Verfahren zur Herstellung von trans-Oestradiol aus eis-Oestradiol. Es ist bekannt, dass bei der Reduktion von ketogruppenhaltigen Sexualhormonen zu den entsprechenden Alkoholen, wie zum Bei spiel bei der Reduktion des Oestrons, des Dehydroandrosterons, des trans-Androste- rons und dergleichen, ein Gemisch isomerer Alkohole entsteht,
das sich nur durch die Stellung der durch Reduktion gebildeten OH- Gruppe am Kohlenstoffatom 17 unterschei det. So entstehen zum Beispiel aus dem Oestron sowohl das cis-Oestradiol, in dem die OH-Gruppe am Kohlenstoffatom 17 und die benachbarte Methylgruppe in eis-Stellung zueinander stehen, als auch das trans-Oestra- diol, in dem diese Gruppen in trans-Stellung zueinander angeordnet sind.
Durch geeignete Auswahl der Reduktionsbedingungen gelingt es zwar, das Gleichgewicht der Reaktionspro dukte zugunsten des einen oder des andern Isomeren zu verschieben. Es ist aber nicht möglich, die Bildung des einen Isomeren voll ständig zu unterbinden. Da sich nun die bei den Isomeren in ihren physiologischen Eigen- schaften wesentlich unterscheiden (so weist zum Beispiel das cis-Oestradiol nur etwa ein Zehntel der Wirksamkeit des trans-Oestra- diols auf),
bedeutet diese bisher unvermeid bare Bildung des weniger wirksamen Isome- ren einen Verlust an physiologischen Einhei ten. Verfahren, diesen Verlust zu vermeiden oder zu verringern, besitzen daher grosse tech nische Bedeutung.
Es ist auch bekannt, die Alkoholgruppe in 17-Stellung der Cyclopentanopolyhydro- phenanthrenderivate durch Oxydation in die Betongruppe überzuführen.
Es wurde nun gefunden, dass man zu einer erhöhten Ausbeute an trans-Isomeren gelangt, wenn man entsprechend vorliegender Erfin dung ein rohes, vorwiegend cis-Oestradiol ent haltendes cis-trans-Oestradiolgemisch unter intermediärem Schutz der phenolischen Hy- droxylgrupl:

<tb> cis-Oestradiol Für die Oxydation eignen sich alle Oxy dationsmittel, die eine sekundäre Alkohol gruppe in eine getogruppe überzuführen ver mögen. Als besonders zweckmässig hat sich dabei Chromsäureanhydrid erwiesen.
Zum intermediären Schutz der phenoli- schen Hydroxylgruppe während der Oxyda tion kann man diese in eine Gruppe umwan deln, die man durch Hydrolyse leicht wieder in die Hydroxylgruppe überführen kann, zum Beispiel durch Überführung in die an organischen oder organischen Ester oder Äther und dergleichen. Als besonders vorteil haft hat sich dabei die Benzoylierung der phenolischen Hydroxylgrruppe im Oestradiol erwiesen.
Beispiel <I>1:</I> 0,36 g eines cis-trans-Oestradiolgemisches, das vorwiegend cis-Oestradiol enthält, wer den nach Schotten-Baumann benzoyliert. Das benzoylierte Produkt wird in 100 cm' Eis essig gelöst und mit 106 mg Chromsäure (1,2 Mol) in 100 cm@ Eisessig tropfenweise bei Zimmertemperatur versetzt. Man lässt über Nacht stehen. Dann giesst man das Reak tionsgemisch in die fünffache Menge Wasser Die folgenden Formelbilder sollen das Ver fahren an Hand des Oestradiols näher erläu tern und äthert aus.
Der Äther wird mit Soda und Wasser gewaschen und anschliessend getrock net. Der Ätherrückstand schmilzt bei 200 bis 210 . Man kristallisiert ihn aus Alkohol um und erhält in einer Ausbeute von 70-80 das reine Oestronmonobenzoat. Dieses Pro dukt wird in Propylalkohol mit metallischem Natrium unter Erwärmen auf dem siedenden Wasserbade verseift und gleichzeitig zum Oestradiol hydriert. Das Hydrierungsgemisch wird nach beendeter Reaktion mit verdünnter Schwefelsäure angesäuert und erschöpfend mit Äther extrahiert. Der Ätherauszug wird getrocknet und durch Abdestillieren vom Lö sungsmittel befreit.
Der hinterbleibende Rück stand wird aus Methanol umkristallisiert, wobei man 85 % der Ausbeute als reines trans- Oestradiol mit den aus der Literatur bekann ten physikalischen Konstanten erhält. Das daneben anfallende cis-Oestradiol wird er neut in den Prozess zurückgeführt.
<I>Beispiel 2:</I> Man verfährt nach Beispiel 1 bis zur Ge winnung des reinen Oestronmonobenzoats. Dieses wird bei 60-80 in Methanol gelöst und in Gegenwart von Nickelkatalysator nach Raney im. eIgeschlossenen Gefäss mit Wasserstoff bis zur Aufnahme von 1,1 1VIol Wasserstoff geschüttelt, wobei man die Tem peratur von 60-80 aufrechterhält. Nach Unterbrechung der Wasserstoffaufnahme kühlt man auf Zimmertemperatur ab, filtriert den Katalysator ab und entfernt aus dem Fil trat das Lösungsmittel durch Eindampfen im Vakuum.
Der hinterbleibende Rückstand, das Oestradiolmonobenzoat, wird mit alkoho lischer Kalilauge verseift. Aus der Versei- fungsflüssigkeit lässt sich nach Ansäueirn mit verdünnter Schwefelsäure das Oestradiol aus- äthern, das, wie in Betspiel 1 beschrieben, in trans,
- und cis-Oes@tradiol getrennt wisrd, wonach man :letzteres erneut dem Verfahren unterwirft.
Process for the production of trans-estradiol from cis-estradiol. It is known that the reduction of sex hormones containing keto groups to the corresponding alcohols, such as the reduction of oestrone, dehydroandrosterone, trans-androsterone and the like, results in a mixture of isomeric alcohols,
which differs only in the position of the OH group formed by reduction on carbon atom 17. For example, both the cis-oestradiol, in which the OH group on carbon atom 17 and the adjacent methyl group are in the cis position, and the trans-oestradiol, in which these groups are in the trans position, arise from the oestrone are arranged to each other.

