Abstract

Patente de Invenção: <B>"ENGENHEIRAMENTO DE TIOESTERASES DE PLANTA E DIVULGAçãO DE TIOESTERASES DE PLANTA TENDO ESPECIFICIDADE DE SUBSTRATO''<D>. Métodos de alterar a especificidade do substrato de tioesterases de acil-ACP de planta, e tioesterases de acil-ACP de planta construída assim produzidas são fornecidos. A porção de dois terços de terminal C de tioesterases de planta é identificada como desejável para tais modificações. As seq³ências e construções e DNA para a expressão de tioesterases construídas, bem como as novas tioesterases produzidas delas são também fornecidas. Tais seq³ências de DNA podem ser empregadas para a expressão das tioesterases construídas em células hospedeiras, particularmente células de semente de plantas de colheira de semente oleoginosa, para a modificação de composição de ácido graxo. Um C12 preferindo a tioesterase de acil-ACP de planta descrito aui pode ser alterado para obter uma tioesterase de planta tendo aproximadamente igual atividade sobre os substratos C14 e C12. Outra modificação da enzima C12 produz uma tioesterase tendo maior atividade sobre o C14 quando comprado aos substratos C12. De particular interesse é uma tioesterase 18:1 de planta na qual a atividade 18:0 relativa foi aumentada. Tais tioesterase de FatA encontram uso para a produção melhorada de estearato em óleos de semente vegetal.
