Abstract

La présente invention concerne un ADN de recombinaison comprenant un promoteur de transcription, une séquence en aval à exprimer, liée de façon opérationnelle avec ce dernier, le promoteur de transcription comprenant une région située en amont du cadre de lecture ouvert d'un gène de Phaffia fortement exprimé, de préférence un gène intervenant dans un processus glycolytique, ou encore mieux, le gène codant la glycéraldéhyde-3-phosphate déshydrogénase. D'autres ADN de recombinaison préférés, selon l'invention, contiennent des promoteurs de gènes codant une protéine ribosomique, ou encore mieux, le promoteur de transcription comprenant une région située en amont du cadre de lecture ouvert codant une protéine représentée par l'une des séquences d'acides aminés décrites dans l'une des séquences portant le numéro d'identification 24 à 50. Selon un autre aspect de l'invention, on décrit une séquence d'ADN isolé codant une enzyme impliquée dans le processus de biosynthèse de caroténoïde de Phaffia rhodozyma, ladite enzyme présentant de préférence une activité sélectionnée parmi l'activité de l'isopenténylpyrophosphate isomérase, l'activité de la géranylgéranylpyrophosphate synthase, l'activité de la phytoène synthase, l'activité de la phytoène désaturase et l'activité de la lycopène cyclase, ou encore mieux, celles qui codent un enzyme présentant une séquence d'acides aminés choisie dans l'une des séquences portant le numéro d'identification 13, 15, 17, 19, 21, 23. L'invention concerne également des vecteurs, des organismes hôtes transformés, des méthodes pour produire ces protéines et/ou caroténoïdes, par exemple l'astaxanthine, ainsi que des méthodes pour isoler des promoteurs fortement exprimés à partir du Phaffia.