It is also known that the alcohol group in the 17-position of the cyclopentanopolyhydrophenanthrene derivatives can be converted into the concrete group by oxidation.
It has now been found that an increased yield of trans isomers is obtained if, in accordance with the present invention, a crude cis-trans-estradiol mixture containing predominantly cis-oestradiol with intermediate protection of the phenolic hydroxyl groups is used:

For the intermediate protection of the phenolic hydroxyl group during the oxidation, it can be converted into a group which can easily be converted back into the hydroxyl group by hydrolysis, for example by conversion into organic or organic esters or ethers and the like. The benzoylation of the phenolic hydroxyl group in the estradiol has proven to be particularly advantageous.
Example <I> 1 </I> 0.36 g of a cis-trans-oestradiol mixture, which contains predominantly cis-oestradiol, who benzoylated according to Schotten-Baumann. The ether is washed with soda and water and then dried. The ether residue melts at 200 to 210. It is recrystallized from alcohol and the pure oestrone monobenzoate is obtained in a yield of 70-80. This product is saponified in propyl alcohol with metallic sodium while warming on a boiling water bath and at the same time hydrogenated to estradiol. After the reaction has ended, the hydrogenation mixture is acidified with dilute sulfuric acid and extracted exhaustively with ether. The ether extract is dried and freed from the solvent by distilling off.
The remaining residue is recrystallized from methanol, 85% of the yield being obtained as pure trans-estradiol with the physical constants known from the literature. The cis-oestradiol that is also produced is fed back into the process.
<I> Example 2 </I> The procedure of Example 1 is followed until the pure oestron monobenzoate is obtained. This is dissolved in methanol at 60-80 and in the presence of a nickel catalyst according to Raney im. Shaken closed vessel with hydrogen until the absorption of 1.1 1VIol hydrogen, the temperature of 60-80 being maintained. After interruption of the uptake of hydrogen, the mixture is cooled to room temperature, the catalyst is filtered off and the solvent is removed from the fil by evaporation in vacuo.
The residue that remains, the estradiol monobenzoate, is saponified with alcoholic potassium hydroxide solution. After acidification with dilute sulfuric acid, estradiol can be extracted from the saponification liquid, which, as described in Betspiel 1, is expressed in trans,
- and cis-Oes @ tradiol separately wisrd, according to which one: submits the latter to the procedure again.
