Verfahren zur Herstellung eines basischen Esters. Im schweizer. Patent Nr. 241782 ist ein Verfahren zur Herstellung von basischen Estern durch Veresterung, von a-substituier- ten Aryloxyessigsäuren mit N-disubstituier- ten Aminoalkoholen beschrieben.
Es wurde nun gefunden, dass man zu den gleichen basischen Estern gelangen kann, wenn man a-substituierte Arylogyessigsäuren in ihre Oxyalkylester bezw. deren reak- 3 tionsfähige Derivate überführt und diese mit sekundären Aminen umsetzt.
Unter reaktionsfähigen Derivaten der ge nannten Oxyalkylester sind insbesondere Ester mit Halogenwasserstoffsäuren,, Aryl- sulfonsäure und dergleichen zu verstehen.
Zur Herstellung der Halogenalkylester der a-substituierten Aryloxyessigsäuren ver fährt man zweckmässig so, dass man auf a-substituierte Aryloxyessigsäuren bezw. ihre Halogenide, Ester oder ihre Anhydride in An- oder Abwesenheit von Kondensations- mitteln Alkylenhalogenhydrine einwirken lässt,
oder dass man Salze von a-substituierten Aryloxyessigsäuren mit Alkylendihalogeni- den oder Alkylenhalogenhydrinen umsetzt und in den erhaltenen Verbindungen gege-. benenfalls vorhandene Hydroxylgruppen durch Halogen ersetzt.
Die a-substituierten Aryloxyessigsäuren sind zum Teil bekannt. Sie -können nach den in der Literatur beschriebenen Methoden er halten werden.
Gegenstand vorliegenden Patentes ist ein Verfahren zur Herstellung eines basischen Esters. Das Verfahren ist dadurch gekenn zeichnet, dass man eine Verbindung der Formel

worin X und Y reaktionsfähige, bei der Re aktion sich abspaltende Reste bedeuten, ein wirken lässt. Die entstandene Verbindung, der. a-(Phenoxy)-cyclopentylessipäure-ss'-di- äthylamino-äthylester, ist eine Base vom Siedepunkt 142-144 bei 0,12 mm Druck.
Sie lässt sich mit anorganischen oder orga nischen Säuren in Salze und ferner auch in quaternäre Verbindungen überführen. Sie soll therapeutische Verwendung finden. <I>Beispiel:</I> 8 Teile Glykolchlorhydrin werden in 100 Teilen Pyridin gelöst. Dazu tropft man unter Rühren 24 Teile a-(Phenoxy)-cyclopen- tylessigsäurechlorid, wobei die Reaktionstem- peratur unter 0 gehalten wird. Dann giesst man auf Eis, stellt kongosauer und äthert aus.
Nach Waschen, Trocknen und Ab- dampfen des Äthers erhält man den a-(Phen- oxy) - cyclopentylessigsäure -<B>ss'-</B> chlor - äthyl- ester.
14 Teile dieser Verbindung werden in 100 Volumteilen absolutem Toluol mit 8 Teilen Diäthylamin versetzt und in Auto klaven 6 Stunden auf 100-120 erwärmt. Nach dem Erkalten zieht man wiederholt mit verdünnter wässriger Salzsäure aus. Die saure wässrige Lösung wird ausgeäthert, dann phenolphtaleinalkalisch gestellt und die ab geschiedene Base in Äther aufgenommen.
Nach dem Trocknen über Pottasche wird der Äther abgedampft. Durch Destillation des Rückstandes erhält man den a-(Phenoxy)- cyclopentylessigsäure-ss' -diäthylamino-äthyl- ester.
Der cl-(Phenoxy)-cyclopentylessigsäure- ss'-chlor-äthylester kann auch aus a-(Phen- oxy)-cyclopentylessigsäure bezw. ihren Sal zen durch Umsetzen mit Äthylendichlorid, oder durch Umsetzen mit Athylenchlorhydrin zum a-(Phenoxy)-cyclopentylessigsäure-ss'- oxy-äthylester und Behandlung des letzteren mit Thionylcblorid hergestellt werden.
Anstatt von a-(Phenoxy)-cyclopentylessig- säure-ss'-chlor-äthylester kann man auch a- (Phenoxy) - cy clopentylessigsäure -ss' - (toluol- sulfogy)-äthylester ausgehen.
Process for the preparation of a basic ester. In swiss. Patent No. 241782 describes a process for the preparation of basic esters by esterification of α-substituted aryloxyacetic acids with N-disubstituted amino alcohols.
It has now been found that the same basic esters can be obtained by converting a-substituted arylogyacetic acids into their oxyalkyl esters or. their reactive derivatives are transferred and these are reacted with secondary amines.
Reactive derivatives of the oxyalkyl esters mentioned are to be understood as meaning, in particular, esters with hydrohalic acids, arylsulfonic acid and the like.
For the preparation of the haloalkyl esters of the a-substituted aryloxyacetic acids ver it is expedient to proceed so that one resp. On a-substituted aryloxyacetic acids. lets its halides, esters or anhydrides act in the presence or absence of condensing agents alkylene halohydrins,
or that salts of α-substituted aryloxyacetic acids are reacted with alkylene dihalides or alkylene halohydrins and mixed in the compounds obtained. any hydroxyl groups present are replaced by halogen.
Some of the α-substituted aryloxyacetic acids are known. They can be obtained using the methods described in the literature.
The present patent is a process for the preparation of a basic ester. The process is characterized in that a compound of the formula

It can be converted into salts and also into quaternary compounds with inorganic or organic acids. It should find therapeutic use. <I> Example: </I> 8 parts of glycol chlorohydrin are dissolved in 100 parts of pyridine. 24 parts of α- (phenoxy) cyclopentyl acetic acid chloride are added dropwise with stirring, the reaction temperature being kept below 0. Then you pour on ice, put Congo sour and ether off.
After washing, drying and evaporation of the ether, the α- (phenoxy) -cyclopentylacetic acid-<B> ss'- </B> chloro-ethyl ester is obtained.
14 parts of this compound are mixed with 8 parts of diethylamine in 100 parts by volume of absolute toluene and heated to 100-120 for 6 hours in an auto. After cooling, it is extracted repeatedly with dilute aqueous hydrochloric acid. The acidic aqueous solution is extracted with ether, then made alkaline with phenolphthalein and the base which has separated off is taken up in ether.
After drying over potash, the ether is evaporated. The a- (phenoxy) -cyclopentyl acetic acid-ss'-diethylamino-ethyl ester is obtained by distilling the residue.
The cl- (phenoxy) -cyclopentylessigsäure- ss'-chloro-ethyl ester can also from a- (phenoxy) -cyclopentylessigsäure bezw. their salts are prepared by reacting with ethylene dichloride, or by reacting with ethylene chlorohydrin to form a- (phenoxy) -cyclopentylessigsäure-ss'- oxy-ethyl ester and treating the latter with thionyl chloride.
Instead of a- (phenoxy) -cyclopentylacetic acid-ss'-chloroethyl ester, a- (phenoxy) -cyclopentylessigsäure -ss' - (toluenesulfogy) ethyl ester can also be used as a starting point.
