Verfahren zür Herstellung des goldbenzimidazol-4-sulfonsauren Natriums. Es ist bekannt, dass Imidazole, die keine zur Salzbildung befähigten sauren oder basi- sehen Gruppen tragen, Metallkomplexver- bindungen zu bilden vermögen, die, wie zum Beispiel die der Berizimidazolreihe, in Wasse; praktisch unlöslich sind.
Es wurde nun gefunden, dass auch solche Imidazole, die keine Merkaptangruppe im Imidazolring enthalten und die mit einem aromatischen oder heterozyklischen Ring system kondensiert oder substituiert sind und ausserdem zum mindesten eine zur Salzbildung befähigte saure oder basische Gruppe im Molekül enthalten, Komplexverbindungen mit Metallen zu bilden vermögen, und dass man durch Darstellung solcher Verbindungen zu Produkten gelangen kann, die sich im Gegen satz zu den bereits bekannten Komplexver bindungen der Imidazolreihe durch ihre Wasserlöslichkeit auszeichnen.
Die Eigen schaften der neuen Verbindungen machen sie für die therapeutische Verwendung, im be- sonderen bei der Bekämpfung der Erreger von Infektionskrankheiten, besonders wertvoll und zeichnen sich gegenüber den Metallver bindungen der Benzimidazole, die eine Mer- kaptangruppe im Imidazolring enthalten, durch ein besonders günstiges Verhältnis von thera peutischer Wirksamkeit zur Giftigkeit aus.
Als Vertreter der oben erwähnten, zur Salzbildung befähigten sauren Substituenten der Imidazole seien beispielsweise die Sulf0.-, Carboxyl- und Sulfoxylatgruppe, der Arsin- säure-, Stibinsäure- und Glycinrest erwähnt. Als zur Salzbildung befähigte basische Grup pen haben sich stickstoffhaltige organische Reste, in denen der Stickstoff basischen Charakter hat, z.
B. Aminoalkyl- oder Amino- cycloalkylgi-uppen, sowie beterozyklische Reste, wie der Pyridyl-, Piperidyl- oder Chinolylrest bewährt. Diese basischen Reste können mit dem Imidazolringsystem unmittel bar oder über Sauerstoff, Schwefel oder Stick stoff hinweg verbunden sein. Die Darstellung der neuen Komplexver bindungen erfolgt nach den für den Aufbau von Verbindungen der genannten Art üblichen Methoden.
Man kann zum Beispiel auf Imid- azolverbindungen mit zur Salzbildung be fähigten sauren oder basischen Gruppen in an sich üblicher Weise Metallverbindungen einwirken lassen und die nach dem Neu tralisieren gebildeten Metallkomplexverbin- dungen in an sich bekannter Weise zur Ab- scheidung bringen.
Man verfährt zum Beispiel derart, dass man Imidazole mit sauren Gruppen der oben gekennzeichneten Art oder ihre Salze in wässeriger Suspension oder Lösung auf Me talloxyde oder Hydroxyde oder Metallsäuren bezw. Metallsalze oder Metalldoppelsalze ein wirken lässt, wobei nur das Kation des Schwermetalles in das Imidazol eintritt und die dabei entstehenden Metallkomplexsäuren durch Basen in Lösung bringt und die ge bildeten neutralen Komplexsalze durch Ein giessen in Fällungsmittel abscheidet.
Hierfür eignen sich besonders mit Wasser misehbare, organische Flüssigkeiten, wie zum Beispiel Alkohol, Methylalkohol, Aceton oder deren Gemische. Anderseits kann man zum Bei spiel die Salze der durch basische Gruppen substituierten Imidazole mit Schwermetall- salzen bezw. Schwermetalldoppelsalzen, Me talloxyden, Metallhydroxyden oder Metall säuren zur Umsetzung bringen. Die gebilde ten Metallverbindungen können durch ge eignete Basen abgeschieden und mit Säuren in leicht und neutral und nahezu neutral lösliche, Salze übergeführt werden.
Zweckmässig verwendet man bei der Dar stellung der neuen Komplexverbindungen der Imidazolreihe einen Überschuss an Komplex bildnern oder setzt ihn nachträglich zu.
Die auf diese Weise erhältlichen Ver bindungen sind<B>je</B> nach der Art des ange wandten Metalles gefärbte oder ungefärbte Pulver von grosser Haltbarkeit, die sich in Wasser mit neutraler oder praktisch neu traler Reaktion lösen.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung des goldbenz- imidazol-4-sulionsauren Natriums, eines hell gelben, in Wasser neutrallöslichen Pulvers, das dadurch gekennzeichnet ist, dass man Benzimidazol-4-sulfonsäure auf ein Goldsalz, worunter auch Doppelsalze zu verstehen sind, in Gegenwart von Natriumhydroxyd einwir ken lässt und das gebildete Goldkomplexsalz durch Eingiessen in ein Fällungsmittel zur Abscheidung bringt'. Hierbei geht von dem verwendeten Goldsalz nur das Gold in den Komplex über.
Das so erhältliche goldbenz- imidazol-4-sulfonsaure Natrium soll als Heil mittel verwendet werden. <I>Beispiel:</I> Eine Lösung von<B>6,5</B> gr Benzimidazol-4- sulfoiisä:ure in<B>6</B> ems <B>5</B> n-Natronlauge und <B>10</B> ems Wasser wird unter Umrühren mit einer Lösung von 4 gr Goldkaliumehlorid in <B>10</B> cms Wasser versetzt, die Reaktions mischung mit<B>5</B> n-Natronlauge neutralisiert und in Alkohol gegossen.
Man erhält das goldbenzimidazol-4-sulionsaure Natrium als ein bellgelbes, in Wasser mit neutraler Re aktion lösliches Pulver.
