T. verwachsene Kristallnadeln vom Schmelzpunkt 184 u. Z.
Process for the preparation of p-aminobenzenesulfonyl-methyl-thiourea. In the main patent, a process for the manufacture of p-aminobenzenesulfonyl-ethyl-thiourea is described, according to which ethyl mustard oil is allowed to act on acetylsulfanilamide and the resulting p-acetyl-aminobenzenesulfonyl-ethylthiourea to p - aminobenzenesulfonyl-ethylthioureate.
It has now been found that p-aminobenzenesulfonyl-methylthiourea is obtained when methyl mustard oil is allowed to act on acetylsulfanilamide and the p-acetylaminobenzenesulfonyl-methylthiourea obtained is saponified to p-aminobenzenesulfonyl-methylthiourea.
As in the process of the main patent, the reaction of the mustard oil with the sulfanilamide are accelerated.
Example: 108 g aoetylsulfanilamide are in 20 g NaOH, 200 cm 'water and 180 cm' den. Dissolved ethanol. In the solution at room temperature 37 g of methyl mustard oil, in 20 cm3 den. Dissolved alcohol, add dropwise. The temperature rises a little. The mixture is then stirred for 16 hours at the usual temperature and then for a further 12 hours at 50.
After cooling, it is diluted with 300 cm3 of water and then the alcohol is distilled off in vacuo at 50. The solution is treated with coal and concentrated with. HCl made Congo acidic. The precipitated substance is filtered off with suction, washed with water, pulverized and stirred well with sodium bicarbonate and water for 1-2 hours. After the undissolved starting material has been filtered off with suction, the filtrate is added dropwise to excess 50% glacial acetic acid while stirring.
The precipitated substance is filtered off with suction, washed with H 2 O and dried. He holds 95 g of p - acetylaminobenzene sulfonyl - methylthiourea as a crude product with a melting point of 185 u. Z. - 66% d. Th. The crude product is not purified any further, but is immediately processed further to give the deacetylated compound by saponifying 72 g of the acetylated compound as usual.
The free p-aminobenzenesulfonyl-methylthiourea is obtained from the potassium salt obtained first by adding dropwise the aqueous potassium salt solution in 50% glacial acetic acid with stirring. After drying, 50 g of substance with a melting point of 182 u. Z. = 81% d. Th. After reprecipitating twice, the substance forms white, e.g. T. fused crystal needles with a melting point of 184 u. Z.
