Process for the preparation of a basic ester. The present patent relates to a process for the preparation of a basic ester. The process is characterized in that a compound of the formula
EMI0001.0003
where X denotes a radical which splits off during the reaction, a compound which donates the β-halopropionyl radical and diethylamine allows them to act on one another.
In carrying out the process, for example, a compound of the above formula can be reacted with a compound which donates the β-diethylamino-propionyl radical.
The resulting new compound, α-cyclopentyl-7benzyl-α-diethylamino-propionate, is a colorless 01 boiling point 148 to 151 at 0.4 mm pressure. It should find therapeutic use. Example 1:
A solution of 175 parts ss is added to a mixture of 180 parts of α-cyclopentylbenzyl alcohol and 110 parts of absolute pyridine in 900 parts of absolute ether while stirring and cooling with ice water Bromopropionic acid chloride was added dropwise in the same amount of absolute ether. After two hours of heating on the water bath, water is added, the ether solution is washed with dilute bicarbonate solution and evaporated.
The crude α-cyclopentyl-benzyl ss'-bromopropionate is added dropwise to a solution of 150 parts of diethylamine in 300 parts of absolute ether with cooling at 20 and heated on the steam bath for 3 hours. After adding a lot of water, it is acidified and etherified. The aqueous solution is made alkaline in the cold with lye and the base that has been deposited is taken up in an organic solvent. The free base boils at 148 to 151 at 0.4 mm pressure.
The α-cyclopentylbenzyl alcohol can also be used in the form of an alcohol and, instead of pyridine, benzene can be used as the solvent.
Example 2: 7.3 parts of ß-diethylamino-propionic acid are refluxed with 12 parts of thionyl chloride for 1 hour and then the excess thionyl chloride is removed in vacuo.
The crystallized residue is taken up in 50 parts of absolute benzene, 10 parts of α-cyclopentylbenzyl alcohol are added in portions and, after the reaction has subsided, refluxed for a further hour. The reaction product is taken up in dilute acid, separated from the benzene, the base is released in the cold with pot ash and taken up in ether. It boils at 152 to 155 at 0.6 mm pressure.
