriummethylat in Methylalkohol gegeben.
Die Mischung blieb im geschlossenen Kolben eine Stunde bei Zimmertemperatur stehen, worauf sie mit Wasser verdünnt -find mit Chloroform extrahiert wunde. Der Chloro- formext.rakt wurde gewaschen, getrocknet und im Vakuum eingedampft.
Der kristalline Rückstand wurde an Aluminiumoxyd chro- matographiert. Die Elution mit Äther-Petrol- ä.ther-Misehimg ergab 45-3-Äthylendioxy- 11,20-diketo-17-isopregnen RIVA, Smp. 121 bis 125 C.
<I>Beispiel</I> A5-3-Äthylendioxy-11,20 - diketo-17-iso- pregnen RIVA.

Der Chloroformextrakt wurde gewaschen, getrock net und unter vermindertem Druck zu einem kristallinen Rückstand eingedampft. Das kri stalline Material wurde in einer Benzol-Pe- troläther-Uischung gelöst, an sauer gewa schenem Aluminiumoxyd chromatographiert und mit Äther-Petroläther eluiert. Aus dem Eluat konnte kristallines 45-3 ,@t.hylendioxy- 11,?0-diketo-isopregnen XIVB erhalten wer den.

Eine Lösung von 165 mg A5-3-Äthylen- dioxy-11,20-diketo-17-isopregnen in 5 ein, Benzol und 2 em3 Methanol wurde mit 3 Gina 2n-Natriummethylat in Methanol vermischt. Nach zweistündigem Stehen bei Zimmer temperatur wurde die Mischung mit Wasser v errniseht und mit Chloroform extrahiert. Der Chloroformextrakt wurde gewaschen, ge trocknet und im Vakuum zu einem kristalli sierten Rücl-,stand eingedampft,
woraus durch Chromatographie in der beschriebenen Weise 45 - 3 - Äthy lendioxy -<B>11,220</B> - diketo - pregnen XIVB mit dem Smp. 181,0-182,5 C gewon nen werden konnte.
Die gleiche Epimerisation wurde auch unter Verwendung von Kaliumearbonat statt des Natiiummethylates durehgeführt.

To a solution of 80 mg of 2,4b-dimethyl-1- (2-tosyloxy-ethyl) r2-acetonyl-7-ethylenedioxy-1,2,3,4,4a, 4b, 5,6,7,8 , 10,10a-dodecahydroplienanthren-4-one in 1.5 cm3 of benzene was poured 0.15 cm3 of a molar solution of potassium tert-butoxide in tertiary butanol. After stirring for 20 hours at room temperature, the mixture was diluted with water and extracted with chloroform.
The chloroform extract was washed, dried and evaporated under reduced pressure to a crystalline residue. The crystalline material was dissolved in a benzene-petroleum ether mixture, chromatographed on acid-washed aluminum oxide and eluted with ether-petroleum ether. Crystalline 45-3, @ t.hylenedioxy-11,? 0-diketo-isopregnene XIVB could be obtained from the eluate.

A solution of 165 mg of A5-3-ethylene-dioxy-11,20-diketo-17-isopregnene in 5 a, benzene and 2 em3 of methanol was mixed with 3 gina of 2N sodium methylate in methanol. The same epimerization was also carried out using potassium carbonate instead of sodium methylate.
