worin X Wasserstoff, Hydroxyl, Alkyl oder sonstige Substituenten, jedoch mindestens ein X eine zur Bildung beständiger, nicht hydro- lysierender Salze befähigte, saure oder basi sche (Cxruppe bedeutet, mit geeigneten Schwer metallverbindungen, insbesondere Schwer- inetallsalzen, neue Komplexsalze von wert vollen Eigenschaften liefern, die in Form der neutralen Salze mit Basen oder Säuren wasserlöslich sind.
Der Reaktionsvorgang ist wahrscheinlich so, dass das Wasserstoffatom der Mercaptogruppe von einem oder mehreren Mercaptopyrimidinen gegen den Schwermetall rest ausgetauscht wird. Zur Bildung bebtän- diger, nicht hydrolysierender Salze befähigte saure Gruppen sind zum Beispiel die Carbo- xyl- und Alkylcarbonsäuregruppeii, die mit Alkalien, Erdalkalien oder Stickstoffbasen neutrale Salze bilden.
Als basische Substi- tuenten der genannten Art kommen haupt sächlich Aminoalkyl- und Alkylaminoalkyl- gruppen in Frage, die mit anorganischen oder organischen Säuren Salze bilden. Diese Gruppen können auch zunächst in nicht freier saurer oder basischer Form vorliegen und gleichzeitig mit der Bildung der Komplex verbindung oder nachträglich einer Aufspal tung, zum Beispiel einer Verseifung unter worfen werden.
Die Neutralisation kann auch durch innere Salzbildung zwischen einem sauren und einem basischen Substituenten des blercaptopyrirriidins ohne besonderen Zu satz einer anorganischen oder organischen Säure erfolgen. Die Ausgangsmaterialien, das heisst also die Mercaptopyrimidine, werden nach den üblichen Methoden erhalten.
Die neuen Verbindungen besitzen thera peutische Wirksamkeit und können in Form der wasserlöslichen neutralen Salze als Injektionspräparate Verwendung finden - sie kommen ausserdem auch als Zwischenprodukte für sonstige Verwendungszwecke in Betracht. Sie sind. durch besondere Stabilität ausge zeichnet. Für therapeutische Verwer)dungs- zwecke sind die Silber- und Goldderivate von besonderem Interesse. Sie haben sich als wirksam bei der Behandlung von Infektions krankheiten erwiesen.
Man hat bereits Metallkomplexverbindun- gen von Mercaptopyrimidinderivaten, zum Beispiel von Oxy- und Aminomercaptopyrinri- dinen dargestellt. Die bekannten Metallkom- plexverbindungen sind jedoch im Gegensatz zu den nach dem vorliegender) Patent er hältlichen Produkten nicht löslich in Wasser und sind mangels Gehaltes an zur Bildung beständiger, nicht hydrolysierender Salze be fähigten Gruppen nicht im Stande, neutrale wasserlösliche Salze zu bilden.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung einer Mercapto- pyrimidinmetallkomplexverbirrdung, das darin besteht, dass man auf N-Allyl-diäthylamir)o- äthyl-thiobarbitursäure ein wasserlösliches Silbersalz zur Einwirkung bringt und die Silber-N-allyl-diäthylamirroäthyl-thiobarbitur- säure mit Hilfe einer Base abscheidet, die jedoch dabei nicht in den Endstoff eintritt. Die Reaktion wird zweckmässig unter Zusatz von Wasser oder einem wasserhaltigen Löse mittel vorgenommen. Als wasserlösliches Silbersalz kann zum Beispiel Silbernitrat Verwendung finden.
Als Base für die Ab scheidung des Reaktionsproduktes hat sich konzentrierter Ammoniak als zweckmässig erwiesen.
Die Silber-N-allyl-diäthylamirroäthyl-thio- barbitursäure bildet ein farbloses Kristall pulver. Es wird in Wasser löslich, ebenso in Alkalilaugen und Säuren unter Bildung der entsprechenden Salze. Es soll therapeu tische Anwendung finden.
<I>Beispiel:</I> 2,9 Gewichtsteile N-Allyl-diäthylamino- äthyl-thiobarbitursäur-e werden mit einer Lösung von 1,7 Gewichtsteilen Silbernitrat in 5 Gewichtsteilen Wasser zusammengegeben und nach erfolgter Lösung mit konzentriertem Ammoniak versetzt, worauf die Silber-N- allyl-diäthy laminoäthyl-thiobarbitursäure, die infolge innerer Salzbildung wasserlöslich ist, auskristallisiert. Man erhält ein farbloses, auch in Alkalilaugen und Säuren lösliches Kristallpulver.
