Description

  

  Verfahren zur Herstellung eines basischen Esters.         Gegenstand    vorliegenden Patentes ist ein  Verfahren zur Herstellung     eines    basischen  Esters. Das Verfahren ist dadurch gekenn  zeichnet, dass man eine Verbindung der  Formel

         worin    X einen bei der Reaktion sich abspal  tenden Rest bedeutet, ein     ss-halogenpropion-          saures    Salz und     Diäthylamin        aufeinander          einwirken    lässt.

   In Ausführung des Verfah  rens kann man zum Beispiel eine     Verbindung     der obigen Formel mit einem     ss-diäthylamino-          propionsauren    Salz umsetzen.  



  Die entstandene neue Verbindung, der       ss'-Diäthylamino-propionsäure    - a -     cyclohexyl-          benzylester,        ist    ein farbloses 01 vom Siede-         Punkt    151 bis 156  bei 0,03 mm Druck. Er  soll therapeutische     Verwendung    finden.    <I>Beispiel 1:</I>  25 Teile     a-Cyclohexyl-benzylbromid    und  18 Teile     ss-brom-propionsaures    Natrium wer  den in<B>150</B> Teilen absolutem     Toluol    6 Stun  den unter     Rückfluss    erhitzt.

   Nach Erkalten       wird    mit Wasser neutral gewaschen, das Lö  sungsmittel im Vakuum entfernt und der zu  rückbleibende     ss'-Brom-propionsäure-a-cyclo-          hexyl-benzylester    im     Höchvakuum    destil  liert. Er siedet bei 139 bis 142  bei 0,1     mm     Druck.

   Durch Umsetzung dieses     ss-Brom-pro-          pionsäureesters    mit einem Überschuss von     Di-          äthylamin    bei 60 bis 70  wird der     ss'-Diäthyl-          amino-propionsäure    -     a-cyclohegyl-benzylester     vom Siedepunkt 151 bis 156  bei 0,03 mm  Druck erhalten.  



  <I>Beispiel 2:</I>  Eine Mischung von 12 Teilen     ss-diäthyl-          amino-propionsaurem    Silber und 10 Teilen       a-Cyclohexyl-benzylbromid    in 100 Teilen ab-           solutem    Alkohol wird 6 Stunden am Rück  fluss erhitzt, dann der Alkohol im Vakuum  entfernt und der Rückstand in verdünnter  Salzsäure aufgenommen. Die filtrierte Lö  sung wird     ausgeäthert,    die     wässrige    Schicht  filtriert     und    unter Eiskühlung mit Pottasche  alkalisch gestellt.

   Daraus     wird    die freie Base  mit Äther     extrahiert,    die     Atherlösung    mit  konzentrierter     Pottaschelösung    gewaschen,  getrocknet und eingedampft. Das erhaltene  Endprodukt     destilliert    bei 151 bis 156      und     0,03 mm Druck und stellt den im Beispiel 1  genannten Ester dar.

         where X denotes a residue which splits off during the reaction, an β-halopropionic acid salt and diethylamine can act on one another.

   In carrying out the process, for example, a compound of the above formula can be reacted with an β-diethylaminopropionic acid salt.



  The resulting new compound, the ss'-diethylamino-propionic acid - a - cyclohexylbenzyl ester, is a colorless oil with a boiling point of 151 to 156 at 0.03 mm pressure. It should find therapeutic use. <I> Example 1: </I> 25 parts of α-cyclohexylbenzyl bromide and 18 parts of sodium β-bromopropionic acid are refluxed in 150 parts of absolute toluene for 6 hours.

   After cooling, the mixture is washed neutral with water, the solvent is removed in vacuo and the α-cyclohexyl-benzyl β-bromopropionate which remains is distilled in a high vacuum. It boils at 139 to 142 at 0.1 mm pressure.

   By reacting this ß-bromo-propionic acid ester with an excess of diethylamine at 60 to 70, the ß-diethylamino-propionic acid - a-cyclohegyl-benzyl ester of boiling point 151 to 156 at 0.03 mm pressure is obtained.



  <I> Example 2: </I> A mixture of 12 parts of ß-diethylamino-propionic acid silver and 10 parts of α-cyclohexylbenzyl bromide in 100 parts of absolute alcohol is refluxed for 6 hours, then the alcohol in Removed vacuum and the residue taken up in dilute hydrochloric acid. The filtered solution is extracted with ether, the aqueous layer filtered and made alkaline with potash while cooling with ice.

   The free base is extracted from this with ether, the ether solution is washed with concentrated potash solution, dried and evaporated. The end product obtained distills at 151 to 156 and 0.03 mm pressure and represents the ester mentioned in Example 1.

PATENT CLAIM: Process for the preparation of a basic ester, characterized in that a compound of the formula EMI0002.0013 where X denotes a residue which splits off during the reaction, an β-halopropionic acid salt and diethylamine can act on one another.

The resulting new compound, the ss'-diethylamino-propionic acid-a-cyclohexylbenzyl ester, is a colorless oil with a boiling point of 151 to 156 at 0.03 mm pressure. SUBSCRIBES 1.

Process according to patent claim, characterized in that a compound of the formula EMI0002.0028 where X denotes a residue which splits off during the reaction, is converted with an β-halopropionic acid salt into the α-cyclohexylbenzyl α-halopropionate and this is reacted with diethylamine. 2.

Method according to claim, characterized in that one is a compound of the formula EMI0002.0035 wherein X is a residue that splits off during the reaction, is reacted with an β-diethylamino-propionic acid salt.
