Verfahren zur Herstellung einer 17 -Methyl-pregnenverbindung. .Es wurde gefunden, dass man zu einer neuen 17-lIetlivl-pregnenverbindung gelangen kann, wenn man ein d5-3-Acyloxy-17a-niethyl- ätio-cliolensäurehalogenicl mit einer Methyl- Metallverbindung reagieren lässt und das er haltene Umsetzungsprodukt nach Verseifung der Aeyloxygruppe in 3-Stellung zum Ai-17a- Methyl-3,
20-dioxo-pregnen (17a-lIethpl.proge- steron) oxydiert.
Das neue Verfahrensprodukt, das 4-1-17a- Methyl-3,20-dioxo-pregnen (17a-Methyl-proge- steron) vom F. = 129-1.30 , ist progestativ hochwirksam. Es soll als lIeilmittel oder als Zwischenprodukt, zur Herstellung von Heil- mitteln finden.
Als Methyl-Metallverbindung verwendet man z. B. eine solche des Magnesiums, Kad miums oder Zinks. Das durch anschliessende Verseifung erhaltene 45-3-Oxy-l7a-methyl-20- oxo-pregnen wird durch Oxydation, beispiels weise mittels Chromsäure in Eisessig, oder durch ein dehydrierendes Mittel, wie Erhitzen mit Kupferpulver, Einwirkung eines Metallalkolio- lats bzw.
-plienolats in Gegenwart eines Ketons wie Aceton oder Cyclohexanon, in das 17a- Methylprogesteron übergeführt. Beispiel:
.'3,5 Gewichtsteile 4 5- 3ss-Acetoxy-1. 7 a- methyl-ätio-eliolensätirechlorid (F. = 137 bis 110(1) werden in 100 Volumteilen Äther gelöst und tropfenweise einer Lösung von Diniethyl- Cadmium (bereitet in üblicher Weise aus 7,6 Gewichtsteilen Magnesittmspänen, 100 Volum- teilen Äther, 30 Gewichtsteilen Methylbromid und 28 Gewichtsteilen Cadmiumchlorid) in Äther zugefügt.
Das Reaktionsgemisch wird anschliessend 1 Stunde unter Rühren zum Sieden erhitzt, dann vorsichtig mit verdünnter Essigsäure zersetzt und mit Äther ausgezogen. Die ätherische Schicht wird mit Wasser und Natriumhydrogenearbonatlösung gewaschen, getrocknet und eingedampft. Das Rohprodukt wird in einem Gemisch von 50 Volumteilen Methanol und 3,0 Gewichtsteilen Kalium hydroxyd 12 Stunden bei 20 aufbewahrt. Das Reaktionsgemisch wird mit Wasser verdünnt, mit Äther ausgezogen und in üblicher Weise aufgearbeitet. Nach zweimaligem Umkristalli- sieren des Rohproduktes aus Aceton werden 2,1.
Gewichtsteile reines 45-3ss-OxT>-17a-methyl- 20-oxo-pregnen vom F. = 180-182 erhalten.
1 Gewichtsteil45-3ss-Oxy-17a-methyl-20- oxo-pregnen wird in einem Gemisch von 35 Voluinteilen Benzol und 20 Volumteilen Toluol gelöst, dann mit 5 Volumteilen Cyclohexanon und 11;2 Gewichtsteilen Aliiminiuni-tert.-but@-- lat versetzt und 1.5 Stunden am Rüekfluss ge kocht. Die Reaktionslösung wird in verdünnte Schwefelsäure gegossen und der ätherische Extrakt in analoger \'eise, wie im vorangehen den Absatz angegeben, aufgearbeitet.
Das Roh produkt reinigt man chromatographiseli. Das erhaltene 17(c-llethyl-progesteron der Formel
