Diese und ihre Salze weisen wertvolle pharma kologische Eigenschaften auf. So besitzen sie eine Hemmwirkung auf die Erregungsübertragung im Zentralnervensystem und können deshalb als Heil mittel bei neurologischen Erkrankungen verwendet werden.
Die neue 1,3,5-Triazinverbindung wird erfin dungsgemäss erhalten, indem man in einem 4,6-Bis- diäthylamino-1,3,5-triazin, das in 2-Stellung eine freie oder verätherte Mercaptogruppe aufweist, diese durch Behandlung mit Hydrazin gegen die Hy- drazinogruppe austauscht. So kann man z.
B. ein 2-Alkylmercapto-4,6-bis-diäthylamino-1,3,5 - triazin, wie 2-Methylmercapto-4,6-bis-diäthylamino-1,3,5- triazin, besonders aber 2-Mercapto-4,6-bis-diäthyl- amino-1,3,5-triazin mit Hydrazin umsetzen. Die Ausgangsstoffe können auch in Form ihrer Salze angewandt werden. Die Umsetzung wird zweckmässig in Anwesenheit von Verdünnungsmitteln, allenfalls auch in Gegenwart von Kondensationsmitteln durch geführt, wobei man ausserdem in Gegenwart von Katalysatoren, wie Kupferpulver, arbeiten kann.
Soweit die für die Durchführung der genannten Reaktion notwendigen Ausgangsstoffe nicht bekannt sind, können sie nach den üblichen Methoden ge wonnen werden. Je nach der Arbeitsweise erhält man das neue Hydrazinotriazin in Form seiner Base oder ihrer Salze.
Aus der Base können therapeutisch verwend bare Salze gebildet werden, wie der Halogenwasser stoffsäuren, Schwefelsäure, Salpetersäure, Phosphor säure, Essigsäure, Propionsäure, Oxalsäure, Äpfel säure, Zitronensäure, Methansulfonsäure, Äthan- sulfonsäure, Oxyäthansulfonsäure, Benzoesäure, Sa- licylsäure,
p-Aminosalicylsäure oder Toluolsulfon- säure. Erhaltene Salze können in üblicher Weise in die freie Base übergeführt werden.
Die neue Verbindung oder ihre Salze können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeuti schen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
<I>Beispiel 1</I> 6 g 2-Mercapto-4,6-bis-diäthylamino-1,3,5-triazin werden in 50 ml absolutem Äthanol mit 6 g Hydra zinhydrat 12 Stunden am Rückfluss gekocht, wobei Schwefelwasserstoffentwicklung eintritt. Das Lö sungsmittel wird auf dem siedenden Wasserbad im Vakuum abdestilliert, der Rückstand mit Wasser ver setzt und mit verdünnter Natronlauge stark alkalisch gestellt. Hierauf schüttelt man dreimal mit Äther aus und wäscht die Ätherlösungen dreimal mit je 50 ml In Salzsäure. Die saure Lösung wird mit verdünnter Natronlauge alkalisch gestellt und mit Äther extra hiert.
Die Ätherlösung wird mit Natriumsulfat ge trocknet und eingedampft. Das 2-Hydrazino-4,6-bis- diäthylamino-1,3,5-triazin bleibt als ölige Flüssigkeit zurück. Mit äthanolischer Salzsäure und Äther zugabe kann es in das kristallisierte Dihydrochlorid vom F. 198-200 verwandelt werden.
Das als Ausgangsmaterial verwendete 2-Mer- capto-4,6-bis-diäthylamino-1,3,5-triazin kann wie folgt erhalten werden: 10 g 2-Chlor-4,6-bis-diäthylamino-1,3,5-triazin werden mit einer Lösung von 4,4 g Natriumhydro- sulfid in 150 ml 95 /oaigem Äthanol im geschlossenen Gefäss 10 Stunden auf 130-140 erhitzt. Das Lö sungsmittel wird abgedampft, der Rückstand mit Wasser versetzt, mit verdünnter Natronlauge alka lisch gestellt und mit Äther extrahiert. Die wässrig alkalische Lösung filtriert man sodann mit Zusatz von Tierkohle und stellt sie mit halbkonzentrierter Salzsäure sauer.
Nun wird mit Chloroform ausge schüttelt, die Chloroformlösung über Natriumsulfat getrocknet und eingedampft. Der Rückstand stellt das 2-Mercapto-4,6-bis-diäthylamino-1,3,5-triazin dar. Aus Essigester umkristallisiert schmilzt es bei 155-156 .
<I>Beispiel 2</I> 7,5 g 2-Methylmercapto-4,6-bis-diäthylamino- 1,3,5-triazin werden in 100 ml absolutem Athanol mit 7 g Hydrazinhydrat 6 Stunden im geschlossenen Gefäss auf 140-150 erhitzt. Das Reaktionsgemisch wird analog Beispiel 1 aufgearbeitet. Man erhält so das im Beispiel 1 beschriebene 2-Hydrazino-4,6-bis- diäthylamino-1,3,5-triazin, dessen Dihydrochlorid bei 198-200 schmilzt.
Das als Ausgangsstoff verwendete 2-Methyl- mercapto-4,6-bis-diäthylamino-1,3,5-triazin kann wie folgt hergestellt werden: 19 g 2 - Mercapto - 4,6-bis-diäthylamino-1,3,5- triazin werden in 200 ml Methanol gelöst und lang sam mit 160 ml einer ätherischen Diazomethan- lösung versetzt. Man lässt über Nacht stehen und dampft die überschüssige Diazomethanlösung ab. Der Rückstand wird am Hochvakuum destilliert. 2-Methylmercapto-4,6-bis-diäthylamino-1,3,5-triazin destilliert bei 135-136 unter 0,25 mm Hg-Druck.
