Verfahren zur Darstellung eines neuen Benzolsulfonylguanidins. Gegenstand des vorliegenden Patentes bil det ein Verfahren zur Herstellung eines neuen Benzolsulfonylguanidins, welches dadurch gekennzeichnet ist, dass man ein Benzolsulfo- iivl-laurylguanidin, das in p-Stellung einen durch Hydrolyse in die Aminogruppe über- fÜhrbaren Substituenten aufweist, mit hydro- lysierenden Mitteln behandelt.
Das so gewonnene p-Amino-benzolsulfo- nyl-lauryIguanidin schmilzt bei 96-97 .
Die neue Verbindung soll therapeutische Verwendung finden.
DasBenzolsulfonyl-laurylguanidin, das in p-Stellung einen durch Hydrolyse in die Aminogruppe überführbaren Substituenten ,veist, kann auf verschiedene Art und <B>i</B> iuf- Weise gewonnen werden. So kann man z. B. auf ein entsprechendes Benzolsulfonsäure- ha.logenid Laurylguanidin einwirken lassen.
Vorteilhaft ist ferner die Umsetzung von Laurylamin mit einem geeigneten S-substi- tuierten Benzolsulfonyl - pseudo - thioharn- ctoff, der z. B. durch Reaktion eines S-sub- slituierten pseudo-Thioharnstoffes mit einem Benzol;snlfonsänrehalogenid erhalten wird.
<I>Beispiel 1:</I> 40 Teile Laurylguanidin - bromhydrat werden in 150 Teilen Aceton gelöst und unter Rühren mit 13,5 Volumteilen lOn- Natronlauge versetzt. Dann werden bei 5 bis 10 33 Teile p-Acetylamino-benzolsulfochlo- rid und gleichzeitig weitere 13,5 Volumteile 10n-Natronlauge zugegeben.
Nach Istündi- gem Rühren bei Zimmertemperatur wird mit 600 Teilen Wasser verdünnt und der ent standene Niederschlag abgenutscht. Der noch feuchte Filterkuchen wird mit 150 Volum- teilen 17 % iger Salzsäure 1 Stunde lang ge kocht, wobei sich das Chlorhydrat des p-Amino-benzolsulfonyl-laurylguanidins ölig abscheidet.
Dieses wird durch Neutralisieren mit Natronlauge unter Kühlen zersetzt und die erhaltene Kristallisation abgenutscht. Man gewinnt so p-Amino-benzolsulfonyl- laurylguanidin, das aus Essigester umkristal lisiert bei 96-97 schmilzt.
<I>Beispiel 2:</I> Man versetzt die Lösung von 37 Teilen Athyl-pseudo-thioharnstoff-hydrobromid in 75 Teilen trockenem Pyridin in Portionen mit 47 Teilen p-Acetylamino-benzolsulfo- chlorid. Unter Selbsterwärmung erhält man eine Lösung, die man noch 1 Stunde auf dem kochenden Wasserbad erwärmt und dann in Wasser eingiesst.
Der dabei sich ausschei dende N- [p-Acetylamino-benzolsulfonyll -S- äthyl-pseudo-thioharnstoff der Formel

wird abgenutscht und mit Wasser gewaschen. Man kann ihn durch Umkristallisieren aus Alkohol reinigen. F. 182 . 301 Teile p-Acetylamino-benzolsulfonyl- S-äthyl-pseudo-thioharnstoff werden mit 185 Teilen Laurylamin 8 Stunden im Ölbad von 180 erwärmt.
Es entweicht Athylmereaptan. Die erhaltene Schmelze wird mit 1000 Tei len 1Mger Salzsäure gekocht, wobei sich ein schweres 01 bildet. Dieses ist das Hydro- chlorid des p-Amino-benzolsulfonyl-lauryl- guanidins. Es wird von der Säure abge trennt, mit heissem Wasser verrührt und die dabei freiwerdende Salzsäure mit Natron lauge neutralisiert. Man erhält eine halbfeste Masse,
die in wenig Essigester warm gelöst wird. Die Lösung wird mit geglühter Pott asche getrocknet, filtriert und längere Zeit bei Zimmertemperatur stehengelassen. Nach 1-2 Tagen erhält man eine schöne Kristal- lisation von p-Amino-benzolsulfonyl-lauryl- guanidin, die abgenutscht und eventuell mehr mals aus Essigester umkristallisiert wird. F. 96-97 .

The p-amino-benzenesulfonyl-laury-Iguanidine obtained in this way melts at 96-97.
The new compound should find therapeutic use.
The benzenesulfonyl-laurylguanidine, which in the p-position has a substituent which can be converted into the amino group by hydrolysis, can be obtained in various ways. So you can z. B. Let act on a corresponding benzenesulfonic acid ha.logenid Laurylguanidine.
It is also advantageous to react laurylamine with a suitable S-substituted benzenesulfonyl - pseudo - thiourine ctoff, the z. B. by reaction of an S-substituted pseudo-thiourea with a benzene; snlfonsänrehalogenid is obtained.
Example 1: 40 parts of laurylguanidine bromide hydrate are dissolved in 150 parts of acetone, and 13.5 parts by volume of 10N sodium hydroxide solution are added while stirring. Then at 5 to 10 33 parts of p-acetylamino-benzenesulfochloride and at the same time a further 13.5 parts by volume of 10N sodium hydroxide solution are added.
After stirring for 1 hour at room temperature, it is diluted with 600 parts of water and the precipitate formed is filtered off with suction. The filter cake, which is still moist, is boiled with 150 parts by volume of 17% hydrochloric acid for 1 hour, during which the chlorohydrate of p-amino-benzenesulfonyl-laurylguanidine is deposited in an oily form.
This is decomposed by neutralization with sodium hydroxide solution while cooling and the crystallization obtained is suction filtered. So p-Amino-benzenesulfonyl-laurylguanidine is obtained, which recrystallizes from ethyl acetate and melts at 96-97.
Example 2: 47 parts of p-acetylamino-benzenesulfochloride are added to the solution of 37 parts of ethyl pseudo-thiourea hydrobromide in 75 parts of dry pyridine. With self-heating a solution is obtained, which is warmed for 1 hour on a boiling water bath and then poured into water.
The thereby precipitating N- [p-acetylamino-benzenesulfonyl -S- ethyl-pseudo-thiourea of the formula

is sucked off and washed with water. It can be purified by recrystallizing it from alcohol. F. 182. 301 parts of p-acetylamino-benzenesulfonyl-S-ethyl-pseudo-thiourea are heated with 185 parts of laurylamine in a 180-degree oil bath for 8 hours.
Athylmereaptane escapes. The resulting melt is boiled with 1000 parts of 1Mger hydrochloric acid, a heavy oil being formed. This is the hydrochloride of p-amino-benzenesulfonyl-lauryl-guanidine. It is separated from the acid, stirred with hot water and the hydrochloric acid released is neutralized with sodium hydroxide solution. A semi-solid mass is obtained,
which is dissolved warm in a little ethyl acetate. The solution is dried with calcined pot ash, filtered and left to stand for a long time at room temperature. After 1-2 days, a nice crystallization of p-amino-benzenesulfonyl-lauryl-guanidine is obtained, which is suction filtered and possibly recrystallized several times from ethyl acetate. F. 96-97.
